Screening for Antioxidant Activity of Plants and Marine Algae and Its Active Principles from Prunus davidiana

  • The antioxidant activity of methanol extracts of plants and marine algae was tested by using 1,1-diphenyl-2-picrylhydrazyl(DPPH). Five plant extracts(Prunus davidiana, Eriobotrya japonica, Artemisia iwayomogi, Stirodella tolyrrhiza and Ulmus davidiana) and two algae (Ecklonia stolonifera and Symphycoladia latiuscula) were found to be the most effective in DPPH radical scavenging activity. The methanol extract obtained from the stems of Prunus davidiana was fractionated with several solvents. The ethylacetate soluble fraction exhibiting the strongest antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Antioxidant flavonoids and flavonoid glycosides were isolated and the most active ones was identified as (+)-catechin by MS, $^1H-NMR$ and $^{13}C-NMR$. Its antioxidant activity was higher than that oil vitamin C.

  • 큰잎자작 변재의 메탄올(methanol, MeOH) 조추출물로부터 silica gel column, CPC 등의 크로마토그라피를 이용하여 4종의 화합물을 단리하였으며, 단리물질에 대한 NMR 등의 기기분석 결과, flavonoid 및 lignan 배당체인 catechin 7-O-𝛽-D-xylopyranoside, lyoniresinol 9'-O-𝛽-D-glucopyranoside, 그리고 diarylheptanoid인 11-oxo-3, 8, 12, 17-tetrahydroxy-9-ene [7, 0]-metacyclophane 및 11-oxo-3, 8, 9, 10, 12, 17-hexahydroxy [7, 0]-metacyclophane으로 각각 동정하였다.

치자(Gardenia jasminoides Ellis fructus) 껍질 추출물의 항산화 활성 (Antioxidant Activity of Peel from Gardenia jasminoides Ellis Fructus Extracted by Various Solvents)

  • 치자 껍질의 항산화 활성을 측정하기 위하여 70% methanol, distilled water (DW) 및 ethyl acetate (EA)의 3가지 용매를 사용한 추출물의 용매 별 flavonoid 함량 및 항산화 활성, 금속 chelating 능력 측정을 통하여 치자 껍질의 기능성 식품 재료로서의 가치를 검토한 결과, 치자 껍질의 proanthocyanidin 함량은 $48.165{\pm}0.811mg\;CE/g\;DW$로 나타났으며, 추출 수율은 70% methanol(36.26%), DW (39.87%), EA (2.88%) 로 나타났다. 추출 용매 별 항산화 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며, control로 사용된 ascorbic acid, BHA, EDTA 보다는 낮은 활성이 관찰되었다. Flavonoid 함량(mg QE/g)은 70% methanol (0.416), DW (0.225), EA(0.212) 순으로 확인되었으며 다른 항산화 분석 및 금속 chelating 능력 실험에서도 이와 유사하게 관찰되어 모든 분석에서 70% methanol 추출물이 가장 활성이 강한 것으로 나타났다. 이상의 결과를 미루어 볼 때 치자 껍질의 용매 별 flavonoid 함량에 따라 항산화능, 금속 chelating 능력이 증가하는 것으로 사료되며, EA 추출물을 제외한 다른 추출물에서의 수율과 항산화 활성은 높은 수준으로 관찰되었다. 따라서 치자의 껍질은 proanthocyanidin과 flavonoid 화합물을 다량 함유하고 있으며 그로 인한 높은 항산화 활성과 생리활성을 가지고 있어 기능성 식품 및 천연항산화제로서의 가치가 매우 기대된다.

광나무 잎의 페놀성 화합물 (Phenolic Compounds of Ligustrum japonicum Leaves)

  • 식물자원으로부터 생리활성 물질의 이용측면에서 국내에서 자생하고 있는 광나무 잎을 대상으로 하여 다양한 생리활성을 가지는 것으로 보고된 페놀성 화합물의 분포를 확인하였다. 총 페놀성 함량은 $0.89{\sim}1.53%$, 축합형 탄닌은 $0.10{\sim}0.13%$였으며, flavonoid는 aglycone인 apigenin, luteolin과 칼럼크로마토그라피를 행하여 분획으로 분리한 후 가수분해하여 이들의 배당체를 확인하였다. 페놀산의 분획 중 유리형 페놀산 화합물에는 tyrosol, t-cinnamic acid, p-Hydroxybenzoic acid, vanillic acid, shikimic acid, protocatecuic acid등이 존재하였다. 에스테르형 페놀산 화합물에는 tyrosol, t-cinnamic acid, ferulic acid, esculetin, caffeic acid, p-coumaric acid, hydroxytyrosol 등이 함유되었다. 결합형 페놀성 화합물에는 tyrosol, t-cinnamic, p-coumaric acid와 미확인 페놀성 물질들이 존재하였다. 따라서 광나무 잎은 xanthine oxidase 활성저해, LDL 산화, 혈소판 응집저해, 항균활성 등 다양한 생리활성을 나타내는 것으로 보고되고 있는 tyrosol, hydroxytyrosol, protocatecuic acid 등 풍부한 페놀성 화합물을 함유하고 있으므로 유용한 식물자원이 될 수 있음을 확인하였다.

Structure Determination of Four Compounds Isolating from Rhizomes of Rhodiola rosea using NMR Spectrometer

  • Rhodiola rosea, also known as gold root or rose root, is a perennial plant in the family of Crassulaceae. The rhizhome of R. rosea has been widely used as a hemostatic, tonic for burns and contusions in traditional Chinese medicine. Recent studies reported its strong antioxidant and adaptogenic properties. In this paper, we attempted to isolate compounds from the methanolic extract of R. rosea rhizomes. Four compounds including one new compound (1), two kaempferol glycosides (3 and 4) were isolated from chloroform and ethyl acetate soluble fraction of R. rosea extract. The structures of 1~4 including relative stereochemistry were determined by MS and NMR analysis.

Chemical Investigation of the Constitutive Flavonoid Glycosides of the Leaves of Crataegus sinaica

  • Leaves of Crataegus sinaica Boiss, contain the new C-glycosyl flavone $3"',4”’-di-O-acetyl-2“-O-{\alpha}-rhamnosylvitexin$, together with the hitherto unknown, dihydroflavonol 3-O-xyloside, (2R:3R)-dihydroquercetin-$3-O-{\beta}-xylopyranoside$. The known compounds (+)-catechin, (-)-epicatechin, vicenin-II, $2"-O-{\alpha}-rhamnosylvitexin$, and $4"'-O-acetyl-2"-O-{\alpha}-rhamnosylvitexin$ were also characterized. Structures were established by conventional methods of analysis and confirmed by $^1H-,\;^{13}C-NMR$, and ESI-MS.

  • 6-Hydroxykynurenic acid and ginkgolide B together with flavonol glycosides and biflavonoids were isolated from the yellow leaves of Ginkgo biloba and identified by means of spectroscopic methods. The correctness of $H{\"{o}}lzl's$ ${13}^C-NMR$ assignments for 6-hydroxykynurenic acid was confirmed by HMQC and HMBC techniques. Based on our present findings, it may be considered that the yellow Ginkgo leaves may contribute to be a source of high medicinal values.

  • Six compounds were isolated from the BuOH soluble fraction of the leaves of Staphylea bumalda. On the basis of spectral data, they were identified as atragalin (1), $2-methyl-5,\;7-dihydroxy-chromone-7-O-{\beta}-D-glucopyranoside$ (2), isoquercitrin (3), nicotiflorin (4), kaempferol 3-neohesperidoside (5), kaempferol $3-O-[{\alpha}-rhamno-pyranosyl-(1\;{\to}\;4)-rhamnopyranosyl-(1\;{\to}\;6)-{\beta}-D-glucopyranoside]$ (6), respectively.

석죽과식물(石竹科植物)의 성분검색(成分檢索) (The Chemical Screening of Caryophyllaceae Plants)

  • From phytochemical screening of the seven species of Caryophyllaceae, that is, one species of Melandrium, and Dianthus and Pseudostellaria, two species of Stellaria and Cerastium the following compounds were identified. 1) aldehyde, sugars, other reducing compounds and glycosides from the above mentioned seven species of Caryophyllaceae were identified by thin layer chromatography method and comparison of Rf values with the standard substances. 2) Saponins, steroids and terpenoids showed positive reaction with the Froth testing in all of seven plants. Five species of plant showed positive reaction with Liebermann-Buchard test. 3) Flavonoid and polyphenolic compounds were identified by the spots in TLC-test and chlorogenic acid showed the same Rf values with the standard substances. 4) The spots of alkaloid were shown at Rf=0.68 (Stellaria aquatica) and at Rf=0.71 (Pseudostellaria palibiniana) and at Rf=0.70, and 0.44 (Dianthus sinensis).

Phenolic Compounds Isolated from Opuntia ficus-indica Fruits

  • On the phytochemical investigation of a 70% ethanol extract of the fruits of Opuntia ficus-indica, (Cacataceae), we could result in the isolation of thirteen phenolic compounds including seven flavonoids (1 - 9) and four simple phenolic glycosides (10 - 13) by column chromatographic methods. Among the isolated compounds, picein (11), androsin (12), and $1-O-feruloyl-{\beta}-{\small{D}}-glucopyranoside$ (13) were isolated for the first time from O. ficus-indica; additionally, this is the first report $benzyl-O-{\beta}-{\small{D}}-glucopyranoside$ (10) from the genus Opuntia. The structures of the compounds were determined by spectral data analysis which included 1D, 2D NMR spectrum and ESIMS.
