Emulsion Graft Polymerization of MMA to Sodium Alginate : Mechanism and Solvent Effect (알긴산 소다에의 MMA유화 그래프트 중합 : 메커니즘 및 용매효과)

  • In order to increase the lower values of % grafting and monomer conversion in the emulsion graft polymerization of methylmethacrylate(MMA) onto sodium alginate (SA; a polymer electrolyte) with alginic acid-g-PMMA, the graft polymerization with water soluble (methanol and acetone) and insoluble solvents was carried out using a varity of solvent amounts and agitating rates. And some physical properties of the graft polymer were also investigated. In the polymerizations with water insoluble solvent, there were pronounced improvements in both % grafting and MMA conversion by the promotion of MMA diffusion from the core to the SA adsorpted on the outer layer of particle and that of MMA complex formation with SA. And the effect was larger for the solvents (cyclohexane, decalin) which do not dissolve PMMA. On the other hand, in the polymerization with water soluble solvent, there was an significant increase in MMA conversion and a considerable increase in the % grafting by the destruction of electrical double layer around the SA chains, abstracting the hydrated water in the layer and the effect was more potential in the case of methanol which is a precipitant of PMMA.

Preparation and Stability of Silyl Adlayers on 2×1-Reconstructed and Modified Si(100) Surfaces (Si(100)-2×1 표면과 개질된 Si(100) 표면 상에서 실릴 (Silyl) 흡착충의 형성과 안정성)

  • Saturation-coverage silyl, $-SiH_3(a)$, overlayers were prepared from $Si_2H_6$ adsorption on three comparative surfaces: clean unmodified; D-precovered; and atomically roughened Si(100). Together with its precursor-mediated adsorption behavior, the surface reactivity of $Si_2H_6$ was found to be the highest on the unmodified Si(100)-$2{\times}1$ surface. This was correlated with its dissociative adsorption mechanism, in which both the $H_3Si-SiH_3$ bond scission and the dual surface $Si-SiH_3(a)$ bond formation require a surface dangling bond 'pair'. The unusually high thermal stability of $-SiH_3(a)$ on the unmodified surface was ascribed to a nearly close-packed $-SiH_3(a)$ coverage of ${\sim}0.9$ monolayer and the consequent lack of dangling bonds on the silyl-packed surface.

Regulation of Phospholipase D by CoCl2 in Human Glioblastoma Cells (인간 교세포주에서 CoCl2에 의한 phospholipase D의 조절기전)

  • Phospholipase D (PLD) is known to play an important role in a variety of cells. However, little is known about $CoCl_2-mediated$ PLD signaling. In this study we demonstrated for the first time that $CoCl_2$ stimulates PLD activity and increases expression of cyclooxygenase-2 (COX-2), which is known to mediate inflammatory reaction. $CoCl_2-induced$ PLD activity was assessed by measuring the formation of $[^3H]$ phosphatidylbutanol (PtdBut), the product of PLD-mediated transphosphatidylation, in the presence of 1-butanol. To study mechanism of PLD signaling induced by $CoCl_2$, U87 human glioblastoma cells were stimulated by $CoCl_2$ and regulators of PLD activity induced by $CoCl_2$ were investigated using several inhibitors of signaling proteins. Moreover, PLD activation by $CoCl_2$ increased not only expression of COX-2 protein but also COX-2 promoter activity. In summary, these results suggest that $CoCl_2$ increases expression of COX-2 protein via PLD in human U87 glioblastoma cells.

Chromophore formation and phosphorylation analysis of constitutively active rhodopsin mutants (Chromophore 형성과 rhodopsin kinase 활성을 이용한 항활성 로돕신 mutant의 분석)

  • G protein coupled receptors (GPCRs) transmit various extracellular signals into the cells. Upon binding of the ligands, conformational changes in the extracellular and/or transmembrane (TM) domains of CPCRs were propagated into the cytoplasmic (CP) domain of the molecule leading to the activation of their cognate heterotrimeric C proteins and kinases. Constitutively active GPCR mutants causing the activation of C Protein signaling even in the absence of ligand binding are of interest for the study of activation mechanism of GPCRs. Two classes of constitutively active mutations, categorized by their effects on the salt bridge between Ell3 and K296, were found in the TM domain of rhodopsin. Opsin mutants containing combinations of the mutations were constructed to study the conformational changes required for the activation of rhodopsin. Rhodopsin chromophore regenerated with 11-cis-retinal showed a thermal stability inversely correlated with its constitutive activity. In contrast, rhodopsin mutants exhibited a binding affinity to an agonist, all-trans-retinal, in a constitutive activity-dependent manner. In order to test whether the conformational changes responsible for the activation of trans-ducin (Gt) are the same as the conformation required for the recognition of rhodopsin kinase, analysis of the mutants were carried out with phosphorylation by rhodopsin kinase. Rhodopsin mutants containing combinations of different classes of the mutations showed a strong synergistic effect on the phosphorylation of the mutants in the dark as similar to that of Gt activation. The results suggest that at least two or three kinds of segmental and independent conformational changes are required for the activation of rhodopsin and the conformational changes responsible for activating rhodopsin kinase and Gt are similar to each other.

Induction of Apoptosis by Methanol Extract of Endlicheria anomala in Human Lung and Liver Cancer Cells (Endlicheria anomala 메탄올 추출물에 의한 인체 폐암세포주와 간암세포주의 자가사멸 유도)

  • Endlicheria anomala, a neotropical plant, is found in northern South America and the Amazon region. It is traditionally used to remove poisons and cure gangrene. According to recent data, this plant has diverse biological properties such as anti-oxidative, anti-inflammatory and anti-melanogenic properties. However, the anti-cancer effect of E. anomala and its molecular mechanisms remain unclear. In this study, we examined the anti-cancer effect and the active mechanism of methanol extract of E. anomala (MEEA) in human lung adenocarcinoma cells (A549) and human liver cancer cells (HepG2). Our data revealed that MEEA showed cytotoxic activity in a dose-dependent manner and induced apoptosis both in A549 and HepG2 cells. We verified evidences of apoptosis via formation of chromatin condensation, apoptotic body and accumulation of cells in the subG1 phase. Following observed apoptosis-related phenomena, we found that the induction of apoptosis by MEEA was associated with the increase of tumor suppressor p53 and cyclin-dependent kinase inhibitor p21 (WAF1/CIP1) expression. Furthermore, MEEA-induced apoptosis was characterized with proteolytic activation of caspase-3, degradation of poly ADP ribose polymerase (PARP), and up-regulation of pro-apoptotic Bax expression. Taken together, these findings indicate that MEEA may have potential cancer therapeutic utility in A549 and HepG2 cells.

Project-based Organization, Embeddedness and Spatial Clustering in the TV Drama Industry (프로젝트 기반 조직의 배태성과 공간적 군집화에 대한 시론적 연구 -드라마 산업을 사례로-)

  • This paper aims to examine the ways in which project-based organizations (PBOs) are embedded in social networks and geographical clustering in the case of TV drama industry. PBOs refer to a variety of temporary organizational forms for the performance of tasks as to integrate diverse and specialized intellectual resources. PEOs as a flexible and innovative mode of organizing knowledge resources are becoming increasingly worthy of attention in emerging the creative economy. Evidence from interviews with core persons of PEOs like producers, directors, and writers reveals that the key operational mechanism of the project form of organization is based on the highly socialized networks via individual's reputation and past experiences. In other words, the project activity in TV drama is embedded in networks which are socially constructed. Also the geographical clustering plays an important role in PBOs and project practices in TV drama are constructed around a high degree of spatial clustering. PBOs are clustering in Kang-nam and Yeouido, where are located in independent production companies and broadcasting stations. It means that the project formation in TV drama requires geographically-clustered networks of human resources, and socially, culturally and geographically embedded latent networks of interpersonal relationships are a necessary condition of POBs in the TV drama industry.

Determination of HONO Concentration and Investigation on Its Formation Mechanism Using a Continuous Measurement System in Indoor Environment of an Apartment (아파트 실내 환경에서 실시간 측정시스템을 활용한 HONO 농도 측정 및 생성기작 연구)

  • Nitrous acid (HONO) can be produced by heterogenous reactions of nitrogen dioxide on surface materials and direct emission from combustion sources. However, a little is known of indoor HONO levels or the relationship between residential HONO, NO, and $NO_2$ concentrations in occupied houses. Therefore, we measured simultaneously NO, $NO_2$, and HONO concentrations in living room of an apartment using continuous analyzers to study the production of HONO (June $22{\sim}30$, 2006). The 4-min average concentrations of indoor NO, $NO_2$, and HONO were 4.3 (range: $0.4{\sim}214.3$), 10.3 ($2.0{\sim}87.3$), and 1.8 ppb ($0.3{\sim}7.7$), respectively. Peak levels of HONO up to 7.7 ppb and 24-hr averages as high as 1.7 ppb were measured. In agreement with previous studies, indoor HONO concentrations increased during operation of an unvented gas range. Examination of the observed kinetics suggests that the secondary production of indoor HONO, possibly as a result of heterogeneous reactions involving $NO_2$ and $H_2O$ is associated with $[NO_2]^2[H_2O]\;(r^2=0.88)$ rather than with $[NO][NO_2][H_2O]\;(r^2=0.75)$. Three combustion experiments at nighttime were also carried out to investigate the effects of vented combustion on the HONO, NO, and $NO_2$ concentrations. It was found to release HONO for $10{\sim}15$ minutes after NO and $NO_2$ source was turned off, and peak values were finally attained. Compared to unvented combustion, peak $NO_2$ and HONO concentrations were 3.2 and 2.0 times lower at weak vented combustion (air flow: $340\;m^3/hr$) and 4.9 and 2.4 times lower at strong vented combustion (air flow: $540\;m^3/hr$), respectively, emphasizing importance of operating ventilation hood fan during combustion to improve indoor air quality.

Genome-based Gene Expression Analysis of EGCG-mediated Cell Transformation Suppression Effect in Mouse Cell line Balb/c 3T3 A31-1-1 (마우스세포주 Balb/c 3T3 A31-1-1에서 Epigallocatechin gallate(EGCG)의 세포암화 억제효과에 대한 유전자발현 해석)

  • Previous studies showed that epigallocatechin gallate(EGCG) have substantial effects of suppressing the N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)-initiated cell transformation process on the bases of foci formation frequency and loss of anchorage dependency. In this study we tried to clarify the molecular mechanism of suppressing the cell transformation process. Mouse cell line balb/c 3T3 A31-1-1 was exposed 2 days to MNNG followed by 15 days 12-O-tetradecanoylphorbol-13-acetate(TPA) treatment for our transformation process. EGCG was added after the time point of 24 hours exposure to TPA and incubated for 19 days. 2029 genes were selected in our transformation process that showed fold change value of 1.5 or more in the microarray gene expression analysis covering the mouse full genome. These genes were found to be involved mainly in the cell cycle pathway, focal adhesion, adherens junction, TGE-$\beta$ signaling, apoptosis, lysine degradation, insulin signaling, ECM-receptor interaction. Among the genes, we focused on the 631 genes(FC>0.5) reciprocally affected by EGCG treatment. Our study suggest that EGCG down-regulate the gene expressions of up stream signaling factors such as nemo like kinase with MAPK activity and PI3-Kinase, Ras GTPase and down stream factors such as cyclin D1, D2, H, T2, cdk6.

Hydrothermally Synthesis Nanostructure ZnO Thin Film for Photocatalysis Application (수열합성법으로 합성된 산화아연 나노 구조 박막의 광촉매적 응용)

  • ZnO has nanostructured material because of unique properties suitable for various applications. Amongst all chemical and physics methods of synthesis of ZnO nanostructure, the hydrothermal method is attractive for its simplicity and environment friendly condition. Nanostructure ZnO thin films have been successfully synthesized on fluorine doped tin oxide (FTO) substrate using hydrothermal method. A possible growth mechanism of the various nanostructures ZnO is discussed in schematics. The prepared materials were characterized by standard analytical techniques, i.e., X-ray diffraction (XRD) and Field-emission scanning electron microscopy (SEM). The XRD study showed that the obtained ZnO nanostructure thin films are in crystalline nature with hexagonal wurtzite phase. The SEM image shows substrate surface covered with nanostructure ZnO nanrod. The UV-vis absorption spectrum of the synthesized nanostructure ZnO shows a strong excitonic absorption band at 365 nm which indicate formation nanostructure ZnO thin film. Photoluminescence spectra illustrated two emission peaks, with the first one at 424 nm due to the band edge emission of ZnO and the second broad peak centered around 500 nm possibly due to oxygen vacancies in nanostructure ZnO. The Raman measurements peaks observed at $325cm^{-1}$, $418cm^{-1}$, $518cm^{-1}$ and $584cm^{-1}$ indicated that nanostrusture ZnO thin film is high crystalline quality. We trust that nanostructure ZnO material can be effectively will be used as a highly active and stable phtocatalysis application.

An Intramural Electronic Voting System Based on Blockchain (블록체인 기반 교내 전자투표 시스템)

  • As security problems of the paper ballot have been emerged on and on, electronic voting with enhanced security and convenience has been introduced in several countries. However, it has not been adopted most of countries because of the problems that come from interdependence and security flaws. Meanwhile, the blockchain technology has high reliability due to the mechanism of mining that miners verify and preserve blocks independently by using P2P formation which does not have a central authority. Furthermore, because each block refers to the hash of the previous block. if any one block is changed, it is very difficult to forge and modify the blockchain because all blocks must be changed. If this technology is applied to the E-voting, integrity, and transparency about the result of the ballot is guaranteed. In this paper, we propose and implement an electronic voting system based on blockchain that improves interdependence, the reliability of excessive TTP and single point of failure come from original electronic voting. Also, we analyze the security and advantage of the proposal system compared with the existing bitcoin-based electronic voting system.
