Four hundred and sixty six isolates of B. cinerea were obtained from infected leaves, stems and fruits of paprika grown in greenhouses or plastic film houses in Gangwon province, Korea, between August and November in 2006 and 2007. These isolates were classified into five representative phenotypes of resistant (R) and sensitive (S) reactions as SSR, SRR, RSS, RRS and RSR according to the responses of isolates against benzimidazole, dicarboximide and N-phenyl-carbamate fungicide in order. The percentage of five phenotypes were 51.3, 2.4, 35.6, 8.1 and 2.6%, respectively. The SSR phenotype (51.3%) was the most common. Among the nineteen fungicide products evaluated to compare their efficacy against gray mold pathogen on the paprika fruit inoculated with fungal mycelia, the mixture of diethofencarb and carbendazim was the most effective followed by iprodione, boscalid, the mixture of iprodione and thiophanate-methyl, fludioxonil, polyoxin-B, fluazinam, the mixture of tebuconazole and tolyfluanid and procymidone; while in the assay methods inoculated with fungal spores, the mixture of tebuconazole and tolyfluanid was the most effective in controlling gray mold followed by boscalid, fludioxonil, the mixture of diethofencarb and carbendazim and the mixture of pyrimethanil and chlorothalonil.
Resistance to pyraclostrobin due to a single nucleotide polymorphism at 143rd amino acid position on the cytochrome b gene has been a major source of concern in red pepper field infected by anthracnose in Korea. Therefore, this study investigated the response of 24 isolates of C. acutatum and C. gloeosporioides isolated from anthracnose infected red pepper fruits using agar dilution method and other molecular techniques such as cytochrome b gene sequencing, polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and allele-specific polymerase chain reaction (PCR). The result showed that four isolates were resistant to pyraclostrobin on agar dilution method and possessed GCT (alanine) codon at 143rd amino acid position, whereas the sensitive isolates possessed GGT (glycine). Furthermore, this study illustrated the difference in the cytochrome b gene structure of C. acutatum and C. gloeosporioides. The use of cDNA in this study suggested that the primer Cacytb-P2 can amplify the cytochrome b gene of both C. acutatum and C. gloeosporioides despite the presence of various introns in the cytochrome b gene structure of C. gloeosporioides. The use of allele-specific PCR and PCR-RFLP provided clear difference between the resistant and sensitive isolates. The application of molecular technique in the evaluation of the resistance status of anthracnose pathogen in red pepper provided rapid, reliable, and accurate results that can be helpful in the early adoption of fungicide-resistant management strategies for the strobilurins in the field.
Colletotrichum species is known as the major causal pathogen of red pepper anthracnose in Korea and various groups of fungicides are registered for the management of the disease. However, the consistent use of fungicides has resulted in the development of resistance in many red pepper-growing areas of Korea. Effective management of the occurrence of fungicide resistance depends on constant monitoring and early detection. Thus, in this study, various methods such as agar dilution method (ADM), gene sequencing, allele-specific polymerase chain reaction (PCR), and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) were applied for the detection of benzimidazole resistance among 24 isolates of Colletotrichum acutatum s. lat. and Colletotrichum gloeosporioides s. lat. The result of the ADM showed that C. gloeosporioides s. lat. was classified into sensitive and resistant isolates to benomyl while C. acutatum s. lat. was insensitive at ≥1 ㎍/ml of benomyl. The sequence analysis of the β-tubulin gene showed the presence of a single nucleotide mutation at the 198th amino acid position of five isolates (16CACY14, 16CAYY19, 15HN5, 15KJ1, and 16CAYY7) of C. gloeosporioides s. lat. Allele-specific PCR and PCR-RFLP were used to detect point mutation at 198th amino acid position and this was done within a day unlike ADM which usually takes more than one week and thus saving time and resources that are essential in the fungicide resistance management in the field. Therefore, the molecular techniques established in this study can warrant early detection of benzimidazole fungicide resistance for the adoption of management strategies that can prevent yield losses among farmers.
Fungicide-resistant Alternaria alternata impede the practical control of the Alternaria diseases in crop fields. This study aimed to investigate cytological fungicide resistance mechanisms of A. alternata against dicarboximide fungicide iprodione. A. alternata isolated from cactus brown spot was cultured on potato-dextrose agar (PDA) with or without iprodione, and the fungal cultures with different growth characteristics from no, initial and full growth were observed by light and electron microscopy. Mycelia began to grow from one day after incubation (DAI) and continued to be in full growth (control-growth, Con-G) on PDA without fungicide, while on PDA with iprodione, no fungal growth (iprodione-no growth, Ipr-N) occurred for the first 3 DAI, but once the initial growth (iprodione-initial growth, Ipr-I) began at 4-5 DAI, the colonies grew and expanded continuously to be in full growth (iprodione-growth, Ipr-G), suggesting Ipr-I may be a turning moment of the morphogenetic changes resisting fungicidal toxicity. Con-G formed multicellular conidia with cell walls and septa and intact dense cytoplasm. In Ipr-N, fungal sporulation was inhibited by forming mostly undeveloped unicellular conidia with degraded and necrotic cytoplasm. However, in Ipr-I, conspicuous cellular changes occurred during sporulation by forming multicellular conidia with double layered (thickened) cell walls and accumulation of proliferated lipid bodies in the conidial cytoplasm, which may inhibit the penetration of the fungicide into conidial cells, reducing fungicide-associated toxicity, and may be utilized as energy and nutritional sources, respectively, for the further fungal growth to form mature colonies as in Ipr-G that formed multicellular conidia with cell walls and intact cytoplasm with lipid bodies as in Con-G.
Effect of the fungicide mepanipyrim on the resistant and sensitive isolates of Botrytis cinerea was studied in vitro and also tested to control Botrytis rot of strawberry, cucumber and grape in the field. These isolates were selected by relative mycelial growth and spore germination on potato dextrose agar(PDA) incorporated with $100{\mu}g\;a.i./ml$ of benomyl and procymidone, respectively, compared to the unamended PDA. Mycelial growth of the selected resistant isolates was significantly inhibited by mepanipyrim but the inhibition rate was similar to other fungicides belong to benzimidazole or dicarboximide, although spore germination was not inhibited even by the higher concentration of mepanipyrim. When the benomyl and procymidone resistant isolates were inoculated to cucumber leaves, lesion development was significantly inhibited with application of $250{\mu}g\;a.i./ml$ of mepanipyrim but not with that of benomyl and procymidone. In addition, when $250{\mu}g\;a.i./ml$ of mepanipyrim was applied to strawberry, cucumber, and grape in the field, the control of Botrytis rot was significantly different from that of the untreated control(Duncan's multiple range test, p<0.05). The results suggested that mepanipyrim might be an alternative fungicide for the control of benomyl- and procymidone-resistant pathogens of Botrytis rot.
This study was performed to investigate the frequency of blue staining fungal species collected from pine trees, Experimental Forests of Kangwon National University in Korea based on their morphological characteristics. In addition the tolerance to fungicide, Woodguard, was assessed to get basic knowledges for preventing blue stain of wood. Totally Leptographium-type fungi were dominated by 79.3% among Ophiostomatoid fungi associated with scolityd bark beetles in pine trees. Leptographium-type Ds-isolates which have unusual morphology were collected as frequency of 17.0%. The most distinct differeneces of these Ds-isolates from L. procerum were the presence of roughened hyphae and flask-shaped conidiophores that have never been mentioned formerly for L. procerum, but since these Ds-isolates formed black concentric rings being a property of L. procerum, the Df-isolates were characterized as Leptographium-type fungi, which are the most common species with the highest frequency by 33.2% in this particular area. According to our experimental results, Leptographium-type Ds- and Df-isolates were very resistant to fungicide, Woodguard, therefore it was suggested that a new method for wood protection from the blue staining fungi should be developed. Exact identification of blue staining isolates collected from pine trees is keep going.
Chemical management of dollar spot in turf may lead to the development of Sclerotinia homoeocarpa populations with reduced fungicide sensitivity. The objective of this study was to investigate resistance of S. homoeocarpa isolates to triazole fungicides and to test cross-resistance among three triazole fungicides. A total of 66 isolates of S. homoeocarpa were collected from 15 golf courses across Korea, and tested via in vitro sensitivity assay against hexaconazole, propiconazole and tebuconazole. $EC_{50}$ values of the isolates to these fungicides were distributed in the range of $0.001-1.1\;a.\;i.\;{\mu}g\;ml^{-1}$. Based on the $EC_{50}$ values, twelve representative strains were selected as sensitive isolates including control and insensitive isolates with respect to each fungicide. At a concentration of $0.1\;a.\;i.\;{\mu}g\;ml^{-1}$ for all fungicides, the selected strains were distinguished as sensitive or resistant isolates with the mycelial growth inhibition rate of 50% as the criterion. The $EC_{50}$ values of resistant strains exposed to hexaconazole, propiconazole and tebuconazole were 20-50 times, 50-70 times, and 77 times greater, respectively, than that of the control strains. Two isolates of S. homoeocarpa S0-41 and Sh14-2-1 showed sensitivity toward all the fungicides used, while two other isolates Sh7-5-1 and Sh2-1-1 showed resistance to all fungicides. Each isolate showed similar resistance to the three types of triazole fungicides, whereby cross-resistance of isolates was confirmed in the present study; all three triazole fungicide combinations displayed significant correlation coefficients equivalent to or greater than 0.8.

In 1999 and 2002, 130 and 258 isolates of Colletotrichum spp. causing red pepper anthracnose were obtained from infected red pepper fruits, respectively. Their responses to 4 protective and 3 ergosterol biosynthesis-inhibiting(EBI) fungicides were investigated by observing their mycelial growth on PDA incorporated with different concentrations of each fungicide. The Colletotrichum isolates obtained in 1999 showed higher $EC_{50}$ values than those isolated in 2002 against three protective fungicides such as dithianon, chlorothalonil, and propineb, whereas the response was reversed toward other protective fungicide, iminoctadine. On the other hand, the isolates of year 1999 were more resistant against three EBI fungicides such as tebuconazole, hexaconazole, and prochloraz than those of year 2002; the $EC_{50}$ values of the former were 1.2-4.4 times higher than those of the latter, The responses of the Colletotrichum isolates toward protective and EBI fungicides were fluctuated according to regions, where the infected fruits were collected. On the other hand, the resis tance of Colletotrichum isolates to protective fungicides increased during monitoring from July to September, However, their responses towards EBI fungicides were not changed.

With 86 isolates of Botrytis cinerea obtained from diseased fruits of tomato, strawberry and cucumber, it was conducted to detect the fungicide resistance of B. cinerea against mepanipyrim through an agar dilution method. FGA medium was used for monitoring the resistance. Among 86 isolates of B. cinerea, resistant isolates were composed by 23.3%, which was different according to regions isolating the pathogen. In accordance with isolation region, the isolation frequency of resistant isolates was as follows; that of Gyeonggi, Gyeongnam/Pusan and chungnam was 28.6%, 33.3% and 12.8%, respectively. The resistant isolates of B. cinerea showed the cross resistance between mepanipyrim and pyrimethanil. Also the pathogenicity of B. cinerea resistant to mepanipyrim was high as like that of sensitive isolates. Because of them, it should be necessary to manage the spraying system of mepanipyrim and the monitoring for the fungicide resistance.
