Similarities of Scaritoxin to Ciguatoxin on the Chromatographic Behaviours (Scaritoxin과 Ciguatoxin의 크로마토그라피상에서의 몇가지 유사성)

  • In studying the structural work on ciguatoxin, parrot fish collected were identified as Scarus sordidus, S. frenatus, S. scaber and S. pectarlis, in which only S. sordidus contained toxic materials. Crude toxins obtained by silicic acid column chromatography, could be separated on a DEAE-cellulose column into two fractions, ST-1(less polar) and ST-2(polar) eluted with chloroform and chloroform-methanol(1:1). Furthermore ST-1 could be changed into ST-2 by repeated chromatography on DEAE-cellulose. Rf values of ST-1 and ST-2 were 0.60-0.75 and 0.30-0.54 on TLC coated with silica gel 60F-254 developed by chloroform-methanol-water-acetic acid (90:9.5:0.2:0.3) mixture. The peaks of ST-1 and ST-2 were not observed on each HPLC chromatogram at low sensitivity(2X), but by bioassay they were detected in the fraction of 24-27ml(less polar toxin, 120ng) and 22-27 ml (polar toxin, 150 ng). Less polar ciguatoxin from morey eel viscera also showed its peak in the same elution volume(25ml). Being subjected to chromatography on basic aluminum oxide (activity grade I) or to alkaline treatment, followed by basic aluminum oxide (activity grade I) chromatography ST-1 toxin was remarkably converted into the polar toxic component supposed to be polar ciguatoxin in both cases. In the latter case, approximately 74% of the residual toxicity was changed into the polar component, accompanied by about 50% loss of the initial toxicity. More than 26% of ST-2 toxicity was transformed into the less polar toxic component supposed to be less polar ciguatoxin on a deactivated aluminum oxide (activity grade V) column.

REARING OF THE EEL ANGUILLA JAPONICA IN RECIRCULATING AQUARIUMS (뱀장어 양식에 관한 연구 (순환여과식 이용))

  • From August 1976 to May 1977, a series of rearing experiments of eels weighing over 5 grams were carried out utilizing indoor recirculating aquariums and the results are summarized as following: 1. The eels were instantly acclimatized in the aquarium when alive Tubifex was fed as food, resulting in the food coefficient of around 5, and the daily growth rate of $2\%$ or so (Table 2). 2. When mackerel flesh was used in combination with flour dough or commercial powdered feed, the food coefficients were 4 to 5 and daily growth rates were between 1 and $2\%$ (Tables 3 and 4). 3. The eels of 12.4-14.7g in average weight which had not shown any growth when fed processed feed, grew normally when they were fed alternately with alive Tubifex and processed feed with the results of 4.3-6.0 in food coefficient and $1.4-2.3\%$ in daily growth rate (Table 5). 4. Experimental processed feed containing North Pacific which fish meal as the main ingredient showed the food coefficient of 1.31-1. 83 as dry material and this means that there is not any significant difference between this experimental feed and the control commercial eel feed, imported front Japan which showed food coefficient of 1.34 and 1.328 (Tables 6 and 7). 5. The feed cost may be reduced by about $45\%$ (based on tile domestic prices in the spring 1977) if this experimental processed feed is used instead of imported commercial feed. 6. Uneven growth is markedly significant in eels, and those which showed retarded growth gave very poor food efficiency as well as poor growth rate until they reach the size of about 30 grams. Thereafter they recovered both the normal food coefficient and growth rate. 7. Individuals which have been showing significant retarded growth may have some inherent physiological factors but this poor growth might also be, more or less, results of some external factors which are considered necessary to be investigated.

A Study of Food Taboos on Jeju Island (I)-Focused on Pregnancy- (제주지역(濟州地域)의 식품금기(食品禁忌)에 관(關)한 연구(硏究) (I)-임신기(妊娠期)를 중심(中心)으로-)

  • Nutrition counselors in Korea often encounter difficulty in their attempt to change village women's attitudes regarding food taboos which are counter to good eating habits. There are a great many food superstitions which are not due to religious influence, but seem to be related to shape and composition of food. Many expectant mothers superstitiously avoid eating certain foods for fear that they may cause mental or physical abnormality in their babies. As was shown in a previous survey (Mo, 1966)of villages in all provinces except Jeju Island, such superstitions were common among pregnant and lactating mothers. Many food taboos and superstitions based on non-scientific and irrational ideas do exist even in modern society, and are a major obstacle to nutritionally adequate food consumption. A study of food taboos among women of Jeju Island was undertaken from November to December of 1976, these results to be compared as well with those of the previous study. There were 73 items found to be prohibited during pregnancy. Of these, 48.7% were of the deaf group, 17.4% fish, 5.5% eggs, 4.7% cereal, and only 2.2% fruit. Of 252% women respondents, 111 (45% ) abstained from eating chicken, duck, and shark because of the belief that they would cause their babies to be born with gooseflesh or shark skin. Many of them avoided rabbit meat for fear that their babies might be born with harelip. It was also feared that a baby would become disfigured if his mother ate duck, goat, dog meat, chicken or duck eggs, or soup made of bones. A common superstition was that highly spiced or salty foods would cause the fetus to be hairless. Squid and octopus were believed to cause babies to have weak bones, or none at all. Most of these food taboos were associated with fears concerning Physical structure and appearance of unborn babies. Other taboos were associated with fear of undesirable behavioral characteristics. For example, some mothers thought that a baby would pinch or bite the mother's breast during the weaning period, if crab meat were eaten during pregnancy. Unevenly sliced rice cake, loach, snake meat and eel were also believed to cause a baby to be ill-tempered. The findings of this study are remarkably similar to those of the previous study conducted by the authour in 1966. Most of the same food taboos, based on non-scientific and irrational reasons, were found on Jeju Island as on the peninsula, and thor were similarly wide-spread. The results of correlational analysis show that the most significant factors related to prevalence of food taboos, are level of education and religious background. Number of food taboos is correlated with level of education. Also, food taboos are least freqent among the Christian woman. Proper nutrition education should he undertaken in order to encourage intake of protein-rich food, particularly during pregnancy when nutritional needs of mother and fetus are great.

Contents of Arsenic in Some Fisheries Caught in Western Coast (서해안 지역 수산물의 비소함량에 관한 연구)

  • In Korea, a small proportion of fishery products are contaminated with a considerable amount of potentially hazardous contaminants. So, there is generally a high risks to consumers. Inorganic contaminants with the greatest potential for toxicity are Sb, As, Cd, Pb, Hg, Se and the Sulfides. As appears to be much more variable than the others. This study was performed to assess the levels of the As in fisheries caught from the waters of the western coast of Korea. The samples included 26 kinds of fishes, 18 kinds of Mollusca, 3 kinds of Crustacea and 7 kinds of salted fishes. The Moisture content of molluscans was significantly higher than that of others (p<0.05). And the As contents of crustaceans were significantly higher than those of fishes, molluscans and salted fishes (p<0.05). The As content of Turban shell was 51.62 ppm, which was the highest among the samples. Also, the As contents of eel, cuttle fish and blue crab were found to be higher than in other fisheries. This variability offers a solution for the reduction of exposure to As through restricting the harvest of aquatic organism from the western coast of Korea.

Study for fishery management measure on gape net with wings with catch composition in the water of Jin-do (진도해역 낭장망의 어획물 실태에 따른 어업관리 방안)

  • In this study, we conducted field surveys to investigate and analyze species and length composition of catch from gape net with wings being set in two areas - Masari, Supomri - located in southern part of Jin-do for understanding fishing characteristics of gape net with wings fishery from March to September of 2011. And with those results, we attempted to propose fishery management strategy to harvest fisheries resources continuously. Catch obtained from field surveys comprised total 78 species, which accounted for 53 species of fish, 20 species of crustacean, 4 species of cephalopod and one of polychaete. As to species composition in weight, it showed that there was 88.5% for pisces, 9.0% for cephalopoda and 2.5% for crustacea. As to catch specifics by area, there was 36 species (596,891 individual, 456,551g) of pisces, 17 species (35,815 individual, 12,909g) of crustacea and 3 species (2,876 individual, 3,004g) of cephalopoda in Masari area. In Supumri area, there was 41 species (396,898 individual, 281,457g) of pisces, 15 species (16,113 individual, 7,772g) of crustacea and 4 species (6,792 individual, 72,329g) of cephalopoda and one polychaeta (36 individual, 11g). Catch of anchovy by month recorded on high level on June, July, September in both areas. When considering size composition by month, it was found that new populations recruit to these fishing grounds on June and September. Most of bycatch species including hairtail (Trichiurus lepturus), dotted gizzard shad (Konosirus punctatus), conger eel (Leptocephalous), Pacific sandlance (Ammodytes personatus), horse mackerel (Trachurus japonicus) were juvenile, and grass puffer (Takifugu niphobles), Kammal thryssa (Thryssa kammalensis), Japanese sardinella (Sardinella zunasi) were also small in size. As a result, it is considered that gape net with wings fishery which is likely to catch juvenile needs to be set fishing prohibit period for conserving above stocks during the period excluding main fishing periods of anchovy or fisheries management such as expanding mesh size for preventing juvenile catch.

  • Preference on protein rich foods of 103 children, aged 4 to 6, of kindergarten located in Masan Jin-hea, Chang-won cities, was conducted April 25 to May 4, 1983. The results are summarized as follows ; 1) Seventy six percent of the subjects were from families having two three children, without grandmothers in the home About 43% of mother had completed high school, 37% had completed middle school. Mothers' ages ranged from 26 to 50 years, with 57.3% in the 31-40 age bracket. About 63% of the fathers were office and government employees, while 77.6% of mothers were unemployed. 2) Over 50% of the children disliked or refused pork, thick beef soup, liver, soybean, oyster, clam, anchovy, croaker, mackerel, loach, hairtail, porgy, flatfish, a walleye, pollack, a dried walleye pollack, a sciaenoid fish. Taste bad, the characteristic fragrance and lack of experience were the main reasons why the children refused these foods. Over 33% of the children were unexperienced liver, ribs of beef. the small intestine of cattle, a loach, an eel. Over 50% liked milk, egg, dried filefish, sausage, a cattlefish, beef, chicken, a crab, shrimp, bean curd. 3) Most of children hoped that their daily meals should be made in prettier, more sabory, and various ways. Mothers also hoped to have practical knowledge of nutrition and its influence on our body.

Multi-Residue Analysis of 18 Dye Residues in Animal Products by Liquid Chromatography-Tandem Mass Spectrometry

  • This study aimed to develop an analytical method for determination of 18 dyes in livestock and fishery products by liquid chromatograph-tandem mass spectrometry (LC-MS/MS). The developed method was validated for linearity, accuracy, limit of quantifications (LOQ) and recovery based on the CODEX guideline (CAC/GL-71). Target matrices (beef, pork, chicken, egg, milk, flatfish, eel, and shrimp) were extracted using acetonitrile (containing 1% of acetic acid) and then, purified with C18 and primary secondary amine (PSA). Calibration linearity was obtained (r2>0.98) and LOQs were 0.002 mg/kg in animal products. The recoveries of dyes were ranged from 63 to 112% and relative standard deviations (RSDs, %) were less than 15%. The residues of 18 dyes were investigated in real samples (n=124) collected from retail markets in South Korea. As a result, a total of seven samples showed positive results for target analytes in fish samples. However, there was no violation according to the maximum residue limits set by the Korean Food Code. The proposed method will be used for routine analysis of dye residues in livestock and fishery products.

Monitoring of Veterinary Drug Residues in Cultured Fishery Products in Chungcheongnam-do Province (충남지역에서 생산되는 양식수산물의 잔류 동물용의약품 모니터링)

  • For this study, we conducted a simultaneous multiresidue analysis of veterinary drugs in cultured fishery products in Chungnam Province in 2018. A total of 115 fishery product samples were obtained from fish farms and fishery production sites located in the province. In all, 29 residual veterinary drugs in the samples were analyzed using a validated liquid chromatography-tandem mass spectrometry (LC-MS/MS) method. As a result, veterinary drug residues were only detected in a small number of the 106 samples (92.2%), and the detection rate was 7.8% (9 of 115 samples). The amounts were also below maximum residual limit (MRL) for fishery products, although one sample exceeded the MRL allowed by the Ministry of Food and Drug Safety and was detected in loach. The nine residual veterinary drugs were detected in 8 samples: loach, eel, catfish, freshwater bream, flatfish, rockfish and shrimp. The detected veterinary drugs were oxolinic acid, enrofloxacin, ciprofloxacin, sulfadiazine, flumequine and oxytetracycline. The most frequently detected antibiotic was oxolinic acid, and enrofloxacin exceeded the MRL in loach sample. Residues of most veterinary drugs were either not detected or were below the MRL, and while the status of fishery products is seen as safe overall, current surveillance efforts over veterinary drugs should be continued.

Induction of Sexual Maturation in Female Eels (Anguilla japonica) Using an Osmotic Pump (Osmotic pump를 이용한 암컷 뱀장어(Anguilla japonica)의 성성숙 유도)

  • For the artificial induction of the sexual maturation of Anguilla japonica, salmon pituitary extract (SPE) is continuously injected into females, and the eggs obtained from artificial sexual maturation are artificially fertilized with sperms and hatched. However, repeated injection of SPE in the abdominal cavity causes tremendous stress in females, which may prevent their complete sexual maturation and reduce the immune system function, ultimately resulting in death. In addition, the poor quality of the ovulated eggs can reduce the hatching and survival rate of larvae. In the present study, sexual maturation of females was induced by inserting an osmotic (OS) pump containing hormone analogs known to effectively induce sexual maturation into the abdominal cavity of female eels, and the effect of the OS pump on the induced sexual maturation was investigated. Our study results showed that the gonadosomatic index (GSI) was significantly higher in the fish subjected to SPE injection than those subjected to human chorionic gonadotropin (hCG), gonadotropin-releasing hormone analog (GnRHa), and methyl testosterone (MT) injections, either separately or in combination. In addition, a histological analysis showed that the oocytes in the SPE OS pump groups were more mature (entered the nuclear shift stage) than those in the other groups. These results suggest that an osmotic pump containing hormone analogs can be used to induce sexual maturation in female A. japonica artificially.

Detection and Control of Bacterial Diseases of Cultured Fishes in Korea (양식어류(養殖魚類)의 세균성질병(細菌性疾病)의 진단(診斷)과 대책(對策))

  • This is a comprehensive study for considering the effective treatment and control program of bacterial disease occurring in common carp, israel carp, color carp, crucian carp, eel and tilapia by clarifying the causes, mechanism of infection and onset and the diagnostic criteria. As a first step, the authors investigated the external views, gross and histopathologic findings of diseased fish using 450 infected fishes obtained from various farmer of Korea. This infection was characterized by hyperemia, hemorrhage and swelling of body surface and fins, congestion of liver, spleen, kidney, inflammation of intestine, hemorrhagic inflammation of various tissues, and necrosis and ulcer of various tissues were accompanied in serious cases. Bacteriologically, Aeromonas hydrophila and Edwardsiella tarda were isoiated from these fishes. Particularly in the regular check on 222 eels, 177 strains were isolated as 29.94% of Aeromonas hydrophila, 48.58% of Edwardsiella tarda and 21.47% of Flexibacter columnaris. Hexibacter columnaris was isolated from corroded gill of eels. The identical disease was occurred by innoculating the isolated Aeromonas hydrophila and Edwardsiella tarda and the identical strains were isolated from infected experimental fishes. The eels which were diagnosed Aeromonas disease from Kwangju, Pusan accompanied hemorrhage, swelling of body surface and fins, inflammation of stomach and intestine containing mucous fluids mixed with the pathogens. Color carp and crucian carp which were innoculated with the isolated 5 strins of Aeromomas hydrorphil died within 3 or 4 days accompanying with the characteristics of Aeromonas disease. Edward disease was characterized by abscesses of body surface, pus formation with concentration on phagocytes. The size of absecsses increased with progression elf disease. There were also various abscesses at internal organ and white nodules appeared in kidney. Histologically, various progressive granuloma were examined without inflammation of intestine. Columnaris disease of eels showed no hemorrhage except slight white body color. In autopsy, most of internal organs appeared normal and there were no septic odors. The only character was corrosion of gills. In order to treat these bacterial diseases, infected fishes must bathe in 20ppm chloramphenicol or kanamycin solution for 1 hour. Besides, medication program in oral ingestion of 75mg/kg chloramphenicol per day continuing for 5 to 7 days. After injecting the formalin treated Aermonas hydrophila antigen into carp, relatively high agglutination titer showed between 3 weeks and 6 weeks. Though this titer decreased from that time, it was continued for 18 weeks. In the case of injecting the formalin treated Edwardsiella tarda antigen into tilapia, the titer also increased. But tilapia which were immersed in the suspension fluid of the formalin treated Edwardsiella tarda showed no increase of the titer.
