The Effects of Korean Traditional Medicine Mixture on Cytokine Level, Food Intake and Nutrition Metabolism of the Cachexia Induced-Mice (혼합한약재가 악액질이 유도된 생쥐의 Cytokine분비 및 식이섭취와 영양대사에 미치는 영향)

  • Cancer cachexia, characterized by weight loss and progressive tissue wasting, has been postulated to be mediated by cytokines. This study was conducted to evaluate the effect of Korean Traditional Medicine (KTM ; mokhyang, jisil, osooyu) mixture on food intake, blood cytokines level and blood nutrients status of the cachexia induced-mice. Thirty male Balb/c mice aged 6-8 weets were blocked into 3 groups that were Normal (no colon26 cells) Control (colon 26 cells) and KTM (colon26 cells + KTM extract mixture) group. In Control and KTM groups, murine adenocarcinoma colon 26 cells were injected subcutaneously to induce cachexia. KTM mice were given 200 ul KTM extract mixture (7%) per day. Half of each groups were sacrificed at the 14 th day to see serum cytokines & nutrients and the others were fed until almost of control group died to see life span. food intake and body weight were decreased significantly in cachexia induced groups. Tumor weight of KTM group was significantly lower than control group. Serum cytokines (IL-1$\beta$ and TNF-$\alpha$) level of cachexia induced groups were increased than those of normal group, and those of KTM group were significantly lower than the level of control group. Total serum protein and serum albumin were higher in KTM group than other groups. TG and fatty acid were lower in cachexia induced groups than normal group. HDL-cholesterol in serum was increased in KTM group. Effect of oral administration of KTM extract mixture on survival time of colon26 bearing mice showed extension of the life span. Overall, this study showed that KTM (mokhyang, jisil, osooyu) extract mixture inhibited the growth of cancer cell, changed the secretion of cytokines induced by colon26 adenocarcinoma in mice, and changed nutrition metabolism.

Optimization of Makgeolli Manufacture Using Several Sweet Potatoes (다양한 고구마를 이용하여 제조한 막걸리의 최적화)

  • The objective of this study was to manufacture three kinds of domestic sweet potato Makgeolli using a mixture design and an optimization technique. The effects of four different manufacture methods, such as simultaneous saccharification and fermentation (SSF) with or without malt and separate hydrolysis and fermentation (SHF) with or without malt were determined. The SSF methods of Makgeolli produced higher alcohol content than that of SHF methods. The sensory score was not influenced by different making methods. Fourteen experimental points were selected, and rice (10~50%), sweet potato (10~50%) and water (40~60%) were chosen as independent variables. The measured responses were sensory preference, total polyphenol content, and DPPH radical scavenging activities. The ratio of the optimum sweet potato Makgeolli mixture formulation was developed as 15.11 (rice): 44.89 (sweet potato): 40 (water) using the optimization technique. The desirability of the optimum mixture formulation was 0.839. Yellow sweet potato Makgeolli using the optimum mixture formulation produced higher soluble sugar content compared to others. Regular sweet potato Makgeolli produced higher pH. The purple sweet potato Makgeolli's total polyphenol content and DPPH radical scavenging activity were measured to be the highest at $771.91{\pm}1.42mg\;GAE/{\ell}$, $131.55{\pm}4.03%$.

Food waste treatment using Bacillus species isolated from food wastes and production of air-dried Bacillus cell starters

  • The objectives of this research were to 1) isolate and identify thermophilic bacteria for food waste treatment; 2) investigate the capability of food waste treatment using Bacillus species; and 3) develop air-dried Bacillus starters for food waste treatment. Five Bacillus species were isolated from food wastes and identified as Bacillus licheniformis (B. licheniformis) G1, Bacillus circulans C2, Bacillus subtilis (B. subtilis) E1, Bacillus vanillea F1, and Bacillus atrophaeus G2 based on 16S rDNA sequencing. Each identified Bacillus and the mixture of Bacillus species were cultivated in the standard food waste at $45^{\circ}C$ for 8 d. Changes in cell count, solid contents, and pH of the food waste were monitored during cultivation. Air-dried Bacillus cell powders were prepared using wheat flour and lactomil as excipients, and the cell count and survival rate were determined. The cell count of B. licheniformis G1 exhibited the highest number among the tested Bacillus (${\sim}10^8CFU/mL$). The greatest reduction in solid contents of food waste was achieved by B. subtilis E1 (22.6%). The mixture of B. licheniformis G1 and B. subtilis E1 exhibited a synergistic effect on the reduction of solid contents. Lactomil was determined as better excipient than wheat flour based on the greatest survival rate of 95%.

Long Term Feeding with Soy Isoflavone and L-Carnitine Synergistically Suppresses Body Weight Gain and Adiposity in High-Fat Diet Induced Obese Mice

  • Objective: We investigated the efficacy of a 12-week supplementation of soy isoflavone with L-carnitine on the development of obesity in high fat-induced obese C57BL/6J mice, which are known as a good model of diet-induced obesity. Methods: We measured body weights, adipose tissue mass, serum/liver lipid profiles and fat cell size/number in C57BL/6J mice fed diets containing either low fat (4%) or high fat (35%), or high fat supplemented with soy isoflavone powder containing 10% isoflavone and L-camitine for 12 weeks. Results: Body weight gain, abdominal adipose tissue and liver weight were lower by 31% 78% and 31.4% respectively, in mice on high fat diet containing soy isoflavone+L-carnitine (SC mixture) compared with high fat diet group. Also, SC mixture improved serum lipid profiles such as total cholesterol (TC), triglycerides (TG), and liver lipid profiles such as total lipids and TG. As subsequent results, this SC mixture prevented high-fat diet from accumulating TG in the liver. The size of fat cell was also significantly decreased in SC mixture fed mice. At the end point of this experiment, our results showed that feeding with soy isoflavone for 12 weeks finally increased camitine palmitoyltransferase 1 (CPT 1) activity through elevating the level of CPT1 expression. Conclusions: This study suggests that long-tenn supplementation with dietary soy isoflavone and L-carnitine is more synergistically beneficial for the suppression of high-fat diet induced obesity by inhibiting liver TG accumulation and the gain in abdominal adipose tissue weight than that with soy isoflavone. The antiobesity effects of SC mixture might be attributed, at least in part, to the induction of fatty acid catabolism by soy isoflavone, genistein.

  • 염분농도에 따른 발아특성은 쪽제비싸리, Perennial ryegrass의 경우 $0.4\%$ 이상 NaCl 농도에서 발아율과 유아 및 유근 길이가 급격히 감소하였으며, Creeping red fescue는 $0.2\%$ 이상 NaCl 농도에서 매우 급격한 생육저하가 일어났다. 외래초종의 초장 분석결과 Mixture III> Mixture IV> Mixture II> Mixture I의 순이었고, 재래 초$\cdot$목본의 경우는 외래초종의 초장이 길수록 길이가 짧아지는 경향을 나타냈으며, Mixture I> Mixture II> Mixture IV> Mixture III의 순이었다.

Effects of Fermented Extracts Zizyphus jujuba Fruits and Leaves Mixture on Hyperlipidemia in Rats Induced by High Fat Diet (대추과육과 대추잎 혼합발효추출물이 고지방식이로 유발된 고지혈증 흰쥐에 미치는 영향)

  • Objective : This study was designed to investigate the effects of non fermented Zizyphus. jujuba mixture (mixed fruit and leaf : ZM) and fermented Z. jujuba mixture(FZM) on fed high-fat diet induced hyperlipidemic rats for development medicinal food. Method : The extracts prepared for Zizyphus. jujuba mixture(ZM) and fermented Z. jujuba mixture(FZM) with Lactobacillus plantarum and Saccharomyces cerevisiae. Experimental group was divided into normal control group(NC group) and high fat diet groups. The high fat diet groups fed high fat diet 2 weeks after was sub-divided into high fat diet control group(HC group), high fat diet positive control group(HPC group), non fermented Z. jujuba mixture extract group(ZM group) and fermented Z. jujuba mixture extract group(FZM group). NC and HC group was orally administerd of 0.9% saline, HPC group administerd lovastatin diluted in 0.9% saline at a dose of 10 mg/kg BW, ZM and FZM groups was administerd each extracts diluted in 0.9% saline at a dose of 300 mg/kg BW once a day on a fixed time for 4 weeks. In the present study we measured organ weight, epididymal fat tissue weight, concentration of serum lipids, hepatic lipids, MDA contents in liver tissue and metabolic variables in serum. Results : ZM and FZM suppressed testis weight loss and FZM decreased epididymal fat tissue weight to level of NC group in high-fat diets. ZM and FZM did not influence on serum cholesterol level, but prominently decreased serum triglyceride concentration compared with HC group, and FZM diminished hepatic triglyceride same as serum. ZM and FZM did not impair liver and kidney function and influence positive effects through by suppression of elevation lipid level. Conclusion : These results suggested that Z. jujuba mixture(ZM, FZM) should be useful developing medicinal food for prevention and improvement of hyperlipidemia and FZM is more suitable agent than ZM.

Effect of Liquid Smoke and Curing Mixture on Quality Characteristics of Chuncheon Dakgalbi during Storage (춘천닭갈비에 훈연액 및 염지제 첨가가 저장 중 품질특성에 미치는 영향)

  • The goal of the present work was to determine the optimal addition amounts of liquid smoke and curing mixture to develop Chuncheon Dakgalbi with improved preference and shelf-life. In the first experiment, Chuncheon Dakgalbi was prepared with different amounts of liquid smoke. In the second experiment Chuncheon Dakgalbi was prepared with various amounts of curing mixture and 0.1% (w/w) liquid smoke. Different amounts of liquid smoke resulted in different aroma patterns, which were observed using an electronic nose, and Dakgalbi with 0.1% (w/w) liquid smoke had the highest score in overall acceptability. The addition of liquid smoke and curing mixture inhibited the growth of bacteria, slowed down the decline in pH, and delayed increased in volatile basic nitrogen contents and lipid oxidation. However, no clear effects were observed on instrumental color. From a consumer preference test, the highest preference score was achieved by added 0.2% (w/w) curing mixture. Liquid smoke and curing mixture extended shelf-life and improved preference of Chuncheon Dakgalbi. Considering the physicochemical, microbiological and consumer preference, it was recommended to add 0.1% (w/w) liquid smoke and 0.2% (w/w) curing mixture to Chuncheon Dakgalbi to enhance shelf-life and preference.

Preventive Effects of a Probiotic Mixture in an Ovalbumin-Induced Food Allergy Model

  • Although there has been a steady increase in the prevalence of food allergies worldwide in recent decades, no effective therapeutic strategies have been developed. Modulation of the gut microbiota composition and/or function through probiotics has been highlighted as a promising target for protection against food allergies. In this study, we aimed to investigate the allergy-reducing effects of a probiotic mixture (P5: Lactococcus lactis KF140, Pediococcus pentosaceus KF159, Lactobacillus pentosus KF340, Lactobacillus paracasei 698, and Bacillus amyloliquefaciens 26N) in mice with ovalbumin (OVA)-induced food allergy. Administration of P5 significantly suppressed the oral OVA challenge-induced anaphylactic response and rectal temperature decline, and reduced diarrhea symptoms. Moreover, P5 also significantly inhibited the secretion of IgE, Th2 cytokines (interleukin (IL)-4, IL-5, IL-10, and IL-13), and Th17 cytokines (IL-17), which were increased in mice with OVA-induced food allergy, and induced generation of CD4+Foxp3+ regulatory T cells. These results revealed that P5 may have applications as a preventive agent against food allergy.

  • This study was designed to evaluate the nutritive value of pearl barley. Forty males and the same number of females of albino rats, the Leuise strain, weighing from 52.1g to 59.1g were divided into eight experimental groups, five males and females were composed of one group. A group: casein 15%+starch 75%+oil 8%+vitamin mixture 1%+salt mixture 1% B group: pearl barley 98%+vitamin mixture 1%+salt mixture 1% C group: rice 98%+vitamin mixture 1%+salt mixture 1% D group: barley 98%+vitamin mixture 1%+salt mixture 1% E group: rice 49%+pearl barley 49%+vitamin mixture 1%+salt mixture 1% F group: rice 73.5%+pearl barley 24.5%+vitamin mixture 1% +salt mixture 1% G group: barley 49%+pearl barley 49%+vitamin mixture 1% +salt mixture 1% H group: barley 73.5%+Pearl barley 24.5%+vitamin mixture 1%+salt mixture 1% The rats were kept in an individual cage and were fed 8 different diets for 7 weeks and fed by ad-libitum feeding method. The results of this study were eluciated as followings. 1. B group was significantly lower in weight gain than the other groups. 2. Polished pearl barley showed higher digestion rate in protein and carbohydrate than brown pearl barley but lower in fat. 3. Liver weight was obserbed significant differences between B group and C group. but no significant differences between the others. 4. B group was significantly lower in the total protein content of liver and A/G ratio in plasma than other groups. 5. B group was also the lowest in the hematocrit and hemoglobin contents among the experimental groups. 6. Free amino acid composition in plasma was similar to those in diet.

Separation of Lactoferrin from Model Whey Protein Mixture by Reverse Micelles Formed by Cationic Surfactant

  • The selective extraction behavior of lactoferrin (Lf) from whey protein mixture was examined using reverse micelles formed by the cationic surfactant, cetyldimethylammonium bromide (CDAB). The major whey proteins, including ${\beta}$-lactoglobulin, ${\alpha}$-lactalbumin and bovine serum albumin, were solubilized from aqueous phase to organic phase while Lf was recovered in the aqueous phase. The solubilization behaviors of the proteins were manipulated by the process parameters such as the pH and salt concentration of the aqueous phase and the surfactant concentration in the organic phase. Efficient forward extraction was achieved with sodium borate buffer (50 mM, pH 9) containing 50 mM KCl and organic phase containing 100 mM CDAB. Based on SDS-PAGE and densitometry, about 96% of the initial Lf remained in the aqueous phase after forward extraction. The dialyzed Lf fully maintained its bacteriostatic activity against E. coli O157:H7.
