Response of Red Sorrel (Rumex acetosella L.) to Several Soil- and Foliar-Applied Herbicides (애기수영의 화학적 방제를 위한 제초제 선발 및 선발 제초제의 살초효과)

  • Red sorrel (Rumex acetosella L.) is a troublesome perennial weed in the alpine grasslands of Kangwon Province of Korea. A number of soil- and foliar-applied herbicides were evaluated for their efficacy of red sorrel control. In greenhouse experiments, no soil-applied herbicides, such as pendimethalin, simazine, alachlor, metolachlor, ethalfluralin controlled red sorrel, however, foliar-applied herbicides, such as glufosinate, paraquat, glyphosate, glyphosate + 2,4-D, dicamba, mecoprop, 2,4-D, bentazone controlled more than 60% of red sorrel 2 weeks after treatments. When dicamba and 2,4-D were applied to red sorrel in different growth stages, the auxin-type of herbicides cold control red sorrel regardless of growth stage. This result implies that the auxin-type herbicides can be applied between early May (early growth stage) and mid lune (before fruit maturation) at Kangwon alpine grasslands. In a field experiment, glufosinate at 1.0 kg a.i, $ha^{-1}$, glyphosate at 3.28 kg a.i. $ha^{-1}$, dicamba at 0.96 kg a.i. $ha^{-1}$, and mecoprop at 150 kg a.i. $ha^{-1}$ controlled more than 80% of red sorrel 4 weeks for treatment, suggesting such herbicides could be applied for red sorrel control at Kangwon alpine grasslands.

  • Over 500 genetically modified organisms (GMOs) have been developed since 1996, of which nearly 44% have glufosinate herbicide-tolerant traits. Identification of specific markers that can be used to identify herbicide-tolerant traits is challenging as the DNA sequences of the gene(s) of a trait are highly variable depending on the origin of the gene(s), plant species, and developers. To develop specific PCR marker(s) for the detection of the glufosinate-tolerance trait, DNA sequences of several pat or bar genes were compared and a diverse combination of PCR primer sets were examined using certified reference materials or transgenic plants. Based on both the qualitative and quantitative PCR tests, a primer set specific for pat and non-specific for bar was developed. Additionally, a set of markers that can detect both pat and bar was developed, and the quantitative PCR data indicated that the primer pairs were sensitive enough to detect 0.1% of the mixed seed content rate.

Analysis of Glyphosate and Glufosinate in Animal Feeds using LC-MS/MS (LC-MS/MS를 이용한 동물 사료 내 글라이포세이트 및 글루포시네이트 분석)

  • The standards for the contents of glyphosate and glufosinate in foods are specific and well categorized. However, the standard of content in animal feeds is relatively inadequate and the classification is insufficient. There is also constant debate about the risk of glyphosate and glufosinate to human health, but the risk to animals has not been well studied. In this study, we established an analytical method in feeds that is estimated to be the path for animals to ingest glyphosate. The solvent extraction was carried out using 25% methanol. After centrifugation, samples were purified using solid phase extraction (SPE) and quantitatively analysed using LC-MS/MS after concentrated. Assessment of validation was conducted through detection limits, accuracy, and precision tests. The detection limits for the established method were 1.8 of ${\mu}g/kg$ of glufosinate and $2.4{\mu}g/kg$ of glyphosate. Accuracy was ranged from 94.4% to 103.4% and precision was range from 1.5% to 7.2%. Glufosinate was detected in one sample ($ND{\sim}8.8{\mu}g/kg$) and glyphosate was detected in all but one sample ($ND{\sim}337.0{\mu}g/kg$) by applying the analytical method to animal feeds (n=13).

Utilization of the bar gene to develop an efficient method for detection of the pollen-mediated gene flow in Chinese cabbage (Brassica rapa spp. pekinensis)

  • To develop an efficient screening method for detection of the transgene in Chinese cabbage (Brassica rapa spp. pekinensis) utilizing Basta spray, optimal conditions for Basta application were examined in this study. Two transgenic Chinese cabbage lines were obtained through Agrobacterium-mediated transformation and used as transgenic positive controls in the Basta screening experiment. Differential concentrations of glufosinate-ammonium were sprayed into three different growth stages of 12 commercial Chinese cabbage cultivars. The results showed that no plants could survive higher than 0.05% glufosinate-ammonium, and plants at the 2-3 leaf stage were most vulnerable to glufosinate-ammonium. On the other hand, no damage was observed in the transgenic control plants. Reliability of the Basta spray method was proven by showing perfect co-segregation of the tolerance to glufosinate-ammonium and the presence of the bar gene in T1 segregating populations of the transgenic lines, as revealed by both PCR and Southern blot analyses. Using the developed Basta screening method, we tried to investigate the transgene flow through pollen dispersal, but failed to detect any transgene-containing non-transgenic Chinese cabbages whose parents had been planted adjacent to transgenic Chinese cabbages in field conditions. However, the transgene was successfully detected using Basta spray from the non-transgenic plants bearing the transgene introduced by hand-pollination. Since the Basta spray method developed in this study is easy to apply and economical, it will be a valuable tool for understanding the mechanism of gene flow through pollen transfer and for establishing a biosafety test protocol for genetically modified (GM) Chinese cabbage cultivars.

Inheritance of Herbicide (glufosinate) Resistance in Transgenic Rice Plant through Anther Culture

  • Haploid system by anther culture allows the development of homozygous lines when doubled. The response of anther culture to Basta (glufosinate) resistance was investigated on transgenic plants (cv. Anjungbyeo) in order to identify inheritance of bar gene associated with Basta. Most of the regenerated transgenic plants were sterile, and only a few plants produced viable seeds ($A_1$) in the greenhouse. The bar gene was analysis by PCR in basta resistant transgenic plant ($TA_0$). The transgenic seeds ($A_1$) were significantly germinated in Basta solution compared with non-transformed seeds. As a result of anther culture, in regenerated haploid plants, segregation ratio was 1:1 in five of eight cross combinations. In diploid plants, segregation ratio was 1:1 in seven of eight cross combinations. Although there was some differences in the cross combinations, most of the combinations had 1:1 segregation ratio which supports the theory. The difference may be a result of the small sample size or the difference of anther culture response caused by genotypic difference. Hence, when many cross combinations were anther-cultured the results would support the theory.

The Effect of Apple Tree Growth and Apple Yield from the Misuse of Non Selective Herbicide, Glufosinate-ammonium (비선택성 제초제 Glufosinate-ammonium 오용이 사과나무의 품종별 생육반응 및 수확에 미치는 영향)

  • This study was conducted to investigate the phytotoxic reaction and the impact on the growth of apple fruit included the yield amount by misuse of non-selective herbicide, glufosinate-ammonium in apple field and for the study, 4 cultivars of apple were tested. The results showed that the highest phytotoxic degree was observed on Sansa-cultivar and then the degree was diminished on Tsugaru, Hongro and Fuji in order in the application year. In one year after application, the highest phytotoxic degree was showed on Fuji and then diminished on Tsugaru, Hongro and Sansa in order. However, in one year after application, the phytotoxic symptom was negligible in comparison with the degree in the year of application and the flowering and fruiting were reduced 30% only in Fuji, but in the other cultivars, the phytotoxic symptoms were not observed. Meanwhile, the residue of glufosinate-ammonium was below the detection limit as 0.04ppm in all tested cultivars and in 2years after application, the phytotoxic symptom was not observed in all cultivars of the apple.

Utility of the APACHE II score as a neurological prognostic factor for glufosinate-intoxicated patients with alert mental status (의식이 명료한 글루포시네이트 중독환자의 신경학적 예후인자로서 APACHE II의 유용성)

  • Purpose: In patients with glufosinate poisoning, severe neurological symptoms may be closely related to a poor prognosis, but their appearance may be delayed. Therefore, this study aimed to determine whether the Acute Physiology and Chronic Health Evaluation II (APACHE II) score could predict the neurological prognosis in patients with glufosinate poisoning who present to the emergency room with alert mental status. Methods: This study was conducted retrospectively through a chart review for patients over 18 years who presented to a single emergency medical center from January 2018 to December 2022 due to glufosinate poisoning. Patients were divided into groups with a good neurological prognosis (Cerebral Performance Category [CPC] Scale 1 or 2) and a poor prognosis (CPC Scale 3, 4, or 5) to identify whether any variables showed significant differences between the two groups. Results: There were 66 patients (67.3%) with good neurological prognoses and 32 (32.8%) with poor prognoses. In the multivariate logistic analysis, the APACHE II score, serum amylase, and co-ingestion of alcohol showed significant results, with odds ratios of 1.387 (95% confidence interval [CI], 1.027-1.844), 1.017 (95% CI, 1.002-1.032), and 0.196 (95% CI, 0.040-0.948), respectively. With an APACHE II score cutoff of 6.5, the AUC was 0.826 (95% CI, 0.746-0.912). The cutoff of serum amylase was 75.5 U/L, with an AUC was 0.761 (95% CI, 0.652-0.844), and the AUC of no co-ingestion with alcohol was 0.629 (95% CI, 0.527-0.722). Conclusion: The APACHE II score could be a useful indicator for predicting the neurological prognosis of patients with glufosinate poisoning who have alert mental status.

Development of Transgenic Soybean Using Agrobacterium tumefaciens (Agrobacterium tumefaciens을 이용한 대두 형질전환체 개발)

  • Agrobacterium tumefaciens-mediated cotyledonary node transformation was used to produce transgenic soybean. Cotyledonary node explants of three cultivars and one genotype were co-cultivated with strains Agrobacterium (LBA4404, GV3101, EHA101, C58) containing the binary vectors (pCAMBIA3301 and pPTN289) carrying with CaMV 35S promoter-GUS gene as reporter gene and NOS promoter-bar gene conferring resistance to glufosinate (herbicide Basta) as selectable marker. There was a significant difference in the transformation frequency depend on bacteria strain. The EHA101 strain of the bacterial strains employed gave the maximum efficiency (3.6%). One hundred-six lines transformed showed the resistance in glufosinate. Histochemical GUS assay showed that at least 11 plants transformed with the GUS gene were positive response. The soybean transformants were obtained from the Thorne (5 plants), 1049 (5 plants) and Bakun (1 plant), respectively. Southern blot analysis and leaf painting assay revealed that the GUS and bar gene segregated and expressed in their progeny.

The Use of Glufosinate as a Selective Marker for the Transformation of Cucumber (Cucumis sativus L.) (오이의 형질전환을 위반 선발마커로서 Glufosinate의 이용)

Histochemical gus and leaf painting assay showed that 15 individual lines were transgenic with the gus and bar gene. Southern blot analysis also revealed that the gus gene was successfully integrated into each genome of transgenic cucumber.

Development of Antibiotics Marker-free Potato Having Resistance Against Two Herbicides (두 가지 제초제에 대하여 저항성을 가지는 항생제 마커-프리 형질전환 감자 육성)

  • This study was conducted to develop an antibiotics marker-free potato (Solanum tuberosum L., cv. Taedong valley) plant having resistance against two herbicides. Agrobacterium tumefaciens strain EHA105, harboring a binary vector plasmid pCAMBIA3300 containing bar gene under the control of a promoter CaMV35S and linked CP4-EPSPS genes driven by CaMV35S promoter, was used in the current study. The leaf segments of newly bred potato variety (cv. Taedong Valley) was co-cultured with Agrobacterium. Then, the regenerated individual shoots were excised and transferred to potato multiplication medium supplemented with 0.5 mg/L phosphinothricin. The shoots were rooted in MS medium without hormone and obtained putative transgenic plant E3-6. Integration of target genes into the E3-6 plant and their expression was confirmed by PCR, Southern analysis, and ELISA test. The tissue necrosis test on young leaf blade and shikimic acid accumulation test using the tissue of E3-6 plant were conducted to investigate the resistance to glufosinate-ammonium and glyphosate, respectively. The transgenic plants (E3-6) simultaneously showed a high resistance to both herbicides. The same results were surely obtained also in the whole plants foliar-treated with alone or mixture of two herbicides, glufosinate-ammonium and glyphosate.
