Studies on Saprogenic Bacteria Isolated from Korean Red Ginseng Extrart Product (홍삼 추출액 파우치 제품으로부터 분리동정된 변패세균에 관한 연구)

  • Five kinds of saprogenic bacteria were isolated from the red ginseng extract product and identified as Bacillus subtilis, Bacillus sp., Paenibacillus sp., Micrococcus sp., and Pseudomonas sp. by 16S rDNA analysis. Some of the isolated strains were able to grow even at $45^{\circ}C$ which are presumed originated from the raw ingredient of red ginseng extract. All of the isolated strains did not show the hemolytic activity, the diarrhea-inducing activity, and the vascular permeability enhancing activity, indicating that these strains are not pathogenic.

Development of Liquid Phase Product from Red Ginseng and Medicinal Herbs for Alcoholic Beverage (홍삼 및 기능성 소재를 이용한 주류첨가용 액상제제 개발)

  • This study was conducted to investigate the effect of crude saponin of red ginseng and aqueous extracts from several medicinal herbs on the activity of alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) in alcohol metabolism. In order to develop a liquid phase product a model product was prepared using functional plants. In case of red ginseng crude saponin, and some medicinal herbs(Puerariae radix, the flower of Puerariae lobata, the fruits of Hovenia dulcis Thunb., Morus alba L.) the relative activity of ADH was significantly increased, which was from 104~114%. The relative activity of ALDH was also significantly increased in the cases of the fruits of Hovenia duicis Thunb., Morus alba L. which was from 102∼106%. To improve of soju flavours by addition of liquid phase product, it is prepared three samples added a given content (0.28%) of chik, ssanghwa and kyungokgo flavour. The total acceptability test showed significant differences among three samples, and it is observed that the sample added ssanghwa flavour get the best grade. As the second acceptability test, the preference of ssanghwa flavoured sample, sample without flavour and a similar product in market was compared. There was no significant difference among three samples.

Current Status of Korean Ginseng Products and Trends in Enhanced Functional Ginseng Products

  • The abolishment of the red ginseng monopoly act by the Korean government in 1996 resulted in a drastic change in the Korean ginseng industry, leading to a significant increase in the market size and consumption of ginseng products. Red ginseng is most popular type, with approximately 74% of harvested fresh ginseng being processed into various red ginseng products. Since 1997, there has been a substantial increase in the cultivation of ginseng for production of red ginseng, which, in turn, has contributed to the proliferation of ginseng processing companies. To investigate the products of ginseng manufacturing businesses, we select 200 companies primarily engaged in ginseng processing or specializing solely in ginseng. Our survey on the status of ginseng industry covered 8 different categories. 1) Root ginseng: There were 66 companies involved in manufacturing red ginseng root, accounting for 33.0% of all surveyed companies. This was followed by black ginseng root with 36 companies (18.0%) and red ginseng fine roots with 22 companies (11%). 2) Red ginseng products: A total of 144 companies were involved in manufacturing red ginseng pouches, making it the most common product category. This was closely followed by 142 companies producing pure(100%) red ginseng extract concentrate. 3) Fermented red ginseng products: Companies producing fermented red ginseng extract concentrate products were the most numerous, totaling 26. Following this, companies producing fermented red ginseng stick and pouch products were next in line. 4) Ginseng products: There were 15 companies involved in the production of ginseng products, with the majority focusing on ginseng tea. 5) Black ginseng products: Companies producing black ginseng extract concentrate were the most numerous, with 31 companies, followed by 26 companies producing black ginseng extract pouches. 6) Taegeuk ginseng products: Only 5 companies were involved in the production of taegeuk ginseng products. 7) Fermented black ginseng, and 8) Ginseng berry products: These categories are manufactured by less than 5 companies each. However, the variety in ginseng berry products suggests the potential for future growth. In the 2000s, a trend emerged with the development of new processed products aimed at enhancing the functional components of red ginseng, and these products have captured the attention of consumers. However, this study primarily focuses on black ginseng, fermented red ginseng/fermented black ginseng, and ginseng berry products as they have exerted a significant influence on the overall ginseng industry.

The Comparison of Ginseng Prosapogenin Composition and Contents in Red and Black Ginseng (흑삼과 홍삼의 인삼 프로사포게닌 성분 비교)

  • The objective of this study is to provide basic information for developing a high-value ginseng product using ginseng saponin and prosapogenin. In order to achieve the proposed objective ginsenoside compositions of Black (BG) and Red (RG) ginseng extract with 95% ethyl alcohol were examined by means of HPLC. The crude saponin and ginsenoside composition of processed ginseng products were analyzed and compared, with BG topping the list with a crude saponin content of 7.53%, followed by RG (5.29%). Ginseng prosapogenin (ginsenosides $Rg_2$, $Rg_3$, $Rg_5$, $Rg_6$, $Rh_1$, $Rh_4$, $Rk_1$, $Rk_3$, $F_1$ and $F_4$) in BG was found to be contained almost 2.6 times as much as that in RG. Ginsenosides $Rg_3$, $Rg_5$, $Rk_1$, $Rh_4$ and $F_4$ in BG in particular were found to be almost 3 times as much as those in RG. $Rg_6$ and $Rk_3$ in BG were also found to be almost 4 times as much as those in RG.

  • Korean ginseng (Panax ginseng Meyer) is a traditional herb used across the world to treat various diseases. Although, red ginseng is this herb's most famous product and has demonstrated diverse pharmacological activities, white ginseng (WG) is another ginseng product that is made fresh and individually regulated in Eastern Asia. Red and white ginseng show different characteristics due to distinct processing steps despite originating from the same plant, and the drug interactions induced by WG have not been well documented. Selegiline is a selective monoamine oxidase (MAO) inhibitor used as an antidyskinetic and antiparkinsonian agent. Here we developed a quantification method for selegiline in mouse plasma using a C8 stationary phase in triple quadrupole-mass spectrometry (LC-MS/MS) with multiple reaction monitoring (MRM). The validated LC-MS/MS method was successfully applied to determine the potential interaction with WG extract (0.1 g/kg/day) pre-administered for 4 weeks. The $AUC_{0-240min}$ of selegiline was altered due to a decrease in the absorption of selegiline with repeated administration of WG extract.

Type-specific Amplification of 5S rRNA from Panax ginseng Cultivars Using Touchdown (TD) PCR and Direct Sequencing

  • Generally, the direct sequencing through PCR is faster, easier, cheaper, and more practical than clone sequencing. Frequently, standard PCR amplification is usually interpreted by mispriming internal or external regions of the target template. Normally, DNA fragments were eluted from the gel using Gel extraction kit and subjected to direct sequencing or cloning sequencing. Cloning sequencing has often troublesome and needs more time to analyze for many samples. Since touchdown (TD) PCR can generate sufficient and highly specific amplification, it reduces unwanted amplicon generation. Accordingly, TD PCR is a good method for direct sequencing due to amplifying wanted fragment. In plants the 5S-rRNA gene is separated by simple spacers. The 5S-rRNA gene sequence is very well-conserved between plant species while the spacer is species-specific. Therefore, the sequence has been used for phylogenetic studies and species identification. But frequent occurrences of spurious bands caused by complex genomes are encountered in the product spectrum of standard PCR amplification. In conclusion, the TD PCR method can be applied easily to amplify main 5S-rRNA and direct sequencing of panax ginseng cultivars.

Biotransformation of Ginseng Extract to Cytotoxic Compound K and Ginsenoside $Rh_2$ by Human Intestinal Bacteria

  • When saponin extracts of dried ginseng and red ginseng were anaerobically incubated with human intestinal microflora, these extracts were metabolized to compound K and ginsenoside $Rh_2$, respectively. However, when these extracts were incubated with commercial lactic acid bacteria, these did not metabolize these ginsenosides to compound K or ginsenoside $Rh_2$. Among some intestinal bacteria isolated from human feces, Bacteroides C-35 and C-36 transformed these saponin extracts to compound K and ginsenoside $Rh_2$, respectively. These bacteria also transformed water extracts of dried ginseng and red ginseng to compound K and ginsenoside $Rh_2$, respectively, similarly with that of the saponin extracts. Among transformed ginsenosides, compound K and 20(S)-ginsenoside $Rh_2$ exhibited the most potent cyotoxicity against tumor cells.

Effects of Dietary Ginseng By-product on Growth Performance and Pork Quality Parameters in Finishing Pigs (인삼부산물 급여에 따른 비육돈의 생산성 및 육질 특성에 미치는 영향)

  • The objective of the present study was to investigate the effect of dietary supplementation of the ginseng by-product on growth performance and meat quality in finishing pigs. The animals used in the experiment were a total of 24 Landrace×Yorkshire and weighted 65.81±2.02kg. The experimental diets were basal diet(CON) and 2.5% ginseng by-product(GBP), which replaced lupin in basal diet. The pigs were allotted at 4 pigs per pen with three replicate pens per treatment by completely randomized design. In growth performance, ADFI was significantly lower(P<0.0001) in GBP than in CON. In plasma biochemical composition, total protein(P<0.01), blood urea nitrogen(P<0.03), glucose(P<0.01), albumin(P<0.02), calcium(P<0.01) and inorganic phosphate(P<0.01) were significantly higher in GBP than in CON. Carcass and meat quality were not significantly different between treatments. Total ginsenoside content on meat was significantly higher(P<0.0001) in GBP than in CON. TBARs was significantly lower in GBP than in CON for 6 days(P<0.03) and 12 days (P<0.06), respectively. Our research indicates that plasma biochemical composition, total ginsenoside content and TBARs were affected when replaced with ginseng by-product. Ginseng by-product in the pig diet increased pig muscle ginsenoside, indicating that ginseng by-product can be used as a feed additive.

Quality Characteristics of Yanggaeng with White, Red and Black Ginseng Powder (백삼, 홍삼, 흑삼 분말을 이용한 양갱 제조 및 품질 특성)

  • Powders prepared from three different varieties of Korean ginseng (white, red and black) are consumed daily by many Koreans as a functional food material. These variants exert a variety of physiological effects. The principal objective of this study was to evaluate the quality characteristics of Yanggaeng prepared with three different types of ginseng powder. We conducted tests for ginsenosides content, Hunter's color values, mechanical characteristics, and sensory evaluation analysis. The ginsenosides of black ginseng were higher than those of white and red ginseng. The lightness of the black ginseng Yanggaeng was significantly less than the white, red, and control samples (p<0.05). We noted significant differences in Hunter's a and b values. With regard to the mechanical properties of the samples, we noted significant differences in gumminess (p<0.05), but no differences in hardness, springiness, chewiness, and cohesiveness were detected. The results of sensory evaluation showed that there were significant differences in the color, flavor, and overall quality of the samples (p<0.05), but there were no significant differences in taste and texture. Overall, red ginseng Yanggaeng appeared to have the most commercial value for improving the Yanggaeng product.

Quality Inspection to the Ginseng Saponins in Commercial Ginseng-extracts (인삼(人蔘)사포닌을 중심(中心)한 인삼정(人蔘精)의 품질조사(品質調査))

  • The quality inspection was made on commercial Korean ginseng extract (Company A, B, C, D, E, F, G and H), particularly on samples available in Seoul market area. The results were as follews: 1) Among the ginseng extract products out of eight different manufactures, the moisture content of D company's product showed 46.5%, and other companys' met with the moisture standard for ginseng extracts. And protein, fat, ash, fiber and total sugar were about $9.87{\sim}21.07%,\;0.46{\sim}1.62%,\;6.55{\sim}7.88%,\;0{\sim}0.15%\;and\;58.58{\sim}76.74%$, respectively. And residue of D and F company products showed 3.25% and 3.61% which exceed the standard, and other company products met with residue test specifications. 2) The contents of ginseng saponins were 16.16% and 13.12%, respectively, for the C and H company products. However, other company's showed below 9%. By fractional distribution of ginseng saponins, it is supposed to be white ginseng and lateral ginseng that were mostly used as raw materials for ginseng extract manufacturing.
