Variation in Fucoidan Contents and Monosaccharide Compositions of Korean Undaria pinnatifida (Harvey) Suringar (Phaeophyta)

  • Three different forms of Undaria pinnatifida, the southern form (U. pinnatifida f. typica), the northern form (U. pinnatifida f. distans), and Samcheok form (recently cultivated strain), were examined for the contents and compositions of fucoidans. Fucoidans were extracted from the dried edible portions of three forms of U. pinnatifida in low pH condition, mainly by ethanol precipitation and CaCl2 treatment. It was shown that Samcheok form contains 1.8 and 3.5 times more fucoidans than the northern and the southern forms, respectively. The monosaccharide compositions of individual fucoidans were also varied. The fucoidans from the southern and the northern forms were shown to be composed of mainly fucose and galactose with the molar percentage ratios of 83.5%:16.5% and 87.4%:12.6%, respectively, indicating that these are F-type fucoidans. The fucoidan from Samcheok form, however, consisted of fucose (62.7%), galactose (32.9%), and small amount of glucose (4.4%). The results of this study showed that both amount and monosaccharide compositions of fucoidans are variable depending on U. pinnatifida forms.

톳과 모자반 fucoidan의 화학적 특성 (Chemical Properties of Fucoidans from Hizikia fusiformis and Sargassum fulvellum)

  • 톳과 모자반으로 부터 fucoidan을 추출, 정제하여 그 성질을 분석, 검토하였다. Fucoidan의 추출 조건은 pH 2.0의 산성 추출액으로 $65^{\circ}C$에서 1시간씩 3회 추출을 하고 cethylpyridinum chloride를 사용하여 부분 정제하였다. 부분 정제 fucoidan의 수율은 톳 $2.51\%$, 모자반 $1.56\%$였다. DEAE-Sephadex A-25 이온 교환 칼럼으로 부분 정제 fucoidan을 분획한 결과 톳, 모자반 모두 3종의 획분으로 분획되었고, 각 획분은 전기 영동상에서 2종 이상의 band가 검출되어 불균일함을 알 수 있었다. 분획한 주획분을 다시 알콜 분별 침전법으로 재분획한 결과 톳은 에타놀농도 $60-70\%$에서 침전된 획분 (P-70)이, 모자반은 에타놀농도 $60-65\%$에서 침전된 획분 (P-65)이 cellulose acetate 전기 영동 및 젤 여과 크로마토그라프상에서 각각 균일성을 나타내었다. 균일하게 정제 된 톳과 모자반 fucoidan의 fucose : galactose : 황산기의 mole 조성비는 각각 1:0.66:2.74와 1:0.24:1.46였으며, 분자량은 톳은 26,000, 모자반은 105,000이었다.

한국산 다시마 및 미역으로 부터 Fucoidan의 추출 및 정제 (Isolation and Purification of Fucoidans from Laminaria religiosa and Undaria pinnatifida in Korea)

  • 국내산 갈조류로부터 생리 기능성이 우수한 수용성 식이섬유 소재를 개발하기 위한 기초자료를 얻기위해 다시마와 미역(엽상체와 포자엽 각각)으로부터 fucoidan을 추출, 정제하여 그 특성을 조사하였다 Fucoidan의 추출 조건은 pH 2.0의 산성 추출액으로 $65^{\circ}C$I에서 1시간 씩 3회 추출을 하였고 cetylpyridinium chloride를 사용하여 부분 정제하였다. 부분 정제 fucoidan의 수율은 다시마 $2.71\%$, 미역포자엽 $6.65\%$, 미역 엽상체$0.40\%$였다. 특히 미역포자엽의 수율이 가장 높은 반면에 미역 엽상체는 가장 낮아 부위에 따른 차이가 매우 큼을 알 수 있었다. DEAE-Sephadex A-25 이온 교환 칼럼으로 부분 정제 fucoidan을 분획한 결과 다시마, 미역포자엽 모두 3종의 fraction으로 분획되었고 각 fraction은 전기 영동상에서 2종 이상의 band가 검출되어 불균일함을 알 수 있었다 이온 교환 칼럼으로 분획한 주fraction을 다시 알콜 분별 침전법으로 재분획한 결과 다시마와 미역포자엽은 에탄올농도 $60-70\%$에서 침전된 fraction이 cellulose acetate 전기 영동 및 gel filteration chromatography상에서 균일성을 나타내었다. 균일하게 정제된 fucoidan의 fucose : galactose : 황산기의 mole 조성비는 다시마, 미역포자엽이 각각 1 : 0.31 : 2.43,1 : 0.87 : 1.99였다. 분자량은 다시마가 31,000, 미역포자엽이 38,000으로 비교적 적었다.

Contrasting Correlation in the Inhibition Response of ADP-induced Platelet Aggregation and the Anti-coagulant Activities of Algal fucoidans Derived from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu)

  • Sulfated fucans are known to have both anti-thrombotic and anti-coagulant activities. In this study, the variation in platelet aggregation and anti-coagulant activities was investigated in vitro with regard to administered dose, molecular weight distribution, sulfate content, and sugar composition in two algal fucoidans from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu). The anti-coagulant activity largely correlated with sulfate content and with molecular weight distribution in a dose-dependent manner. However, both fucoidans demonstrated inhibitory responses to ADP-induced platelet aggregation in dose- and structure-dependent manners that contrasted with the anti-coagulant activity. Neither molecular weight distribution nor sulfate content greatly affected platelet-aggregation inhibition (PA-inhibition) by the fucoidan fractions, whereas anti-coagulant activity was sensitive to these structural factors. Interestingly, an E. bicyclis fucoidan fraction exhibited almost complete PA-inhibition at a treatment dose of 500 mg/mL while retaining weak anti-coagulant activity. In conclusion, these observations suggest that fucoidan may be a useful anti-thrombotic or anti-platelet agent in various arterial thrombotic disorders, including post-vascular intervention with controlled bleeding complications, due to its anti-coagulant modulating activity.

Physicochemical Properties and Antioxidant Effects of Fucoidans Degraded by Hydrogen Peroxide under Electron Beam at Various Irradiation Doses

  • Fucoidans were degraded by hydrogen peroxide under the electron beam (2.5 MeV) with various radiation doses (5 kGy, 10 kGy, 15 kGy, and 20 kGy) at room temperature. The degradation property was analyzed with a gel permeation chromatography (GPC-MALLS) method. An average molecular weight of fucoidan decreased from 99,956 at the irradiation dose of 0 kGy to 6,725 at the irradiation dose of 20 kGy. The solution viscosity of fucoidans showed a similar pattern to the molecular weight change. The number of chain breaks per molecule (N) increased with increasing the irradiation dose and concentration of hydrogen peroxide. The radiation yield of scission value markedly increased with increasing the irradiation dose up to 15 kGy. Also a 10% hydrogen peroxide concentration was more efficient than that of 5%. The structures of degraded fucoidan samples were studied with Fourier transform infrared spectroscopy (FT-IR). The results showed that the degradation process did not significantly change the chemical structure or the content of sulfate group. The sulfur content of each sample was determined with an Elemental Analyzer. With increasing the concentration of hydrogen peroxide, the ratios of sulfur/carbon, hydrogen/carbon, and nitrogen/carbon slightly decreased. The antioxidant activities of fucoidans were investigated based on hydroxyl radical scavenging activities. The ability of fucoidan to inhibit the hydroxyl radical scavenging activity was depended on its molecular weight.

  • 국내산 미역포자엽, 다시마, 톳, 모자반에서 추출한 부분 정제 fucoidan의 유동특성을 측정하였다. 겉보기 점도는 모자반, 다시마, 톳, 미역포자엽 순으로 높았으며 특히 전단속도 1,500 1/s에서 미역포자엽의 용액 $(5\%)$의 겉보기 점도는 2.2mPa.s로 매우 낮았다. 전시료 모두 유동지수 1이하의 의가소성 유체의 성질을 나타내었으며 시료의 농도가 증가함에 따라 의가소성이 증가하였다. 온도에 따른 활성화 에너지는 전단속도 1,500 1/s에서 톳 $8.269\times10^6$, 다시마 $7.498\times10^6$, 미역포자엽 $7.141\times10^6$, 모자반 $6.677\times10^6\;J/kg.mol$로 낮아 온도 의존성이 적었다. 또한 설탕 첨가시는 부분 정제 fucoidan의 점도가 증가한 반면에 소금 첨가시는 변화가 없었다.

Structural characterization and anti-inflammatory activity of fucoidan isolated from Ecklonia maxima stipe

  • Enzyme-assisted hydrolysis is frequently used as a cost-effective and efficient method to obtain functional ingredients from bioresources. This study involved the enzyme-assisted hydrolyzation and purification of fucoidan from Ecklonia maxima stipe and the investigation of its anti-inflammatory activity in lipopolysaccharide (LPS)-induced RAW 264.7 cells. Fucoidans of Viscozyme-assisted hydrolysate from E. maxima (EMSFs) harvested in Jeju, Korea. Structural and chemical characterizations were performed using fourier transform infrared spectroscopy, scanning electron microscope, and monosaccharide analysis. Among fucoidans, EMSF6 was rich in fucose and sulfate and had a similar structural character to commercial fucoidan. EMSF6 showed a strong inhibitory effect on nitric oxide generation in LPS-induced RAW 264.7 cells and significantly decreased the production of LPS-induced pro-inflammatory cytokines, including interleukin-6, interleukin-1β, and tumor necrosis factor α. The anti-inflammatory potential of EMSF6 was mediated through the downregulation of inducible nitric oxide synthase and cyclooxygenase-2 expression. Thus, fucoidans from E. maxima stipe are promising candidates for functional food products.

  • 국내산 주요 갈조류인 미역, 다시마, 톳, 모자반 정제 fucoidan의 기본 결합 구조 및 황산기 결합 위치를 확인하기 위하여 정제 fucoidan및 이를 methanolysis하여 얻은 고분자 획분을 각각 methylation 및 FT-IR 측정을 하였다. 다시마, 톳, 모자반, 미역포자엽 정제 fucoidan의 주 결합은 $(1\rightarrow3)$ linked fucose였으며 미역포자엽에는 $(1\rightarrow3)$ linked galactose도 상당량 존재하였다. 황산기의 대부분은 fucose와 galactose의 C-4의 위치에 결합되어 있는 것으로 추정된다.

해조류를 이용한 후코이단의 생산 (Production of Fucoidan Using Marine Algae)

  • Fucoidans, polysaccharides that are composed of fucose, uronic acids, galactose and xylose, are one of the main constituents of brown seaweeds. For the past decade algal fucoidans have been extensively studied due to their biological activities including anticoagulant, antiinflammatory, antitumor, contraceptive and antiviral activities. Even though fucoidan is known as a highly functionality derivative, its industrial applications have been limited because of low inclusion efficiency and high cost of manufacturing. Furthermore the sea-weed smell of fucoidan has been a limiting factor for the application especially in food and cosmetic area. The potential application of various extraction technology and deodorization of sea-weed smell is highly required for a wider application of fucoidan. As an alternative extraction process, enzymatic hydrolysis process or ultra-high pressure treatment has been investigated for the improvement of production yield and bioefficacy of fucoidan. More intensive research on the fucoidan production technology and its application in nutraceutical and cosmoceutical area is needed.

Antioxidant Activity of Sulfated Polysaccharides Isolated from Sargassum fulvellum

  • Sargassum fulvellum, a marine brown alga, is a popular low priced edible plant in Korean markets. The polysaccharide fraction of the alga was separated and investigated for its radical scavenging activities and the results compared with those of commercial fucoidans (Fucus vesiculosus and Undaria pinnatifida), BHA and ${\alpha}$-tocopherol. The polysaccharide fraction of S. fulvellum showed a promising DPPH radical scavenging activity than did other fucoidans. Moreover, the sample exhibited a dose-dependent activity on hydrogen peroxide scavenging activity in the V79-4 cell line. Interestingly, all the tested polysaccharide counterparts were more potent NO. scavengers than were the commercial antioxidants, BHA and ${\alpha}$-tocopherol. The sulfated polysaccharide of S. fulvellum had an approximate molecular weight of 529 kDa and mainly consisted of fucose and galactose, and minor amounts of mannose, rhamnose and xylose.
