Classification and Genetic Variation Analysis Among Formae Speciales of Fusarium oxysporum by Using Recombinant DNA Probes (재조합 DNA probe에 의한 Fusarium oxysporum 분화형간의 분류 및 유전적 변이 분석)

  • Five formae speciales of Fusarium oxysporum in Korea were examined using RFLP analysis to find the possibility for classification and analyze genetic variations. DNAs from F. oxysporum f. sp. lycopersici, cucumerinum, fragariae, garlic and sesami were used with three recombinant probes such as pFC46, pFC52 and pFC57. Distinct differences among five formae speciales of this fungus were detected in RFLP band patterns based on southern hybridization of genomic DNA using each recombinant clone, which was a repetitive copy probe. Strains belong to four formae speciales could be very stable in genetic variation except f. sp. sesami which has more variation than the others based on the RFLP analysis. They formed their own cluster which has high similarity within the same formae specialis resulted from the UPGMA analysis for genetic relationship analysis and each cluster represented its own formae specialis. The method using three recombinant DNA probes could be a good tool for classification of formae speciales in F. oxysporum.

  • The positive active material for polymer film-battery was prepared by using polyphenlenediamine(PPD) synthesized in our lab. and 2,5-dimercapto-1,3,4-thiadiazole(DMcT) with various mixture ratio. The transference measurement of surface morphology and thermal stability of the prepared composite film was carried out by using SEM and TGA, respectively. Electrochemical property and electrical conductivity of the composite film were also measured by using cyclic voltammetry and four-probe method in dry box, respectively. The thermal stability of prepared composite film was up to $200^{\circ}C$. The electrical conductivity of the composite film increased and showed the highest value(about 3 S/cm) when doped at 0.4% $LiCIO_4$ solution. And we could confirm that DMcT was effective on reactivation of PPD through cyclic voltammogram.

DC Potential Drop Method for Evaluating Material Degradation

  • The remaining life estimation for the aged components in power plants as well as chemical plants are very important because mechanical properties of the components are degraded with in-service exposure time in high temperatures. Since it is difficult to take specimens from the operating components to evaluate mechanical properties of components, nondestructive techniques are needed to evaluate the degradation. In this study, test materials with several different degradation levels were prepared by isothermal aging heat treatment at $630^{\circ}C$. The DC potential drop method and destructive methods such as tensile and fracture toughness were used in order to evaluate the degradation of 1Cr-1Mo-0.25V steels. In this result, we can see that tensile strength and fracture toughness can be calculated from resistivity and it is possible to evaluate material degradation using DC potential drop method, non-destructive method.

Grain Size Dependence of Ionic Conductivity of Polycrystalline Doped Ceria

  • Conductivities of polycrystalline ceria doped with several rare earth oxides were measured by AC admittance and DC four probe method. The conductions were separated into grain and grain boundary contributions using the complex admittance technique as well as grain size dependence of conductivity. The grain size dependence of polycrystalline conductivity, which can be adequately described by the so-called brick layer model, appears to give a more reliable measure of the grain conductivity compared to the complex admittance method. Polycrystalline resistivity(1/conductivity) increases linearly with the reciprocal of grain size. The intercept of resistivity vs. inverse grain size plot gives a measure of the grain resistivity and the slope gives a measure of the grain boundary resistivity. It was also noted that errors involved in the analysis of experimental data may be different between the complex admittance method and the impedance method. A greater resolution of the spectra was found in the complex admittance method, insofar as the present work is concerned, suggesting that the commonly used equivalent circuit may require re-evaluation.

Fine YBaCuO Powder Prepared by Sol-gel Method

  • In order to get proper original suspension for the electrophoresis process of making YBaCuO films, YBaCuO superconductor powder was prepared with the Sol-gel method. The composition of the powder was analyzed by X-ray diffraction, which was identified as YBa$_2$Cu$_3$O$\sub$7-x/(Yl23) phase. The form of YBaCuO single particle was showed to be spherical by scanning electronic microscope and its size was among 0.2-1 $\mu\textrm{m}$. The critical transition temperature (T$\sub$c/) and the critical current density (J$\sub$c/) were measured with the four-probe, method. The T$\sub$c/ was about 91 K, and the J$\sub$c/ was 5-30 A/$\textrm{cm}^2$.

  • An experimental study was performed to investigate the turbulence intermittency measuring methods across the boundary layer in the transition region. A single type hot-wire probe was used to measure instantaneous streamwise velocities in laminar, transitional and turbulent boundary layer To estimate wall shear stresses on the flat plate, near wall mean velocities are applied to the principle of CPM. Distribution of intermittency factor is obtained by dual-slope method and compared to the results of four methods,$\'{u},\;\{U}$, TERA and M-TERA method. In these methods, M-TERA shows a good agreement in the near wall region. However, the result of M-TERA method shows that intermittency factor is underestimated in the outer part and outside of the boundary layer and the dimensional constant of M-TERA method should be changed appropriately depending on measuring point.

  • High Temperature Superconducting (HTS) power cables are capable of transmitting bulk power without any loss compared to conventional copper cables. The major challenge in the design of such HTS cables is the high stresses (electro-thermal/electro-mechanical) developed at high voltages, high currents and cryogenic temperatures. The safe and reliable operation of HTS cables involves lots of instrumentation for monitoring, measurement, control and safe operation. In principle, a four probe method for resistance (RTD PT-100) is used for temperature measurements at various locations of HTS cable. The number of connecting leads required for this is four times that of the number of sensors. The present paper discusses a novel way of connecting 128 RTD sensors with the help of only 14 leads using a cold electronics based multiplexer board. LabVIEW 11.0 software was used for interfacing and displaying the readings of all the sensors on computer screen.

  • It is well known that thin film growth and surface morphology can be substantially modified by ion-bombardment during the deposition. This is particularly important in case of thin-film deposition at low temperatures where the film growth occurs under highly nonequilibrium conditions. An attractive way to promote crystalline growth and surface morphology is deposition of additional energy in to the surface of the growing film by bombardment with hyperthermal particles. We were deposited crystalline Ti and TiN thin films on Si substrate by magnetron sputtering method with grid. Its thin films were highly smoothed and dense as increasing grid bias. In order explore the benefits of a bombardment of the growing film with high energetic particles. Ti and TiN films were deposited on Si substrates by an unbalanced magnetron sputter source with attached grid assembly for energetic ion extraction. Also, we have studied the variation of the plasma states by Langmuir probe and Optical Emission Spectroscopy (OES). The epitaxial orientation. microstructual characteristics. electrical and surface properties of the films were analyzed by XRD. SEM. Four point probe and AFM.

  • Indium-tin oxide (ITO) films were deposited on the glass substrate by the reactive -ionized cluster beam deposition(ICBD) method. In the oxygen atmosphere, indium cluster formed through the nozzle is ionized by the electron bombardment and is accelerated to be deposited on the substrate. And tin is simultaneoulsy evaporated from the boron-nitride crucible. The chracteristics of films were examined by the X-ray photoelectron spectroscopy(XPS), glancing angle X-ray diffractrion(GXRD) and the electrical properties. were measured by 4-point-probe and Hall effect measurement system . From the XPS spectrum , it was found that indium and tin atoms combined with the oxygen to form oxide$(In_2O_3, SnO_2)$. In the case of films with high tin-concentration, the GXRD spectra show that the main $In_2O_3$ peak of (222) plane, but also sub peaks((440) peak etc.) and $SnO_2$ peaks were detected. From that results, itis concluded that the heavily dopped tin component (more than 14 at. %) disturbs to form $In_2O_3$(222) phase. Four-point-probe and Hall effect measurement show that, in the most desirable case, the transmittance of the films is more then 90% in visible range and its resistivity is $$\rho$=3.55 \times10^{-4}\Omega$cm and its mobility is $\mu$=42.8 $\textrm{cm}^2$/Vsec.
