Development of Fresh Cheeses and Whey Drinks Using Milk Components (우유 성분을 이용한 생치즈와 유청 음료의 개발)

  • In order to save foreign currency and to domesticize the dairy products, various fresh cheeses and whey drinks were developed and some physicochemical, microbiological and sensory evaluation were performed. The yield of fresh cheese was 22.3%, while that of whey 77.7%. The pH-values of fresh cheeses were $5.90{\sim}6.49$, while those of whey drinks $6.07{\sim}6.49$, and fermented whey drinks $3.97{\sim}4.91$. The acidities of fresh cheeses were $0.09{\sim}0.26%$, while those of whey drinks $0.09{\sim}0.36%$. The contents of solid substances, protein and lactose in fresh cheeses were $25.67{\sim}34.18%$, $7.45{\sim}9.11%$ and $3.61{\sim}4.14%$, while those of whey drinks $7.39{\sim}7.70%$, $0.88{\sim}0.94%$ and $4.93(\sim}6.17%$, respectively. The lactic acid contents of whey drinks varied from $0.01{\sim}0.38%$, where the content in the fermented sample was the highest. The general colony counts of fresh cheeses were $0{\sim}30/g$, while those of whey drinks $0{\sim}80/ml$. The psychrotrophs counts of fresh cheeses were $0{\sim}20/g$, while those of whey drinks $0{\sim}60/ml$. Lactic acid bacterial counts in both products were not detected except for $97{\sim}401{\times}10^8/ml$ in fermented whey drinks. E. coli and fungi were not detected in both products. In sensory evaluation of both products, the strawberry added fresh cheese was the best of fresh cheeses, while the garlic added fresh cheese was the worst. Pure whey drink was the best of whey drinks, while the ginseng added whey drink was the worst.

  • This study was performed to investigate the high yielding potential of Pleurotus ostreatus (Fr.) Quel derived from intermating and intramating of three strains: Local strain, Nonggi 201, and Pleurotus florida Eger. All the strains tested were tetrapola mating systems, and hybridable. Intermating system resulted in more fresh sporophores than those from intramating. The highest yield of fresh sporophores of P. ostreatus was obtained from Nonggi 201 crossed with P. florida, when cultivated on the rice straw substrates.

Cell-Recycle Continuous Fermentation of Enterococcus faecalis RKY1 for Economical Production of Lactic Acid by Reduction of Yeast Extract Supplementation

  • Both lactic acid productivity and cell growth were linearly correlated with yeast extract supplementation in batch fermentation. During conventional continuous operation, although fresh feed was introduced into the bioreactor with a significantly low dilution rate (0.04 $h^{-1}$), the amount of yeast extract employed was not enough to maintain the growth of microorganism. However, when the fresh feed contained 100 g/l glucose and 2 g/l yeast extract during cell-recycle continuous operation at a dilution rate of 0.04 $h^{-1}$, more than 90 g/l lactic acid was continuously produced, with the average productivity of 3.72 $g/l{\cdot}h$. In this experiment, 82 g of yeast extract (77% of reduction yield) could be reduced for the production of 1 kg of lactic acid compared with batch fermentation of a similar volumetric productivity.

Co-inoculation of Burkholderia cepacia and Alcaligenes aquatilis enhances plant growth of maize (Zea mays) under green house and field condition

  • The synergistic effect on phosphate solubilization of single- and co-inoculation of two phosphate solubilizing bacteria, Burkholderia cepacia (C1) and Alcaligenes aquatilis (H6), was assessed in liquid medium and maize plants. Co-inoculation of two strains was found to release the highest content of soluble phosphorus (309.66 ?g/mL) into the medium, followed by single inoculation of B. cepacia (305.49 ?g/mL) and A. aquatilis strain (282.38 ?g/mL). Based on a plant growth promotion bioassay, co-inoculated maize seedlings showed significant increases in shoot height (75%), shoot fresh weight (93.10%), shoot dry weight (84.99%), root maximum length (55.95%), root fresh weight (66.66%), root dry weight (275%), and maximum leaf length (81.53%), compared to the uninoculated control. In a field experiment, co-inoculated maize seedlings showed significant increases in cob length (136.92%), number of grain/cob (46.68%), and grain weight (67.46%) over control. In addition, single inoculation of maize seedlings also showed improved result over control. However, there was no significant difference between single inoculation of either bacterial strains and co-inoculation of these two bacterial strains in terms of phosphate solubilization index, phosphorous release, pH of the media, and plant growth parameters. Thus, single inoculation and co-inoculation of these bacteria could be used as biofertilizer for improving maize growth and yield.

Effects of Temperature and Light intensity on Growth and Yield of Condonopsis lanceolata (온도 및 光條件이 더덕의 생육과 수량에 미치는 영향)

  • The present study was performed to obtain a basic data of cultivation for Condonopsis lanceolata. Various temperatures, light, and DIF were treated during the whole plant growth. The growth of aerial part was most remarkable between $20^{\circ}C$ and $25^{\circ}C$, but inhibited by means of shading treatment. The fresh weight of subterranean part was heaviest(16.6g) at $15^{\circ}C$. Also, plant height increased when a constant temperature and +DIF were treated. Leaf width and length was not related to temperature and DIF treatment. The fresh weight of subterranean part increased in +DIF-treated sample at $25^{\circ}C$.

Weed control treated with salt and seawater in organic agricultural upland

  • Weed control is the most important issue in organic farming systems that limit crop growth and their yield. Field experiments were conducted in organic soybean (Glycine max Merrill) to evaluate the weed suppression effects of salt and seawater treatment. Weed population and fresh weight were monitored after 6 weeks of salt and seawater treatments. The most important weeds were Digitaria sanguinalis, Portulaca oleracea, Tradescantia reflexa and Chenopodium album var. centrorubrum, but also 6 other species were observed in soybean arable field. Soybean crops under seawater or their solids application were well grown. The results treated with salts and seawater indicate decreases by 13.4~30.8% in weed density and by 18.0~43.2% in their fresh weight and soil hardness increases of up to 2.1-fold. Salt and seawater provided good additional weed control, but they were caused a serious problem in deterioration of soil physical properties.

Optimization of Maca (Lepidium meyenii) Extraction for Natural Beverage Development using Enzyme Treatment (효소처리에 의한 천연 마카음료 개발을 위한 최적 추출 조건)

  • The purpose of this study was to establish the best optimized extraction condition for the optimal development of fresh maca beverage using low temperature extraction and enzyme treatment. Low temperatures were applied to prevent heat-related nutritional loss during the extraction process. Best extraction conditions were investigated based on the ratio of maca to water, the ratio of enzymes, extraction temperature and time, and agitation. The optimal enzyme conditions were also examined after the treatment of cellulase:pectinase mixture to maintain the original color and flavor, as well as to increase the extraction yield. When cellulase:pectinase was 1:1, the extraction rate ranged from 77.84 to 79.29%. In addition, the best extraction rate was found when maca was mixed with twice volume of water and incubated at $45^{\circ}C$ ($84.05{\pm}0.32%$) with 90 rpm ($87.13{\pm}0.46%$) agitation for 3 hours ($84.73{\pm}0.29%$). Furthermore, sensory evaluation showed a high score in flavor, sweetness, and overall acceptability after adding 3% jujube concentrate into a fresh maca beverage.

Improvement of Biomass Degradation by Fenton Oxidation and Reusability of the Fenton Oxidation Solution (펜톤산화에 의한 바이오매스 분해향상과 펜톤산화 용액 재사용 평가)

  • In this study, the reusability of the Fenton oxidation solution was evaluated to reduce the cost of the pretreatment process. Biomass was sequential subjected to Fenton oxidation-hydrothermal treatment and enzymatic hydrolysis to produce monosaccharides. The liquid solution recovered after Fenton oxidation contained OH radicals with a concentration of 0.11 mol/L. This liquid solution was reused for a new Fenton oxidation reaction. After Fenton oxidation, hydrothermal treatment was performed under the same conditions as before, and 9.34-13.63 g/L of xylose was detected. This concentration was slightly lower than that of a fresh Fenton oxidation solution (16.51 g/L) but was higher than that obtained by hydrothermal treatment without Fenton oxidation (2.72 g/L). The degradation rate during hydrothermal pretreatment involving Fenton oxidation was 36.02%, which decreased (29.24-31.05%) slightly when the liquid solution recovered after Fenton oxidation was reused. However, the degradation rate increased compared to that measured from hydrothermal treatment without Fenton oxidation (15.21%). Moreover, the yield after enzyme hydrolysis decreased in the following order: fresh Fenton oxidation-hydrothermal treatment (89.64%) > Fenton oxidation with reused solution-hydrothermal treatment (74.84%) > hydrothermal treatment without Fenton oxidation (32.05%).

Effect of Seeding Date on Forage Yield and Chemical Composition of Kenaf in Jeju (제주지역에서 파종기에 따른 양마의 사료수량 및 조성분 변화)

  • This study has been conducted from April 25, 1999 to June 25, 1999 in Jeju Province in order to determine the influence of seeding date on growth, yield and chemical composition of Kenaf (Hibiscus cannabinus L.) Delaying the seeding date from April 25 to June 25 decreased the plant height of Kenaf from 278.5 cm to 205.7 cm. As for the number of leaves, number of withering leaves, number of branches, and stem diameter decreased as the seeding date was delayed. As the seeding date was delayed, fresh forage yield decreased from 98.5MT/㏊ to 45.9MT/㏊, dry matter yield from 20.7MT/㏊ to 8.2MT/㏊, crude protein (CP) yield from 2.9MT/ha to 1.3MT/ha and total digestible nutrients (TDN) yield form 1l.6MT/㏊ to 5MT/㏊. However, both leaf yield and stem yield were nearly the same tendency. Delaying the seeding date from April 25 to June 25 increased crude protein contents of leaves from 21.5% to 24.4%, crude fat contents from 5.2% to 6.1%, nitrogen free extract (NFE) contents form 39.9% to 41.2% and TDN contents from 64.3% to 69.7%. However, delaying seeding date decreased crude fiber contents from 20.8% to 17.5%, and crude ash contents from 7.9% to 7.0%. Based on the these findings, optimum seeding date for forage production of Kenaf seems to be about 25 April in atmospheric phenomena and volcanic ash soils of Jeju island.

Influence of shading and polyethylen vinyl mulching on growth and yield of Cinidium officinale Makino (차광(遮光), 비닐피복(被覆)이 궁궁이(천궁(川芎))의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • The study was carried out to elucidate the influenced of shading and mulching with polyethylen vinyl on growth and yield of Cinidium officinale Makino, and improve the cultivation method of those medicinal plants in northern area of Gyeong-buk province, Korea. Top fresh weight of Cinidium officinale Makino was increased by both 35 % and 55% shading compared to the non-shadingand decreased by 75 % shading, but that of Ligusticum chuanxing Hart. Was decreased by the shading tested The summer wilt of Cinidium officinale Makino severely occurred in non-shading but greatly reduced by 55 % or 75 %shading, while no difference was observed in Ligusticum chuanxing Hort, between the shading and the non-shading. Dry root yield Cinidium officinale Makino was increased by 17 % and 19 % in 35 % and 55 % shading levels, respectively, compared to the dry root yield(254kg/10a) in non-shading. However, it was decreased in Ligusticum chuaxing Hort. by the shading. Emergence ratio of Cinidium officinale Makino was increased by black polyethylen vinyl mulch but decreased by transparent polyethylen vinyl mulch, compard to non-mulch. Polyethylen vinyl mulch increa­sed the top fresh weight of Cinidium officinale Makino Especialy black polythylen vinyl mulch was very effective on increasing the top fresh weight. Dry root yield of Cinidium officinale Makino was increased by 32 % and 30 %, respectively, compared to the non-shading.
