홍삼추출물 및 농축물의 마이야르 갈색화반응 촉진에 미치는 아미노산 및 당의 영향 (Effects of Amino Acids and SLlgars on the Maillard Brou'nine Reactions during Extraction and Concentration of Red Ginseng)

  • Browning intensity is a major factor to estimate the quality of red ginseng or red ginseng products. The Maillard type of browning reaction proceeds nonenzymatically during extraction and concentration of red ginseng. The present studies were carried out to investigate the effects of amino acids and sugars on the browning reaction during extraction and concentration of red ginseng. Red ginseng was pulverized to 115 mesh and then tenfold (v/w) of water was added to the powder to make the substrate of red ginseng. Solution (0.1 M) of fourteen amino acids and of folly silgars were added to the substrates of red ginseng powder and these were then extracted and concentrated to examine their browning intensities. Amino acids were more effective than sligars in acrelerating the browning reaction. Acceleration of the browning reaction in the concentrate was in the order of arginine> histidine>glycine>alanine>lysine phenyl alanine>aspartic acid>lelicine>threonine>gllitamic acid>tyrosine>valine>istleucine>methionine for amino acids, and was glucose>frlictose >silcrose, maltose for sugars.

Impulse-Vacuum System을 이용한 홍삼엑스의 추출 (Extraction of Red Ginseng Extract by Impulse Vacuun System)

  • 추출 시간을 5시간으로, 추출 온도를 33-5$0^{\circ}C$로 설정한 진공역적방식 조건과 8$0^{\circ}C$의 고온의 다중단 추출방식 간의 조사포닌 함량비교 결과, 고온의 다중단 추출방법으로 제조한 흥삼엑스의 조사포닌 함량이 8.1-8.2%인데 비하여, 저온 및 단시간 추출인 진공역적 추출방법은 조사포닌 함량이 5.4-5.9%로 조사포닌 함량은 낮은 반면에 사포닌 Rg$_1$ 및 Re의 분해가 거의 없는 것으로 나타났다. 또한 진공역적 추출방법이 다중단 추출방식보다 수율에서도 15-20% 높은 경향을 나타내었다. 진공역적추출 엑스의 색상은 기존 다중단 추출 방식으로 추출된 엑스와 유사하였고, 유동성은 보다 향상된 것으로 나타났다.

추출조건이 홍삼엑기스의 당과 유리당의 조성에 미치는 영향 (Effect of Extracting Conditions on Some Factors Affecting the Sugar Composition of Red Ginseng Extract)

  • The quality of concentrated red ginseng extract, which was prepared from Korean red ginseng tails, was studied with respect to the changes in the ethanol concentration in the range of 0-90% and temperature of 70-10$0^{\circ}C$ during 1-5 times of extraction. Each extraction time was taken 8 hours at given temperature. The ratio of free sugars to total sugar changed significantly with the number of extraction and ethanol concentrations. An increase in reducing sugar and free sugars and a decrease in extracted soluble starch were fond as ethanol concentration increased. Extraction temperature was found little effect on extractability of sugars and their ratios. Analysis of free sugars in red ginseng extract identified 6 frdd sugars such as rhamnose, xylose, fructose, glucose, sucrose and maltose, including sucrose to be major.

인삼 조사포닌의 조제 방법 개선 (Improved Method for the Preparation of Crude Ginseng Saponin)

  • This stuffy was carried to establish a new efficient method for the preparation of edible crude ginseng saponin. The conventional butanol extraction and resin adsorption methods were compared for the contents of total crude ginseng saponin and major ginsenosides. Seventy- percent methanol extract was applied to Diaion HP-20 column and the resin was washed with Hn and eluted with absolute methanol. The methanol elute was dried in vivo and analyzed for its ginsenosides. Use of ethanol instead of methanol to make edible crude ginseng saponin gave a similar result. Butanol extraction was performed by the conventional method. The final aqueous layer from butanol extraction was passed through Diaion HP-20 column followed by elution with methanol and Diaion HP-20 passed fraction was extracted with butanol to recover remaining components, respectively, in order to determine saponin loss. TLC and HPLC qualitatively and quantitatively monitored Ginsenosides, respectively. Loss of ginsenosides was higher in butanol extraction method than in Diction HP-20 adsorption method. In addition, saponin fractions prepared by Diction HP-20 adsorption method showed higher content of each ginsenoside, showing 8.2% higher purity than that of butanol extracted fraction. From these results, we propose the resin adsorption method as a new efficient measure for the preparation of crude ginseng saponin, which is edible by using spirit instead of methanol.

인삼 꽃의 물 추출 온도 및 추출 시간이 진세노사이드 함량 및 품질에 미치는 영향 (Effects of Extraction Temperature and Time on Ginsenoside Content and Quality in Ginseng (Panax ginseng) Flower Water Extract)

  • In this study, ginseng flower water extracts were analyzed to set up the ginsenoside content and quality optimization condition. The highest total ginsenoside content among the ginseng flower water extracts was 67.44mg/g which was extracted at $85^{\circ}C$ for 3 hours. In addition, the ginsenoside content decreased according to the increased extraction temperature and time. The highest total content of $Rb_2$ and Re was 37.42mg/g at $75^{\circ}C$ for 6 hours. Total content of $Rb_2$ and Re decreased according to the increased extraction temperature and time. The highest prosapogenin ($Rg_2$ + $Rg_3$ + $Rh_1$) content among the total of ginseng flower water extracts was 18.58mg/g which was extracted at $95^{\circ}C$ for 12 hours. The sweetness, absorbance were increased according to the increased extraction temperature and time. But pH was decreased according to the increased extraction time.

인삼 다당체 추출 공정 개발을 위한 인삼의 추출 조건 및 원료에 따른 총당 변화 (Analysis of Total Sugar by Extraction Condition and Material to Develope the Extraction Process of Ginseng Polysaccharide)

  • 인삼 속에 다량으로 함유되어 있는 다당체 성분인 진산에 대한 효능이 연구되고 있는 시점에서 산업적으로 이용할 수 있는 대량 추출공정을 위한 추출조건을 분석하였다. 인삼의 종류는 홍삼(5년근)이 $32\%$로 총당의 함량이 가장 높았으며 금산 수삼(5년근)은 $31\%$$1\%$ 정도 조금 적게 나타났다. 그러나 홍삼 제조 공정에 따른 효율을 비교해 본 결과 금산 수삼(5년근)과 별 차이가 없어 금산 수삼을 사용하였다. 수삼부위 중 주근, 최적온도 $80^{\circ}C$, 최적 추출시간 6시간 추출 시 총당의 추출 효율이 가장 높았다. 또한 총당의 추출효율을 높이기 위해 인삼 추출물에 protease, ${\alpha}-amylase$, cellulase 효소 처리했을 때 추출 효율이 $20\%$까지 높은 것으로 나타났으며, 인삼 추출액에 주정 알콜농도 $70\%$, 동결건조 방법이 총당 회수율이 높았다.

  • This study compared the contents of ginseng prosapogenin depending on the extracting conditions of Ginseng Radix palva(Panax ginseng) to provide basic information for developing Ginseng Radix palva-based functional foods. Our findings show that the content of crude saponin peaked at 18 hours of extraction and when extracted twice at $100^{\circ}C$ (GRP-18). However, the content of total saponin reached its height at 6 hours of extraction at $100^{\circ}C$ (GRP-6) and when extracted twice. On the other hand, the content of ginsenoside $Rg_3$, $Rg_5$ and $Rk_1$ from Red and Black ginseng reached their heights at 18 hours of extraction, followed by 72 hours and 15 hours of extraction at $100^{\circ}C$. And at $100^{\circ}C$ the main prosapogenin of the content of Black ginseng ginsenoside $Rg_5$ and $Rk_1$ reached their heights at 18 hours of extraction, followed by 72 hours and 15 hours of extraction.

Ginsenoside composition of Panax ginseng flower extracts obtained using different high hydrostatic pressure extraction conditions

  • Ginsenosides are active constituents of ginseng (Panax ginseng) that have possible anti-aging, physiological and pharmacological activities, such as anti-cancer and anti-inflammatory effects. Although the ginseng root is generally used more often than the aerial parts for medicinal purposes, the flowers also contain numerous ginsenosides, including Rb2, Rc, Rd, Re and Rg1. Therefore, an extract from the flowers of the P. ginseng could have the pharmacological efficacy of bioactive compounds including ginsenosides. The high hydrostatic pressure extraction (HHPE) is a method that is used for the efficient extraction of bioactive compounds from plant materials. In this study, we compared the yield of ginsenosides from ginseng flowers under different conditions of extraction pressure and time of HHPE. The results indicate that the total yield of the ginsenosides improved as the pressure increased from 0.1 to 80 MPa and treatment duration increased to 24 hours. In addition, the ginsenoside extracts from HHPE at 80 MPa, which possessed a higher total ginsenoside concentration, decreased the viability of the primary human epidermal keratinocytes (HEKs) significantly than the ginsenoside extracts from HHPE at 0.1 MPa. Collectively, we found that the method of HHPE that was performed for 24 hours at 80 MPa showed the highest yield of ginsenosides from the flowers of P. ginseng. In addition, our study provides a foundation for the efficient extraction of ginsenosides, which had a potent bioactivity, from flowers of P. ginseng through HHPE.

Microwave를 이용한 인삼 중의 잔류 농약 추출 (Extraction of Pesticide Residues in Ginseng by Microwave)

  • 인삼시료에 있는 quintozene, tolclofos-methyl 그리고 procymidone 등의 농약성분을 추출하기 위한 방법으로서 microwave를 이용할 수 있을 지의 여부와 인삼을 가공제품으로 만들기 전에 microwave를 이용하여 잔류농약 성분을 신속히 추출하여 제거시킬 수 있는지 알고자 하였다. 인삼분말 시료에 hexane을 추출용매로 이용한 microwave 추출의 최적 조건은 power 150 watts, 추출시간 2분이었고 그 추출 효율은 Soxhlet으로 7시간 동안 가열 환류 추출한 결과를 지수 100으로 하였을 때 quintozene 100, tolclofos-methyl 92, procymidone 100으로 거의 동일한 수준으로 나타났다. 그리고 microwave 추출법을 사용함으로써 실제 Soxhlet 추출법에서 사용된 용매양의 최소 7배 이상 감소시킬 수 있었고 추출 시간 또한 7시간에서 2분으로 훨씬 단축되었다. 인삼 엑기스 제조 전 잔류농약 성분의 제거 실험에 있어서 45%나 60% ethyl alcohol을 용매로 사용하고 microwave로 power 150 watts 추출시간 2분 이내로 1회 추출하여 quintozene 100%, tolclofos-methyl 69%, procymidone 83%정도의 제거효과를 얻었다.

고려홍삼의 총 페놀성환의 정량 및 그 추출조건 (Determination of Total Phenolic Compounds from Korean Red Ginseng, and Their Extraction Conditions)

  • 본 연구에서는 홍삼에 함유되어 있는 총 phenolic화합물을 정량하기 위하여 Folin-Denis방법을 사용하였으며, 이 때 정량에 방해를 줄 수 있는 물질에 대해서 조사하였고 홍삼분말로부터 phenolic 화합물 추출조건을 조사하였다. 아미노산중에서는 tyrosine, cystein 및 tryptophan이 영향을 미쳤고, 여러 가지 유기산, 당류 및 ginsenosides는 거의 영향을 미치지 않았다. 비타민 중에서는 vitimln B$_{12}$, d-biotin, D-pantothenic acid, niacinamide, nicotinic acid, vitamine A palmitate, vitamine D$_3$ 등을 제외한 비타민은 발색에 영향을 미쳤다. 홍삼으로부터 phenolic 화합물을 추출하기 위한 추출조건은 60% ethanol을 사용해서 80。C에서 1-2시간씩 3회 정도 추출이 적당하다고 판단된다.
