Distillation and Quality Characteristics of Medicinal Herb Wines (약용주의 증류와 품질특성)

  • Korean general medicinal herbs-sasam, gilkyung, jakyak, danggwi, hwangki, and chunkung-were added In the normal brewing procedure as a raw material or in the distilling procedure as a packing material. The distillates from the former procedure and those from the latter procedure were compared in quality and distillation properties. As distillation proceeded, pH of the medicinal herb wine distillate and the control(not added herbs) distillates were decreased, whereas that of the herb packing distillate was increased slowely of $0.05{\sim}0.97$ during $1{\sim}4$ fractions and decreased remarkably of $0.92{\sim}0.98$ afterward. Average pH was the highest of 5.70 in jakyak and lowest of 4.37 In gilkyung. Absorbances of the herb Packing distillate were decreased rapidly of $0.60{\sim}1.59$ in the $1{\sim}4$ fractions but slowely of $0.19{\sim}0.54$ in the next fractions. During distillation both fractional alcohol concentration of the distillates and distillation rate were decreased. Their values were decreased more slowly than the control. Distillation rates of medicinal herb wine distillate were varied by medicinal herb varieties and alcohol concentration of fermented wine. Danggwi and control showed the highest average distillation rate as $0.12\;m{\ell}/sec$ and gilkyung the lowest value as $0.073\;m{\ell}/sec$. Maximum concentration of index component, paeoniflorin of jakyak was observed as 293 mg% in the 5th fraction of herb packing distillate and decrusin of danggwi as 3514 mg% In the 1st fraction of herb packing distillate. The extraction rate was 41.3% for paeoniflorin and 20.5% for decrusin. From sensory evaluation, the highest overall Qualify was observed in the medicinal herb wine distillate of hwangki added wine, the next in those of danggwi and jakyak added wine.

Optimization of Culture Conditions for Xylitol Production by A Mutant of Candida parapsilosis (Candida parapsilosis 돌연변이주에 의한 Xylitol 생산조건의 최적화)

  • Effect of culture conditions such as pH, temperature, agitation speed and oxygen transfer rate on xylitol production from xylose by Candide parapsilosis ATCC 21019 mutant was investigated in a jar fermentor. The initial concentration of xylosr was fixed at 50 g/l in this experiment. When pH was increased, cell growth and xylose consumption rate were increased, but maximum xylitol production was shown in the range of pH 4.5 and 5.5 with a yield of 0.68 g/g-xylose. The optimal temperature for xylitol production was determined to be $30^{\circ}C$. Considering the importance of dissolved oxygen tension, for xylitol production, the effect of oxygen transfer rate coefficient $(k_La)$ on fermentation parameters was carefully evaluated in the range of $20{\sim}85\;hr{-1}\;of\;k_La$ (corresponding to $100{\sim}300$rpm of agitation speed). The xylitol production was maximized at $30\;hr^{-1}\;of\;k_La$(150 rpm). A higher oxygen transfer rate supported better cell growth with lower xylitol yield. It was determined that maximum xylitol concentration, xylitol yield and productivity was 35.8 g/l, 71.6% and $0.58\;g/l{\sim}hr^{-1}$, respectively, at $30\;hr^{-1}\;of\;k_La$ In order to further increase xylitol productivity, ferementation using the concentrated biomass(20 g/l) was carried out at the conditions of pH 4.5, $30^{\circ}C$ and $30\;hr\;1$ of oxygen transfer rate. The final xylitol concentration of 40 g/l was obtained at 18 hours of culture time. From this result, it was calculated that xylitol yield was 80ft on the basis of xylose consumption and volumetric productivity was $2.22\;g/l{\sim}hr$ which was increased by $3{\sim}4$ fold compared with $0.5{\sim}0.7\;g/l-hr$ obtained in a normal fermentation condition.

Lipid Composition of Improved Rape Seed on the Content of Erucic acid (에루신산에 대(對)한 개량(改良) 유채종자(油菜種子)의 지질구성(脂質構成) 성분(成分))

  • In order to analyze the lipid composition ana chemical characteristics of rapeseed oil in relation to content of erucic acid, three recommendable cultivars(MOK-PO 70, MOK-PO DANGYO 12 and 14) seeds were used. The composition of lipid was confirmed by column chromatography and TLC, and the composition of fatty acid in total and neutral lipid was determined by GLC, and the results were as follows. 1. The crude fat was 44.0% to 45.3% in all cultivars. 2. The iodine value, saponification value, acid value and nonsaponifiable content were 100 to 101, 176 to 184, 0.7 to 0.9 and 1.0 to 1.2, respectively, with no remarkable difference among three cultivars. 3. The content of neutral lipid was 95.3% to 96.3% of total lipid, and others were compound lipids. 4. The content to triglyceride was 92. 1% to 92.5% of total lipid. Diglyceride and sterol ester were contained 1.1% and 1.2%, respectively. There was a small quantity of free fatty acid and free sterol and monoglyceride was little. 5. The erucic acid was the highest (26.4%), in MOK-PO DANGYO 12 among the composition of fatty acid, while those of MOK-PO 70 and MOK-PO DANGYO 14 were so small as 1.0% and 3.9%. The content of erucic acid was decreased with the increase of oleic acid, the content of oleic acid in MOK-PO DANGYO 12 was 30%, 63% in MOK-PO 70 and, 60% in MOK-PO DANGYO 14. The content of linoleic acid and linolenic acid showed no remarkable difference but that of eicosenoic acid was 11.7% in MOK-PO DANGYO 12 as the highest. The ratio of the fatty acid content in eluted neutral lipid and the total oil showed no remarkable differences.

  • The anti-obese, hypolipidemic and hepatoprotective effects of post-fermented green tea by Monascus pilosus was tested with mice fed with high-fat diet for 7 weeks. The body weight gain and feed efficiency ratio (FER) in normal control group (NC), CHA (2% non-fermented green tea powder supplemented high-fat diet group) and mCHA (2% green tea powder post-fermented by M. pilosus supplemented high fat diet group) groups were significantly lower than those of high fat diet control group (HC). Epididymal fat weight in mCHA and NC were significantly lower than HC. The hepatic lipid peroxide was dramatically higher in HC than that of NC and was significantly lower in CHA and mCHA. In addition, dehydrogenase type activity of xanthine oxidoreductase in HC was lower than that of NC, but significantly higher than CHA and mCHA. In histopathological findings, hepatic fat accumulation in HC was higher than that of NC, CHA and mCHA. Antiobese, hypolipidemic and antifatty liver effect of green tea powder post-fermented by M. pilosus was slightly higher than that of non-fermented green tea. In conclusion, the constituents of green tea fermented by M. pilosus has been proven to not only inhibit obesity and hyperlipidemia but also decrease the hepatic fat accumulation in high fat diet-induced obese mice.

A Study on the Cosmeceutical activities of Prunus Sargentii R. (산벚나무(Prunus sargentii R.) 수피의 화장품활성에 관한 연구)

  • Prunus sargentii R. of Rosaceae familiy, has been reported to have radical scavenging activity and anti-inflammatory effect. On these facts, biological activity and safety test were conducted to evaluate biological activities of the extracts of P. sargentii R. as a potential pharmaceutical ingredient. The electron donating ability of its ethanol extracts at a 500 ppm level showed 92%, which was higher than that of hot water extract (59%), the superoxide dismutase (SOD)-like activity of the water extract of P. sargentii R. was about 50%, the ethanol extract of P. sargentii R. was about 40% at 1,000 ppm concentration. Xanthine oxidase inhibition by the water extract of P. sargentii R. was about 40% and that by the ethanol extract was 60% respectively at 500 ppm concentration. From the measurement on lipid oxidation, the $Cu^{2+}$ chelating effect of the ethanol extract was higher than that of hot water extract. The $Fe^{2+}$ chelating effect was also shown to be about 80% at a 500 ppm concentration in both hot water extract and ethanol extract. The tyrosinase inhibition effect related to skin-whitening was 26% by hot water extract and 20% by ethanol extract respectively at a 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 96% in ethanolic extract at a 500 ppm. Clear zones formed by P. sargentii R. against the human skin-resident micro-flora such as Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli and Propionibacterium acnes indicated that antimicrobial activity of the ethanol extract was higher than that of the hot water extract.

Prediction of Evapotranspiration from Grape Vines in Suwon with the FAO Penman-Monteith Equation (FAO Penman-Monteith 공식을 이용한 수원지역 포도 수체 증발산량 예측)

  • Food and Agricultural Organization (FAO) Penman-Monteith (PM) equation is one of the most widely used equations for predicting evapotranspiration (ET) of crops. The ET rate and the base crop coefficients ($K_{cb}$) of the two different grape vines (i.e., Campbell Early and Kyoho) cultivated in Suwon were calculated by using the FAO PM equation. The ET rate of Campbell Early was $2.41\;mm\;day^{-1}$ and that of Kyoho was $2.22\;mm\;day^{-1}$ in August when the leaf area index was 2.2. During this period, the $K_{cb}$ of Campbell Early based on the FAO PM equation was on average 0.49 with the maximum value of 0.72. On the other hand, the $K_{cb}$ of Kyoho was averaged to be 0.45 with the maximum value of 0.64. The seasonal leaf area index for two grape cultivars was measured as 0.15 in April, 0.5 in May, 1.4 in June, 2.2 in July-September, and 1.5 in October. The $K_{cb}$ of Campbell Early showed a seasonal variation, changing from 0.03 in April to 0.11 in May, 0.31 in June, 0.49 in July-September, and 0.33 in October. The magnitudes and the seasonality of $K_{cb}$ of Kyoho were similar to those of Campbell Early.

Improvement of Water Quality Using Ultra Filtration System in Artificial Seed Production of Olive Flounder, Paralichthys olivaceus (넙치 인공종묘생산에 있어 막분리 여과 시스템을 이용한 수질환경의 개선)

  • Water quality, bacterial phase and fish growth rate were analyzed in the process of artificial seed production of flounder (Paralichtys oliraceus) larvae to investigate the water quality in rearing tank using Ultra Filtration System (UES). Sand Filtration System (SFS) and Ultra Filtration System (Ins) were set up in the experimental group. For the analysis of water quality, pH, salinity, DO, SS, COD, $NH_{4}^{+},\;NO_{2}^{-},\;NO^-,\;DIN$ (dissolved inorganic nitrogen) and DU (dissolved inorganic phosphate) were measured. There was no data difference between SFS group and UES group in most analysis items, but the UEs group showed low salinity and low 55 values, such that salinity was $33.5\%_{\circ}$ in SES group and $30.2\%_{\circ}$ in WS group and 55 was 15.5 mL/L in SES group and 7.0 mL/L for UPS group. For changes in bacterial phase and TBC (Total Bacterial Counts), in SES group, 6$\times$10^{5}CFU/mL in seawater decreased to the ratio of about 116, and TBC, Genus Vibrio and bacteria in the Genus Acinetobacter and Genus Micrococcus sharply increased after nine days, while stable bacterial phase was maintained low in UES group during the experiment except for Genus Ajteromonas. In the growth of the larvae, fish length was 17.0 mm (SGR 14.0) in the SES group and 18.8 mm (SGR 14.3) in the UFS group. It is concluded that when water is supplied for artificial seed production with WS, stabilization of water quality condition and inhibition of bacterial multiplication are possible. When production environment becomes stable, stable growth of fish becomes possible by reduction of environmental stress.

Solubility Improvement of Cuttle Bone Powder Using Organic Acids (유기산처리에 의한 갑오징어갑 분말의 가용성 개선)

  • As a pan of a study on effective use of seafood processing by-products, such as cuttle bone as a calcium source, we examined on the kind of organic acid (acetic acid and lactic acid), reaction concentration (mole ratio of calcium to mole of organic acid), reaction temperature $(20\~60^{\circ}C)$ and reaction time (6$\~$24 hours) as reaction conditions for the solubility improvement of cuttle bone powder. The high soluble cuttle bone powder was also prepared from the optimal reaction conditions and partially characterized. From the results on examination of reaction conditions, the high soluble cuttle bone powder was prepared with 0.4 in mole ratio of a calcium to mole of a acetic acid at room temperature for 12 hours, Judging from the patterns of IR and X-ray diffraction, the main component of the high soluble cuttle bone powder was presented as a form of calcium acetate, and a scanning electron micrograph showed an irregular form. The soluble calcium content in the high soluble cuttle bone powder was $5.3\%$ and it was improved about 1,380 times compared to a raw cuttle bone powder. For the effective use of the high soluble cuttle bone powder as a material for a functional improvement in processing, it should be used after the calcium treatment at room temperature for about 1 hour in tap water or distilled water. from these results, we concluded that it is possible to use the high soluble cut시e bone powder as a material for a functional improvement in processing.

Growth Inhibition of Diatom, Chaetoceros calcitrans by Marine Bacteria, Shewanella sp. SR-14 - Effects of Marine Bacteria on the Changes of Fatty Acid Composition of Diatoms - (해양세균, Shewanella sp. SR-14에 의한 규조류 Chaetoceros calcitrans의 증식저해 - 해양세균이 규조류의 지방산 조성변화에 미치는 영향 -)

  • In the previous reports, the authors isolated two strains of marine bacteria, Shewanella sp. SR-14, which has Chaetonros sp. growth inhibition activity, and Vibrio alginolyticus, that did not affect growth of the alga. In the present study, fatty acid compositions of diatoms, Chaetoceros calcitrans and Skeletonema costatum, and marine bacteria, Shewanella sp. SR-14 and V. alginolyticus, were analyzed. Changes of fatty acid composition in the diatoms grown with the marine bacteria were also determined. Major fatty acids of Sbewanella sp. SR-14 were 16:1n-7 $(29.4\%)$ and 16:0 $(19.2\%)$ during incubation in peptone broth at $20^{\circ}C$ for 3 days. The compositions of V. alginolyticus detected were 16:0 $(23.7\%),$ 16:1n-7 $(27.7\%)$ and 18:1n-7 $(21.0\%).$ C. calcitrans consisted of 16:1n-7 $(33.3\%),$ 16:0 $(27.1\%)$ and 14:0 $(12.1\%).$ S. costatum mainly contained 16:1n-7 $(28.9\%),$ 16:0 $(21.6\%)$ and 20:5 $(19.8\%).$ When halves of cell numbers of C. calcitrans were moribund cells by Shewanella sp. SR-14, the C. calcitrans and S. costatum simultaneously cultured with the bacteria were harvested by filtration with GE/D glass microfibre filter. In the fatty acid composition of both diatoms, saturated fatty acid contents in both diatoms grown with Shewanella sp. SR-14 were decreased, but unsaturated fatty acid contents were increased. The differences were greater in C. calcitrans than those in S. costatum. During the growth of diatoms with V. alginolyticus, C. calcitrans showed increase of saturated fatty acid contents and decrease of unsaturated fatty acid contents; however, S. costatum did not show sharp difference in fatty acid content. In this study, Shewanella sp. SR-14, which showed growth inhibition activity against C. calcitrans, influenced on the changes of fatty acid contents in the diatom. It was suggested that increased unsaturated fatty acid was synergistically activated algal growth inhibition activity of Shewanella sp. SR-14.

Improving Performance of Recommendation Systems Using Topic Modeling (사용자 관심 이슈 분석을 통한 추천시스템 성능 향상 방안)

  • Recently, due to the development of smart devices and social media, vast amounts of information with the various forms were accumulated. Particularly, considerable research efforts are being directed towards analyzing unstructured big data to resolve various social problems. Accordingly, focus of data-driven decision-making is being moved from structured data analysis to unstructured one. Also, in the field of recommendation system, which is the typical area of data-driven decision-making, the need of using unstructured data has been steadily increased to improve system performance. Approaches to improve the performance of recommendation systems can be found in two aspects- improving algorithms and acquiring useful data with high quality. Traditionally, most efforts to improve the performance of recommendation system were made by the former approach, while the latter approach has not attracted much attention relatively. In this sense, efforts to utilize unstructured data from variable sources are very timely and necessary. Particularly, as the interests of users are directly connected with their needs, identifying the interests of the user through unstructured big data analysis can be a crew for improving performance of recommendation systems. In this sense, this study proposes the methodology of improving recommendation system by measuring interests of the user. Specially, this study proposes the method to quantify interests of the user by analyzing user's internet usage patterns, and to predict user's repurchase based upon the discovered preferences. There are two important modules in this study. The first module predicts repurchase probability of each category through analyzing users' purchase history. We include the first module to our research scope for comparing the accuracy of traditional purchase-based prediction model to our new model presented in the second module. This procedure extracts purchase history of users. The core part of our methodology is in the second module. This module extracts users' interests by analyzing news articles the users have read. The second module constructs a correspondence matrix between topics and news articles by performing topic modeling on real world news articles. And then, the module analyzes users' news access patterns and then constructs a correspondence matrix between articles and users. After that, by merging the results of the previous processes in the second module, we can obtain a correspondence matrix between users and topics. This matrix describes users' interests in a structured manner. Finally, by using the matrix, the second module builds a model for predicting repurchase probability of each category. In this paper, we also provide experimental results of our performance evaluation. The outline of data used our experiments is as follows. We acquired web transaction data of 5,000 panels from a company that is specialized to analyzing ranks of internet sites. At first we extracted 15,000 URLs of news articles published from July 2012 to June 2013 from the original data and we crawled main contents of the news articles. After that we selected 2,615 users who have read at least one of the extracted news articles. Among the 2,615 users, we discovered that the number of target users who purchase at least one items from our target shopping mall 'G' is 359. In the experiments, we analyzed purchase history and news access records of the 359 internet users. From the performance evaluation, we found that our prediction model using both users' interests and purchase history outperforms a prediction model using only users' purchase history from a view point of misclassification ratio. In detail, our model outperformed the traditional one in appliance, beauty, computer, culture, digital, fashion, and sports categories when artificial neural network based models were used. Similarly, our model outperformed the traditional one in beauty, computer, digital, fashion, food, and furniture categories when decision tree based models were used although the improvement is very small.
