Formation of Brain Tumor and Lymphoma by Deregulation of Apoptosis Related Gene Expression in VP-SV40 T Ag Transgenic Mice

  • The neuropeptide vasopressin (VP) is a nine- amino acid hormone synthesized as preprohormone in the cell bodies of hypothalamic magnocellular neurons. The tumor in magnocellular neurons of the hypothalamus is associated with disfunctions of the cell bodies, leading to the diabetes insipidus. In order to study with the diabetes insipidus caused by a defect in VP synthesis and its secretion, we have produced the transgenic mice regulated by vasopressin promoter inserted to SV40 T antigen coding sequence (pVPSV.IGR2.1). One transgenic line expressing high levels of SV40 T antigen was propagated. The founder and all transgene positive adult animals have appeared with shorten mortality or apparent phenotypic abnormalities, including immune complex disease, and eventually die between 4 and 8 months of age. The mRNA and protein of SV40T antigen transgene were detected in brain of fetus as well as in brain, spleen, lung and lymph node in moribund at the age of 20 weeks. Histological analysis of transgenic mice showed that tumor developed in brain similar to primitive neuroectodermal tumors (PNET) in man. We also detected lymphomas in spleen and lymph node, and consequent tumor formation in various tissues of the transgenic mice. In pVPSV.IGR2.1, 21% mice showed brain tumor (PNET) at 5 weeks and 100% mice showed brain tumor after 15 weeks. In addition, Expression of apoptosis related genes (Bcl-28 & Bax) was increased over their age in mice with PNET as compared to control mice. Apoptosis related gene expression might be deregulated in mice with brain tumor. However, transgenic mice were not developed with the diabetes insipidus. These mice represent the first disease model to exhibit primitive neuroectodermal tumor in brain, as well as a unique model system for exploring the cellular pathogenesis of lymphomas.

신두리 지역의 고사구(古砂丘)에 대한 OSL 연대 측정 (A Luminescence Dating for a Relict Dune from the Sindu Dunefield)

  • 태안반도 신두 해안사구지대의 중앙부에서 고사구층으로 판단되는 퇴적물의 퇴적시기를 파악하기 위해 OSL 연대측정을 실시하였다. 조사 지점 최하부에서의 사구사 퇴적은 대략 680$\sim$730년 전으로, 1.5m 깊이에서는 480$\sim$610년 전으로 나타났다. 반면 1.3m부근의 암갈색 퇴적층을 사이에 두고 1.2m 깊이 퇴적층에서의 산출연대는 123$\sim$151년 전으로, 최상부 퇴적물의 연대는 약 68년인 것으로 나타났다. 한편, 사구 표면으로부터 1.5m와 3m 깊이에서 채취한 두개의 시료는 200년 정도의 연대 차이가 있다. 이에 의하면 사구사의 집적율은 연간 약 0.75cm로 나타났다. 본 조사에서 연대를 추정한 사구사 퇴적층은 1개 지점에 불과하지만, 이를 토대로 볼 때, 지난 1,000년간 상당한 양의 사구사가 퇴적되었거나 재이동되었고, 적어도 한번의 토양화과정이 진행되었음을 알 수 있다. 이러한 사실은 지금까지 플라이스토세나 홀로세 초기로 알려진 고사구층의 형성 연대에 대한 연구가 다시 시도되어야 함을 시사한다.

극소 저출생 체중아에서 조성한 장루의 복원 경험 (Experience with Enterostomy Closure in Very Low Birth Weight Infants)

  • The survival of Very Low Birth Weight (VLBW) infants has been improved with the advancement of neonatal intensive care. However, the incidence of accompanying gastrointestinal complications such as necrotizing enterocolitis has also been increasing. In intestinal perforation of the newborn, enterostomy with or without intestinal resection is a common practice, but there is no clear indication when to close the enterostomy. To determine the proper timing of enterostomy closure, the medical records of 12 VLBW infants who underwent enterostomy due to intestinal perforation between Jan. 2004 and Jul. 2007 were reviewed retrospectively. Enterostomy was closed when patients were weaned from ventilator, incubator-out and gaining adequate body weight. Pre-operative distal loop contrast radiographs were obtained to confirm the distal passage and complete removal of the contrast media within 24-hours. Until patients reached oral intake, all patients received central-alimentation. The mean gestational age of patients was $26^{+2}$ wks ($24^{+1}{\sim}33^{+0}$ wks) and the mean birth weight was 827 g (490~1450 g). The mean age and the mean body weight at the time of enterostomy formation were 15days (6~38 days) and 888 g (590~1870 g). The mean body weight gain was 18 g/day (14~25 g/day) with enterostomy. Enterostomy closure was performed on the average of 90days (30~123 days) after enterostomy formation. The mean age and the mean body weight were 105 days (43~136 days) and 2487 g (2290~2970 g) at the time of enterostomy closure. The mean body weight gain was 22 g/day after enterostomy closure. Major complications were not observed. In conclusion, the growth in VLBW infants having enterostomy was possible while supporting nutrition with central-alimentation and the enterostomy can be closed safely when the patient's body weights is more than 2.3 kg.

울릉도 및 독도 근해의 불볼락(Sebastes thompsoni)의 연령과 성장 (Age and Growth of the Goldeyes Rockfish Sebasetes thompsoni in the Ulleungdo and Dokdo Areas)

  • We studied the characteristics of age and growth of the Goldeyes rockfish Sebasetes thompsoni in the Ulleungdo and Dokdo areas by using 730 otoliths from February 2013 to February 2014. The lowest value of the marginal index was found in June once a year. The spawning period was estimated as April. Therefore, the duration from fertilization to the complete formation of the first annual growth ring is fourteen months. From the parameters calculated using the weighted average length of the year ring formation, the growth of S. thompsoni was demonstrated by the von Bertalanffy growth equation as $L_t=34.68(1-e^{-0.2113(t+0.7683)})$ for females and $L_t=29.79(1-e^{-0.2163(t+1.7964)})$ for males, $L_t=32.84(1-e^{-0.25(t+0.57)})$ for combined sex. Comparing with growth performance index(${\Phi}$) from these 4 offshore areas, parameter of S. thompsoni in Ulleungdo area was 2.4087, that of in Tong-yeong was 2.2835, that of in Aomori was 2.4077 and in Niigata was 2.3927. As a result, the value of Dokdo and Ulleungdo areas showed the highest.

한국 남서부의 열수점토광상과 주변암에 대한 K-Ar 연대 측정 (K-Ar ages of the hydrothermal clay deposits and the surrounding igneous rocks in southwest Korea)

  • 한국 남서부의 점토광상들과 주변암에 대한 K-Ar 연대측정 결과, 점토광물이 75.1∼81.4 Ma, 백악기 화산암류(황산층)가 81.4∼86.4 Ma, 백악기 화강암류는 77.1∼81.5 Ma의 연대로 나타났다. 이 결과는 점토광상들이 백악기 후기의 산성마그마티즘과 성인적으로 밀접한 관계가 있음을 지시한다. 월각산 화강암은 K-Ar 흑운모의 연령이 140.g∼144.8 Ma로 밝혀져, 이의 관입연대는 쥬라기로 판명되었다. 이곳의 점토광상들과 백악기 화강암류의 밀접한 관계는 점토광상들이 화산활동에 수반된 열수작용에 의한 것이 아니라 화강암류의 관입에 따른 열적 효과(열수 순환)에 기인된 열수변질작용으로 형성되었다.

성인의 평생학습 참여동기와 만족도 및 학습성과가 사회적자본 형성에 미치는 영향 (Effects of Adults' Participation Motives for Lifelong learning, and their Satisfaction and Learning Outcomes on Social Capital Formation)

  • 본 연구에서는 성인 평생학습 참여동기와 만족도 및 학습성과가 사회적자본 형성에 미치는 영향 분석을 위해 D시의 평생교육기관 및 시설에서 평생교육참여 경험이 있는 만 20세 이상의 성인학습자 354명을 대상으로 회귀분석, 중다회귀분석 및 경로분석을 실시하였다. 연구결과, 성인의 평생학습 참여동기는 사회적자본 형성에 유의한 영향을 미치는 것을 알 수 있었으며, 만족도와 학습성과에 유의한 영향을 미치는 것을 알 수 있었다. 또한 성인의 만족도와 학습성과는 성인의 사회적자본 형성에 유의한 영향을 미치는 것을 알 수 있었으며, 성인의 평생학습 참여동기와 사회적자본 형성 사이에서 유의한 매개효과가 있음을 확인할 수 있었다. 본 연구결과는 평생교육의 이론적 실천적 함의로서 평생교육 발전에 많은 시사점을 제공할 것이다.

Differential Expression of Adipocyte-Related Molecules in the Distal Epididymal Fat of Mouse during Postnatal Period

  • The epididymal fat of mouse is a part of visceral fat deposit and is divided into the distal or proximal part based on its histochemical characteristics. Even though the formation of the epididymal fat pad begins before the birth, a detailed adipogenic procedure of the epididymal fat has not been revealed. The epididymal fat pad becomes enlarged and expended with age, and expressional changes of numerous genes are associated with the maturation of fat tissues. In the present research, expressional patterns of adipose tissue-related genes in the distal epididymal fat of mouse at 2, 5, 8, and 12 months of postnatal age were determined by a quantitative real-time polymerase chain reaction (PCR) analysis. The lowest transcript levels of fatty acid binding protein 4 (Fabp4), lipoprotein lipase (Lpl), delta like non-canonical Notch ligand 1 (Dlk1), peroxisome proliferator-activated receptor gamma (Pparg), leptin (Lep), adiponectin (Adipoq), and resistin (Retn) were detected at 2 months of age, except fatty acid synthase (Fasn) showing the lowest level at 5 months of age. Even though expression of Lep and Fabp4 were gradually increased until 12 months of age, significant increases of Pparg and Adipoq transcript levels were continued until 8 months of age. The transcript levels of Lpl, Rent, Dlk1, and Fasn were significantly increased at 8 months of age, compared with those at 2 months of age. The current findings suggest that the expansion of the distal epididymal fat of mouse during postnatal period would be companied with differential expression of various adipocyte-associated molecules.

충주지역 북서부 옥천변성대의 층서 및 지질구조 (Stratigraphy and Geological Structure of the Northwestern Okcheon Metamorphic Belt Near the Chungju Area)

  • 충주지역 북서부 옥천변성대는 문주리층, 대향산규암, 향산리돌로마이트, 계명산층이 분포하고 있으며 현재까지 층서적 관계가 논란이 되고 있는 지역이다. 충주지역에 분포하는 북서부 옥천변성대에 대한 층서적 연구를 위해 충주호일대에 두 지점의 노두를 선정하여 단면에 대한 정밀기재 및 인근지역에 대한 야외조사를 실시하였다 문주리층과 대향산규암의 경계는 정이적인 변화가 인지되나 경계부는 층리면과 평행한 단층이 발달한다 대향산규암과 향산리돌로마이트는 정합적인 것으로 판단된다. 향산리돌로마이트와 계명산층은 단층을 경계로 접하고 있다. 연구지역의 북부에는 $41^{\circ}/280^{\circ}$의 습곡축을 가지는 습곡이 분포하는 것이 확인되었으며, 두 층의 경계에 발달하는 단층은 충상단층일 가능성도 있으나 습곡이 형성되는 동안 나타나는 단층일 가능성도 있다. 본 연구에서 확인된 점이적인 암상변화와 기존 연구 결과를 볼 때 경계부의 단층은 습곡이 형성되는 동안 층리면과 평행하게 발달하는 단층으로 판단된다 향산리돌로마이트와 계명산층의 경계부에 발달하는 단층은 층리면과 평행하게 발달하며, 돌로마이트가 반복적으로 나타나고 있는 점을 볼 때 충상단층으로 판단된다. 두 층이 충상단층 관계일 경우 상부의 계명산층이 문주리층 아래에 놓이게 된다. 하지만 계명산층과 문주리층은 비슷한 암상분포를 보이며, 신원생대의 절대연령을 나타내는 점을 볼 때 두 층이 동일한 층일 가능성이 있을 것으로 생각되며 충상단층에 의해 반복적으로 분포하는 것으로 판단된다. 이러한 결과를 종합해 볼 때 충주지역 북서부 옥천변성대의 층서는 하부로부터 계명산층/문주리층, 대향산규암, 향산리돌로마이트의 정합적 관계로 판단된다.

Comparison of Ammonia-Oxidizing Bacterial Community Structure in Membrane-Assisted Bioreactors Using PCR-DGGE and FISH

  • The ammonia-oxidizing bacterial (AOB) communities in three membrane bioreactors (MBRs) were monitored for 2 months after an acclimation period in order to investigate the influence of sludge age and medium type on AOB changeability and its connection with nitrification effectiveness. One MBR with a sludge age of 4 days was fed with a synthetic medium, whereas the other two with sludge ages of 8 and 32 days were fed with landfill leachate. The research revealed that landfill leachate can be effectively treated in an MBR with a higher sludge age for longer periods of time and that this improvement in performance was correlated with an increase in AOB biodiversity. Interestingly, the medium type has a stronger influence on AOB biocenosis formation than the sludge age.

철원 및 연천 지역 용암대지 현무암의 형성 시기 및 형성 과정 (Formative Age and Process on Basalt of Lava Plateau in the Cheolwon and Yeoncheon Areas, Central Korea)

  • The Cheolwon-Pyeonggang Lava Plateau on the Chugaryeong tectonic valley is one of the most extensive volcanic areas in central Korea. However, formative age and process of the plateau still remains a controversial issue. This study presented OSL ages on the upper and lower sedimentary layers of basalt from four sites in the Cheolwon and Yeoncheon areas and estimated age and process of plateau formation based on sedimentary- and chrono-stratigraphy and topographic analysis. The results suggested that most of the initial topography of the plateau on the Cheolwon and Yeoncheon areas had been almost completed before approximately 90 ka. However, the last lava flow around Jangheung-ri, Cheolwon, seemed to occur until 20-30 ka and had led to complete the present plateau, while the last lava flow in the Jeongok area, Yeoncheon, was estimated to occur at approximately 40 ka.
