In analysis of DNA viruses from the contaminated shellfish using PCR, preparation method of template DNA is an important factor to get enough copy number of viruses. In this study, we evaluated the efficiency of PCR template of Megalocytivirus (sT50mg-D) DNA obtained from 50 mg digestive gland homogenate of oyster using commercial method, and compared with that obtained from 5 g of the same tissues (T5g-D) after PEG precipitation procedures of virus. Both templates DNA suspended in the same volume of distilled water showed positive results by primary PCR with 35 cycles, and the presence of Megalocytivirus was confirmed in oysters collected from cultured farms in Korea. Moreover, PCR with sT50mg-D allowed us to discriminate the contaminated oyster individually, that can not be done in PCR with T5g-D prepared from the mixture of three different individual oyster to get 5 g digestive gland homogenate. In quantitative analysis with real time PCR, Megalocytivirus concentrations in 50 ${\mu}l$ templates prepared using 0.5~50 mg of one positive sample were appeared in the range 6.14E+00~1.2E+02/${\mu}l$. We were not able to get positive result using template DNA contained less than 6.14E+00 copies. Consequently, 2-step PCR performed with DNA extracts from oyster homogenate of small amount (sT50mg-D) i) was enough to detect the contaminated Megalocytivirus in shellfish, ii) allowed us to do the analysis for individual shellfish rather than mixture of several shellfish and iii) showed the presence of Megalocytivirus in oyster from Korea.
The purpose of this paper is to compare China's and Korea's marine fisheries industries in order to present the main features of China's marine fisheries and show the comparative advantages they have in production. The results indicate that China's marine fisheries have the following features. (1) The marine proportion of the fisheries industry economic activity is less than 60%. Fishing is 71.3% of that marine activity. (2) The production trends of marine fisheries history in China can be classified as follows: 1) an early growth period, 2) a deliberation/consolidation period, 3) a second growth period, 4) a third growth period, and 5) a fourth growth period. The growth rate has rapidly increased recently. (3) Fish production is over 70% of marine fishing fisheries, the next major product is crustacea. The production of shellfish occupies over 70% of marine aquaculture, seaweed production however, is only 22% of total marine aquaculture. (4) The licensed area for marine aquaculture in China is 586.3 ha and that area is 5.4 times larger than that of Korea. The allotted area for shellfish aquaculture is 60% of marine aquaculture, production areas of crustaceans occupy 27.3%, fish has 7 1%, and seaweed production only 5.7% of allocated marine aquaculture areas. (5) The proportion of power vessels for marine fisheries of China's total power vessel fleet is around 65%, and the marine fisheries portion of non - powered vessels constitutes only 12%. The highest proportion of power vessels engaged in marine fisheries activities is between 10 tonnes to 100 tonnes. (6) The portion of marine fishery workers of all fishery industry employees is 22%, and 70% of them are full - time workers. Of marine fishery workers, 64% are in the fishing sector, 22%, aquaculture workers, and the number of employees in marine fisheries is increasing every year. The analysis of China's fishery industry in the production competitiveness indicates as follows : (1) The licensed areas in marine aquaculture, number of fishing vessels, number of marine fishing workers in China's fishery industry are much more than those of Korea's. Therefore China is much more competitive than Korea in the quantity of production side. However, licensed areas for seaweed aquaculture are more extensive in Korea than China. In China, the number of power vessels of between 10 tonnes and 100 tonnes, the licensed shellfish aquaculture areas, and the number of fishing workers within the fisheries industry are much more than those of Korea. (2) It is estimated that the licensed areas in marine aquaculture, number of medium sized power vessels, number of marine fishery workers will be increased as the quantity of production factors grow in China. (3) At present, yield per Ha. in marine cultures is very low in China. Therefore it is estimated that aquaculture techniques have only been diffused recently in China. Yield of fish per Ha especially is much lower than that of Korea. So the level of aquaculture techniques seems much lower than that of Korea. (4) China is behind Korea in production technique, however the number of HP per boat in China is lower than that of Korea. Therefore, China is much more competitive than Korea in Costs. (5) Average fish catches per marine fishery worker in China is only 1/3 that of Korea's, and average marine aquaculture production in China is only 1/2 that of Korea. Therefore we can say Korea is more competitive than China in efficiency. The average income of marine fishery workers in China is higher that that of other Chinese industries. However, the competitiveness of the fisheries industry in China will be increased as more capital is invested and advanced techniques are developed.
One hundred twenty healthy college students and housewives from urban middle income household were investigated in terms of food consumption patterns, nutrient intake and dietary fiber intake. Total food intake was slightly lower in college students(1059.9g/d) than in housewives(1160.g/d) . However, the ration of animal food to total food was considerably higher in college students(23.7%) than in housewives(16.8%). College students tended to consume more dairly products, meat & products, and cereals & grain products but less vegetables , fruits, fish and shellfish than housewives. Although nutrient intake was not much different (except for total calories, fats, and ascorbic acid), dietary fiber intake was slightly and significantly lower in college students(14.9g/d, 8.3g/1000kcal) than in housewives(16.9g/d, 103g/1000kcal) . That is partially due to college students irregular food habits and the change in their meal patterns from traditional to western diets. There was a significantly positive correlation between dietary fiber intake and thiamin, riboflavin, niacin and crude fiber intake.
Clam, Tapes japonica, has been widely used and occupies an important position in the markets. More than thirty kinds of fermented fish and shellfish are presently available in the markets in korea. This study was attempted to establish the basic data for evaluating the taste compounds of the clam. The free amino acids as taste compounds during the fermentation of the clam, Tapes japonica, were analyzed by an amino acid autoanalyzer. Alanine (17.0%), glycine (15.5%), taurine (13.9%) were high in amount in the fresh clam and then glutamic acid, arginine, tyrosine, asparagine, lysine, threonine, serine and leucine were the next in order. Such amino acids as methionine, phenylalanine, histidine and cystine were low in amounts. The total free amino acid nitrogen in the fresh clam was 45.5% of its extract nitrogen. During the fermentation of the clam, the kinds of the free amino acids were the same with the fresh samples although the amounts were changed. It is believed that glutamic acid, aspartic acid, glycine, alanine, lysine, and leucine may play an important role as the taste compounds in the fermented clam.
Minamata disease made its first appearance in the world at Minamata City, Kumamoto Prefecture, in May 1956. In 1962 methyl mercury poisoning through the placenta was found for the first time in the world. This was called congenital Minamata disease. In all cases the clinical symptoms were consistent with those of cerebral palsy. The time and place of outbreak were the same as those for Minamata disease. Their mothers had eaten fish and shellfish during pregnancy. The principal symptoms of congenital Minamata disease are mentalretardation ($100\%$); primitive reflexes ($100\%$); disturbance of coordination ($100\%$); dysarthria ($100\%$); limb deformation (100%); growth disorders ($100\%$); nutritional disorders ($100\%$); chorea-athetose ($95\%$); and hypersalivation ($95\%$). However, today, when the world is polluted by mercury in various places and at various levels, the data we need is not represented by those severe cases, but rather by the chronic milder type. Even in Minamata, the issue of Minamata disease has not been resolved. And likewise, on a global scale the problem of Minamata disease is not yet over.
The causative agent for the tunic softness syndrome of the cultured ascidian Halocynthia roretzi from Jan 1999 to Feb 2009 was identified using virus isolation and polymerase chain reaction (PCR). The pathogenicity of the isolated virus MABV UR-1 strain was determined by experimental infection trials. The cytopathic effects was observed in CHSE-214 cell line at a level 5.1% (4/78) in normal ascidian and 1.8% in abnormal ascidian showing tunic softness syndrome signs. MABV gene was detected in 16.8% (18/107) of normal and 13.1% (5/38) of abnormal organisms by PCR. The ratio of MABV isolation and gene detection was similar level in normal and soft tunic diseased ascidian. Based on the VP2/NS junction region sequences, eight strains of virus isolated from ascidian, were included in the same genogroup with MABV which is originally isolated in wide ranges of marine fish and shellfish species. The UR-1 strain caused 60% mortality (36.5% mortality in control group) by immersion infection and 37% mortality (same mortality in control group) in injection infection indicating no significant differences in infected and control groups. These results suggest that ascidian can act as reservoir of the MABV, and this virus is not directly related with the ascidian mortality.
The study examined overuse of the fishery resource. Influence of fishing activity was estimated by application of Schaefer model's. Fishing efforts that produced the maximum sustainable yield were determined in the model, allowing the effect of overfishing to be assessed. In the model, a wide variety of fish species as well as crustaceans and shellfish were susceptible to overfishing, while mollusks were not. Overfishing by modern techniques exacted a greater toll than more traditional methods. The results of the modeling study suggest that the 'Buy bag' input-control system of fisheries resource management warrants consideration, as does modernization, expansion and strengthening of self-control management of the fishery resource. Finally, more effective efforts in dissemination of policy information and education concerning the fishery resource are needed.
Objectives: The purpose of this study was to analyze the factors affecting Bisphenol A (BPA) exposure in children and adolescents using the results of the Korean National Environmental Health Survey (KoNEHS) cycle 3. Methods: A total of 2,380 subjects (n=571, 887, and 922 for 3-5, 6-11, and 12-17 years of age, respectively) were analyzed using an environmental exposure survey and environmental chemical substances concentration levels. Univariable linear regression analysis was performed to determine associated variables such as sex, age, income level, housing type, secondhand smoke time, cup noodles and canned food consumption, seafood consumption, new furniture (within the previous six months), drinking water type, and consumption of herbal medicines. Variables with p-values of less than 0.2 were extracted from the results and a multivariable linear regression analysis was performed using stepwise selection. Results: Univariable linear regression analysis showed positive associations between BPA concentration levels and variables including sex, age, secondhand smoke time, new furniture (within the previous six months), renovated living space (within the previous six months), fish and shellfish consumption, plastic-bottled drink consumption, and herbal medicine. As a result of performing multivariable linear regression analysis, the lower was the age the higher was the concentration of BPA levels. Additionally, women showed higher BPA levels than those of men. The more frequently fish was consumed, the higher was the BPA concentration. Moreover, higher BPA concentrations were observed when taking herbal medicine. Conclusions: The main factors affecting BPA concentration levels were age, gender, and consumption of fish and herbal medicine.
In this study, we have compared the genome of marine birnavirus (MABV) detected from starry flounder Platichthys stellatus and olive flounder Paralichthys olivaceus. A molecular analysis based on the nucleotide sequence (433 bases) of VP2-NS-VP3 region revealed that MABV (08-KU) from starry flounder showed 98% similarity with MABV Y6 isolated from Yellowtail Seriola quinqueradita in Japan (Accession no: AY283781) and with other aquabirnaviruses identify more than 76%. Comparison with MABV strains (06-KP, 08-KC) from olive flounder and MABV Y6 strain showed 97-98% sequence identities. Phylogenetic analysis was performed in order to examine the relationship among previously determined aquatic birnaviruses isolates showed that MABV and IPNV strains were classified into seven clusters. Three isolates from starry flounder and olive flounder in this study, belong to the genogroup VII including MABV Y6 strain and other aquabirnaviruses isolated from marine fish and molluscan shellfish in Japan. This report is the first description of a MABV from starry flounder in Korea.

The purpose of this study is to investigate and analyze the relationships between tidal cycles, surface landforms, and sediments of tidal flats and their resident life. The study areas are Uengdo Ri in the Garolim Bay with wide the mud flat, Doou Ri in the Youngwang tidal flat which is developed sand flat and salt field, and Gomso Bay which is developed aquaculture industry. For resident around tidal flats, sea working takes precedence over farming, and main earnings gets from tidal flats. Resident life around the tidal flats is deeply related to tidal cycles, and low water level of spring tide becomes, they are most active and get more earnings than usual day, so it seems to be periodic markets. It is usually developed shellfish aquafarms in the mud flat, the stow net fishery in the sand flat, and salt fields in the mixed flat near the coast. Also a tidal flat has specialization of economic activity, and is divided into salt field and fish-farming in the supratidal zone, shellfish aquafarm in the intertidal zone, and oyster and porphyra culture in the subtidal zone.
