Analysis of antioxidant and anti-inflammatory effects of Mongolian wild lingonberry and blueberry, and identification of their bioactive compounds (몽골 야생 링곤베리와 블루베리의 항산화, 항염증 효과 및 생리활성 물질 분석 연구)

  • The Mongolian lingonberry and blueberry are two essential food sources found in Mongolia. This study investigated the antioxidant and anti-inflammatory effects of methanol extracts from Mongolian lingonberry (LBE) and blueberry (BBE). Compared to the LBE, the BBE showed higher total phenolic, flavonoid, and anthocyanin contents, as well as antioxidant capacities. The LBE and BBE inhibited the mRNA expression of pro-inflammatory genes, including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and cyclooxygenase (COX-2) in lipopolysaccharide-stimulated RAW 264.7 macrophage cells. In addition, the LBE and BBE inhibited NADPH oxidase-2 (Nox2) mRNA expression, indicating that they have cellular antioxidant capacities. Anthocyanin derivatives of the LBE and BBE were analyzed using LC-QTOF/MS. Six anthocyanins were identified in the BBE, while one was detected in the LBE. Our findings demonstrate that the anthocyanin-rich LBE and BBE could be used as functional food sources in Mongolia.

Antioxidant Activity of Korean Gomchwi (Ligularia fischen) Extracts (국내산 곰취(Ligularia fischen) 추출물의 항산화 활성)

  • In this study, Gomchwi (Ligularia fischen) derived from Taebaek-si, Gangwon-do was extracted with 70% ethanol (LFE) and antioxidant activity was measured. The following experimental techniques were used to evaluate the antioxidant efficacy of LFE. Total phenolic contents, ABTS/DPPH radical scavenging analysis, cell viability assay, NO assay, and quantitative real-time PCR technique. The content of polyphenol and flavonoid was each 113.97±0.37 mg GAE/g or 29.22±2.06 mg QE/g in LFE. DPPH radical scavenging activity was measured to be 25 ㎍/㎖ 11.26±0.95%, 50 ㎍/㎖ 17.12±0.63%, 100 ㎍/㎖ 29.54±0.36%, 250 ㎍/㎖ 68.31±0.28%, 500 ㎍/㎖ 75.12±0.05%, and 1000 ㎍/㎖ 75.75±1.57%. In addition, ABTS radical scavenging activity was identified as LFE 25 ㎍/㎖ 13.75±0.21%, 50 ㎍/㎖ 26.71±0.20%, 100 ㎍/㎖ 56.92±0.22%, 250 ㎍/㎖ 91.30±0.12%, 500 ㎍/㎖ 93.40±0.02, and 1000 ㎍/㎖ 93.19±0.04%. There was no significant cytotoxicity of LFE. NO production was significantly decreased to LFE 50 ㎍/㎖ 79.40±2.64%, 100 ㎍/㎖ 55.01±5.36%, and 200 ㎍/㎖ 30.93±3.11%. Also, the NOS2 gene expression was significantly reduced to LFE 50 ㎍/㎖ 0.94±0.11, 100 ㎍/㎖ 0.59±0.05, and 200 ㎍/㎖ 0.32±0.04. This result objectively confirmed the antioxidant effect of Gomchwi. We will continue to conduct in-depth research. Therefore, it is believed that the possibility of using Gomchwi as a cosmetic and functional food material can be established.

Acid treatment effects on the contents of quercetin glycosides and aglycone in red onion powder (산처리에 의한 적양파 분말의 quercetin 배당체와 aglycone의 농도변화)

  • Flavonoids are bioactive plant metabolites that have a range of beneficial effects on human health. Quercetin 4'-glycoside (Q4'G), quercetin 3,4'-diglycoside (Q3,4'G), and quercetin aglycone (QA) are the main flavonoids found in onions. QA, in particular, is likely to have a greater biological effect than glycosides. To develop an onion extract with high quercetin content, the optimal extraction conditions for red onion powder containing the outer layer of the onion were determined. The effects of acid treatment on the concentration of quercetin glycosides and QA were evaluated. The flavonoids of red onion powder were optimally extracted under 60-70% ethanol at 70℃ for 2 h. The deglycosylation of Q3,4'G and an increase in Q4'G content occurred within 6 h of 0.2% acetic acid treatment. The QA content and deglycosylation of Q4'G eventually peaked at 24 h. In addition, QA content and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity were highly correlated, with a correlation coefficient of 0.90.

Antioxidant and Antiproliferative Activities of the Halophyte Angelica japonica Growing in Korean Coastal Area (한국 연안지역에 서식하는 갯강활의 항산화 및 암세포증식 억제 활성)

  • This study evaluated the antioxidizing and antiproliferative effects of Angelica japonica extract and its solvent-partitioned fractions. A dried sample of the halophyte A. japonica was extracted twice using methylene chloride (CH2Cl2) and extracted twice again using methanol (MeOH). The combined crude extracts were then fractionated by solvent polarity into distilled water (water), n-butanol (n-BuOH), 85% aqueous methanol (85% aq.MeOH), and n-hexane fractions. The antioxidant activities of the crude extracts and their solvent-partitioned fractions were assessed according to their DPPH radical and peroxynitrite scavenging abilities, formation of intracellular reactive oxygen species (ROS), DNA oxidation, NO production, and ferric reducing antioxidant power (FRAP). The crude extract showed significant antioxidant activity in the overall antioxidizing bioassay systems. Among solvent-partitioned fractions, good antioxidant activities were observed in n-BuOH and 85% aq.MeOH fractions and significantly correlated with the polyphenol and flavonoid contents of the samples. Furthermore, all samples tested, including the crude extract, not only showed cytotoxic effects against human cancer cells (AGS, HT-29, MCF-7, and HT-1080) but also prevented cell migration in a dose-dependent manner in the wound healing assay using HT 1080. Among the solvent-partitioned fractions, the 85% aq.MeOH fraction most effectively inhibited the invasion of HT-1080 cells. Therefore, these results suggest that A. japonica may be a potential antioxidizing and antiproliferative agent.

Antioxidant and Antibacterial Activities of Extracts from Different Parts of Sophora japonica L. (회화나무 부위별 추출물의 항산화 및 항균활성)

  • In this study, the antioxidant and antibacterial activities of Sophora japonica extracts were investigated to determine the potential of S. japonica as a functional food and medicinal materials. S. japonica was divided into flowers, fruits, and branches, and ethanol extraction was used. The total polyphenol and flavonoid contents were significantly higher in the flower and fruit extracts than in the branch extracts, but the ABTS and DPPH radical scavenging activity and ORAC value were higher in the branch extracts. Among the ethanol extracts of S. japonica, branch extracts showed strong antibacterial activity against Porphyromonas gingivalis, and the MIC was 0.2 mg/ml. Branch extracts showed bacteriostatic activity against P. gingivalis at a concentration of 0.4 mg/ml or less and bactericidal activity at a concentration of 0.6 mg/ml or more. Biofilm biomass production and cell growth of P. gingivalis in the culture medium treated with the branch extract at a concentration of 0.2-2.0 mg/ml were significantly decreased in a concentration-dependent manner. In addition, the mRNA expression of fimA and mfa1 associated with fimbriae formation in these cultures was suppressed in a concentration-dependent manner. Based on these results, S. japonica branch extracts can be used as functional food and medicinal materials, as demonstrated by their antioxidant and antibacterial activities against P. gingivalis and the inhibition of biofilm formation resulting from P. gingivalis.

Analysis of Chlorogenic Acid Content and Biological Activities of Aralia elata Ethanol Extract (두릅 에탄올 추출물의 Chlorogenic acid 함량 분석 및 생리활성)

  • This study was conducted to quantify chlorogenic acid content and evaluate biological activity, such as antioxidant, antibacterial, anti-inflammatory, and digestive enzyme activity of Aralia elata ethanol extract (AEE). The SC50 of DPPH and ABTS radical scavenging activities of AEE were 4.79±0.05 mg/mL, 5.79±0.05 mg/mL; total polyphenol and total flavonoid contents were 170.0±1.8 mgGAE/g, 105.5±4.1 mgQE/g, respectively. Nitric oxide (NO) was increased in RAW 264.7 cells and Caco-2 cells with treatment of LPS, and production of NO was inhibited by AEE in a concentration-dependent manner. Production of NO was reduced by 60.0±1.1% in RAW 264.7 cells and 50.7±2.8% in Caco-2 cells at of AEE. Similarly, the production of inflammatory cytokines (TNF-α, IL-1β and IL-6) was inhibited in a concentration dependent manner. Antibacterial activity increased as the dose concentration of AEE increased, and the MIC was 75 mg/mL for L. monocytogenes, and 100 mg/mL for S. typhimurium and H. pylori. In addition, amylase and protease enzyme activity was observed in AEE and increased enzyme activity was observed according to the concentration of the extract. AEE contained 7.06±0.01 mg/g of chlorogenic acid. As a result of the experiment, it is judged that it can be used as basic data for the development of health food using Aralia elata.

Physicochemical Properties and Antioxidant Activities of gochujang with lotus leaf powder

  • This study examined that have excellent antioxidant effect and expand the base of consumption of lotus leaves. We added 1%, 3%, and 5% of lotus leaf powder to traditional gochujang. pH was the highest in the gochujang with lotus leaf powder 1%(p<0.001). The moisture was significantly lower as the addition of lotus leaf powder increased(p<0.01). The viscosity was the lowest with lotus leaf powder 5% was found to be 15.61 dPa·s(p<0.001). In chromaticity, the L value was the highest in the control, the viscosity of gochujang became darker and lessened(p<0.001). The a value and b value showed the highest in the control(p<0.001). The salinity was the lowest in the gochujang with lotus leaf powder 3%(p<0.001). The sugar content decreased as the more amount of lotus leaf. Total phenol and total flavonoid contents was the highest in the gochujang with lotus leaf powder 5%. The DPPH radical scavenging ability was higher as the more amount of lotus leaf. Reducing power and α-glucosidase inhibitory activity was the highest in the gochujang with lotus leaf powder 3%. The gochujang with lotus leaf powder can be expected to have higher antioxidant activities and to be a health functional food.

Antioxidant Activities of Protaetia brevitarsis Larvae Fermented by Lactobacillus acidophilus (유산균 발효가 흰점박이꽃무지(Protaetia brevitarsis) 유충의 항산화활성에 미치는 영향)

  • This study was carried out to evaluate the effect of fermentation by Lactobacillus acidophilus on the antioxidant activity of Protaetia brevitarsis larvae fed with mushroom substrates (king oyster mushroom). The total polyphenol content of the P. brevitarsis larvae extracts (PLEs) (93.33±0.98 mg GAEs/extract g) was higher than that of the fermented P. brevitarsis larvae extracts (FPLEs) (65.02±1.32 mg GAEs/extract g). The flavonoid contents of the PLEs and FPLEs were 18.3±1.57 QEs mg/extract g and 17.69±0.95 QEs mg/extract g, respectively. The DPPH radical scavenging activity showed no significant difference between the PLEs and FPLEs at a concentration of 2-4 mg/ml. However, at a concentration of 8 mg/ml or more, the DPPH radical scavenging activity of the FPLEs was higher than that of the PLEs. The reducing power of the FPLEs was also higher than that of the PLEs, and more than twice as high at a concentration of 1.6 mg/ml or more. The ORAC value of the FPLEs (79.22±0.72 μM TEs/extract g) was higher than that of the PLEs (74.34±0.37 μMTEs/extract g). A WST-1 assay of the RAW 264.7 cells indicated that the PLEs and FPLEs showed no cytotoxicity.

Comparison of the Antioxidant and Mineral Properties of Korean Adzuki Bean (Vigna angularis L.) Leaves and Seeds (국내 팥 육성계통의 잎과 종실의 무기질 및 항산화 특성 비교)

  • This study compared the antioxidant and mineral properties of the leaves and seeds of fifteen Korean adzuki bean (Vigna angularisL.) breeding lines. This study was conducted in an attempt to expand the use of Korean adzuki bean leaves. The potassium, calcium, magnesium, and sodium contents of the leaves were significantly higher than the seeds, in particularly, the potassium content. The leaves had approximately 3.3 times higher potassium content than the seeds. For instance, the potassium content of YA1317 leaves was 21% higher than that of Arari. The total polyphenol content and ABTS activity of Adzuki bean leaves were significantly higher than the seeds, as opposed to the total flavonoid content and DPPH scavenging activity. Among the 15 breeding lines, YA1402 had 1.2~3.2 times higher antioxidant content and activity as compared to the Arari variety. It was concluded that adzuki bean leaves had higher mineral content, antioxidant component and activity as compared to the seeds. Therefore, adzuki bean leaves could be used an ingredient for dishes and as a medicine.

Evaluation of Evodiae Fructus Extract on the Chronic Acid Reflux Esophagitis in Rats (오수유(吳茱萸) 추출물이 만성 역류성 식도염 흰쥐에 미치는 효능 평가)

  • Objective : Reflux esophagitis (RE) is a disease that caused gastric acid reflux and inflammation due to unstable gastroesophageal sphincter, as increasing worldwide respectively. This study was conducted to evaluate the effect of Evodiae Fructus (EF) extract on chronic reflux esophagitis in rats. Methods : The EF was measured antioxidant activity, such as total polyphenol and total flavonoid contents, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and 2, 2'-azinobis-3-ethyl-enzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity. Rats were divided into 3 groups; Nor (normal group), Con (chronic acid reflux esophagitis rats treatment with water), EF (chronic acid reflux esophagitis rat treatment with EF 200 mg/kg body weight group). A surgically-induced chronic acid reflux esophagitis (CARE) model was established in SD rats, and treated with water or EF 200 mg/kg body weight for 14 consecutive days. Results : Administration of EF to rats of induction of chronic acid reflux esophagitis was found to reduce esophagus tissues injury. Reactive oxygen species (ROS) and produces peroxynitrite ($ONOO^-$) levels of esophagus tissues were significantly decreased in EF compared to Con group. As results of esophagus protein analyses, EF effectively reduce inflammatory-related factors ($NF-{\kappa}Bp65$, $p-I{\kappa}B{\alpha}$, iNOS, $TNF-{\alpha}$, IL-6), and increase anti-oxidant enzyme (Nrf2, HO-1, SOD, catalase, GPx-1/2). Conclusions : These results suggest that EF administration comfirmed that decreased esophagus tissues injury, oxidantive stress, anti-inflammation effect, and increased anti-oxidant effect. Therefore, EF was the potential to be used as a natural therapeutic drug.
