The aim of this study is to elucidate the biochemical properties of manno-oligosaccharides (MOS) hydrolyzed by extracellular enzyme of Bacillus N3. We strived to characterize the biochemical properties of MOS since N3 can effectively hydrolyzed natural polymannans such as galactomannan (GM) and konjac (glucomannan, KM), respectively. The hydrolysis of GM and KM was applied by the strain N3 in terms of reducing sugars and the highest production of reducing sugars was estimated to be about 750 mg/L and 370 mg/L respectively, which were quantified after 7 days of cultivation in the presence of both substrates. Hydrolysates derived from the hydrolysis of KM showed the significant antioxidant activity based on DPPH and ABTS radical scavenging activity with increasing of tyrosinase inhibitory activity. On the other hand, hydrolysates derived from the hydrolysis of GM showed only ABTS radical scavenging activity without showing significant changes on tyrosinase inhibitory activity. Our data suggest that those biological characteristics may be depend on the primary structure and the size of MOS, which may be useful as potent additives for diet foods.
A thermophilic bacterium producing the extracellular cellulase-free xylanase was isolated from soil and has been identified as Bacillus sp. The optimal growth temperature was 50$\circ$C and the optimal pH, 7.0. Under the optimal growth condition, maximal xylanase production was 2.2 units/ml in the flask culture. The enzyme production was induced by xylan and xylose, but was repressed by sucrose or trehalose. The partially purified xylanase was most active at 70$\circ$C. It was found that the enzyme was stable at 65$\circ$C for 10 hours with over 75% of the activity. The enzyme was most active at pH 7.0 and retained 90% of its maximum activity between pH 5.0 and pH 9.0 though Bacillus sp. was not grown on alkaline conditions (>pH 8.0). In addition, the activity of xylanase was over 60% at pH 10.0. At the ambient temperature, the enzyme was stable over a pH range of 5.0 to 9.0 for 10 h, indicating that the enzyme is thermostable and alkalotolerant. The activity of xylanase was completely inhibited by metal ions including Hg$^{2+}$ and Fe$^{2+}$, while EDTA, phenylmethylsulfonyl fluoride (PMSF), $\beta$-mercaptoethanol and SDS didn't affect its activity. The enzyme was also identified to exert no activity on carboxymethylcellulose, laminarin, galactomannan, and soluble starch.
Guar gum, a storage polysaccharide galactomannan was administered to 11 patients with type-II diabetes mellitus for 7 days and 3 weeks. They took 5 grams of guar gum 30 mins before each of three meals daily. The dinner 2-h postprandial values of their blood glucose were significantly lowered (P<0.05) after their guar treatment for 7 days compared with before taking guar gum. The 2-h postprandial values of blood glucose were significantly lowered(P<0.05) after 3 weeks of gual treatment compared with before taking guar gum. In an oral glucose tolerance test, their blood glucose values were significantly lowered at 120 mins(P<0.02) and 180 mins(P<0.05) after guar treatment. Total-lipid(P<0.01) and triglycerides(P<0.02) of their blood were significantly decreased and HDL-cholesterol(P<0.02) was significantly increased after guar treatment. HbA1C was significantly reduced (P<0.05) from 11.3% to 10.1% The body weight, total-cholesterol and insulin activity of the patients after guar treatment were not significantly changed and the satiety ratings of the patients with guar treatment was not significantly changed and the satiety ratings of the patients with guar treatment was not significantly changed, however, the subjects that answered from 'Want to eat but can wait' to 'No desire to eat' were 81.1%. It is concluded that guar gum improves their carbohydrate and lipid metabolism in Korean type-II diabetic subjects with high fiber diets.

A 45-year-old-male who had underlying ulcerative colitis and presented with fever and dry cough. Initially, the patient was considered to have invasive aspergillosis due to a positive galactomannan assay. He was treated with amphotericin B followed by voriconazole. Nevertheless, the patient deteriorated clinically and radiographically. The lung biopsy revealed eosinophilic pneumonia, and ELISA for Toxocara antigen was positive, leading to a diagnosis of pulmonary toxocariasis. After a 10-day treatment course with albendazole and adjunctive steroids, the patient recovered completely without any sequelae. Pulmonary toxocariasis may be considered in patients with subacute or chronic pneumonia unresponsive to antibiotic agents, particularly in cases with eosinophilia.
A highly thermostable ${\beta}-(1-4)-glucanase$ (NA23_08975) gene (fig) from Fervidobacterium islandicum AW-1, a native-feather degrading thermophilic eubacterium, was cloned and expressed in Escherichia coli. The recombinant FiG (rFiG) protein showed strong activity toward ${\beta}-{\small{D}}-glucan$ from barley (367.0 IU/mg), galactomannan (174.0 IU/mg), and 4-nitrophenyl-cellobioside (66.1 IU/mg), but relatively weak activity was observed with hydroxyethyl cellulose (5.3 IU/mg), carboxymethyl cellulose (2.4 IU/mg), and xylan from oat spelt (1.4 IU/mg). rFiG exhibited optimal activity at $90^{\circ}C$ and pH 5.0. In addition, this enzyme was extremely thermostable, showing a half-life of 113 h at $85^{\circ}C$. These results indicate that rFiG could be used for hydrolysis of cellulosic and hemicellulosic biomass substrates for biofuel production.
Lung granulomas are uncommon in Thailand. The disease typically develops from an occupational environment and is mostly caused by infection. Herein is a case report of a female patient, aged 48, working as a nurse in an Accident and Emergency Department at a hospital. Eighteen years prior to admission the patient was diagnosed with myasthenia gravis and pulmonary tuberculosis. The chest X-ray and CT scans showed a solitary pulmonary nodule in the lower left lung. The patient received an open thoracotomy with a left lobectomy. Granulomatous and nonseptate hyphae were found in the pathology diagnosis. The patient was thus diagnosed as having a lung granuloma. The galactomannan antigen test was positive. The solitary pulmonary nodule-found from the use of a Polymerase Chain Reaction (PCR) test-was an Aspergillus spp. The fungus culture was collected from air samples. The air samples were collected by the impaction technique using a microbial air sampler. Three types of Aspergillus spp. were found as well as Penicillium spp. and Monilia sitophila. The Aspergillus spp. was a match for the patient's disease. The patient was diagnosed as having a lung granuloma possibly Aspergillus nodule which was caused by airborne Aspergillus spp. from the occupational environment.
Four hundred and fifty tilapias ($6.77{\pm}0.23$ g) were assigned randomly to six groups to evaluate the feasibility of the tested antibacterial peptides (ABPs) and oligosaccharides as substitutes for antibiotics. The control group was fed with a commercial tilapia diet; other five groups were fed with the same commercial diet supplemented with konjac glucomannan (KGLM), cluster bean galactomannan (CBGAM), and three animal intestinal ABPs derived from chicken, pig and rabbit at 100 mg/kg respectively. After 21 days of feeding, growth, disease resistance, and in vivo anti-adherence were determined. Furthermore, the inhibitory effect of tested agents on adhesion of Aeromonas veronii biovar sobria (A.vbs) strain BJCP-5 to tilapia enteric epithelia in vitro was assessed by cell-ELISA system. As a result, the tested agents supplemented at 100 mg/kg show significant benefit to tilapia growth and disease resistance (p<0.05), and the benefit may be correlated with their interfering in the contact of bacteria with host mucosal surface. Although none of the tested agents did inhibit the growth of BJCP-5 in tryptic soy broth at $100{\mu}g/ml$, all of them did inhibit the adhesion of A.vbs to tilapia enteric epithelia in vivo and in vitro. In vitro mimic assays show that three ABPs at low concentrations of $25{\mu}g/ml$ and $2.5{\mu}g/ml$ have the reciprocal dose-dependent anti-adherence effect. The inhibition of ABPs may be correlated with a cation bridging and/or receptor-ligand binding, but not with hydrophobicity. The KGLM and CBGAM inhibited the adherence of BJCP-5 to tilapia enteric epithelia with dose-dependent manner in vitro, and this may be through altering bacterial hydrophobicity and interfering with receptor-ligand binding. Our results indicate that the anti-adherence of the tested ABPs and oligosaccharides may be one of the mechanisms in promoting tilapia growth and resistance to A.vbs.
새로 분리된 곰팡이 Trichoderma erinaceum DG-312의 균사체 배양에 의해 갈락토만난(GMs)을 생산하여 쥐에서 항염증 활성을 조사하였다. 5 리터 교반 발효조에서 3일 동안 배양에 의해 얻어진 균사체 및 갈락토만난의 최대 농도는 각각 9.44 g/l 및 2.72 g/l 이였다. Sepharose CL-6B 젤 크로마토그라피와 다당체 성분 분석 결과, crude 갈락토만난의 조성은 주로 mannose $(74.9\%)$와 galatose $(24.1\%)$로 구성된 이종의 다당체인 것으로 밝혀졌다. Trichoderma erinaceum DC-312로부터 생산된 갈락토만난은 아세트산으로 처리한 쥐에서 체중(kg)에 대하여 100 또는 200 mg의 양으로 처리하였을 때 농도에 비례하여 현저한 항 염증 활성을 갖는 것으로 나타났다. 갈락토만난의 항염증 활성을 나타내는 모세혈관 투과도(vascular permeability) 및 발작 반응(writhing response)의 저해 수치는 각각 $60.6\%$ 및 $62.5\%$로 나타났다. 따라서 이런 갈락토만난의 항염증 및 발작반응 저해에 효과적인 특성을 이용한다면 치료 목적을 위한 의약물질 개발의 잠재성을 보여준다.
Paenibacillus woosongensis의 유전체 부분 염기서열로부터 mannanase를 코드하는 것으로 유추되는 open reading frame을 중합효소연쇄반응으로 증폭하여 대장균에 클로닝하고 염기서열을 결정하였다. Mannanase 유전자는 man26AT로 명명하였으며 1,053 아미노산으로 구성된 단백질을 코드하는 3,159 뉴클레오티드로 이루어졌다. 아미노산 잔기배열을 분석한 결과 Man26AT는 glycosyl hydrolase family 26의 mannanase와 상동성이 높은 활성영역, 탄수화물 결합영역 CBM27과 CBM11로 구성되어 있었다. Man26AT의 아미노산 배열은 P. ihumii의 유추된 mannanase와 상동성이 81%이고 다른 Paenibacillus 속 균주의 여러 mannanases와 57% 이하의 상동성을 보였다. man26AT 유전자를 함유한 재조합 대장균의 균체 파쇄상등액은 $55^{\circ}C$와 pH 5.5에서 최대의 mannanase 활성을 보였고, $50^{\circ}C$에서 1시간 열처리한 후에 80% 이상의 잔존활성을 보였다. Man26AT는 locust bean gum (LBG) galactomannan과 konjac glucomannan에 대한 분해활성이 유사하였으며, carboxymethylcellulose, xylan과 para-nitrophenyl-${\beta}$-mannopyranoside는 분해하지 못하였다. Man26AT에 의해 mannotriose, mannotetraose, mannopentaose와 mannohexaose 등의 만노올리고당이나 LBG로부터 공통의 최종 가수분해 산물로 mannose, mannobiose와 mannotriose가 생성되었다. 또한 mannotriose 보다 큰 만노올리고당이 LBG와 guar gum의 분해산물로 각각 생성되었다. 그러나 Man26AT는 mannobiose를 분해하지는 못하였다. 활성염색을 통해 Man26AT는 균체 내에서 3개 이상의 크기가 다른 활성 단백질로 분해된 것이 확인되었다.
(1) 콩과 식물인 guar (Cyamopsis tetragonolobus)의 발아종자(發芽種子)에서 세 가지의 다른 다당류가수분해효소(多糖類加水分解酵素)인 ${\beta}-1$, 4-mannanase A,B,C 를 추출(抽出), 유안염석(硫安鹽析), 이온교환(交換)크로마토그래휘 및 젤여과법(濾過法)에 의하여 분리(分離), 정제(精製)하였다. (2) 이들 효소(酵素)는 작용최적(作用最適) pH, 급속이온의 영향, 분자량(分子量), ior nut mannan A에 대한 Michaelis 항수(恒數), 가수분해(加水分解)의 한도(限度) 및 생성물등(生成物等) 여러 가지 점(點)에서 다르다. (3) ${\beta}-1$, 4-Mannanase A 와 C 는 기질(基質)인 ${\beta}-1$, 4-mannanase 에 random으로 작용(作用)하여 과당류(寡糖類)를 생성(生成)하는 endo형(型) 효소(酵素)임이 밝혀졌다. 한편 ${\beta}-1$, 4-mannanase B 는 같은 기질(基質)의 비환원말단기(非還元末端基)로 부터 수차로 작용(作用)하여 단당류(單糖類)를 생성(生成)하는 exo형(型) 효소(酵素)로 추리(推理)된다. (4) Guar 종자중(種子中)의 galactomannan 인 guaran에 이들 정제효소(精製酵素)를 작용(作用)시킨 결과(結果)로부터 기질(基質)의 구조(構造) 및 guar 종자(種子)의 발아중(發芽中)에 가지는 역할(役割)과 관련시켜 논의(論議)하였다.
