Changes in volatile compounds of persimmon vinegar during aging (숙성기간에 따른 감식초의 휘발성 성분 변화)

  • To investigate the changes in volatile compounds of persimmon vinegar (PV) during aging, PVs aged for 3-25 years were analyzed using solid-phase microextraction-gas chromatography-mass spectrometry. In total, 90 volatile compounds were identified, and acids and esters were the most abundant, followed by ketones and alcohols. The concentration of acetic acid ranged from 8,925.0 to 26,132.3 ng/mL, depending on the aging period. Ethyl acetate concentration ranged from 32.5 to 50,681.7 ng/mL and ethanol was present only in PV aged for 3 years (PV-3). The volatile compound content was the highest in PV-3. The volatile profiles of PV aged for 22 years (PV-22) and 25 years (PV-25) were considerably similar. Acids were the most predominant in PV-15, PV-22, and PV-25, whereas esters were abundant in PV-3. The contents of acids, alcohols, and esters decreased, while the aldehyde and ketone content increased with aging.

Xiang Study: an association of breastmilk composition with maternal body mass index and infant growth during the first 3 month of life

  • BACKGROUND/OBJECTIVES: This study aimed to establish a mother and child cohort in the Chinese population, and investigate human breastmilk (HBM) composition and its relationship with maternal body mass index (BMI) and infant growth during the first 3 mon of life. SUBJECTS/METHODS: A total of 101 Chinese mother and infant pairs were included in this prospective cohort. Alterations in the milk macronutrients of Chinese mothers at 1 mon (T1), 2 mon (T2), and 3 mon (T3) lactation were analyzed. HBM fatty acid (FA) profiles were measured by gas chromatography (GC), and HBM proteomic profiling was achieved by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS). RESULTS: During the first 3 mon of lactation (P < 0.05), significant decreases were determined in the levels of total energy, fat, protein, and osteopontin (OPN), as well as ratios of long-chain saturated FA (including C16:0, C22:0 and C24:0), monounsaturated FA (including C16:1), and n-6 poly unsaturated FA (PUFA) (including C20:3n-6 and C20:4n-6, and n-6/n-3). Conversely, butyrate, C6:0 and n-3 PUFA C18:3n-3 (α-linolenic acid, ALA) were significantly increased during the first 3 mon (P < 0.05). HBM proteomic analyses distinguished compositional protein differences over time (P = 0.001). Personalized motherinfant analyses demonstrated that HBM from high BMI mothers presented increased total energy, fat, protein and OPN, and increased content of n-6 PUFA (including C18:3n-6, C20:3n-6 and n-6/n-3 ratio) as compared with low BMI mothers (P < 0.05). Furthermore, BMI of the mothers positively correlated with the head circumference (HC) of infants as well as the specific n-6 PUFA C20:3n-6 over the 3 time points examined. Infant HC was negatively associated with C18:0. CONCLUSION: This study provides additional evidence to the Chinese HBM database, and further knowledge of FA function. It also helps to establish future maternal strategies that support the healthy growth and development of Chinese infants.

Gossypii Semen oil alleviates memory dysfunction in scopolamine-treated mice (면화자 정유의 기억력 손상 완화 효과)

  • Objectives : Gossypium arboreum (cotton) is traditionally used to treat various health disorders. However, anti-amnesic effect of G. arboreum has not been reported. The objective of this study was to investigate in-vivo the anti-amnesic effects along with in vitro antioxidant and acetylcholinesterase (AChE) inhibition potential in G. arboreum seed essential oil. Methods : The essential oil of G. arboreum obtained by solid phase microextraction (SPME) techniques were identified by gas chromatography-mass spectroscopy (GC-MS). 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay were performed to determine the antioxidant activity at various concentrations (312.5, 625, 1250, 2500, 5000, 10000 ㎍/㎖. Y-maze, passive avoidance and Morris water maze tests were carried out to evaluate improved effect on scopolamine (1 mg/kg)-induced memory dysfunction at the dose level of 50, 100 and 200 mg/kg. Donepezil (5 mg/kg) was used as a positive drug control. We performed acetylcholinesterase (AChE) activity assay in ex vivo. Results : Five volatile compounds were identified in G. arboreum. The assays of DPPH and ABTS revealed that G. arboreum increased antioxidant activity in a dose-dependent manner. G. arboreum ameliorated the percent of spontaneous alternation in the Y-maze test, shortened step-through latency in the passive avoidance test, and increased swimming time in the target zone in the Morris water maze test. In addition, G. arboreum inhibited the AChE activity. Conclusions : Based on these findings, G. arboreum may aid in the prevention and treatment of learning and memory-deficit disorders through antioxidant and AChE inhibitory activities.

Seasonal Concentration of Polycyclic Aromatic Hydrocarbons (PAHs) in Residential Areas Around Petrochemical Complexes and Risk Assessment Using Monte-Carlo Simulation (석유화학단지 주변 주거지역 다환방향족탄화수소(PAHs)의 농도와 Monte-Carlo 모의실험을 통한 위해성평가)

  • Background: Polycyclic aromatic hydrocarbons (PAHs) are generated in petrochemical complexes, can spread to residential areas and affect the health of residents. Although harmful PAHs are mainly present in particle phase, gas phase PAHs can generate stronger toxic substances through photochemical reaction. Therefore, the risk assessment for PAHs around the petrochemical complex should consider both particle and gas phase concentrations. Objectives: This study aimed to investigate the concentration characteristics of particle and gas phase PAHs by season in residential areas around petrochemical complexes, and to assess the risk of PAHs. Methods: Samples were collected for 7 days by seasons in 2014~2015 using a high volume air sampler. Particle and gas phase PAHs were sampled using quartz filter and polyurethane foam, respectively, analyzed by GC-MS. Chronic toxicity and probabilistic risk assessment were performed on 14 PAHs. For chronic toxicity risk assessment, inhalation unit risk was used. Monte-Carlo simulation was performed for probabilistic risk assessment using the mean and standard deviation of measured PAHs. Results: The concentration of particle total PAHs was highest in autumn. The gas phase concentration was highest in autumn. The average gas phase distribution ratio of low molecular weight PAHs composed of 2~3 benzene rings was 85%. The average of the medium molecular weight composed of 4 benzene rings was 53%, and the average of the high molecular weight composed of 5 or more benzene rings was 9%. In the chronic toxicity risk assessment, 7 of the 14 PAHs exceeded the excess carcinogenic risk of 1.00×10-6. In the Monte-Carlo simulation, Benzo[a]pyrene had the highest probability of exceeding 1.00×10-6, which was 100%. Conclusions: The concentration of PAHs in the residential area around the petrochemical complex exceeded the standard, and the excess carcinogenic risk was evaluated to be high. Therefore, it is necessary to manage the air environment around the petrochemical complex.

Emission Rates of Biogenic Volatile Organic Compounds from Various Tree Species in Korea (II): Major Species in Urban Forests (국내 수종별 BVOCs 방출량(II): 도시 숲 주요 수종)

  • In this study, the isoprene and terpene emissions from 32 major urban tree species were investigated. We conducted sampling using a dynamic enclosure system between June and July 2021. Seedlings aged < three years were enclosed in a chamber consisting of a 400 L transparent Tedlar bag. The air flow from the outlet of the chamber was sampled using Tenax-filled sorbent tubes under standard conditions (temperature: 30°C; PAR: 1,000 μmol/m2/sec). A thermal desorption gas chromatography/mass spectrometry system was used to analyze the following 38 biogenic volatile organic compounds: isoprene, monoterpenes, sesquiterpenes, oxygenated monoterpenes, and oxygenated sesquiterpenes. Isoprene emitters included Quercus mongolica, Salix koreensis, Robinia pseudoacacia, and Salix chaenomeloides. Monoterpene emitters included Pinus strobus, Cedrela sinensis, and Cercis chinensis. The monoterpene emission profiles were dominated by á-pinene, myrcene, camphene, and limonene. The predominant oxygenated monoterpene and oxygenated sesquiterpene were eucalyptol and caryophyllene oxide, respectively. For all species, the contributions of sesquiterpenes and oxygenated sesquiterpenes were relatively low.

A Scientific Analysis of Decorative Metal Foil Used in Pouch for the Sutra Embroidered with a Sun and Moon Design Designated as National Folklore Cultural Heritage (국가민속문화재 일월수 다라니 주머니 금속 장식지의 과학적 분석)

  • Through scientific analysis, this study identified the material characteristics of metal foil decorating the border line and knotting of the National Folklore Cultural Heritage 'Pouch for the Sutra Embroidered with a Sun and Moon Design'. Through Scanning Electron Microscope-Energy Dispersive Spectroscopy results, it was estimated that silver (Ag) and sulfur (S) were present in the metal foil, and silver leaf was also attached to the medium. S may discolor Ag from yellow to black depending on its concsentration and contact time. Yellow color could not be identified in metal foil at present. But there existed an example of the preparation of a gold-colored flat silver thread; therefore, further research is needed to estimate the original color. The lamella was reddish brown on the back. Aluminum, silicon, and iron were also detected and were the main components found in red soil. This is believed to be the red adhesive in traditional flat gold thread and is considered to be an adhesive-related component of the metal foil. From the gas chromatography mass spectrometry results, the adhesive component was confirmed to be animal glue.

Determination of the Effect of Trimethylamine Reduction in Egg Yolk Following Supplementation of Laying-Hen Feed with Riboflavin

  • The intensity of fishy odor in eggs, which differs depending on the poultry type and individual perception, can be due to many factors including trimethylamine (TMA) which has been identified as the main. Notably, riboflavin can increase the activity of flavin-containing monooxygenase 3, the enzyme responsible for converting TMA into odorless trimethylamine-N-oxide. This study aimed to analyze the TMA content in egg yolk, evaluate its contribution to fishy odor, and develop a method to prevent this undesired odor. Solid-phase microextraction-gas chromatography/mass spectrometry was used to detect and quantify volatile compounds in egg yolk from hens fed a standard TMA-rich diet and hens fed a riboflavin-supplemented diet. To compare the relative content of volatile substances between eggs, a correlation study was performed using an electronic nose. Higher concentration of TMA (1.06 ± 0.12 mg/kg) was detected in egg yolks obtained from hens fed a normal diet than those fed a riboflavin-supplemented diet. Overall, this study suggests that riboflavin affects the quantity and quality of volatile substances, including TMA, present in eggs and we expect these findings to improve the quality and reduce the fishy odor of eggs.

The study on the measurement of formaldehyde in saliva and urine by GC-MS (가스크로마토그래프-질량분석기에 의한 타액 및 뇨 중 포름알데하이드 분석법 연구)

  • A gas chromatography-mass spectrometric method was developed for the determination of formaldehyde in urine and saliva. In a 20 mL glass tube, 0.2 mL of urine or saliva was taken. Further, 1.8 mL of 0.1 M HCl, 0.1 mL of 2,000 mg/L 2,4-dinitrophenyl hydrazine and $20{\mu}l$ of 500 mg/L acetone-$d_6$ as internal standard were added in the tube and sealed tightly with cap. The solution was shaken for 20 min at room temperature and extracted using 4 mL of toluene. The extract was concentrated and redissolved with $100{\mu}l$ of acetonitrile, and then measured by gas chromatography-mass spectrometer (selected ion monitoring). The detection limit was 2.0 ng/mL and 0.5 ng/mL in saliva and urine, respectively. The calibration curves showed good linearity with r = 0.997 and 0.998 for saliva and urine, respectively. The method was used to analyze formaldehyde in rat urine after oral exposure. The developed method may be use ful to the monitoring for formaldehyde exposure in human.

Determination of new antifouling agents in seacoasts in Korea by gas chromatography-mass spectrometry (GC/MS를 이용한 한국연안의 새로운 방오제 분석)

  • Antifouling agents including tributyltin (TBT) compound and its derivatives have been used for many years, but the usage of TBT in Korea was legally restricted in 2003, due to its significant environmental impact. Following this, many new alternative antifouling agents have been used. In this experiment, four major antifouling agents were selectively analyzed to study their release in seawater and tidal flats on the Korean Peninsula. These new antifouling agents were extracted from the seawater and tidal flats using a liquid-liquid extraction method and microwave extraction, respectively. The measured concentrations of Irgarol 1051, Sea-Nine 211, Dichlofluanid and Chlorothalonil ranged from N.D.$-23.80ng/{\ell}$, N.D.$-15.30ng/{\ell}$, N.D.$-61.69ng/{\ell}$ and N.D.$-4.19ng/{\ell}$ in the seawater samples and from N.D.-159.45 ng/g, N.D.-476.57 ng/g, N.D.-59.79 ng/g and N.D.-21.27 ng/g in the tidal flat samples, respectively. Interestingly, these new antifouling agents were not detected in any area in the tidal flats at Pusan, whereas a certain amount of them was found in the seawater.

Survey of nitroso-compounds level derived from additives in metal-working fluids (유통 수용성 금속가공유 중 니트로 화합물 함유 실태)

  • Nitrite which can be derived from water for dilution of metal working fluid can induce nitroso compounds which can be classified as carcinogen, if it co-exists with ethanolamines added for pH control in metal working fluid. The survey of nitrite, nitrate and nitroso-compounds level in 42 metal-working fluids collected from 17 factories was done by ion chromatography and gas chromatography with mass detector. Diluted metal working fluid showed higher level of nitrite and nitrate compared with raw fluid. Nitrite was detected in 11 (52%) samples among 21 diluted solution. Three (14%) samples showed over German recommendation level ($20{\mu}g/mL$).N-nitrosodiethanolamine(NDELA) was detected in 18 samples among 21 diluted solution. Seven (33%) samples showed over German recommendation level ($5{\mu}g/mL$). The concentration of NDELA was correlated with nitrite ion ($R^2=0.453$, n=19).
