Juvenile Hormone Titers and Juvenile Hormone Esterase Activity during Larval Stage of the Chestnut Gall Wasp, Dryocosmus kuriphilus Yasumatsu (밤나무 혹벌 유충의 유약호르몬 함량과 유약호르몬 에스테라제 활성)

  • The juvenile hormone(JH) titers and juvenile hormone esterase (JHE) activities were mea¬sured in larval homogenates of the chestnut gall waL,J, Dryocosmus kuriphilus Yasumatsu, parasiting a susceptible and two resistant chestnut ( Cheuk-Pa, and Dan- Tak) varieties by GLC, Galleria wax test and Liquid scintilation counter. JH of the chestnut gall wasp was identified as JH- I. Their juvenile hormone titers were 35,800 GU/g(Cheuk-Pa), 30,900 GU/g (Dan-Tak), and 28,600 GU/g(susceptible variety). The juvenile hormone esterase activities were 1.48 n mole/min/ml(Cheuk-Pa), 1.63 n mole/min/ml(Dan- Tak), and 1.89 n mole/mini ml(susceptible variety). JH titer activity of the chestnut gall wasp parasiting resistant varie¬ties were higher than that from susceptible, whereas their JHE activity was higher in those from susceptible variety than those from resitant varieties. JH titer and JH specific esterase activity was inversely proportional.

Distribution of pine needle gall midge, Thecodiplosis japonensis Uchida et Inouye (Diptera: Cecidomyiidae), infestations on Japanese red pine, Pinus densiflora S. et Z. (소나무에서의 솔잎혹파리 피해 분포)

  • Distribution of pine needle gall midge infestations was analyzed from data collected in young Japanese red pine stands during 1992, and 1995-1996 in Kangwon-do. No significant differences in percentages of infested needle pairs were found among trees and between terminal and lateral shoots within a tree. However, the mean percentages of infested needle pairs increased significantly from the lower crown to the upper. Percentages of infested needle pairs on sample units, consisting 1 terminal and 2 lateral shoots, in the midcrown were best predictors of whole-tree percentages than were other crown levels. Therefore, a sample unit consisting of 1 terminal shoot and 2 lateral shoots per branch were fixed from the midcrown level. Number of tree and sample unit combinations needed to estimate pine needle gall midge infestations with given two levels of precision were determined.

Use of Sprinkler System for Control of the Pine Needle Gall Midge, Thecodiplosis japonensis Uchida et Inouye (분무장치(噴霧裝置)를 이용(利用)한 솔잎혹파리 방제(防除)에 관(關)한 연구(硏究))

  • Experiments were conducted to evaluate the efficacy of ground and foliar sprinkling of natural water for control of the pine needle gall midge, Thecodiplosis japonensis, by disturbing the reproductive behavior of adults such as copulation and oviposition. Diurnal ground spray for whole area application and foliar spray for single tree treatment were tested at day time during the period of adult emergence. The results obtained are as follows : 1. The population density of overwintering larvae in soil was not affected by the water spray during the adult emergence period. 2. Spraying the water showed no effect on the whole period and number of adult emergence, but retarded early emergence and facilitated Late emergence. The difference in the emerging time by 50% emergence day was approximately 7 days. 3. Ground spray was significantly different from control in gall formation rates but was not sufficient for effective control. As a single-tree treatment, the foliar spray was found to be highly effective for control of the pine needle gall midge, Average gall formation of spray plots and untreated ones were 2.6% and 38%, respectively ; control value exceeded 90%. 4. Ground spray of water was not effective in reducing larvae in gall of needle. Foliar spray, however, reduced 45% of larval numbers. 5. Foliar spray accelerated shoot growth by 15%-18%. 6. Foliar spray was highly effective for the control of sucking insects, such as mites and aphids on pine trees.

  • Olive flounder Paralichthys olivaceus larvae and early juveniles were reared for 43 days after hatching in order to observe the effects of starvation during development and metamorphosis. Morphological, histological and biochemical measurements were made to assess the nutritional condition during growth and starvation from pre-matamorphic through post-metamorphic phases. Two groups of fish were compared ; one with sufficient food supply and one under continuous starvation until death. Among morphometric analyses, both ratios of body height at anus/head height and pre-/post-anal lengths appeared to be sensitive to starvation during which substantial reduction was observed within a day of food deprivation. Histological variables as intestinal and rectal epithelial heights and gall bladder volume changed significantly with onset of starvation. The gut epithelial heights of starving fish decreased with advances in starvation, although they fluctuated during mid-metamorphic phase. In contrast, gall bladder volume increased remarkably soon after starvation. Ontogenetic changes in both gut epithelial height and gall bladder volume were evident, those associated with settlement and/or completion of metamorphosis. Abrupt decrease in the RNA/DNA ratios of starving fish were found right after onset of starvation. Even in the fed fish marked fluctuations in its ratios during metamorphosis were observed, evident by decreasing from late-metamorphic to post-metamorphic stages. These findings suggest that a combination of morphologically and histologically sensitive characteristics, and biochemical measurement could be utilized as a measure to evaluate nutritional condition related to starvation in wild olive flounder larvae and juveniles.

  • Agrobacterium vitis causes a crown gall disease in grapevine and that is one of the major hindrances for the wide cultivation and production of grapevine. We studied the possibility of biological control using selected biological control agent. One isolate from the infected soil, named as strain 27, was able to inhibit the biovar 1; A. tumefaciens C58 and Ach5, biovar 2; A. rhizogenes 13264, and biovar 3; A. vitis, in vitro and in vivo test. The putative biological control agent, A. radiobacter strain 27 was carrying the plasmid and the size of isolated plasmid was very similar to that of pAgK84 of A. radiobacter K84.

Isolation of Entomopathogenic Fungi for Infection to the Pine Gall Midges, Thecodiplosis japonensis from the Forest Soil in Korea (삼림 토양으로부터 솔잎혹파리 감염 사상균의 분리)

  • To develop a microbial pesticide for the control of pine gall midges. Thecodiplosis japonensis, entomopathogenc fungi were isolated from 233 soil samples in the damaged region of Thecodiplosis japonenesis, and identified with Beauveria spp. 29 strains and Paecilomyces spp. 2 strains. The morphology of entomopathogenic fungi was observed by scanning electron miroscope. In addition, the toxicity of entomopathogenic fungi was observed by scanning electron microscope. In addition, the toxicity of entomopathogenic fungi isolated from soil samples was determined by bioassay against Thecodiplosis japonensis larvae. The result showed that toxicity of relatively pathogenic strains, Beauveria spp. SFB-168-2 was 82.9%, suggesting that Beauveria spp. SFB-168-2 is effective entomopathogenic fungi for the control of pine gall midges.

  • This study was intended to know the histological damage of the gall on the leaves, peduncles and calyx of Lycium chinense $M_{ILL}$ formed Eriophyes kuko $K_{ISHIDA}$ during 1962 to 1965. The results are summarized as follows; 1. Eriophyes kuko $K_{ISHIDA}$ fed on only the Palisade layers of the loaves of Lycium chinense $M_{ILL}$ were injured by boring. 2. From the 1st to 10th day, some parts of the spongy layer of the Lycium chinense $M_{ILL}$ leaves were injured, but variation were not significant in this period. 3. From the 15th day, Eriophyes kuko $K_{ISHIDA}$ injured not only the spongy layer of Lycium chinense $M_{ILL}$. leaves but also the great part of e chloroplast of the loaves, and the sponginess caves were appeared inside of the galls.

  • The pine needle gall midge, Therodiplosis japonensis Uchida et Inouye, is the most important insect pest. It requires two different habitats for the development; on trees and under the ground. The habitat specific mortality rates ere $30\~40\%,\;and\;50\~60\%$ for the respective habitats. The key developmental stage is the prepupa, and the key mortality factor is the moisture contents of the soil and its variability. Since the insect is an exotic, the population status is the periphery and in the source of infestation are considerably different. Such a difference in habitats and the population status of the insect should be considered in relation to suppression of the insect. The control strategies should be directional and rational based on the reality of the pest status. There have been substantial information on the control methods of the pine needle gall midge, and each control method has an important place, but none has always provided a satisfactory solution to the many problems associated by this insect. These methods should be applied to a system based on the ecology of the insect. There should be continued support for directed effort on the development of operational management systems for the insect: specifically, estmation of the critical economic injury level, and of the absolute density of the insect.

  • Using scanning electron microscope, we studied the adult genital segments of pine needle gall midge, Thecodiplosis Japonensis, Uchida et Inouye Male: There were two pairs of hypoproct a pair of claspettes and aedeagus, Hypoproct were surrounded with many setae and sensilla. Clasperres which was projected below hypoproct was composed of two aegments and had many sensilla Aedeagus was long and flattened and possesed some sensilla at the terminal. Female :Ovipositor was segmented into three parts. Each segment had a different externla sturcture; particularly, the 2nd one was covered with reversed boat-like attachments and did not show any sensillum.

Antioxidant and Antiobesity Activities of Various Color Resources Extracted from Natural Plants (천연식물로부터 추출한 색소성분의 항산화 및 항비만 활성)

  • The objective of this study was to investigate the antioxidant and antiobesity activities of various color resources extracted from natural plants such as, clove, persimmon, gall nut, amur cork, gardenia, safflower, and annatto. Total phenolic content was the highest in gall nut extract (2,441.45 mg/kg) followed by clove extract (1,346.48 mg/kg). DPPH, and ABTS radical scavenging activities and ferric reducing antioxidant power (FRAP) were also higher in gall nut extract. ${\alpha}$-Glucosidase inhibitory activity ($IC_{50}$) was highest in persimmon extract ($22.83{\mu}g/mL$) followed by gall nut extract. ${\alpha}$-Amylase and lipase inhibitory activities were also higher in persimmon extract (49.45% and 61.01%, respectively). Lipid accumulation in 3T3-L1 cells was lower in persimmon, clove, and annatto extracts (81.54%, 83.36%, and 85.70% at $20{\mu}g/mL$, respectively). Triglyceride content in 3T3-L1 cells was lowest in clove extract (66.11%) followed by persimmon extract (88.88%). The results of this study suggest that gall nut extract has the highest antioxidant activity, whereas persimmon and clove extract show the antiobesity activities by inhibition of digestive enzymes and fat accumulation in 3T3-L1 cells. These extracts are useful materials for the development of antioxidant and antiobesity functional foods.
