Pathogenic Diversity of Ascochyta rabiei Isolates and Identification of Resistance Sources in Core Collection of Chickpea Germplasm

  • Ascochyta blight caused by Ascochyta rabiei (Pass.) Lab. (Telomorph: Didymella rabiei) (Kov.) is one of the most important fungal diseases in chickpea worldwide. Knowledge about pathogen aggressiveness and identification resistance sources to different pathotypes is very useful for proper decisions in breeding programs. In this study, virulence of 32 A. rabiei isolates from different part of Iran were analyzed on seven chickpea differentials and grouped into six races based on 0-9 rating scale and susceptibility/resistant pattern of chickpea differentials. The least and most frequent races were race V and race I, respectively. Race V and VI showed highly virulence on most of differential, while race I showed least aggressiveness. Resistance pattern of 165 chickpea genotypes also were tested against six different A. rabiei races. ANOVA analysis showed high significant difference for isolate, chickpea genotypes and their interactions. Overall $chickpea{\times}isolate$ (race) interactions, 259 resistance responses (disease severity ${\leq}4$) were identified. Resistance spectra of chickpea genotypes showed more resistance rate to race I (49.70%) and race III (35.15%), while there were no resistance genotypes to race VI. Cluster analysis based on disease severity rate, grouped chickpea genotypes into four distinct clusters. Interactions between isolates or races used in this study, showed the lack of a genotype with complete resistance. Our finding for virulence pattern of A. rabiei and newly identified resistance sources could be considerably important for integration of ascochyta blight resistance genes into chickpea breeding programs and proper decision in future for germplasm conservation and diseases management.

  • 2005년은 전년에 비해 기온이 낮았고 강수량이 적었다. 벼 흰잎마름병, 세균성벼알마름병, 벼 깨씨무늬병, 벼 이삭도열병, 배나무 겹무늬병 등이 2004년에 비해 증가하였다. 특히 벼 깨씨무늬병은 전년에 비해 4배 이상 병 발생이 증가하였다. 벼 이삭도열병이 전년 0.7%에서 2005년 2%로 증가된 것은 출수기 동안 평균기온은 높았고 강수일수, 강수량 및 습도가 높아 잎도열병균이 이삭으로의 전파를 촉진한 것으로 추정되었다. 기타 조사된 병들은 전년에 비해 감소 혹은 유사한 수준이었다. 벼 잎도열병, 시설참외 시들음증상, 흰가루병 및 노균병은 전년에 비해 크게 감소되었다.

  • 2004년은 전년에 비해 기온이 높았고 강수량이 적었다. 벼 못자리 병, 잎도열병 발생 필지, 잎집무늬마름병, 흰잎마름병과 고추 역병, 탄저병 및 바이러스병, 시설참외의 급성 시들음 증상, 흰가루병 등이 2003년에 비해 증가하였다. 특히 벼잎집무늬마름병과 흰잎마름병은 전년에 비해 2배 이상 병 발생이 증가하였다. 벼 이삭도열병이 전년 $3\%$에서 2004년 $0.7\%$로 감소된 것은 출수기 동안 평균기온은 높았으나 강수일수, 강수량 및 습도가 낮아 잎도열병균이 이삭으로의 전파를 억제한 것으로 추정되었다. 고추역병은 전년에 비해 크게 감소되었다. 그리고 발표하지 않은 다른 병에 의한 피해는 다소 감소 혹은 유사한 수준이었다.

Sclerotinia homoeocarpa에 의한 잔디동전마름병(Dollar spot)의 약제 저항성균 발생 및 방제 (Occurence of Chemical Resistance and Control of Dollar Spot Caused by Sclerotinia homoeocarpa in Turfgrass of Golf Course)

  • Antifungal activity of 20 chemicals registered to turfgrass diseases was evaluated. Among the chemicals, iprodione, benomyl, iprodione+thiram, pencycuron+tebuconazole, hexaconazole, and iprodione+thiophanate-methyl exhibited high antifungal activity to the dollar spot fungus. All isolates were greatly inhibited by the chemicals at the concentration over 32ppm($\mu\textrm{g}$/ml). However, sensitivity of the isolates to chemicals was varied at the lower concentration as 8 and 16 ppm as follows. The isolate originated from Ora golf course was resistant to iprodione and iprodione+thiophanate methyl, Gonjiam isolate to iprodion, Youngpyung isolate to iprodione+thiram, and Dogo isolate to iprodione+thiram, pencycuron+tebuconazole, and tebuconazole. It was found from this study that the varied chemical resistance among the isolates was positively related to the application time of the chemical in the golf course. Consequently, when a chemical was applied more often times than the others to the golf course, the fungal isolates originate from the field showed higher resistance to the former chemical. Effects of the chemicals on control of the dollar spot was evaluated in the field. All tested chemicals revealed over 70% disease control efficacy, however, mepronil+propiconazole was the best showing 83% control efficacy and followed by fenarimo, iprodione, terbuconazole, thiram, and thiophanate-methyl.

Genetic diversity of Fusarium graminearum from rice in Korea

  • Fusarium graminearum (telomorph:Gibberella zeae), an important fungal pathogen of cereal crops with ubiquitous geographic distribution, produces mycotoxins on diseased crops that has threaten human and animal health. Recently severe epidemics of scab diseases of barley and rice by this fungus occurred in Korea, causing serious economic losses. To determine genetic diversity of F. graminearum from rice in Korea, a total of 269 isolates were obtained from Southern part of Korea during 2001-2002. A phylogenetic tree of the isolates was constructed by using amplified fragment length polymorphism (AFLP). Population structure of the rice isolates consists of a single lineage (lineage 6). Frequency of female fertility among these Isolates was relatively low (37%) compared to that among lineage 7 isolates from Korean corn. PCR amplification using chemotype specific primers derived from Tri7 and Tri13 genes at the trichothecene biosynthesis gene cluster revealed that most isolates (260) were NIV chemotype;9 isolates were identified as DON chemotype by Tri13 but as either NIV chemotype or unknown by Tri7. The result of chemical analysis also supported the chemotype determination;all of the NIV chemotype isolates produced NIV, whereas the 9 isolates produce either DON or no toxin.

The glyoxysomal nature of microbodies complexed with lipid globules in Botryospheria dothidea.

  • The glyoxysomal nature of microbodies was determined in Botryosphaeria dothidea hyphae based on morphology and in situ enzyme characteristics by transmission electron microscopy and cytochemistry. Bound by a single membrane, microbodies had a homogeneous matrix and varied in size ranging from 200 to 400 m in diameter. Microbodies had crystalline inclusion(s) which consisted of parallel arrays of fine tubules in their matrices. Microbodies and lipid globules were frequently placed in close association with each other, forming microbody-lipid globule complexes in hyphae. The cytochemical activities of catalase and malate synthase were localized in matrices of microbodies, showing intense electron-density of the organelle. In addition, the immunogold labeling detected the presence of catalase in multivesicular bodies and hyphal cell walls as well as in matrices and crystalline inclusions of microbodies, supporting the enzyme secretion through cell walls. Meanwhile, isocitrate Iyase was localized only in matrices of microbodies. These results suggest that microbodies, particularly complexed with lipid globules, in the fungal hyphae are functionally defined as glyoxysomes, where glyoxysomal enzymes are biochemically active for the glyoxylate cycle to be a metabolic pathway in gluconeogenesis.   • Variable systemic diseases affect larynx and vocal fold and result in voice change. Asthma and chronic obstructive pulmonary disease make increase of intra-abdomimal pressure followed by reflux of gastric acid, which stimulate vagal-bronchopulomary reflex aggravating cough and respiratory disturbance. Fungal laryngitis in the general population is extremely rare, but can occur in immunocompromised AIDS patients. Although, initially, empirical antifungal therapy for candidiasis is often given without biopsy, diagnostic direct laryngoscopy and biopsy is imperative if a substantial clinical response is not rapidly achieved. In the highly active anti-retroviral therapy era, HIV-positive patients are living longer and are at higher risk for developing non-AIDS-defining malignancies. The incidence of head and neck cancer (HNC) which is related with human papilloma virus infection has increased. The survival is significantly lower among the AIDS-HNC patients with CD4 counts ${\leq}200cells/{\mu}L$. Rheumatoid arthritis (RA) cause voice disturbance by developing cricoarytenoid joints fixation or nodule on vocal fold. Post-menopausal voice disorder (PMVD) is caused by decreased secretion of estrogen-progesterone resulting in decrease of fundamental frequency (F0). Hormonal replacement therapy is helpful to reduce F0 decrease. RA and PMVD result in slight voice change, but it could crucial in professional voice user.

Aggressiveness of Three Snow Mold Fungi on Creeping Bentgrass Cultivars under Controlled Environment Conditions

  • Snow molds are the most important winter diseases of turfgrass in the United States and Canada. Eight isolates of three snow mold fungal species (three isolates of Typhula ishikariensis, three of T. incarnata, and two of Microdochium nivale) were collected from infected turfgrasses on golf courses. The isolates were evaluated for their relative aggressiveness on three cultivars (L-93, Penncross, and Providence) of creeping bentgrass (Agrostis palustris) under the same controlled conditions. Four plant ages (15, 19, 23 and 27 week-old plants from germination to inoculation) were evaluated for their susceptibility to the three pathogens and for the recovery of the plants. Regardless of age or cultivar of the host plant, M. nivale was found to be more aggressive and faster to infect and colonize than Typhula species. After three weeks recovery, M. nivale-inoculated plants showed higher disease severity than plants inoculated with the two Typhula species. Plants infected by Typhula species displayed no significant difference in disease severity. As creeping bentgrass plants get older, the severity of disease caused by three snow molds gradually decreases. This effect was observed in all cultivars tested, suggesting expression of age-related resistance as the bentgrass plants matured.

Effect of a Bacterial Grass Culture on the Plant Growth and Disease Control in Tomato

  • This study aimed to investigate the plant growth-promoting and biocontrol potential of a grass culture with Paenibacillus ehimensis KWN8 on tomato. For this experiment, treatments of a chemical fertilizer (F), a bacterial grass culture (G), a 1/3 volume of G plus 2/3 F (GF), and F plus a synthetic fungicide (FSf) were applied to tomato leaves and roots. The result showed that the severity of Alternaria solani and Botrytis cinerea symptoms were significantly reduced after the application of the bacterial grass culture (G and GF) and FSf. In addition, root mortality in G and GF was lower compared to F. Tomato plants treated with G or GF had better vegetative growth and yield compared to F. Application of G affected the fungal and bacterial populations in the soil. In conclusion, treatment with a bacterial grass culture decreased disease severity and increased tomato growth parameters. However, there were no statistically significant correlations between disease occurrence and tomato yields. This experiment presents the possibility to manage diseases of tomato in an environmentally friendly manner and to also increase the yield of tomato by using a grass culture broth containing P. ehimensis KWN38.

Production of transgenic Alstroemeria plants containing virus resistance genes via particle bombardment

  • Transgenic Alstroemeria plants resistant to Alstroemeria mosaic virus (AlMV) were generated through RNA-mediated resistance. To this end, the friable embryogenic callus (FEC) of Alstroemeria was induced from the leaf axil tissue and transformed with a DNA fragment containing the coat protein gene and 3'-nontranslated region of AlMV through an improved particle bombardment system. The bar gene was used as a selection marker. More than 300 independent transgenic FEC lines were obtained. Among these, 155 lines resistant to phosphinothricin (PPT) were selected under low stringent conditions. After increasing the stringency of PPT selection, 44 transgenic lines remained, and 710 somatic embryos from these lines germinated and developed into shoots. These transgenic shoots were then transferred to the greenhouse and challenged with AlMV. In total, 25 of the 44 lines showed some degree of resistance. PCR analysis confirmed the presence of the viral sequence. Virus resistance was observed at various levels. Establishment of an efficient transformation system for Alstroemeria will allow inserting transgenes into this plant to confer resistance to viral and fungal pathogens. Accordingly, this is the first report on the production of a transgenic virus-resistant Alstroemeria and lays the foundation for alternative management of viral diseases in this plant.
