Quality Characteristics of Bread Added with Gastrodia elata Blume Powder (천마분말을 첨가한 식빵의 품질 특성)

  • Gastrodia elata blume (GEB) is considered to be a useful herbal medicine in oriental countries for the treatment of headache, migraine, dizziness, childhood convulsion, epilepsy, rheumatism, hypertension, neuralgia and neurological disorders. This study was carried out to investigate the quality of bread added with the powder of GEB. The possibility of GEB wheat flour mixture as bread was studied by adding 0%, 0.5%, 1.0%, 1.5%, 2.0% of GEB powder to wheat flour. In Farinograph data, the dough stability decreased with the increase of GEB powder. Granular size of starches ranged from $36\;{\mu}m\;to\;60{\mu}m$, and the shape of them showed a long oval figure. Amylograph showed that the increase in the ratio of GEB on the doughs slightly elevated in the maximum viscosity. The loaf volume of 0.5% powder increased by 10.2% but that of 2.0% decreased by 16.8%. The moisture content was 43.57% in the control but it increased as the powder addition. The colors of crust and crumb were not significantly different among L, b and ${\Delta}E$, but 'a' value in crumb was increased as the powder addition. The addition of the powder had no significant effect on bread texture. In sensory evaluation, the moistness increased as the increase of the powder addition. The control bread was most excellent, and the bread made by mixing additives were better than just 0.5% GEB-wheat flour in terms of quality.

Phylogenetic relationships of Armillaria spp. on the basis of ITS region sequences (rDNA의 ITS 부위 염기서열 분석에 의한 Armillaria 속 수집 균주의 유전적인 유연관계 분석)

  • Armillaria spp are well known as a symbiotic fungus with Gastrodia elata. This study was carried out to identify and analyze the genetic relationships among 83 strains of Armillaria spp.. The amplified internal transcribed spacer(ITS) region of the rDNA was about 500~750 bp long and identified by 9 strains; A. mellea, A. tabescens, A. ostoyae, A. gallica, A. novae-zenlandia, A. cepistipes, A. nabsnona, A. gemina, A. sinapina. Sequence analysis showed that 52% of strains were different with original identification. A. gallica, A. cepistipes and A. gemina were so close phylogenetic relationship, that was difficult to classify using ITS region. In A. gallica, 12 strains including ASI10104 were showed a close phylogenetic relationship with A. gallica, A. cepistipes and A. gemina. ASI10017 and ASI10114 were classified as the A. sinapina group, ASI10045 was the A. borealis group, ASI10002 and ASI10025 were the A. ostoyae group. So more studies need for more accurate identification and determine the phylogenetic relationships of Armillaria spp.

Effects of Heat Treatment and Selected Medicinal Plant Extracts on GABA Content after Germination (열처리와 한약재 추출물이 발아현미의 GABA 함량에 미치는 영향)

  • The purposes of this work were to study germination conditions on $\gamma$-aminobutyric acid (GABA) contents and to develop a simple and effective technique for the production of germinated brown rice with an enhanced GABA levels. The samples were subjected to heat treatments at 40 and $45^{\circ}C$ before the soaking step. Moreover, four medicinal plant extracts including Schizandra chinensis, Dimocarpus longan Lour, Angelica dahurica, and Gastrodia elata Blume were used as soaking and germinating media. GABA levels were enhanced in the germinated brown rice compared to the non-germinated brown rice and the highest GABA contents were observed in heat treatment at $35^{\circ}C$. The brown rice soaked in Gastrodia elata Blume extract showed the highest GABA contents compared to the control sample. These results demonstrate that the GABA levels during germination could be significantly enhanced by the heat treatment and the treatment of medicinal plant extracts in the soaking step.

Moisture Contents Setting according to Growth Stages of when the Cultivation of Gastrodia elata in Indoor Facilities (천마 실내시설재배 시 생육단계별 수분함량 설정)

  • 천마(Gastrodia elata)는 연중생산을 위해 실내시설 재배 시 생육모델을 구죽하고, 생육단계에 따른 온도, 수분, $CO_2$ 등 환경 조건 설정이 필요하다. 본 연구는 천마의 생육단계 중 괴경형성기와 괴경비대기의 수분함량을 설정하여 최적의 환경조건을 찾기 위해 수행하였다. 먼저 괴경형성기 수분함량 공급은 괴경형성기에 -20kPa, -30kPa, -40kPa로 처리하여 120일간 배양한 뒤, 괴경비대기를 -40kPa로 고정하여 60일간 배양하였다. 반면, 괴경비대기 수분함량 공급은 괴경형성기를 -30kPa로 고정하여 120일간 배양한 뒤 괴경비대기에 -20kPa, -30kPa, -40kPa, -50kPa로 처리하여 60일간 배양하였다. Tensiometer(토양수분장력계)기를 설치하여 수분을 공급하였고, FDR센서 (UbiMas, CoCo sensor, Frequency domain reflectometry type)를 배양토의 깊이 5 cm와 15 cm에 2개를 설치하여 평균값으로 수분함량을 측정하였으며, 전체수량, 성마율, 종마율 등을 조사하였다. FDR센서로 수분함량을 측정한 결과, -20 kPa은 43.3%, -30 kPa은 34.7%, -40 kPa은 29.8%, -50 kPa은 25.3%로 측정되었다. 괴경형성기 수분함량 처리 후 수확기의 상자 당 전체수량은 -30 kPa일 때 985 g으로 가장 많았고, -40 kPa일 때 912 g, -20 kPa일 때 703 g으로 처리간의 유의적인 차이를 보였다. 성마율은 수분함량처리별 각각 25, 34, 30% 이었고, 종마율은 수분함량처리별 각각 53, 73, 65%로 나타났다. 따라서 -30 kPa 처리구가 다른 처리구에 비해 전체수량, 성마율, 종마율 등이 유의적으로 우수하였다. 괴경비대기 수분 함량 처리 후 수확기의 상자 당 전체수량은 -40 kPa일 때 992 g으로 가장 많았고, -50 kPa일 때 955 g, -30 kPa일 때 903 g, -20 kPa일 때 686 g 순으로 나타났다. -30 kPa에서 -50 kPa 사이에서는 전체 수량의 유의성 차이는 없었다. 성마율은 수분함량처리별 각각 20, 30, 35, 33%이었고, 종마율은 수분함량처리별 각각 45, 65, 75, 68%로 나타났다. 따라서 -40 kPa 처리구가 다른 처리구에 비해 전체수량, 성마율, 종마율 등이 유의적으로 우수하였다. 반면 -20 kPa 처리구는 과도한 수분으로 천마가 오히려 부패될 수 있는 환경조건이 조성됨에 따라 성마율, 종마율 등 전체적인 수량 감소에 영향을 미친 것으로 판단되었다.

Monitoring of Drying Conditions for Gastrodia elata Blume by Response Surface Methodology (반응표면분석에 의한 천마의 건조조건 모니터링)

  • The Chunma (Gastrodia elata Blume) requires a simple pre-treatment, due to inconvenience or preservation, toxicities, and pungencies in intact state. Various drying conditions were examined to establish the physicochemical properties of Chunma by the response surface methodology (RSM). Weter activity and total phenolic acid contents of Chunma dried extracts were optimal at $79.56^{\circ}C$ and $79.64^{\circ}C$ for 7.17 hr and 8.74 hr, respectively, for their characteristics. Electron-donating ability and nitrite-scavenging activity were optimal at 79.79 and $7.99^{\circ}C$ for 8.57 and 8.07 hr. We generated a regression equation for each variable and then superimposed the optimal condition in acquiring values for each frying process factor, physicochemical properties, resulting in that the optimal drying condition was at $75{\sim}80^{\circ}C$ for $8{\sim}9 hr$.

Effect of Gastrodia Elata BL Water Extract on Human Cerebral Blood Flow using Transcranial Doppler (천마추출물이 정상인의 뇌혈류에 미치는 영향)

  • Background and objective: Gastrodiae Rhizoma (GR), the rhizoma of Gastrodia elata BL., is one of the popular drugs to treat headache, dizziness, blackout, numbness of limbs, hemiplegia, facial paralysis, dysphrasia, and infantile convulsions. It has been reported that it provides an antihypertensive effect and lowers cerebrovascular resistance in animal experiments. However, there has been no data about these effects with human subjects. In this study, the author examined the effect of Gastrodiae water extracts on blood pressure and cerebrovascular reactivity in human subjects. Methods: We selected 16 normal volunteers, who were divided into 2 groups: Gastrodiae extract administration group and placebo (creamy powder) group. Using transcranial Doppler ultrasound, we monitored changes of mean flow velocity and breath-holding induced CO2 reactivity of middle cerebral artery in both groups. Mean blood pressure, heart rate and PETCO2 were measured using Compact Anesthesia Monitor. In both groups, all evaluation was performed during basal condition, and repeated at 30, 60, and 90 min after administration. Results: Gastrodiae extract decreased CO2 reactivity after administration, reaching the lowest level at 90 minutes $(-29.1\%\;vs.\;basal\;level)$, which showed significant difference compared with the placebo group (p = 0.004). In the placebo group, the pulse rates tended to decrease over time (at 90 minute, $-5.2\%$ vs. basal level) while in the Gastrodiae group the values showed nearly no change, which showed significant difference between both groups (p = 0.036). However, the changes of mean blood pressure and mean flow velocity did not show significant difference between both groups. Conclusion : This study demonstrated that Gastrodiae extract significantly decreased breath-holding induced CO2 reactivity. This result suggests that the clinical effect of Gastrodiae extract might be caused by increasing cerebral blood flow via dilation of cerebral resistant vessels instead of antihypertensive effect.

  • The flora of Bongamsa (Temple) area, Mungyeong were listed 552 taxa; 99 families, 303 genera, 468 species, 1 subspecies, 70 varieties and 13 forms. Based on the rare plants (IUCN), total 12 taxa; Crypsinus hastatus (Polypodiaceae), Eranthis stellata (Ranunculaceae), Cnidium tachiroei (Umbelliferae), Monotropa hypopithys (Pyrolaceae), Rhododendron micranthum (Ericaceae), Salvia chanroenica (Labiatae), Gastrodia elata (Orchidaceae), etc. Based on the endemic plants, total 11 taxa; Asarum chungbuensis, Aconitum pseudolaeve, Corydalis maculata, Lonicera subsessilis, Aster koraiensis, Saussurea seoulensis, etc. Based on the specific plants by floral region were total 48 taxa (8.7% of all 552 taxa of vascular plants); 2 taxa in class V, 5 taxa (Cimicifuga heracleifolia, Wistaria floribunda, Elsholtzia angustifolia, etc.) in class IV, 10 taxa (Betula davurica, Spiraea chinensis, Acer barbinerve, Vaccinium koreanum, Syringa reticulata var. mandshurica, etc.) in class III, 7 taxa (Potentilla dickinsii, Viola orientalis, Ostericum melanotilingia, Melica nutans, Veratrum parulum, etc.) in class II, 24 taxa (Camptosorus sibiricus, Cephalotaxus koreana, Betula schmidtii, Ilex macropoda, Tilia amurensis, Triadenum japonica, Lobelia sessilifolia, etc.) in class I. Based on the naturalized plants, total 21 taxa (Silene armeria, Oenothera lamarckiana, Symphytum officinale, Erechtites hieracifolia, etc.) and naturalization rate was 3.8% of all 552 taxa of vascular plants. The flora of importance in the Bongamsa, rare plants were Crypsinus hastatus, Eranthis stellata, Gastrodia elata, Monotropa uniflora, Monotropa hypopithys, Rhododendron micranthum and Cnidium tachiroei, Iris ensata var. spontanea in intermediate marsh and alien species classify into planted species of Bongamsa, area for investigation of afforestation and artificial forests, introduced of naturalized plants.

Production of 4-Hydroxybenzyl Alcohol Using Metabolically Engineered Corynebacterium glutamicum (대사공학에 의해 개발된 코리네박테리움 글루타미컴에 의한 4-히드록시벤질 알코올 생산)

  • 4-Hydroxybenzyl alcohol (4-HB alcohol) is one of the major active components of Gastrodia elata Blume, with beneficial effects on neurological disorders such as headache, convulsive behavior, and dizziness. Here, we developed a metabolically engineered Corynebacterium glutamicum strain able to produce 4-HB alcohol from 4-hydroxybenzoate (4-HBA). First, the strain APS963 was obtained from the APS809 strain via the insertion of aroK from Methanocaldococcus jannaschii into the NCgl2922-deleted locus. As carboxylic acid reductase from Nocardia iowensis catalyzes the reduction of 4HBA to 4-hydroxybenzaldehyde (4-HB aldehyde), we then introduced a codon-optimized car gene into the genome of APS963, generating the GAS177 strain. Then, we deleted creG coding for a putative short-chain dehydrogenase and inserted ubiCpr encoding a product-resistant chorismate-pyruvate lyase into the pcaHG-deleted locus. The resulting engineered GAS355 strain accumulated 2.3 g/l 4-HB alcohol with 0.32 g/l 4-HBA and 0.3 g/l 4-HB aldehyde as byproducts from 8% glucose after 48 h of culture.

Neuroprotective and Anti-oxidant Effects of Gastrodiae Rhizoma Extracts against Hydrogen Peroxide-induced Cytotoxicity in SH-SY5Y Cells (산화적 스트레스에 대한 천마 추출물의 신경세포 보호 및 항산화 효과)

  • We recently reported that Gastrodia elata extracts (GEE) had an effects to protect against lipopolysaccharide-induced cognitive impairment in vivo model. In this study, we investigated the neuroprotective effects and the mechanism of action of GEE in hydrogen peroxide (H2O2)-induced cell death of SH-SY5Y human neuroblastoma cell. The SH-SY5Y cells were divided into five groups, including control(non-treated group), 100 μM H2O2, 100, 200, 500 ㎍/㎖ GEE+ 100 μM H2O2 groups. Pre- and co-treatment with GEE prevented cell death induced by 100 μM H2O2 for 24 h in SH-SY5Y cells. Our findings also showed that anti-oxidants enzymes (Cu/Zn superoxide dismutase, Mn superoxide dismutase, catalase) were up-regulated by 100 μM H2O2. But GEE suppressed H2O2-induced anti-oxidants enzymes decrease in a dose-dependent manner. Treatment with GEE also inhibited phosphorylation of eukaryotic initiation factor-2α (eIF-2α) and p38 by H2O2. Taken together, the neuroprotective effects of GEE in terms of recovery of antioxidant enzymes expression, down-regulation of eIF-2α and p38 phosphorylation, and inhibition of cell death are associated with reduced oxidative stress in SH-SY5Y cells.
