차광정도와 접종목 흔용이 천마의 수량에 미치는 영향 (Effects of Shading and Mixing Ratio of Recycling Spawn bed Logs on the Yield of Gastrodia elata Blume)

  • 천마 재배에서 차광방법 개발과 접종목 재활용의 가능성을 검정하여 저 비용, 고 생산의 재배법을 확립하기 위한 실험을 수행하였다. 비가림을 하지 않은 노지재배에서는 종마의 수량이 성마의 수량 보다 높았다. 차광 방법에 따른 천마의 수량은 $95\%$ 차광망 1겹, 2겹 및 무차광순 이었으며, 접종목 혼용에 따른 천마의 수량은 생목만 사용하였을 경우에 가장 높았고, 다음으로는 생목과 재활용목을 2:1로 및 1:1 혼용처리 처리 순이었다. 접종목 혼용에 따른 수익성 비교에서 $95\%$ 차광막 1겹 처리시 생목과 재생목을 2:1로 혼용한 경우는 생목만 사용한 경우 수익의 $81\%$이었다. 재배법 개선을 통해 수량성을 높일 수 있다면 생목과 재생목을 적당한 비율로 섞어 재배할 경우 비용 절감과 환경보호 차원에서 유용한 방법으로 사료된다.

수확시기와 자마의 특성별 천마의 아미노산 함량 변화 (Change of amino acids contents of Gastrodia elata Blume with harvest times and seed tuber)

  • 천마는 난초과 식물로서 예로부터 약용으로 사용되어 왔으며, 봄과 가을에 수확을 한다. 본 연구에서는 천마의 수확시기와 자마의 특성에 따른 아미노산의 함량을 조사하였다. 결론적으로 수확시기에 따른 총 아미노산 함량에는 변화가 없었지만 각 아미노산의 함량은 차이가 있음을 확인하였다. 가을에 수확한 천마에서는 아스파라긴이 19%로 함량이 제일 높았으며, 필수 아미노산으로는 발린인 10%를 차지하였다. 봄에 수확한 천마에서는 세린이 23%로 가장 높은 함량을 보였으며, 필수 아미노산 중에는 아르기닌(10%)로 가장 높은 함량을 보였다. 자마의 특성별 아미노산 함량을 조사 했을 때, 유성증식 천마가 무성증식 천마에 비하여 총 아미노산 함량이 높은 것으로 나타났다. 유성증식 천마의 필수 아미노산은 가을에 수확한 무성증식 천마와 함량이 비슷한 것으로 나왔으며, 비필수 아미노산은 봄에 수확한 무성증식 천마와 함량이 비슷한 것으로 나왔다. 본 연구를 통하여서 천마의 다양한 재배 환경에 대한 아미노산 함량의 변화를 알 수 있었으며, 더 나아가 자연 식품 재료로서의 가치가 높아질것으로 판단이 된다.

랫드에서 헛개나무와 천마 혼합 추출물의 에탄올 섭취에 의한 숙취 제거 효과 (Eliminatory Effect of Mixed Extract of Hovenia Dulcis Thunb and Gastrodia Elata on Ethanol-Induced Hangover in Rats)

  • To investigate an eliminatory effect of mixed extract of Hovenia dulcis Thunb, Gastrodia elata and Alnus japonica etc., on the ethanol-induced hangover, 12 hr-fasted male Sprague-Dawley rats weighing 220±20 g were given mixed extract (10 mL/kg, p.o.) and administered ethanol at a dose of 3 g/kg bw (25% in distilled water) orally 30 min postdosing. Blood was collected from caudal artery at 0.25, 0.5, 1, 2, 4, 6, 8, 10, and 12 hr and then the animals were sacrificed at 24 hr after the ethanol treatment. From 0 to 12 hr, the administration of mixed extract significantly decreased the area under the serum alcohol concentrations-vs.-time curves by 21 % compared with control group. In these experiments, liver function indices, such as alanine aminotransferase, aspartate aminotransferase and sorbitol dehydrogenase activities, showed unaltered results in all treated groups compared with the normal group. These results suggest that oral intake of the mixed extract may be effective on elimination of ethanol-induced hangover.

선행된 천마식이가 당뇨유발백서의 항산화효과 및 병태생리학적 인자에 미치는 영향 (Effect of Prior Gastrodia elata Blume diet on Diabetic Mellitus Induced Rats at Antioxidant Effect and Pathophysiology factor)

  • The purpose of this study was investigation to examine the prevention diabetic mellitus and treatment effect on Gastrodia Elata Blume(GEB) dieted at prior and after induced diabetic application. Prior induced diabetic 3 weeks ago application GEB dieted. It is to analysis changes in body weight, blood glucose, SOD, CAT and histopathological findings. For the fingding significantly concentration diabetic rats were divided into 3 different experimental groups and each groups were induced diabetic. Experimental group Ⅰ (STZ-induced diabetic rats; n=10), experimental group Ⅱ (after induced DM and GEB dieted rats; n=10), experimental group Ⅲ (Prior GEB dieted thereafter DM induced; n=10). Prior and After GEB dieted application was that body weight, blood glucose were increase in experimental group Ⅱ, Ⅲ. Specially, group Ⅲ was significantly change than group Ⅱ at 1st weeks. and the level of CAT were significantly decrease in experimental group Ⅱ, Ⅲ than group Ⅰ. but SOD level was increase in experimental group Ⅱ, Ⅲ than group Ⅰ. In histological observation; group Ⅰ showed decrease in the intensity and incidence of vacuolations, cellular infiltration and hypertrophy of in liver and kidney. The Gomori's stain result, group Ⅰ showed disruption ${\beta}$-cell in pancrease.

황금과 천마의 학습 및 기억에 미치는 영향 (Effect of Scutellaria baicalensis and Gastrodia elata on Learning and Memory Processes)

  • Learning and memory are essential requirements for every living organism in order to cope with environmental demands, and cholinergic systems are known to be involved in learning and memory. Scutellaria baicalensis (SB) and Gastrodia elata (GE) as a traditional Oriental medicine have been clinically used to treat or prevent memory deficits, including Alzheimer's disease. In the present study, we investigated the effects of SB and GE on learning and memory in the Morris water maze task and the central cholinergic system of the rats with excitotoxic medial septum lesions. In the water maze test, the animals were trained to find a platform at a fixed position over 6 days and then received a 60-s probe trial in which the platform was removed from the pool on the 7th day. Ibotenic lesion of the medial septum (MS) impaired their performance in the maze test (latency of acquisition test on the 3rd day, $27.6{\pm}$4.4 sec vs. $61.7{\pm}17.7$ sec; retention test, $7.9{\pm}1.3%$ vs. $5.7{\pm}1.0%$: sharn vs. ibotenic lesioned groups, respectively) and reduced choline acetyltransferase (ChAT) - immunoreactivity in the MS and the hippocarnpus, which is a marker for degeneration of the central cholinergic system (number of cells, $21.1{\pm}1.1$ vs. $13.2{\pm}1.3$: sham vs. ibotenic lesioned group). Daily administrations of SB (100mg/kg, p.o.) and GE (100mg/kg, p.o.) for 21 consecutive days produced significant reversals of ibotenic acid-induced deficit in learning and memory. These treatments also reduced the loss of cholinergic immunoreactivity in the MS and the hippocarnpus induced by ibotenic acid. These results demonstrated that SB and GE ameliorated learning and memory deficits through effects on the central nervous system, partly through effect on the acetylcholine system. Our studies suggest an evidence of SB and GE as treatment for Alzheimer's disease.

  • Scutellariae baicalensis GEORGt Acorus gramineus SOLAND and Gastrodia elata BLUME are traditional medicines used in the treatment of incipient stoke. In this study we investigated their effects on various aspects of neuronal differentiation in single or composite forms. Water-extracts of these medicines showed neuroprotective effects on cultured rat cortical neurons in normoxia and hypoxia. To understand the mechanism for neuroprotection we carried out various cell biological assays. They stimulated initial differentiation of neuronal development (transition from stage 1 to 2), and increased the number of spines and the length and number of dendritic processes. These effects were best manifested in the experimental group, which were given a mixture of the three kinds of extracts (p<0.01). To assess improvement of brain functions we carried out Morris water-maze tests for the mice that were fed on these extracts instead of water for 4 weeks. The experimental groups, especially those which were given the mixture of the three kinds of extract, showed significant (p<0.01) enhancement in memory as early as one day after the learning trial. These results indicate that these three kinds of extracts have synergistic effects on neuronal protection and improvement of brain functions.

Kainic acid로 유도된 생쥐의 간질 발작에 대한 천마 메탄올 추출물의 항경련 효과 연구 (Anti-convulsant Effects of Methanol Extract of Gastrodia Elata on Kainic Acid-induced Epilepsy Mouse Model)

  • Kainic acid (KA) is a excitatory agonist causing epileptic seizure and excitotoxicity in the hippocampus. Gastrodia Elata (GE) is known to have anti-convulsant and anti-oxidant effects. This study was investigated a possible role of GE in suppressing epileptic seizure using KA-induced epilepsy mouse model. Eight-week-old male C57BL/6 mice were administrated GE (50 or 500 mg/kg) once a day for 5 days, and then injected KA (30 mg/kg) intraperitoneally. Behavioral changes in mice by KA were evaluated for 90 minutes immediately after the KA administration. Six hours after the KA administration, their brains were harvested and the expressions of glutamate decarboxylase 67 (GAD-67) and K+-Cl- cotransporter 2 (KCC2) in the hippocampus of the mice were measured by immunohistochemistry.GE delayed the onset of epileptic seizure after KA administration, suppressed the severity of the seizure and decreased the number of severe seizures dose dependently. Moreover, GAD-67 and KCC2 expressions in the cornu ammonis (CA) 1 and CA3 of 500 mg/kg GE administrated mice were significantly increased compared to those in KA-treated mice.GAD-67 and KCC2 play an important role in regulating GABAergic system. Our results suggest that GE has anti-convulsant effect against KA-induced epileptic seizure through enhancing GABAergic system.

Isolation and Identification of Succinic Semialdehyde Dehydrogenase Inhibitory Compound from the Rhizome of Gastrodia elata Blume

  • In our search for the anticonvulsant consitutent of Gastrodia elata repeated column chromatographies guided by activity assay led to isolation of an active compound, which was identified as gastrodin on the basis of spectral data. Brain succinic semialdehyde dehydrogenase (SSADH) was inactivated by preincubation with gastrodin in a time-dependent manner and the reaction was monitored by absorption and fluorescene spectroscopic methods. The inactivation followed pseudo-first-order kinetics with the second-rate order constant of $1.2{\times}10^{3} M^{-1} min^{-1}$. The time course of the reaction was significantly affected by the coenzyme NAD^{+}$, which affected complete protection against the loss of the catalytic activity, whereas substrate succinic semialdehyde failed to prevent the inactivation of the enzyme. It is postulated that the gastrodin is able to elevate the neurotransmitter GABA levels in central nervous system by inhibitory action on one of the GABA degradative enzymes, SSADH.

천마의 흑질 내 도파민성 신경세포 보호 효과에 대한 단백체학적 분석 (Proteomic Analysis for Neuroprotective Effect of Gastrodia elata Blume in the Substantia Nigra of Mice)

  • Objectives : Parkinson's disease (PD) is a neurodegenerative disorder threatening the quality of life and highly occurred in over 65 years old. Gastrodia elata Blume (GEB), a traditional medicine used for the treatment of headache and convulsion, has been reported to have neuroprotective effect. This study was designed to investigate the neuroprotective effect of GEB and the proteomic changes in the substantia nigra (SN) of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated mice. Methods : Male eleven-week-old C57BL/6 mice were intraperitoneally injected with 30 mg/kg of MPTP at 24-h intervals for 5 days. Two hours after the daily MPTP injection, the mice were orally administered 800 mg/kg of GEB extract, which continued for 7 days beyond the MPTP injections, for a total of 12 consecutive days. Two hours after the final GEB administration, the brain samples were collected, and dopaminergic neuronal death and proteomic changes in the SN were evaluated. Results : GEB prevented the MPTP-induced dopaminergic neuronal death and regulated the expression of 11 proteins including thimet oligopeptidase, T-complex protein 1, glycine tRNA ligase, and pyruvate kinase isozymes M1. Conclusions : GEB prevents MPTP-induced dopaminergic neuronal death by regulating the proteins in the SN.

발효 천마 추출물의 생리 활성 (Physiological Activities of Fermented Gastrodia elata Blume Extracts)

  • 천마를 다양한 용매(물, 에탄올 및 70%) 및 다양한 농도 (0.725, 1.25, 2.5 및 5 mg/mL)로 추출하고 추출된 물질을 젖산균과 유용미생물(Effective Microorganisms)로 발효시킨 다음, 이들 물질들이 화장품 소재로 활용 가능한가를 확인하였다. 물, 에탄올 및 70% 에탄올로 추출한 물질들을 각각 0.725, 1.25, 2.5 및 5 mg/mL 농도에서 실험한 결과, flavonoid, polyphenol 및 DPPH free radical scavenger 등과 같은 항산화물질은 에탄올 추출물에 가장 많이 함유되어 있었고, DPPH radical 소거능은 유용미생물로 발효시킨 천마를 에탄올로 추출한 경우 1.25 mg/mL에서 $27.08{\pm}0.5%$이었으나 2.5 mg/mL에서는 $27.08{\pm}0.5%$로 증가하였다. 미백 활성을 측정할 수 있는 tyrosinase 활성 저해 실험에서는 EtOH 추출물을 젖산균으로 발효시켰을 때 가장 저해 효과가 우수하였고(0.725 mg/mL에서 $39.1{\pm}0.4%$, 2.5 mg/mL에서 $62.8{\pm}1.5%$), RAW 264.7 Cells을 이용한 세포 생존율 실험 결과 물, EtOH, 70% EtOH 추출물과 이들의 발효 물질 모두 85%이상의 세포 생존율을 나타내 세포의 염증에 대한 안정성을 확인하였다. ROS 생성으로 인한 항산화 실험에서는 양성 대조군으로 RAW 264.7 cells을 lipopolysaccharide로 자극시켜 활성화된 세포에 비발효와 발효에 따른 추출물을 농도별로 처리하였을 때 유효미생물로 발효시켰을 경우가 가장 항산화성이 높은 것으로 확인하였다. 이상과 같은 연구결과 천마 추출 및 발효 물질은 화장품용 항산화제, tyrosinase 활성 저해제 및 항염증제로 사용될 수 있음을 알았다.
