고려홍삼의 당뇨병성 신장병증 개선 효과 (Renoprotective Effects of Korean Red Ginseng)

  • 자연발생적 고혈압 흰쥐에 STZ로 당뇨를 유발한 고혈압-당뇨 질환모델에서 고려홍삼 조사포닌분획과 비사포닌 분획간의 신장보호 활성을 비교해 보았다. STA 65mg/kg용량을 투여한 후 3일에 네 그룹으로 나누어 본 실험에 임하였다. 신장비대에 대한 저해 활성을 비교해 보았을 때 GTS와 GNS 각각 12, 13% 억제 활성을 나타내었고 두 군간에는 유의적인 차이를 보이지 않았다. 한편 요 중 요소질소의 배설은 GNS 투여군에서는 69% 증가(p<0.05)한 반면 GTS 투여군은 31%만이 증가하였다. 혈중 포도당, 콩 콜레스테롤 및 중성지방의 경우는 고혈압-당뇨 대조군과 비교시 홍삼투여군 두군 모두에서 유의한 차이를 발견할 수 없었다. 반면 oxidative stress를 저해하는 효소인 SOD 활성에 있어서는 GNS투여군에서 대조군에 비해 17%유의하게 상승하는 활성을 보여주었다. 신장비대의 지표인 TGG-$\beta$1, fibronectin그리고 upstream의 MEPK인 p-38, ERK 등의 발현을 그룹간 비교해 본 결과 모든 지표에서 비사포닌 분획이 사포닌 분획에 비해 우수한 억제활성을 나타내었다. TGF-$\beta$1의 mRNA 및 단백질의 발현을 살펴보았을 때 GNS 투여군은 각각 33%, 91% 억제활성을 나타낸 반면 GTS 투여군은 각각 5%, 65% 억제활성을 나타내었다. Fibronectin 단백질의 발현에서도 GNS 투여군은 87% 억제 활성을 나타낸 반면 GTS 투여군은 28% 저해활성을 나타내었다. 본 실험의 결과를 종합해보면 고려홍삼의 신장보호 활성은 사포닌 분획보다는 비사포닌 분획에 함유된 성분에 의해 일어나는 효과가 아닌가 추측된다.

  • We have been isolating various physiologically active substances from non-saponin fraction of Korean Red Ginseng These are adenosine, gyro-glutamic acid, dencichine and acidic polysaccharide. Adenosine and gyro-glutamic acid are loom to inhibit epinephrine-induced lipolysis in fat cells and stimulate the insulin-mediated lipogenesis. In addition to these actions, adenosine was found to inhibit both norepinephrine- and histamine-induced aorta constriction, and pyre·glutamic acid inhibits angiotensin-converting enzyme. Dencichine stimulated histamine-induced aorta constriction. Finally, acidic polysaccharide was found to inhibit both lipolytic and anorexigenic actions of Toxohormone-L. Based on these experimental results, I presented a brief review on these compounds isolated from non- saponin fraction of Korea Red Ginseng.   • This study was undertaken to determine the effects of ginseng total saponin (GTS) on stress- induced analgesia (SIA) in mice. intermittent foot shock (FS)-SIA was antagonized not by on but by naloxone in the tail flick FS-SIA which was not antagonized by naloxone in the T.F. test. On the other hand, GTS did not antagonize the continuous FS-SIA naloxone antagonized in the T.P. test. Also GTS antagonized psychological (PSIF)-SIA which was not antagonized by naloxone in the T.F. test. However, GTS did not antagonize the PSY-SIA which naloxone antagonized in the T.P. test. Forced swimming (FSIP)-SIA was not affected by both GTS and naloxone. These results suggest that the antapeonisms of intermittent FS-SIA in the T.F. test, continuous FS-SIA and PSY-SIA by GTS are mediated by non-opioid mechanisms but the antagonism of intermittent FS-SIA in the T.P. test by GTS is mediated by opioid mechanism.

초임계 유체로 추출된 산삼 부정 배양근 오일의 효모균 처리에 의한 비사포닌계 지방산 함량 변화 및 Collagenase 및 Elastase 저해 활성 증대 (Changes in Non-saponin Fatty Acid Content and Increases in Inhibitory Activities of Collagenase and Elastase by Treatment with Saccharomyces cerevisiae of the Supercritical Fluid Extracted Oil of the Adventitious Roots Culture of Wild Mountain Ginseng)

  • Background: To obtain useful cosmetic resources, this study aimed to determine the non-saponin fatty acid and inhibitory activities of collagenase and elastase by treatment of Saccharomyces cerevisiae in supercritical fluid extracted oil of the adventitious root culture of wild mountain ginseng. Methods and Results: We performed supercritical fluid extraction at various conditions such as pressure, temperature, time, and use of co-solvents, unlike the n-hexane extraction for the adventitious roots culture of wild mountain ginseng. The non-saponin-fatty acid obtained from the oil of the adventitious roots culture was incresed by treatment with S. cerevisiae. The supercritical fluid extraction was conducted using gas chromatography. Non-saponin-fatty acid content, in the oil of adventitious roots culture of wild mountain ginseng treated with S. cerevisiae for 2 days were three times higher than that in the control. In addition, the oil of the adventitious roots culture treated with S. cerevisiae was investigated for the anti-wrinkle effect by using collagenase and elastase. The oil of adventitious roots culture treated with S. cerevisiae exhibited higher collagenase and elastase inhibitory activities than those in the control. Conclusions: Supercritical fluid extracted oil of the adventitious roots culture of wild mountain ginseng treated with S. cerevisiae was found to have decreased ratio of saturated fatty acids and incresed ratio and content of unsaturated fatty acids increased. Furthermore, it showed anti-wrinkle effects in vitro.

Immunomodulatory Effects of Non-saponin Red Ginseng Components on Innate Immune Cells

  • for 4 h, tumoricidal activity was significantly increased. However, NSRG had effects on tumoricidal activity of macrophages at low concentration (1 ug/ml), which was not related to the production of nitric oxide. The mitogen response of lymphocytes to LPS and ConA in the spleen did not show significant differences between the control and NSRG-treated cells, whereas LPS-induced blastogenesis was slightly increased at 100 ${\mu}g/ml$ (p < 0.05). These results suggest that NSRG has differential effects on innate immune response and could be useful as immunotherapy for cancer treatment.

묘삼경의 근형성에 미치는 식물 생장 조절물질의 영향 (Effects of Phytohormone on the Root Formation of Stem Cuttings in Panax ginseng C.A. Meyer)

  • This study was carried out to obtain the basic information about the large scale propagation of ginseng (Panax ginseng C.A. Meyer). Therefore, the stem cuttings of 1-year old ginseng, treated with various concentrations of plant growth regulators for 5 seconds (quick dipping) and 24 hours (prolonged soaking), were cultured. The root formation of stem cuttings was varied with the concentrations, kinds, and treatment methods of plant growth regulators. Besides normal-looking roots various malformed roots were observed. In the prolonged soaking method, the culture of stem cuttings, treated with 10 ppm of IBA or NAA, resulted in profuse root regeneration. And stem cuttings, in quick dipping method, treated with 2000 ppm of IBA or NAA resulted in more excellent root regeneration. In general, IBA was more vigorous for the root formation than NAA, The treatment with 50 ppm kinetin or 100 ppm BA brought good result for the retardation of senescence of stem cuttings and BA treatment was more effective than kinetin. As for the saponin content of roots derived from stem cutting culture, the roots, formed by non-treatment of growth regulators, were higher in saponin content than those formed by treatment of growth regulators.

Effect of Ginseng Total Saponin on Bovine Adrenal Tyrosine Hydroxylase

  • Ginseng total saponin (GTS) can modulate dopaminergic activity at both presynaptic and postsynaptic dopamine receptors (Kim et al, 1998). The present study investigated t he effect of GTS on the bovine adrenal tyrosine hydroxylase (TH), which catalyze L/tyrosine to DOP. GTS inhibited the bovine adrenal TH by 42.4, 51.5 and 55.3% at concentrations of 40, 80 and 100${\mu}g$/ml, respectively. The IC50 value of GTS was 77.5${\mu}g$/ml. GTS exhibited noncompetitive inhibition with a substrate L-tyrosine. The Ki value was 155${\mu}g$/ml.

CA 및 MA 저장이 수삼 및 홍삼의 외관품질 및 사포인 조성에 미치는 영향 (Effect of Controlled Atmosphere and Modified Atmosphere Storage on the Apparent Quality and Saponin Component of Fresh and Red Ginseng)

  • During the controlled atmosphere storage (CA), fresh ginseng showed good appearance in quality, and other deterioration of freshness was not observed until 12 weeks. On the other hand, MA storage had kept freshness only in treatment of 1 until 8 weeks. There was no significant difference between treated and non-treated sample with preservatives, and not treated sample was not infected with various different fungi. Moisture contents and hardness of ginseng in all treatments were not changed much until 12 weeks, and surface shrinkage did not occur either. But shear stress increased somewhat in all treatments after 12 weeks. The granule of microstructure in tissue diminished slightly. The apparent Quality of red ginseng was good until 4 weeks of treatment. But as time passed, white skin and wrinkled skin were generated and darkened in its color. B-1 in CA and E-1 in MA were found to be the most favorable one. The content of crude saponin did not change significantly during storage of CA or MA by preservation conditions and period. Though a small increase in saponin content from 4.92% to 5.43% was recognized in B-1, which was treated with preservative and 6.0% In B-2, control, this could rather explain increment of soluble component by butanol. Thus, there was no change in total contents of ginsenoside pattern and composition of each content. The Rbl content in B-1 and B-2 were 0.98%, and 0.97%, respectively, whereas that of control was 0.96%. E-1 of MA, treated with preservative was 5.32% after 12 weeks, but was 5.73% in control, indicating that ginsenosides pattern was quite similar to that of CA storage.
