Chemical Investigation on an Endophytic fungus Gibberella moniliformis JS1055 Derived from a Halophyte Vitex rotundifolia

  • Chemical investigation of the ethyl acetate extract of Gibberella moniliformis JS1055 endophytic fungus derived from a halophyte, Vitex rotundifolia, led to the isolation of nine compounds including 7-butyl-6,8-dihydroxy-3(R)-pent-11-enylisochroman-1-one (1), 7-butyl-6,8-dihydroxy-3(R)-pentylisochroman-1-one (2), 7-butyl6,8-dihydroxy-3(R)-pentylisochroman-1-one (3), $5{\alpha},8{\alpha}$-epidioxyergosta-6,9(11),22-trien-3-ol (4), ergosterol peroxide (5), tetradecanoic acid (6), 8-O-methylfusarubin (7), nicotinic acid (8) and adenosine (9). They were identified by extensive spectroscopic data analysis including 1D, 2D ($^1H-^1H$ COSY, HSQC, HMBC) NMR, and ESIMS. All the isolates (1-9) are reported for the first time from this endophytic fungus.

다양한 염료의 탈색이 가능한 목재부후균 분리 (Isolation of a Wood-rotting Fungus to Decolorize a Wide Range of Structurally Different Synthetic Dyes.)

  • Twenty-one different fungi were tested for their ability to decolorize a wide range of structurally different dyes. Twenty fungal strains were isolated from fruiting bodies which were collected at the Kwangneung National Arboretum, Korea. One fungal strain were isolated from a rotting wood at Soongsil University, Korea. Nine kinds of dyes were used: three anthraquinone dyes and six azo dyes. The five fungal strains, Laetiporus sulphureus, Polyporus arcularius. Auricularia polytricha, Stereum ostrea, and Bjerkandera sp. UK-l showed decolorization ability. Except Auricularia polytricha, the four fungal strains were wood rotting fungi, and belonged to Aphyllophorales. Bjerkandera sp. UK-I, which was a white rot fungus, could decolorize all kinds of dyes tested in this study, indicating this fungus is one of candidates for applying in biological methods of dye waste treatment.

Penicillifer diparietisporus: a New Record from Field Soil in Korea

  • A fungus was isolated from field soil collected from Daegu, Korea. The colony of the isolated fungus showed short, branched, and light to dark yellow pigments with hyaline, yellowish red to orange brown aerial mycelia. In addition, the fungus produced solitary to aggregated perithecia, ovoid to pyriform, short neck, and asci as well as biseriately arranged ascospores. Phylogenetic analysis using the internal transcribed spacer region and translation elongation factor $1-{\alpha}$ sequences and morphological characteristics identified the isolated fungus as Penicillifer diparietisporus, which belongs to the family Nectriaceae. To our knowledge, this is the first report of Penicillifer diparietisporus in Korea.

  • 특정수도품종에 대한 도열병균의 침입률은 접종전처리온도와 균주의 조합보다는 공시균주나 온도의 단독요인에 의한 영향이컸다. 이병성품종은 그 품종이 어떤 온도조건하에서 자랐던지 또는 병원성이 다른 어떤 균주로 접종이 되었던지 이병성으로 남아 있었으며 접종전에 기주가 다른 온도조건하에서 자랐을 경우, 도열병균의 특정균주 또는 Race의 병원성발현은 다르게 나타나는 것이 관찰되었다.

  • Phenolic compounds from 7 species of naturalized invader species and Korean wild plants were analyzed by high performance liquid chromatography. Eleven phenolic compounds including benzoic acid were identified. The extract of naturalized plants was significantly more inhibitory to seed germination and seedling growth of the both naturalized and Korean wild plants. The content of total phenolic compounds in each extract were 43.5 mg/l in Ailanthus altissima as the maximum amount and 25.5 mg/l in Phytolacca americana as the minimum. Phytotoxic substances of ethanol extracts was investigated for antifungal activity against 23 selected fungus species. The antifungal activity of Phytolacca americana showed the greatest clear zone of 23 mm in Aspergillus awamori and its activity had an effect against 6 fungus species. Ailanthus altissima formed the greatest clear zone of 26 mm in Erwinia carotovora sub. sp. carotovora and had an effect agsinst 2 fungus species.

고정화된 백색부후균을 이용한 염료의 탈색에 관한 연구 (A Study on Decolorization of Dyes with an Immobilized White Rot Fungus Irpex lacteus)

  • Decolorization of congo red, rhodamlne B was investigated by the white rot fungus Irpex lacteus welch has biodegrading capability of various recalcitrants. White rot fungus Irpex lacteus is Immobilized by PVA-freezing method. An Immobilized Irpex lacteus decolorizes 91% of congo red In 8 days under culture with glucose 2%(Initial cone.). It also showed 70% of decolorization at 3 days In the state of putting MnSO4 1mM. But, rhodauune B has no significant differences about decolorization among different mixture ratio of Irprx lacteus with PVA, concentration of carbon, nitrogen and manganese sulfate.

Trichodermamide A and Aspergillazine A, Two Cytotoxic Modified Dipeptides from a Marine-Derived Fungus Spicaria elegans

  • Two known modified dipeptides, trichodermamide A (1) and aspergillazine A (2), were isolated from an ethyl acetate extract of the metabolite of a marine-derived fungus Spicaria elegans, and were found to have a weak cytotoxic effect on three cancer cell lines P388, A-549, and HL-60 agreed. To our knowledge, this is the first report on the isolation of compounds 1 and 2 from the fungus Spicaria elegans and their cytotoxic effect.

  • An undescribed black root rot of soybean, caused by Cylindrocladium crotalariae, was observed in Suweon area. The diseased plants showed yellowing at the top and dry rot at the root. Lesions of roots and stems in the soil were red to brown and main roots were cracked. Although not observed the disease in the field, leaves of inoculated test plants in the greenhouse exhibited circular, brown lesion surrounded by chloratic halos. The fungus was recovered in culture from the infected stem and root, and the perithecia of Calonectria crotalariae were demonstrated to be present as well as the cylindrocladium state. The fungus was pathogenic to the root, stem, petioles and leaves of soybean. The probable source of primary inoculum was microsclerotia formed in infected soybean root and stem from the previous season's soybean debris. Black root rot by this fungus was considered to be one of detrimental factors to the maximum yield of soybean. From the morphological and physiological characteristics and pathogenic behaviors, this fungus was identified as Cylindrocladium(Calonectria) crotalariae.

High Density Cell Culture of Mucor sp. KCTC 8405P for Production of ${\gamma}-Linolenic$ Acid in Fed-Batch Culture

  • In order to produce ${\gamma}-linolenic$ acid by Mucor sp. KCTC 8405P. the fungus was cultivated in fed-batch culture with two phases. i.e., growth in yeast-like form and induction to hyphal growth by pH shift of the culture medium during cultivation. The synchronous growth of the fungus into the appropriate sizes was important for the high density cell culture of this dimorphic fungus. Dissolved oxygen concentration in the medium did not affect degree of unsaturation of fatty acids and ${\gamma}-linolenic$ acid content. Under the culture conditions applied in this experiment. the fungus was found to produce 100 g/l dry mycelia containing 40% of the lipids, where ${\gamma}-linolenic$ acid comprised about 9% of the total extractable fatty acids.

Isolation, Cultivation, and Antifungal Activity of a Lichen-Forming Fungus

  • A lichen-forming fungus was successfully isolated by discharged spore method from Korean lichen (Hetero-dermia sp.) and cultivated in pure culture. The isolate JR0012 inhibited mycelial growth of several plant-pathogenic fungi. Mycelial growth of the four Pythium spp. tested was completely inhibited. Potato dextrose broth was found to be the medium favorable for large-scale production of antibiotics from the isolate. Anti-fungal substances produced in axenic culture were partially purified. This is the first report in Korea of lichen-forming fungus successfully isolated and which exhibited strong antifungal activity against plant-pathogenic fungi, especially the four Pythium spp..
