Partial Characterization of Proteases from Culture Filtrate of Mycobacterium tuberculosis

  • Two proteases were partially characterized from culture filtrate of Mycobacterium, tuberculosis KIT110. Their molecular weights were approximately 200 and 180 kDa, respectively and they exhibited similar enzymatic characteristics. These enzymes were inhibited significantly by EDTA and to some extent by EGTA. Their activity was enhanced by $Ca^{2+}$ and $Mg^{2+}$ to some degree. However, $Cu^{2+}$ and $Ag^{2+}$ completely inhibited the enzyme activity at the concentration of 2.5 and 5 mM, respectively. The optimal pH was 7.0 and optimal temperature was around $40^{\circ}C$. These enzymes were rapidly inactivated at $80^{\circ}C$. Therefore, they were heat-labile, neutral metalloproteases. These enzymes exhibited antigenicity shown by their reacting with sera from the partients with pulmonary tuberculosis. These enzymes were able to degrade serum proteins including hemoglobin, bovine serum albumin, lysozyme and immunoglobulin G and structural matrix protein such as type I collagen. Therefore, these enzymes may be thought to contribute to tissue necrosis and pathogenesis during infection.

Characterization of a Myostatin-like Gene from the Scallop Patinopecten yessoensis

  • Myostatin (GDF8) is a growth factor that limits muscle tissue growth and development in vertebrates. We isolated a myostatin-like gene (Py-MSTN) from the marine invertebrate, the scallop Patinopecten yessoensis. Py-MSTN was highly expressed in the adductor muscle and in the gill unexpectedly. Amino acid analysis showed that Py-MSTN has 49% amino acid sequence identity and 64% similarity to human myostatin (Hs-MSTN), and 42% identity and 61% similarity to myoglianin, the only invertebrate homolog. These results indicated that Py-MSTN may be functionally similar to the vertebrate MSTN than the invertebrate homolog. Phylogenetic analysis suggested that Py-MSTN is an ancestral form of vertebrate MSTN and GDF11 and does not belong to other $TGF-{\beta}$ family members. Molecular modeling showed that Py-MSTN exhibits a similar tertiary structure to mammalian BMP7, a member of $TGF-{\beta}$ family. In addition, the amino acid residues which contact extracellular domain of the receptor were relavively conserved. Given these results, we propose that Py-MSTN is a functionally active member of the $TGF-{\beta}$ family and is involved In muscle growth and regulation.

Increased Expression of Nitric Oxide Synthases in Left and Right Ventricular Hypertrophy

  • The present study was aimed to explore pathophysiological implications of nitric oxide in the development of left and right ventricular hypertrophy. To induce selective left and right ventricular hypertrophy, rats were made two-kidney, one clip (2K1C) hypertensive and treated with monocrotaline (MCT), respectively. Six weeks later, the hearts were taken and their ventricular tissue mRNA and protein expression of endothelial constitutive isoform of nitric oxide synthase (NOS) were determined by reverse transcription-polymerase chain reaction and Western blot analysis, respectively. In 2K1C hypertensive rats, the expression of NOS mRNA was increased in parallel with its proteins in the left ventricle, but not in the right ventricle. In MCT-treated rats, the expression of NOS mRNA and proteins were proportionally increased in the right ventricle, but not in the left ventricle. These results suggest that the expression of NOS is specifically increased in association with the ventricular hypertrophy, which may be a mechanism counteracting the hypertrophy.

Changing Salinity Affects Hematological and Histological Response in Hybrids and Hybrid Triploids between River Puffer, Takifugu obscurus and Tiger Puffer, T. rubripes

  • We investigated the physiological responses of the river puffer, Takifugu obscurus, the tiger puffer, T. rubripes, a hybrids of the two (river puffer${\times}$tiger puffer) and a hybrid triploids to acute changes of salinity from 30 psu to 0 psu and from 0 psu to 30 psu. The blood and plasma factors of each species were elevated for 48, 72, or 96 hrs, and thereafter decreased due to hyper-osmoregulation and hypo-osmoregulation. In hyper-osmoregulation and hypo-osmoregulation, the cortisol concentration of river puffer, hybrids, hybrid triploids and tiger puffer increased for 12 or 48 hrs, and decreased thereafter. Chloride cells in the gill filaments of each species increased with increasing salinity, and melano-macrophages in the kidney tissue of each species increased with decreasing salinity. In conclusion, the hematological and stress responses of the hybrids were between those of the river puffer and tiger puffer, and the hematological responses of the hybrid triploids were higher than those of the other groups. The stress response of the hybrids was more sensitive than that of the hybrid triploids. In all groups, the histological responses of kidney in hyper-osmoregulation were more sensitive than those in hypo-osmoregulation.

Detection of herpesviral hematopoietic necrosis virus (Cyprinid herpesvirus 2, CyHV-2) from goldfish, Carassius auratus (L.) in Korea

  • In April and June 2014, mortalities of goldfish occurred at Korea. The principal signs included pale gills, severely enlarged and softened spleen and kidney and red liver. Moreover, the histopathological characteristics observed mainly in hematopoietic tissues of kidney, gill lamellae and intestine. The predominant histopathological changes were severe necrosis of hematopoietic tissue. In addition, nucleus exhibiting peripherally displaced chromatin were particularly abundant in the kidney of affected fish. The histological examination led to hypothesize the implication of a virus in the mortality. The hematopoietic necrosis herpesvirus (Cyprinid herpesvirus 2, CyHV-2) DNA polymerase gene successfully detected in DNA extracted from kidney and spleen of affected fish using PCR assay and showed 100% identity with already deposited CyHV-2 DNA polymerase gene in NCBI. Artificial infection trials using affected tissues filtrates gave cumulative mortalities of 30% for virus injected goldfish. In the present study, CyHV-2 was detected and identified as the causative pathogen of the epizootic in goldfish in Korea.

Determination of arsenic and mercury in longtail tuna (Thunnus tonggol) collected from Terengganu waters: risk assessment of dietary exposure

  • Despite the beneficial aspect of fish consumption, bioaccumulation of toxic metals such as arsenic (As) and mercury (Hg) can enhance the health risk for the consumers. Arsenic and Hg concentrations were measured in edible tissue and two targeted organs, namely gill and liver of longtail tuna species (Thunnus tonggol) from Terengganu waters, including Kuala Besut, Kuala Terengganu, Dungun and Kemaman. The concentration of As and Hg were analysed by using inductively coupled plasma- mass spectrometry. The mean concentrations of both elements were significantly different (p < 0.05) among the locations and targeted organs. The hierarchy of As and Hg mean concentrations in muscle samples were Dungun > Kuala Besut > Kemaman > Kuala Terengganu. The mean concentration of As in all samples, including muscle, exceeded the permitted level set by Malaysia Food Act. Estimate Weekly Intake (EWI) was conducted to assess the health risk effect, and 63 kg was used as the average body weight of Malaysian adults. However, the EWI values show that the weekly intake of As and Hg does not exceed the provisional tolerable weekly intake limit suggested by Food and Agricultural Organization for the United Nations and is considered safe to be consumed.

Physico-chemical Characteristics and In situ Fish Enclosure Bioassays on Wastewater Outflow in Abandoned Mine Watershed (폐광산 지역의 유출수에 대한 이.화학적 수질특성 및 Enclosure 어류 노출시험 평가)

  • The objectives of this study were to evaluate the physico-chemical water quality, trophic and tolerance guilds in the control ($C_o$) and impacted streams of the abandoned mine, along with the ecological health, using a multimetric health model and physical habitat conditions of Qualitative Habitat Evaluation Index (QHEI), during the period of three years, 2005~2007. Also, eco-toxicity ($EE_t$) enclosure tests were conducted to examine the toxic effects on the outflows from the mine wastewater, using the sentinel species of Rhynchocypris oxycephalus, and we compared the biological responses of the control ($C_o$) and treatment (T) to the effluents through a Necropybased Health Assessment Index ($N_b$-HAI). Tissue impact analysis of the spleen, kidney, gill, liver, eyes, and fins were conducted in the controlled enclosure experiments (10 individuals). According to the comparisons of the control ($C_o$) vs. the treatment (T) in physicochemical water quality, outflows from the abandoned mine resulted in low pH of 3.2, strong acid wastewater, high ionic concentrations, based on an electrical conductivity, and high total dissolved solid (TDS). Physical habitat assessments, based on Qualitative Habitat Evaluation Index (QHEI) did not show any statistical differences (p>0.05) in the sampling sites, whereas, the $M_m$-EH model values in a multimetric ecological health ($M_m$-EH) model of the Index of Biological Integrity (IBI), using fish assemblages, were 16~20 (fair condition) in the control and all zero (0, poor condition) in the impacted sites of mine wastewater. In addition, in enclosure eco-toxicity ($EE_t$) tests, the model values of $N_b$-HAI ranged between 0 and 3 in the controls during the three years, indicating an excellent~good condition (Ex~G), and were >100 (range: 100~137) in the impacted sites, which indicates a poor condition (P). Under the circumstances, organ tissues, such as the liver, kidney, and gills were largely impaired, so that efficient water quality managements are required in the outflow area of the abandoned mine watershed.

Histopathological changes in fingerlings of Japanese flounder, Paralichthys olivaceus, with severe scuticociliatosis (스쿠지카섬모충에 중감염된 넙치 (Paralichthys olivaceus) 치어에 대한 병리조직학적 관찰)

  • In order to elucidate the patterns of tissue damage evoked by the scuticociliatids, eighteen fingerlings of Japanese flounder, Paralichthys olivaceus, heavily infected with an unidentified scuticociliatid were histopathologically examined. Skin layer with the underlying musculature were severely necrotized due to the infestation of the ciliates. However in the early lesions, both dermis and myofibres remained relatively intact compared with other surrounding loose connective tissues. Mild damages were found in more dense tissues. One or more scuticociliatids were recognized in the blood and lymph vessels of the loose connective tissues with or without destructive changes. Many of nerve trunks or ganglia were also parasitized with less marked histological damage in the parenchyma. Dura and its adjacent tissues in the spinal cord were severely necrotized with massive accumulation of the ciliates in subdural space. The parasitic invasion in the central nervous system was usually confined to the cortical region. In the gill, variable degenerative changes were occurred due to the invasion of the ciliates recognized in the blood vessels of branchial arches or primary filaments. From these results, it was strongly suggested that the scuticociliatids are very actively penetrated into the deep tissues mainly through the severe destruction of the loose connective tissue components and that the vascular system could play a role in the rapid distribution of the ciliates to the remote tissues or organs.

Anti-diabetic and Anti-Inflammatory Effects of Water Extract of Ligustrum japonicum Leaves in db/db Mouse (당뇨병 동물모델에서 여정엽(女貞葉) 추출물의 항당뇨 및 항염증 효과)

  • Objectives : In this study, we investigated the anti-diabetic and anti-inflammatory effects of water extract from leaves of Ligustrum japonicum (WLJ) in db/db mouse. Methods : The db/db mice were treated orally with WLJ (300 mg/kg/day) for 10 weeks to examine the long-term effects on hyperglycemia and glomerular tissue as well as biochemical and functional abnormalities in the kidney. Results : WLJ treatment markedly reduced plasma levels of glucose, triglyceride, creatinine, and systolic blood pressure in diabetic db/db mouse. Treatment of WLJ significantly increased plasma level of high density lipoprotein (HDL)-cholesterol. We also found that overexpressions of vascular cellular adhesion molecule (VCAM)-1 and endothelin (ET)-1 were observed in aortic tissue of db/db mouse, whereas, WLJ suppressed both expression of VCAM-1 and ET-1 in aorta. In renal tissue, overexpressions of ICAM-1 and TGF-${\beta}1$ were found in untreated db/db mouse, however, significantly decreased those levels by WLJ treatment. The insulin immunoreactivity of the pancreatic islets remarkably increased in WLJ treated db/db mouse compared with untreated db/db mouse. Taken together, WLJ treatment ameliorated hyperglycemia and hyperlipidemia via improvement of insulin secretion and lipid metabolism, respectively. Furthermore, WLJ treatment also ameliorated hypertension via inhibition of inflammatory process in vascular and renal tissues. Conclusions : Ligustrum japonicum has an anti-diabetic and anti-inflammatory effects in db/db mouse. Thus, these results suggested a beneficial effect of Ligustrum japonicum in treatment with diabetes and diabetic vasculopathy.

A comparative analysis of cell cycles in diploid and induced triploid tissues in marine medaka(Oryzias dancena) (해산송사리(Oryzias dancena) 2배체와 유도 3배체 조직의 세포주기 비교)

  • The aim of this study was to conduct a comparative analysis of the diploid and induced triploid cell cycles in marine medaka, Oryzias dancena, tissues. The mean fraction of cells in the G1, S, and G2+M phases was 85.5%, 7.6%, and 6.9%, respectively, in the tail fin tissues of diploid fish and 91.2%, 3.6%, and 5.2%, respectively, in those of induced triploid fish. The mean fraction of cells in the G1, S, and G2+M phases were 78.4%, 10.6%, and 11.0%, respectively, in the liver tissues of diploid fish; 86.2%, 5.9%, and 7.9%, respectively, in those of induced triploid; 79.3%, 9.4%, and 11.3%, respectively, in the gill tissues of diploid fish; and 85.7%, 5.4%, and 8.9%, respectively, in the induced triploid fish. The differences among the tissues were statistically significant within both the diploid and induced triploid fish (p<0.05). Mitosis was more active in each tissue of the diploid fish than in the corresponding tissues of the induced triploid fish and mitosis was more active in the liver and gill tissues than in the tail fin tissues in both the diploid and induced triploid marine medaka.
