Correlation of Expression of p53, Cylcin D1 and Galectin-3 in Papillary Carcinoma and Follicular Carcinoma

  • The thyroid is the organ that has the greatest risk of malignant tumors among the endocrine tumors. The papillary carcinoma occupies 80% of the entire thyroid tumors. Immunohistochemical staining of galectin-3 has usually been used in differentiating papillary carcinoma and follicular carcinoma. The p53 gene of the cell cycle is a tumor suppressor gene acting in on the control points. The cyclin D1 genes in the cell cycle, involved in the implementation of G1 and S phase, plays an important role in the progression of thyroid tumors. This research compares and analyzes correlation between papillary carcinoma, follicular carcinoma, p53, cyclin D1 and galectin-3 gene expression patterns. In a total of 30 cases from papillary carcinoma, 21 cases from p53 (70%), 27 cases in galectin-3 (90%), and 26 cases in cyclin D1 (86.7%) showed positive rate. The galectin-3 staining investigated, showed a significant difference between a papillary carcinoma and a follicular carcinoma. Follicular carcinoma from 15 cases, p53 in 13 cases (86.7%), galectin-3 in 5 cases (33.3%) and cyclin D1 in 12 cases (80%) showed a positive rate. The cyclin D1 in follicular carcinoma and staining between the p53 that had correlation was also investigated. In this study, as the examples of the expression of the 27 cases of galectin-3 (90%) in papillary carcinoma and 5 cases in follicular carcinoma (33.3%) indicate, it was concluded that there is a difference in the expression on both carcinoma. In addition, cyclin D1 and p53 has a positive rate in follicular carcinoma, when cyclin D1 in 12 cases (80%), there was a significant correlation that was investigated. Distinguishing between papillary carcinoma and follicular carcinoma can be identified by the expression of galectin-3. It is considered to get results that are more accurate in follicular carcinoma diagnosis depending on whether the cyclin D1 and p53 is expressed or not.

갑상선 미세 유두암종에서 Galectin-3, Cytokeratin 19와 HBME-1의 발현 (Immunohistochemical Expression of Galectin-3, Cytokeratin 19 and HBME-1 in Papillary Microcarcinoma of the Thyroid Gland)

  • 갑상선 미세유두암종은 영상 기술의 발달로 더욱 그 빈도가 높아질 것이 예상되므로 진단의 정확도를 높이는 것이 무엇보다 중요하다. 갑상선 미세 유두암종으로 진단 받은 37예를 대상으로 galectin-3, cytokeratin 19와 HBME-1 분자 표지자들을 면역조직화학적 염색을 시행한 결과 cytokeratin 19에서 1예를 제외하고 모든 예에서 갑상선 종양 세포에서 발현하였다. Galectin-3는 갑상선 종양 세포의 세포질에 강하게 염색되었으며 cytokeratin 19는 세포질과 세포막을 따라 강하게 염색되었다. HBME-1은 종양 세포의 세포막에 강하게 염색되었으나 3가지 분자 표지자 모두 정상 갑상선 조직에서는 염색되지 않았다. 이상의 결과에서 볼 때 galectin-3, cytokeratin 19와 HBME-1은 병리조직학적 소견과 함께 갑상선 미세 유두암종을 진단함에 있어 보조적 도움이 되는 표지자로 생각된다.

넙치, Paralichthys olivaceus 발생단계별 galectin-1 유전자의 발현 분석 (Gene analysis of galectin-1, innate immune response gene, in olive flounder Paralichthys olivaceus at different developmental stage)

  • 선천성 면역반응은 감염성 질병에 대한 저항력이 부족한 초기 발달단계의 자치어에게 중요하다. 특히 lectin은 선천성 면역반응에 많은 기여를 하고 있으며 종류 또한 다양하다. Lectin의 종류 중 하나인 galectin은 어류 점액 내에 존재하며 선천적으로 외부 병원체로부터 숙주를 보호하는 기능을 지니고 있다. 하지만 어류 galectin과 초기발생단계 시기와 관련된 연구들은 부족한 실정이다. 본 연구는 넙치에서 발현되는 galecin-1을 통하여 자치어의 초기발생단계별 그리고 성어의 조직별 발현을 조사하였다. 초기발생단계별 galectin 발현을 조사한 결과, 부화 전 수정란의 낭배기부터 galectin-1 유전자의 발현이 시작되었으며, 부화 후 25일까지 서서히 증가하였다. 부화 후 30~35일에 galectin-1 유전자의 발현량이 급격하게 증가하였고, 그 이후의 galecin-1 유전자의 발현량은 서서히 감소하였다. 성어 (5개월령, 29개월령)의 조직별 galectin 발현을 조사한 결과, 근육, 장, 지느러미, 위, 눈 조직 순서로 높게 발현하였고, 이외의 조직에서는 발현이 나타나지 않았다. 초기발생단계에서 galectin-1의 발현이 증가되는 것은 낭배기부터 넙치의 근육, 지느러미 그리고 눈 조직이 형성되고 galectin의 분비가 시작되어 기능을 나타내기 때문인 것으로 추측된다. 부화 후 30일부터 galectin의 발현량이 많이 증가하는 것은 장과 위 조직이 활성화됨에 따라 점액의 분비량이 증가하여 galectin의 발현량 또한 증가하는 것으로 판단된다. 이로써 넙치 초기발생단계 시기의 galectin-1 유전자의 발현패턴과 galectin이 점액 내에 다량 포함되어 있는 것을 확인하였으며, galectin의 발현은 선천적 면역반응으로 넙치의 초기발생단계 시기와 성어기에 중요할 것으로 판단된다.

향나무 추출물의 Galectin-3 저해에 의한 암 전이 억제 효과 (Anti-Metastatic Activity of Juniperus chinensis Extract by Galectin-3 Inhibition)

  • 향나무 에탄올 추출물에 대한 암 전이 억제 효과를 측정하기 위하여 세포 수준에서 암 전이에 밀접히 관련되어 있는 galectin-3에 대한 억제 효과를 colon 26-M3.1 lung carcinoma에서 측정하였다. 향나무 추출물 $5{\mu}g/m{\ell}$, $50{\mu}g/m{\ell}$$500{\mu}g/m{\ell}$ 처리 시, 농도 의존적으로 galectin-3에 대한 저해 효과를 나타내었다. 한편, 암 전이 관련 있는 또 다른 단백질로 MMP 효소에 대한 직접적인 억제효과를 측정한 결과, 향나무 추출물 $5{\mu}g/m{\ell}$, $50{\mu}g/m{\ell}$$500{\mu}g/m{\ell}$ 처리 시 직접적으로 MMP-1, MMP-2 및 MMP-9 효소에 대해서 농도의존적인 저해 효과는 나타났으나, 강한 활성으로 평가되지는 못하였다. Colon 26-M3.1 lung carcinoma을 이용한 in vivo mouse 모델에서 향나무 추출은 농도 의존적으로 전이 억제 효과를 나타내었고, MMP-2 활성화 물질인 relaxin과 병용투여 시, relaxin의 기능에 영향을 받지 않는 것으로 나타났다. 따라서 in vivo mouse 모델에서 나타난 암 전이 억제 효과는 galectin-3 저해로 인한 효과로 사료된다.

자기면역성뇌척수염 척수조직에서 galection-3의 발현 (Expression of galectin-3 in the spinal cords of Lewis rats andNOD mice with experimental autoimmune encephalomyelitis)

  • The aim of this study was to evaluate the expression of galectin-3, one of beta-galactoside-binding proteins, in the experimental autoimmune encephalomyelitis(EAE) model of Lewis rats or non-obese diabetic (NOD) mice. Western blot analysis showed that galectin-3 was weakly expressed in the spinal cords of complete Freund's adjuvant(CFA) immunized control rats. In EAE, however, galectin-3 expression was significantly increased at the peak stage(days 14 post-immunization), while it was decreased slightly at the recovery stage(day 21 post-immunization). Immunohistochemical analysis showed that galectin-3 was detected in some macrophages in demyelinating lesions of NOD mice, while galectin-3 was immunoreacted in some inflammatory cells in the perivascular cuffing in rat EAE lesions. Collectively, it is postulated that the expression of galectin-3 is significantly increased in response to neuroimmunological stimulation in the central nervous system, whereas it is weak in normal rats and mice.

Binding Affinities of Carbohydrate-Conjugated Chlorins for Galectin-3

  • Carbohydrate-conjugated chlorins were synthesized for use as biosensors for the detection of the galectin-3 cancer marker. We used ELISA, SDS-gel electrophoresis, and Bradford assays to examine the binding of galectins to d-(+)-galactose- and b-lactose-conjugated chlorins. The binding affinities of these conjugated chlorins for galectin-3 were quantified using fluorescence spectroscopy. The fluorescence emission of the carbohydrate-conjugated chlorins decreased as the amount of galectin-3 in the binding reaction increased over a limited concentration range, indicating that carbohydrate-conjugated chlorins are potentially useful fluorescence biosensors for the galectin-3 cancer marker.

Mesenchymal Stem Cells Ameliorate Fibrosis by Enhancing Autophagy via Inhibiting Galectin-3/Akt/mTOR Pathway and by Alleviating the EMT via Inhibiting Galectin-3/Akt/GSK3β/Snail Pathway in NRK-52E Fibrosis

  • Background and Objectives: Epithelial-Mesenchymal transition (EMT) is one of the origins of myofibroblasts in renal interstitial fibrosis. Mesenchymal stem cells (MSCs) alleviating EMT has been proved, but the concrete mechanism is unclear. To explore the mechanism, serum-free MSCs conditioned medium (SF-MSCs-CM) was used to treat rat renal tubular epithelial cells (NRK-52E) fibrosis induced by transforming growth factor-β1 (TGF-β1) which ameliorated EMT. Methods and Results: Galectin-3 knockdown (Gal-3 KD) and overexpression (Gal-3 OE) lentiviral vectors were established and transfected into NRK-52E. NRK-52E fibrosis model was induced by TGF-β1 and treated with the SF-MSCs-CM for 24 h after modelling. Fibrosis and autophagy related indexes were detected by western blot and immunocytochemistry. In model group, the expressions of α-smooth muscle actin (α-SMA), fibronectin (FN), Galectin-3, Snail, Kim-1, and the ratios of P-Akt/Akt, P-GSK3β/GSK3β, P-PI3K/PI3K, P-mTOR/mTOR, TIMP1/MMP9, and LC3B-II/I were obviously increased, and E-Cadherin (E-cad) and P62 decreased significantly compared with control group. SF-MSCs-CM showed an opposite trend after treatment compared with model group. Whether in Gal-3 KD or Gal-3 OE NRK-52E cells, SF-MSCs-CM also showed similar trends. However, the effects of anti-fibrosis and enhanced autophagy in Gal-3 KD cells were more obvious than those in Gal-3 OE cells. Conclusions: SF-MSCs-CM probably alleviated the EMT via inhibiting Galectin-3/Akt/GSK3β/Snail pathway. Meanwhile, Gal-3 KD possibly enhanced autophagy via inhibiting Galectin-3/Akt/mTOR pathway, which synergistically ameliorated renal fibrosis. Targeting galectin-3 may be a potential target for the treatment of renal fibrosis.

소포성 갑상샘 유두암종의 세침흡인 세포검사의 정확도와 Galectin-3 면역염색의 유용성 (The Diagnostic Accuracy of Fine Needle Aspiration Cytology and the Diagnostic Usefulness of Galectin-3 Immunostaining for the Follicular Variant of Papillary Thyroid Carcinoma)

  • The cytologic diagnosis of the follicular variant of papillary thyroid carcinoma (FVPTC) has become one of the common causes of false negative diagnoses when performing fine needle aspiration cytology (FNAC) of the thyroid gland. We retrospectively reviewed all the aspirates for which a diagnosis of FVPTC had been made based on the surgically excised specimens, regardless of the cytologic diagnosis. 145 FNACs was performed in 135 patients. The cytologic diagnoses were categorized as 2 unsatisfactory specimens (1.4%), 16 benign (11.0%), 49 atypical (33.8%) and 78 malignant lesions (53.8%). The tumor cells consistently showed significant nuclear overlapping, irregular nuclei and fine chromatin in all cases; however, nuclear grooves and inclusions were scarce. Galectin-3 immunostaining was performed on the cell blocks of 65 cases and this was positive for 45 cases (69.2%). The results of our study demonstrate that the determination of minimal cytologic criteria is needed to raise the sensitivity of detecting FVPTC by FNAC, and galectin-3 immunostaining is useful to make decisions on the surgical treatment of cytologically atypical thyroid nodules.

Biophysical Feature, Crystallization and X-ray Crystallographic Studies of Toxascaris leonina Galectin

  • Galectins are generally believed to be potential candidates for use in the development of novel antiinflammatory agents or as selective modulators of the immune response. In particular, galectin-9 exhibits some of the extracellular functions, including cell aggregation, adhesion, chemoattraction, activation, and apoptosis. Tl-galectin (Tl-gal, galectin-9 homologue gene) was isolated from an adult worm of the Toxascaris leonina. The full-length Tl-gal gene, which was incorporated into pET-28a, was overexpressed in E. coli and purified by nickel affinity and gel filtration chromatographies. The purified Tl-gal was crystallized using the hangingdrop vapor-diffusion method. The crystal belonged to the tetragonal space group $P4_1$, with unit-cell parameters of a = b = $75.7\AA$ and c = $248.4\AA$. The crystals were obtained at $20^{\circ}C$ and diffracted to a resolution of $3.0\AA$. The asymmetric unit contained four molecules of Tl-gal, which gave a crystal volume per protein mass (Vm) of $2.8\AA^3Da^{-1}$ and a solvent content of 54.1%.

성숙마우스의 난소에서 황체형성호르몬수용체와 galectin-3의 상반된 면역조직화학적 발현분포 (Opposite Localization of Luteinizing Hormone Receptors and Galectin-3 in Mature Mouse Ovaries)

  • 본 연구는 성숙마우스의 난소에서 황체형성호르몬수용체(luteinizing hormone receptor, LHR)와 beta-galctoside-binding animal lectin인 galetin-3 (Gal-3)의 발현분포를 면역조직화학방법으로 평가하였다. LHR은 활동(성숙) 황체에서 강하게 발현되는 반면에, Gal-3의 발현은 퇴행중인 황체와 폐쇄난포에서 높게 나타났다. 또한, 임신중인 마우스의 황체에서는 LHR의 면역반응이 강하게 나타났으나, Gal-3의 면역반응은 현저히 낮았다. 위의 결과를 종합해 볼 때, 성숙마우스의 난소에서 LHR과 Gal-3는 난소의 특정한 시기에 따라 다른 분포양상을 나타냄을 알 수 있었으며, 이들 각각의 단백질의 발현으로 보아 황체의 형성과 퇴행에서 상반된 역할을 할 것으로 여겨진다.
