야생 잎새버섯 물추출물 및 유기용매 분획물의 생리활성 탐색 (Studies on the biological activity of water extract and solvent fractions of wild Grifola frondosa)

  • 야생 잎새버섯 추출물을 이용하여 심혈관계 질환과 관련된 기능성 식품 개발을 위한 기초자료를 얻기 위해 준비한 물 추출물과 유기용매 분획물의 혈전용해활성과 트롬빈저해활성, acetylcholinesterase 저해활성, 항산화활성을 확인하였다. 10 mg/mL의 농도에서 물 추출물과 물 분획물은 각각 0.93 plasmin unit와 0.73 plasmin unit의 높은 혈전용해활성을 나타냈으며, 부탄올 분획물은 79.60%의 트롬빈저해활성을 나타냈다. 클로로포름 분획물은 85.88%의 높은 acetylcholinesterase 저해활성을 나타냈고, 물 추출물의 항산화 활성은 39.81%로 비교적 낮게 나타냈다. 부분 정제된 효소를 포함하고 있는 물 분획물은 섬유소원의 Bβ chain를 모두 분해시켰지만, Aα chain은 느리게 분해시키고 γ chain 과는 반응하지 않았다. 실험 결과로부터 야생 잎새버섯 추출물은 높은 혈전용해활성과, 트롬빈저해활성, acetylcholinesterase 저해활성을 나타내 심혈관계 질환 예방을 위한 제약과 기능성식품 개발에 이용 가능할 것으로 기대된다.

제조 추출물의 혈전 용해 활성에 대한 실험적 연구 (An Experimental Study on the Thrombolytic Activities of Holotrichia Extracts)

  • 1. Objectives This study was performed to find the activities and characteristics of purified thrombolytic enzymes from Holotrichia extracts. 2. Methods In the first time, a coarse enzyme fluid was made by using the freedried Holotrichia extracts. After manufacturing total soluble proteins and purifing enzymes, it was evauluated the activities and characteristics of this enzyme's dissolving capability to fibrin and thrombus. This study was taken using azocasein assay, fibrin-plate method, native-PAGE and fibrin zymography. 3. Results A soluble proteins were efficiently extracted form freezedried Holotrichia extracts. And, this purified enzyme had a ten times fibrinolytic capability compare with ustulation Holotrichia sample. In native PAGE and fibrin zymography, Holotrichia extracts showed the respectable fibrinolytic activity. Also, It had higher thrombolytic activities compared with general thrombolytic enzyme 'plasmin'. In experiment of various protease inhibitors of the purified enzyme from Holotrichia extracts on the azocaseinolytic activity, the enzyme was strongly inhibited by EDTA ${\cdot}$ EGTA, and weakly by APMSF ${\cdot}$ PMSF ${\cdot}$ TPCK. 4. Conclusion Holotrichia extracts has the thrombolytic activities, and it will operate directly th fibrin-clot and thrombus.

아위버섯 열수추출물 및 유기용매 분획물의 생리활성 효과 (Physiological Activities of Hot Water Extract and Solvent Fractions of Pleurotus ferulea)

  • 아위버섯으로부터 성인병 치료와 예방 관련된 생리활성물질을 확인하고 이 물질의 추출을 위한 최적 조건을 확립하기위해 아위버섯 열수추출물과 유기용매 분획물을 사용하여 성인병 관련 생리활성을 측정하였다. 아위버섯 열수추출액에서는 작은 항산화효과. 트롬빈저해효과, 혈당강하효과가 확인되었지만 혈전용해효과는 나타나지 않았다. 그러나 에틸아세테이트 분획물과 부탄올 분획물에서 86.79%와 87.82%의 높은 항산화효과가 확인 되었으며, 에틸아세테이트 분획물에서 0.08 plasmin unit의 작은 혈전용해활성을 확인할 수 있었다. 클로로포름 분획물과 에틸아세테이트 분획물에서 74.90%와 71.88%의 높은 트롬빈저해효과와 함께 부탄올 분획물에서 49.67%의 혈당강하효과를 확인하였다. 따라서 아위버섯의 에틸아세테이트 분획물과 부탄올 분획물은 혈관계 질환을 위한 기능성 식품의 개발에 이용할 수 있을 것으로 판단된다.

Bacillus subtilis Natto가 생산하는 Nattokinase의 항혈전 및 피브린 용해능 효능평가 (The Evaluation of Antithrombotic and Fibrinolytic Activities of Nattokinase from Bacillus subtilis Natto)

  • We previously reported that Ultra nattokinase$^{(R)}$ showed high fibrinolytic activity and revealed antithrombotic effect in rat blood plasma based on its ability to suppress collagen-induced platelet aggregation. This research was carried out to verify the clot lysing activity and blood flow enhancing effects of Ultra nattokinase$^{(R)}$ via monitoring and comparing the antithrombotic effects in rat artery between oral administration of Ultra nattokinase$^{(R)}$ and maltodextrin. SD rats were fed with 1.11 mg/kg of Ultra nattokinase$^{(R)}$ for 4 weeks. The effect on arterial thrombosis was then evaluated using an antithrombotic model after induction by $FeCl_3$. Detected fibrinolytic activity was proportional to the content of Ultra nattokinase$^{(R)}$ and statistical extents of the antithrombotic activity was enhanced strongly twice rather than control group. The PT and the aPTT, however, showed only a small difference between two groups. The results suggest that Ultra nattokinase$^{(R)}$ can effectively treat thromboembolism and enhance blood flow, and that Ultra nattokinase$^{(R)}$ can also prevent venous occlusion by aiding clot lysis.

Improvement of the Functional and Organoleptic Properties of the Fermented Soymilk with Bacillus subtilis 2829PNU015 in Addition of Corni fructus Extracts

  • To improve the functional and organoleptic properties of the fermented soymilk with Bacillus subtilis 2829PNU015 which has chungkukjang like flavor, functional materials from the edible plant extracts with well-known health promoting effects were examined by fibrinolytic activity and sensory evaluation. Among five samples such as Corni fructus, garlic, purple sweet potato, aged garlic, and pumpanini were tested, Corni fructus showed the highest fibrinolytic activity. The appearance and taste of the fermented soymilk (FS) was varied to the amount of Corni fructus added, and the highest acceptability of FS was observed from FS containing 1% Corni fructus extracts ($10^{\circ}$Brix) in it. Best way of applying Corni fructus to the fermented soymilk (FFS) was found to be incubating the mixture of soymilk and Corni fructus with Bacillus subtilis. FFS demonstrated dose dependent radical scavenging activities and these activities were found to be higher than those of FS at designated concentration.

죽엽의 생리활성 및 죽엽분말 첨가 냉면의 제조 (Biological Activities of Bamboo Leaf and Quality Characteristics of Buckwheat Cold Noodle Using Bamboo Leaf Powder as a Functional Ingredient)

  • This study was conducted to determine the usefulness of bamboo leaf powder as a functional ingredient for buckwheat cold noodles. The anti oxidative and fibrinolytic activities of bamboo leaf powder were very high, and were further maintained or improved after strong heat treatment. We prepared buckwheat cold noodles using 0%, 3% or 5% of bamboo leaf powder. Only the cohesiveness of the noodle having 5% bamboo leaf powder was lower than that of the noodles contained 0% and 3% bamboo leaf powder. The L-, a- and b-values were significantly different between noodles which contained bamboo leaf powder and those which did not. Nevertheless, the amount of bamboo leaf powder didn't affect the color parameters of buckwheat cold noodles. According to sensory evaluation, the addition of bamboo leaf powder improved the score of color and texture of buckwheat cold noodles, but overall acceptance was not significantly affected. Cooked volume, cooking loss and turbidity of buckwheat cold noodle containing 5% bamboo leaf powder were lower than the others, indicating that adding 5% bamboo leaf powder prevented the softening of the noodles with moist heat. We concluded that the application of bamboo leaf for the production of buckwheat cold noodle will be a good way to intake the biologically active ingredients for good health.

Changes in Physiochemical Properties during the Fermentation of Doenjang Prepared with Black Soybeans

  • A physicochemical assessment of Doenjang (traditional fermented soyfood) prepared with Korean black soybeans (Glycine max) was carried out. The T-N rate increased slowly during storage up to 120 days and the A-N rate increased up to 80 days of ripening and then decreased slightly. The caseinolytic activity increased slowly during storage up to 80 days and then decreased after 80 days. In addition, the fibrinolytic and ${\beta}-glucosidase$ activities increased up to 80 and 30 days and then decreased. Genistin and daidzin concentrations gradually decreased with increased fermentation time. However, genistein and daidzein slowly increased with fermentation time. Genistein and daidzein reached maximum concentrations (316.8 and $305.2{\mu}g/g$, respectively) and plateaued thereafter. The anthocyanins increased greatly during fermentation up to 50 days and then remained constant between 50 and 90 days. Polyphenol contents showed a slight increase up to 80 days and then slowly decreased. The DPPH and ABTS radical scavenging activities increased linearly during storage up to 50 days, reached about 28.9% and 2.17 mg/g, respectively, and then slowly decreased. At 20 days of fermentation, macrophage-stimulating activity of the extract showed a maximum activity.

  • In order to produce a high quality and functional black bean Chungkugjang, a bacterium which has potent enzyme activities(protease:124.8 U/$m\ell$, $\alpha$-amylase: 78.2U/$m\ell$, glucoamylase; 13.9U/$m\ell$, ), was isolated by using the halo zone method and identified by morphological, cultural and biochemical properties, and then finally confirmed by GP-microplate identification system as Bacillus megatrium SMY-212. The isolated SMY-212 system was also high in the formation of mucoid material and fibrinolytic activity.

산야초를 이용한 기능성 발효음료개발 및 생리활성 연구 (Development of Functional Beverage (SanYa) from Fermented Medical Plants and Evaluation of Its Physiological Activities)

  • 지금까지의 연구에 의하면 산야 발효액에 대한 보고가 미비하므로 본 연구에서는 산야의 기능성에 관한 연구를 위하여 산야 발효액으로 여러 가지 생리활성과 면역활성에 대하여 분석하였다. 우선, 산야의 항산화 활성을 측정하기 위하여 DPPH 활성과 플라보노이드 함량을 측정하였다. 그 결과 DPPH법을 통해 측정한 산야 발효액의 항산화력은 94.3%의 radical 소거능을 나타내었다. BHA와 비교 했을 때 유사한 항산화력이 측정되었는데, 이것은 항산화 활성이 높은 것으로 산야의 항산화력에 관한 높은 이용가치를 의미한다. 산야 발효액의 총 플라보이드 함량은 4.646 mg/g으로 높았으며, 이는 높은 항산화능과 연관성이 있는 것으로 나타났다. 산야 발효액과 면역활성과의 연관성은 NO 합성률과 LPS에 의해 유도되는 NO 합성 저해률 분석으로 조사되었다. 그 결과, 산야 발효액은 무려 11배의 높은 NO 합성율을 보였다. 또한 LPS에 의해 유도된 NO 합성 저해율은 56%로 나타나 산야의 높은 면역효과를 증명하고 있다. 혈전분해능에 대한 산야 발효액의 분석 결과는 혈전용해제로 알려져 있는 plasmin보다 높은 활성을 보이고 있다. SOD 유사활성은 42.2%로 비교적 높은 SOD 유사활성을 나타내었고, xanthine oxidase 저해활성은 75.6%로 높게 나타났다. 항고혈압 측정실험에서는 73.5%의 저해율을 나타내어 ACE 저해 활성이 뛰어난 것으로 나타났다. 이상의 결과는 산야의 우수한 생리활성을 증명하고 있고, 항산화력, 면역활성 및 항고혈압 효과가 높은 것으로 나타나 기능성 음료의 소재로서 그 활용도가 높을 것으로 판단된다.

적포도주들의 발효와 후 발효 중 심혈관 관련 Angiotensin I 전환효소 저해활성과 혈전용해활성 및 $\beta$-secretase 저해 활성의 변화 (Changes of Angiotensin I-Converting Enzyme Inhibitory Activity, Fibrinolytic Activity and $\beta$-Secretase Inhibitory Activity of Red Wines During Fermentation and Post-Fermentation)

  • 본 연구는 4종류의 한국산 포도를 이용하여 포도주를 제조한 후 이들의 발효와 후 발효중의 심혈관 관련 angiotensis I 전환효소 저해 확정과 혈전 용해 활성 및 항치매성 $\beta$-secretase 저해활성을 조사하였다. 발효 10일 후 모든 시료 포도주들의 항고혈압성 엔지오텐신 전환효소(ACE)저해활성은 $38.6%{\sim}58.8%$ 이었다. 그러나 후발효가 진행됨에 따라 ACE저해활성은 증가하여 세리단(Vitis hybrid) 포도주가 후발효 120일 후 최고인 76.9%에 도달하였다. 혈전용해활성은 모든 시료 포도주들에서 미약하거나 없었다. 발효 10일 후, 켐벨어리(Vitis labrusca B) 포도주가 54.8%의 가장 높은 항치매성 $\beta$-secretase저해 활성을 보였으나 후발효 120일 후에는 10% 미만으로 현저하게 감소되었다. 결론적으로 본 연구에서는 세리단 포도를 S. cerevisiae K-7 효모로 $25^{\circ}C$에서 10일간 발효 시킨 후 $4^{\circ}C$에서 120일간 후발효 시켜서 고부가가치의 생리 기능성을 가진 세리단 적포도주를 제조하였다.
