To prepare chitosan-based polymeric amphiphiles that can form nanosized core-shell structures (nanoparticles) in aqueous milieu, chitosan oligosaccharides (COSs) were modified chemically with hydrophobic cholesterol groups. The physicochemical properties of the hydrophobized COSs (COSCs) were investigated by using dynamic light scattering and fluorescence spectroscopy. The feasibility of applying the COSCs to biomedical applications was investigated by introducing them into a gene delivery system. The COSCs formed nanosized self-aggregates in aqueous environments. Furthermore, the physicochemical properties of the COSC nanoparticles were closely related to the molecular weights of the COSs and the number of hydrophobic groups per COS chain. The critical aggregation concentration values decreased upon increasing the hydrophobicity of the COSCs. The COSCs efficiently condensed plasmid DNA into nanosized ion-complexes, in contrast to the effect of the unmodified COSs. An investigation of gene condensation, performed using a gel retardation assay, revealed that $COS6(M_n=6,040 Da)$ containing $5\%$ of cholesteryl chloroformate (COS6C5) formed a stable DNA complex at a COS6C5/DNA weight ratio of 2. In contrast, COS6, the unmodified COS, failed to form a stable COS/DNA complex even at an elevated weight ratio of 8. Furthermore, the COS6C5/DNA complex enhanced the in vitro transfection efficiency on Human embryonic kidney 293 cells by over 100 and 3 times those of COS6 and poly(L-lysine), respectively. Therefore, hydrophobized chitosan oligosaccharide can be considered as an efficient gene carrier for gene delivery systems.
Leucine-responsive regulatory protein (Lrp)는 대장균 (Escherichia coli)에서 발견된 '글로벌 조절자(global regulator)'로서 Lrp-regulon이 leucine에 의하여 상이한 형태의 조절 양상을 나타낸다. 6XHis-tag 시스템으로 제조한 야생형 Lrp (Lrp Wt)와 돌연변이 Lrp (Lrp R145W) 단백질을 정제하여 그들의 생화학적 성질을 조사하였다. 이들은 gel retardation assay를 통하여 ilv 오페론의 프로모터 영역 consensus 염기서열인 21bp의 이중가닥 DNA와 결합하여 복합체를 형성하는 것을 확인 하였다. 형광성 아미노산인 tryptophan을 지닌 Lrp R145W은 단백질의 농도가 증가함에 따라 형광이 커졌으며, 아미노산 leucine에 의하여 형광성의 변화가 관찰되었다. 즉 1 ${\mu}M$의 Lrp R145W 단백질에 leucine을 첨가하여 결합시키면 약 20 ${\mu}M$까지는 형광이 증가하다가 그 이상의 농도에서는 감소하는 양상을 얻었다. 이들 실험 결과는 leucine과 Lrp의 결합 양상 및 구조변이에 관한 심층연구에 있어서 Lrp의 고유 형광성이 요긴하게 쓰일 수 있음을 시사한다.
Cationic liposomes have been actively used as gene delivery vehicle because of their minimal toxicity, but their relatively low efficiency of gene delivery is the major disadvantage of these vectors. Recently, cysteine residue incorporation to HIV-1 Tat peptide increased liposomemediated transfection compared with unmodified Tat peptide. Therefore, we designed a novel modified Tat peptide having a homodimeric (Tat-CTHD, Tat-NTHD) and closed structure (cyclic Tat) simply by using the disulfide bond between cysteines to develop a more efficient and safe nonviral gene delivery system. The mixing of Tat-CTHD and Tat-NTHD with DNA before mixing with lipofectamine increased the transfection efficiency compared with unmodified Tat peptide and lipofectamine only in MCF-7 breast cancer cells and rat vascular smooth muscle cells. However, cyclic Tat did not show any improvement in the transfection efficiency. In the gel retardation assay, Tat-CTHD and Tat-NTHD showed more strong binding with DNA than unmodified Tat and cyclic Tat peptide. This enhancement was only shown when Tat-CTHD and Tat-NTHD were mixed with DNA before mixing with lipofectamine. The effects of Tat- CTHD and Tat-NTHD were also valid in the experiment using DOTAP and DMRIE instead of lipofectamine. We could not find any significant cytotoxicity in the working concentration and more usage of these peptides. In conclusion, we have designed a novel transfection-enhancing peptide by easy homodimerization of Tat peptide, and the simple mix of these novel peptides with DNA increased the gene transfer of cationic lipids more efficiently with no additional cytotoxicity.
양이온성 천연고분자 물질로서 유전자와 효과적으로 복합체를 형성할 수 있는 저분자량 수용성 키토산(LMWSC)에 유전자 발현 효율을 높이기 위해 histidine을 컨쥬게이션 시킨 유전자 전달체를 제조하였다. 히스티딘-LMWSC의 제조는 무수프탈산으로 LMWSC의 아민기를 보호한 후 히스티딘과 결합한 다음 무수프탈산을 제거하는 반응으로 제조하였다. 제조한 유전자 전달체의 물리화학적 특성은 FT-IR과 NMR 스펙트럼을 통하여 확인하였고, 전기영동 결과로부터 본 실험에서 제조된 유전자 전달체와 DNA가 복합체를 형성하였다는 것을 알 수 있었다. 또한 유전자 전달체의 입자크기는 DLS를 이용하여 측정한 결과 히스티딘-LMWSC-DNA 복합체입자의 크기는 100~200 nm임을 확인하였다. 이로부터 히스티딘을 함유하는 LMWSC가 유전자 전달체로서 사용될 수 있음을 확인하였다.
본 연구에서는 세포 독성을 감소시키고 in vivo에서 표적화를 향상시키기 위하여 methoxypoly(ethylene glycol) (mPEG) 및 folate가 도입된 poly(ethylene imine)(PEI)를 합성하였다. mPEG 말단에 카복실 그룹을 도입하여 EDC/NHS에 의해 활성화시킨 후 PEI의 아민기와의 반응으로 mPEG-PEI를 합성하였다. Folate의 카복실 그룹을 EDC/NHS으로 활성화시킨 후 mPEG-PEI의 아민기와의 반응으로 PEG-folate-graft-PEI를 합성하였다. 합성된 고분자의 화학적 구조는 $^1H-NMR$과 H-n을 이용하여 확인하였다. 합성된 고분자와 DNA가 정전기적 인력에 의해 완전한 복합체의 형성을 확인하기 위해 여러 가지 N/P charge 비율로 agarose gel 전기영동 및 형광을 측정하였고, 2 이상의 N/P charge 비율에서 완전한 복합체를 형성함을 확인하였다. 복합체의 크기는 광산란장치 및 AFM으로부터 $100\~300nm$임을 관찰하였다. 이러한 결과들로부터 합성된 양이온성 고분자와 DNA가 복합체를 잘 이루어지는 것을 관찰하였고 유전자 전달체로서의 가능성을 평가하고자 하였다.
비바이러스성 DNA 전달체 중에서 최근 양이온성 고분자와 양이온성 리피드들은 많이 연구되어 왔다. 본 연구에서는 비바이러스성 유전자 전달체로서 양이온성 고분자인 폴리에틸렌이민(PEI)을 리피드에 수식하여 더 효과적인 전달체를 제조하고자 하였다. 새로운 양이온성 리피드(PEI-DSPE)는 1,2-디아실-sn-글리세로-3-포스포에탄올아민(DSPE)과 PEI를 이용하여 고수율로 합성하였다. 1,2-디스테아로일-sn-글리세로-3-포스포콜린(DSPC), L-$\alpha$-포스파티딜콜린(소이-하디드로제네티드) (HSPC), 콜레스테롤(CHOL) 및 합성된 PEI-DSPE를 이용하여 리포좀을 제조하였다. 리포좀 제조는 리피드 함량에 대해 양이온성이 도입된 PEI-DSPE의 양을 증가시키며 수행하였다. 제조된 리포좀의 크기는 PEI-DSPE의 첨가량이 증가할수록 감소하였으며 리포좀의 표면전하 또한 양의 값으로 증가됨을 관찰할 수 있었다. PEI-DSPE 양적 변화에 따라 제조된 리포좀을 이용하여 DNA의 복합체 형성에 관한 결과 사용된 리포좀의 함량이 증가할수록 DNA와의 복합체 형성이 용이함을 전기영동 및 형광 측정을 통해 관찰할 수 있었으며 형성된 복합체가 사용된 리포좀 함량이 증가할수록 양이온적 성질이 증대됨을 관찰할 수 있었다. 그러므로 본 연구에서 합성한 PEI-DSPE를 사용하여 제조된 리포좀이 유전자 전달체로서의 가능성을 갖고 있음을 확인할 수 있었다.
$crp^{\ast}$ 유전자가 도입된 대장균 MK2001($crp^{{\ast}1}$}, cya::km)을 숙주로 사용하여 mal 유전자 발현을 촉진시키는 유전자의 하나인 sfs1(sugar fermentation stimulation)의 구조해석 결과에 의하면, 잠정적인 sfs1의 promoter 영역에는 CRP 단백질과의 결합영역으로 보이는 염기배열이 존재하였다. 본 실험에서는 sfs1 유전자의 cAMP-CRP에 의한 발현 조절을 확인하고자, lacZ 와의 융합 유전자를 작성하였다. 작성된 융합 유전자는 cya 결손주인 Tp2010에서 cAMP의 첨가에 의해 ${\beta}-galactosidase$ 활성이 크게 증가하였으며, Western blotting의 실험에서도 같은 결과를 나타냈다. in vivo에서 발현이 확인된 전사산물은 cAMP에 의해 전사 촉진이 일어났으며, CRP의 결합부위로 예상되는 DNA 영역은 cAMP가 존재하면 CRP 단백질과 결합하는 특성을 나타내었다. 이상의 결과로 보아, sfs1 유전자의 발현은 UMP-CRP에 의한 전사촉진 현상을 받는 것으로 나타났다.
