Two tobacco cultivars (Nicotiana tabacum L.), Bel-B and Bel-W3, tolerant and sensitive to ozone, respectively, were grown in a greenhouse supplied with charcoal filtered air and exposed to 200 ppb ozone for 4 hr. Effects on chlorophyll fluorescence, net photosynthesis, and stomatal conductance are described. Quantum yield was calculated from chlorophyll fluorescence and the initial slope of the assimilation-light curve measured by the gas exchange method. Only the sensitive cultivar, Bel-W3, developed visual injury symptoms on up to 50% of the $5^{th}$ leaf. The maximum net photosynthetic rate of ozone-treated plants was reduced 40% compared to control plants immediately after ozone fumigation in the tolerant cultivar; however, photosynthesis recovered by 24 hr post fumigation and remained at the same level as control plants. On the other hand, ozone exposure reduced maximum net photosynthesis up to 50%, with no recovery, in the sensitive cultivar apparently causing permanent damage to the photosystem. Reductions in apparent quantum efficiency, calculated from the assimilation-light curve, differed between cultivars. Bel-B showed an immediate depression of 14% compared to controls, whereas, Bel-W3 showed a 27% decline. Electron transport rate (ETR), at saturating light intensity, decreased 58% and 80% immediately after ozone treatment in Bel-B and Bel-W3, respectively. Quantum yield decreased 28% and 36% in Bel-B and Bel-W3, respectively. It can be concluded that ozone caused a greater relative decrease in linear electron transport than maximum net photosynthesis, suggesting greater damage to PSII than the carbon reduction cycle.
Inhalational anthrax is caused by B. anthracis, a virulent sporeforming bacterium which secretes anthrax toxins consisting of protective antigen (PA), lethal factor (LF) and edema factor (EF). LF is a Zn-dependent metalloprotease and is the main determinant in the pathogenesis of anthrax. Here we report the identification of a lead small-molecule inhibitor of anthrax lethal factor by screening an available synthetic small-molecule inhibitor library using fluorescence-based high-throughput screening (HTS) approach. Seven small molecules were found to have inhibitory effect against LF activity, among which SM157 had the highest inhibitory activity. All theses small molecule inhibitors inhibited LF in a noncompetitive inhibition mode. SM157 and SM167 are from the same family, both having an identical group complex, which is predicted to insert into S1' pocket of LF. More potent small-molecule inhibitors could be developed by modifying SM157 based on this identical group complex.

Also the field of lighting source belong to environmental control. And in the future the control will be strong. In the mercury free lighting source system the Xe gas lamp is one type. And the Ne:Xe mixing gas lamp improvements firing voltage of Xe gas lamp. Purpose and subject of this study are understand, efficiency, ideal of Ne:Xe plasma which mercury free lamp. Before ICP was designed, basic parameters of plasma, which are electron temperature and electron density, were measured and calculated by langmuir probe data. Property of electron temperature and electron density were confirmed by changing ratio of Ne:Xe.

Culture conductivity and on-line NADH fluorescence were used to measure cellular growth in plant cell suspension cultures of Podophyllum hexandrum. An inverse correlation between dry cell weight and medium conductivity was observed during shake flask cultivation. A linear relationship between dry cell weight and culture NADH fluorescence was obtained during the exponential phase of batch cultivation In a bioreactor under the pH stat (pH 6) conditions. It was observed that conductivity measurement were suitable for biomass characterisation under highly dynamic uncontrolled shake flask cultivation conditions. However, if the acid/alkali feeding is done for pH control the conductivity measurement could not be applied. On the other hand the NADH fluorescence measurement allowed online-in situ biomass monitoring of rather heterogenous plant cell suspension cultures in bioreactor even under the most desirable pH stat conditions.
Effect of iron on the extracellular proteolytic activity of live Uronema marium was determined by fluorescence polarization (FP) method. Supplementation of 0.5 and 5.0 μM iron significantly increased caseinolytic activity of live U. marinum. In contrast, supplementation of 50 μM iron showed no significant differences in FP values compared to the control. The present result suggests that iron in cultured water or skin tissue of olive flounder may influence on the penetration and establishment of U. marinum, correlating with modulation of extracellular protease activity of the ciliates.

Proteolytic activity of live Uronema manum was analyzed by fluorescence polarization (FP) technique. Protease activity was measured by a decrease in FP value using fluorescein isothiocynate (FITC)-casein as a protein substrate. The results demonstrated an inverse linear relationship between fluorescence polarization (FP) values and live ciliate concentration over the range $1\times10^4\;to\;2\times10^5$ cells/well. However, the FP values of $10-10^3$ live parasites were not different significantly from that of control. Time-dependent decrease in FP value was shown in the wells containing live U. marinum. In the present study, FP assay had the benefit to provide measurements of substrate hydrolysis by live parasites in real-time, and did not require separations, precipitations, or transfers of reaction mixture.
In order to find the cellular interaction factors of the Heliothis armigera nuclear polyhedrosis virus capsid protein VP39, a Heliothis armigera cell cDNA library was constructed. Then VP39 was used as bait. The host actin gene was isolated from the cDNA library with the yeast two-hybrid system. This demonstrated that VP39 could interact with its host actin in yeast. In order to corroborate this interaction in vivo, the vp39 gene was fused with the green fluorescent protein gene in plasmid pEGFP39. The fusion protein was expressed in the Hz-AM1 cells under the control of the Autographa californica multiple nucleopolyhedrovirus immediate early gene promoter. The host actin was labeled specifically by the red fluorescence substance, tetramethy rhodamine isothicyanete-phalloidin. Observation under a fluorescence microscopy showed that VP39, which was indicated by green fluorescence, began to appear in the cells 6 h after being transfected with pEGFP39. Red actin cables were also formed in the cytoplasm at the same time. Actin was aggregated in the nucleus 9 h after the transfection. The green and red fluorescence always appeared in the same location of the cells, which demonstrated that VP39 could combine with the host actin. Such a combination would result in the actin skeleton rearrangement.
To investigate the effects of strong acidic electrolytic water on the chloroplast, barley leaves were treated with strong acidic electrolytic water(pH 2.5). And to investigate the effects of weak acidic electrolytic water on the chloroplast development, etiolated barley leaves were treated with weak acidic electrolytic water(pH 6.5) during greening period. Chl contents, Fo, Fv, and Chl fluorescence quenching coefficient in barley leaves were measured during and after treatment of acidic electrolytic water. The following results were obtained. Chl a, b, and carotenoid were decreased with treatment of strong acidic electrolytic water. Chl contents were significantly decreased than that of the control after 5 min. These results provide evidence that the strong acidic electrolytic water dissimilate the Chl and so that the value of Fo was slightly increased. The strong acidic electrolytic water damaged PS II because Fo was increased and Fv, Fm, and Fv/Fm ratio were decreased. qP, qNP and qE were decreased. On the other hand qI was increased than that of the control. But Chl content and Chl fluorescence patterns were a little changed as the pH increase over 4.0 Chl a, b, and carotenoid were increased with treatment of weak acidic electrolytic water during greening period. Chl contents were significantly increased than that of control after 12 hours greening. These results provide evidence that the weak acidic electrolytic water accelerated the chlorophyll synthesis. And the weak acidic electrolytic water accelerated PS II development because Fv, Fm, qP and Fv/Fm ratio were increased than that of the control.
Winter cereals are acclimated during wintering, and thereafter their freezing resistance is increased. In order to analyze inheritance and heritabilities for photoinhibition of photosynthesis by high light intensity under low temperature, and to evaluate the relationship between low temperature-induced photoinhibition and winter survival, 4 parental half diallel crosses were used. The detached leaves of 7-8cm long from plants grown for 35 and 55 days were placed on wet filter paper and placed in trays at 5$^{\circ}C$ cold room with 1,200 $\mu$mol $m^{-2}$$s^{-1}$ PPFD. Chlorophyll fluorescence was measured with a chlorophyll fluorescence system after dark adaptation for 30 min. The Fv/Fm of 35day old plants was reduced from 0.714 in the control leaves to 0.409 and 0.368 following photoinhibitory treatment of 6h and 8h and the CVs were increased from 0.8% to 22.2-22.3%. The Fv/Fm of 55-day old plants was reduced from 0.775 in the control leaves to 0.485 and 0.439 following photoinhibitory treatment of 10h and 12h, respectively. According to half diallel cross analysis, Reno and Dongbori 1 (highly resistant to photoinhibition) was dominant, but Oweolbori (susceptible to photoinhibition) was recessive, and photoinhibition showed partial dominance with highly additive gene action. Dongbori 1 showed the greatest GCA effects for photoinhibition, and GCA/SCA ratios (8.7-22.3 times) indicated that the additive variance for the character was more important. Winter survival in barley crosses was positively correlated with resistance to photoinhibition and significantly fitted by linear regression ($R^2$=0.751$^{**}$-0.779$^{**}$). The chlorophyll fluorescence measured by Fv/Fm has been found to be highly inheritable and very useful in evaluating relative levels of freezing resistance in barley.ley.
The effects of change in injection pressure on spray structure in high temperature and pressure field have been investigated. The analysis of liquid and vapor phases of injected fuel is important for emissions control of diesel engines. Therefore, this work examines the evaporating spray structure using a constant volume vessel. The injection pressure is selected as the experimental parameter, is changed from 22 MPa to 112 MPa using a high pressure injection system (ECD-U2). Also, we conducted simulation study by modified KIVA-II code. The results of simulation study are compared with experimental results. The images of liquid and vapor phase for free spray were simultaneously taken by exciplex fluorescence method. As experimental results, the vapor concentration of injected fuel is leaner due to the increase of atomization in the case of the high injection pressure than in that of the low injection pressure. The calculated results obtained by modified KIVA-II code show good agreements with experimental results.
