Polysaccharide isolated from fermented barley extract activates macrophages via the MAPK and NF-κB pathways (보리발효추출물로부터 분리한 다당의 대식세포 활성화 및 신호 전달)

  • Barley has nutritional benefits due to its high dietary fiber content; therefore, the intake of whole barley grains is recommended. However, barley is often consumed in the fermented form because of the improved texture and digestibility. The present study was designed to elucidate the intracellular signaling pathway for macrophage activation by the polysaccharide BF-CP from fermented barley. BF-CP is a neutral polysaccharide, composed of neutral sugars, including glucose (70.7%), xylose (11.4%), and arabinose (9.0%). BF-CP exhibited macrophage-stimulatory activity by inducing the production of interleukin (IL)-6, tumor necrosis factor $(TNF)-{\alpha}$, and nitric oxide in RAW 264.7 macrophages. Further, BF-CP treatment strongly increased the IL-6 and $TNF-{\alpha}$ gene expression in a concentration-dependent manner. Signal transduction experiments using immunoblotting showed that BF-CP phosphorylated mitogen-activated protein kinases (MAPKs), such as c-Jun N-terminal kinase, extracellular signal-regulated kinase, and p38, and nuclear factor $(NF)-{\kappa}B$, in RAW 264.7 cells in a concentration-dependent manner. These results suggest that BF-CP activates the macrophages via MAPK and $NF-{\kappa}B$ pathways, and also induces an increase in the production of cytokines.

The Regional Folk Plants in Inland of Gyeongsangbuk-do (I) (경상북도 내륙지역의 민속식물 I)

  • This study was carried out to classify the regional folk plants in the inland of Gyeongsangbuk-do. According to the survey results from 582 survey sheets of 126 residents at 85 places of 17 counties and cities from March to October, 2009, the folk plants in the inland of Gyeongsangbuk-do consisted of a total 254 taxa; 76 families, 207 genera, 225 species, 2 subspecies, 26 varieties, and 1 form. The use by its usage was: 216 taxa; edible, 82 taxa; medicinal, 30 taxa; dye, 3 taxa; aroma, 4 taxa; spice, 4 taxa; ornamental, 1 taxa; fiber, 2 taxa; oil, 1 taxa; starch, 5 taxa; fuel, 9 taxa; others, respectively, so the edible use is the highest. The most useful part was the leaf, followed by stem and root. The folk plants of 3 head families from Yeongyang and Cheongsong were 42 taxa. The consistency comparison between the scientific name and the local name were the highest in the 50's and the lowest in 80's.

A Study on the Development of a Dry P0SCO E&C Fire Board Method with High Fire Resistance (건식화 P0SCO E&C Fire Board 공법 개발에 관한 연구)

  • The present study was to develop a dry PFB method similar to the existing gypsum board construction method in order to apply the existing wet PFB method that uses fire.resistant adhesive. It was found that the existing wet method can produce concrete compressive strength of 80MPa and fire resistance of 3 hours with 30mm PF boards. The goal of development in this study was fire resistance of 3 hours through dry construction of 15mm fire resistant boards. The results of fire resistance test showed an increase in thermal durability and thermal strain. It is believed that inorganic fiber reduces thermal strain, and lowers heat insulation performance by 15% or less. This suggests that heat insulation performance was improved by the change in the inner composition of PF board resulting from the adjustment of Al:Si mol ratio, high temperature molding, and dry curing. According to the results of fire resistance test, when the dry PF method was applied, the temperature of the main reinforcing bar was 116$^\circ$C in 15mm, 103.8$^\circ$C in 20mm, and 94$^\circ$C in 25mm, and these results satisfied the current standards for fire resistance control presented by the Ministry of Land, Transport and Maritime Affairs. When a 3 hour fire resistance test was performed and the external properties of the specimen were examined, the outermost gypsum board hardly remained and internal PF board maintained its form without thermal strain.

THE EFFECTS OF DECALCIFIED FREEZE-DRIED BONE AND SYNTHETIC BONE GRAFTS ON REGENERATION OF ALVEOLAR BONE DEFECTS IN DOGS (탈회동결건조골과 합성골이식재가 치조골 결손부 재생과정에 미치는 영향)

  • The purpose of this study was to compare effects of various bone grafts on periodontal regeneration of alveolar bone defects in dogs. Seven adult dogs aged 12 to 18 months were used in this study. Experimental alveolar bone defects were created surgically with a #1/2 round bur at the furcation area of the buccal surface of the mandibular 3rd, 4th premolars and 1st molar. Each experimental alveolar bone defects were grafted with dense hydroxyapatite, natural coral, and decalcified freeze-dried bone, and respectively divided into DHA, NC, DFDB group. An area without bone graft was divided into control group. At 1,2,4,6, and 12 weeks, dogs were serially sacrificed and specimens were prepared with Hematoxylin-Eosin stain and Mallory stain for light microscopic evaluation. The results of this study were as follows : 1. In control group, the matrix change of granulation tissue was observed at 1 week. And in experimental groups, the appearance of connective tissue around graft materials was loosely formed at 1 week, but densely formed at 2 weeks. 2. In every group, the slight formation of new trabecular bone was seen from remaining bone at 1 week. 3. The DHA and NC particles were gradually encapsulated by new trabecular bone from remaining bone, and the osteoid tissue was directly induced from DFDB particles. 4. The presence of osteoblasts was first observed at 1 week in control group and at 2 weeks in NC group, but at 6 weeks in DHA group. 5. In DHA group, the resorption of particles was not observed until 12 weeks. But in NC and DFDB group, the particles were resorbed at 6 weeks and replaced by new bone. And the amount and size of particles were reduced, and their border represented irregular form. In summary, in three experimental groups the inflammatory or foreign body reaction were slight, but the regeneration of new osteoid tissue and the matrix change of dense connective tissue fiber were observed. Especially, NC and DFDB materials were considered as the biocompatible graft materials which were effective in the regenertion of new bone.

Vascular Plant Species in the Southern Sejong (세종시 남부일대의 관속식물상 연구)

  • The flora of Sejong were summarized as 531 taxa including 104 families, 315 genera, 462 species, 3 subspecies, 59 varieties and 7 form. The rare plants were 8 taxa including Nepeta cataria L. and Aristolochia ontorta Bunge. The Korea endemic plants were 9 taxa including Populus tomentiglandulosa T. B. Le and Clematis trichotoma Nakai.. The specific plants were 42 taxa including Phytolacca esculenta Van Houtte(V), Corydalis decumbens (Thunb.) Pers(IV), Poncirus trifoliata Raf(III), Pseudostellaria coreana (Nakai) Ohwi(II), Cyrtomium fortunei J.Sm.(I). The naturalized plants were 49 taxa including Sicyos angulatus L, Ambrosia trifida L, Physalis angulata L. Panicum dichotomiflorum Michx., Galinsoga parviflora Cav, and Erigeron philadelphicus L. Especially, Sicyos angulatus L., Phytolacca americana L. and Bidens frondosa L. have been spread throughout the country are required to manage. The naturalized index(NI) and urbanized index(UI) were 8.7% and 15.3%. Useful plants incuding Edible was 199 taxa, Pasture was 193 taxa, Medicinal was 178 taxa, Ornamental was 82 taxa, Timber was 25 taxa, Fiber was 18 taxa, Stain was 9 taxa. Useful plants will be important materials for plant design in recreational and healing forests.

Exploration of suitable rice cultivars for close mixed-planting with upland-adapted cereal crop

  • In semi-arid countries such as Namibia, the flooding unexpectedly happens in a rainy season, causing losses in the yield of upland-adapted cereal crop. In flooding conditions, rice roots sequentially form aerenchyma and a barrier to radial oxygen loss (ROL), and oxygen is released into the rhizosphere near the root tips. Iijima et al. (2016) and Awala et al. (2016) reported that close mixed-planting with rice can mitigate the flood stress of co-growing upland-adapted cereal crop by modifying their rhizosphere microenvironments via the oxygen released from the rice roots. Moreover, by using the model system of hydroponic culture, it was confirmed that oxygen from rice roots was transferred to co-growing upland-adapted cereal crop in close mixed planting system (Kawato et al., 2016). However, it is not sure whether the ability of oxygen release varies among rice cultivars, because Kawato et al. (2016) used only one japonica cultivar, Nipponbare (Oryza sativa). The objective of this study was to compare the ability of oxygen release in rhizosphere among rice cultivars. The experiment was conducted in a climate chamber in Kindai University. We used 10 rice cultivars from three different rice species (O. sativa (var. japonica (2), var. indica (3)), Oryza glaberrima Steud. (2) and their interspecific progenies (3)) to compare the ability of oxygen release from the roots. According to the method by Kawato et al. (2016), the dissolved oxygen concentration of phase I (with shoot) and phase II (without shoot) were measured by a fiber optic oxygen-sensing probe. The oxygen released from rice roots was calculated from the difference of the measurements between phase I and phase II. The result in this study indicated that all of the rice cultivars released oxygen from their roots, and the amount of released oxygen was significantly correlated with the above-ground biomass (r = 0.710). The ability of oxygen release (the amount of the oxygen release per fresh root weight) of indica cultivars (O. sativa) tended to be higher as compared with the other cultivars. On the other hand, that of African rice (O. glaberrima) and the interspecific progenies tended to be lower. These results suggested that the ability of oxygen release widely varies among rice cultivars, and some of indica cultivars (O. sativa) may be suitable for close mixed-planting to mitigate flood stress of upland-adapted cereal crop.

TISSUE CHANGE AFTER EMBEDDING GELATIN MATRIX IMPLANT(FFIBREL®) IN SUBCUTANEOUS TISSUE OF RATS;HISTOLOGIC, IMMUNOHISTOCHEMICAL AND SCANNING ELECTRON MICROSCOPIC STUDY (백서의 피하조직에 Gelatin Matrix Implant (Fibrel®) 매식시 조직변화에 관한 연구)

  • GMI (Fibrel${(R)}$) is one of the dermal filling substances which have been successfully used for the treatment of depressed cutaneous scar and wrinkles. It's major components are; Gelatin powder, which provides a framework for the clot to form and remains stable under the scar, and ${\varepsilon}$-aminocaproic acid, which inhibits the production of fibrinolysin, and Plasma, which provides the necessary ingredients for collagen synthesis. GMI has advantages of low immunogenicity and increased longevity. It has been known to induce fibroblast activity and promote new collagen synthesis. We used 34 Sprague-Dawley rats which were bred under the same condition and duration. 18 of experimental animals were undergone cardiac puncture, and their blood were collected, centrifugated, and stored in freezer. Out of 16 animals, control group were injected with 2ml plasma into the subcutaneous tissue of Lt. scapular, while experimental group were implanted of 2 ml GMI into the Rt. same area. Experimental animals were sacrificed at the 3rd day, 5th day, 1st week and 2nd week respectively after implantation of GMI. To observe the histopathologic change of GMI and surrounding tissue reaction of GMI, we had examined with H&E staining, immunohistochemical staining with vimentin, ${\alpha}$-SMA, S-100 under LM and SEM. The obtained results were as follows ; 1. In LM study, the inflammatory cell infiltrations and granulation tissue formation were observed, and muscle tissues were well attached with adipose tissues in the control group. In the experimental group, inflammatory cell infiltrations had been observed by the 2nd week and irregular adipiose tissues and well differentiated mesenchymal tissues were examined. 2. In immunohistochemical study, the experimental group of ${\alpha}$-SMA study, there were a prominent positive response on endothelial development of granulation tissues and mesenchymal tissues compare with the control group. In vimentin study, positive response on mescenchymal fibroblast continued to 2nd week, but negative in the control group. In S-100 study, both groups were positively responded on irregular adipose tissues. 3. In SEM study, collagen fibers were embedded by the plasma by the 5th day in the control group, and in the 3rd day experiment GMI were resorved but communited with collagen fiber till the 1st week. Collagen fibers were infilt-rated into GMI at the 2nd week and the infilltrated GMI were conglomerated with the mature adipose cells and the collagen fibers. From the above results, GMI implantation in the subcutaneous tissue of Sprague-Dawley rat, the mild infiltration of inflammatory cells were showed till 2nd week and the granulation tissues were observed. GMI were nearly resorbed till 2nd week, but well attached with adipose tissue and collagen fibers. The endothelium and fibroblasts were actively proliferated. Adipose tissues and mesenchymal tissue cells were observed. As already expressed, GMI showed resorptive change in course of time without any early immune reaction, and seemed to induce fibroblast activity and promote new collagen synthesis.

An Experimental Study on the Effect of Cytoskeletal Changes or Osteoblastic Cell Activities (골모세포유사세포의 cytoskeletal change가 세포 활성에 미치는 영향에 관한 실험적 연구)

  • The cytoskeleton has been shown to form a network, connecting the extracelluar matrix via integrin with the nucleus and the cytoplasmic constituents of the cell. It is therefore assumed that the cytoskeleton may mediate signals generated by perturbations originating in the matrix. The purpose of this study is to examine the effect of cytoskeletal change on osteoblastic cell activities. The author cultured osteoblastic cells obtained from neonatal mouse calvaria. The cells were teated with cytochalasin B(CB) or colchicine (COL) at four concentrations for 3 hours and after another 24 hours the conditioned media was collected and assayed for prostaglandin $E_2\;(PGE_2)$, interleukin-6(IL-6), tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and matrix metalloproteinase-1(MMP-1). In addition, the cytoskeletal protein actin were observed by immuno-fluorescence. The results were as follows: 1. The production of $PGE_2$ showed the tendency to be increased in CB-treated group. $PGE_2$ was increased in COL-treated group dose-dependantly, 2. IL-6 production, in CB-treated group, was increased, except at 1.0 ${\mu}g/ml$. IL-6 was induced in COL-treated group. 3. TNF-$\alpha$ production was increased in CB-treated group, except at 1.0 ${\mu}g/ml$, and in COL-treated group, that was increased. 4. The MMP-1 production was decreased in CB-treated soup and was not changed in COL-treated group, which could be selectively visualized by immunoblotting with monospecific antibody. 5. The cytoskeletal actin stress fibers were disappeared and the cells showed to be rounded in CB-treated group. These results indicated that there are a relationship between the cytoskeletal rearrangements and osteoblastic cell activities, especially in release of paracrine/autocrine factors, such as $PGE_2$, IL-6, and TNF-$\alpha$.

Chemical Cleaning of Iron Stains on Ceramics (화학세척제를 이용한 도자기의 철산화물 제거 특성 연구)

  • To remove metal stains of the ceramics, chemical cleaning is essential case by case. This study investigated the removal characteristics of iron stains by oxalic acid and citric acid including their application methods of soaking and poultice. The soaking method in cleaning agents showed removal process by color difference and released iron contents from iron stains on ceramics. Iron stains were removed successfully from ceramics, which soaked in oxalic acid for 60 hours. However, it is recommendable to soak in 0.25M oxalic acid for one to three hours because most iron stains were disappeared in 3 hours soaking. Citric acid is less effective than oxalic acid in removing iron stains because of heavy molecular weight and low acidity. Poultices (bentonite, sepiolite, activated carbon fiber and celite) with oxalic acid were applied on contaminated ceramics. After ten hours, iron stains on ceramics were removed successfully by poultice. Among them, bentonite and sepiolite have better application. Therefore, sepiolite with 0.25M oxalic acid was applied on the iron stains of whiteware and celadon from Ma Island, and then stains were removed. However, it is judged that the application methods can be varied according to the form and depth of contaminant. In addition, the residues of poultice on the ceramics will be considered for preventing contamination.

Effect of Level of Crude Protein and Use of Cottonseed Meal in Diets Containing Cassava Chips and Rice Straw for Lactating Dairy Cows

  • The effects of different levels of crude protein (CP) and cottonseed meal replacing for soybean meal in cassava chips and rice straw-based diets for mid-lactating cows (100-150 day in milk [DIM]), were studied using 32 multiparous Holstein Fresian crossbred dairy cows. Diets containing 10.5, 12.5, 13.7, 14.4% CP of the rations and 0, 12.1, 14.9, 17.8% cottonseed meal were fed to cows for 60 days. Thirty two cows were randomly divided into four dietary treatments using a Randomized complete block design. Four dietary treatments were offered in the form of total mixed ration (TMR) with concentrate to roughage (chopped rice straw) at 60:40 and offered ad libitum. Dry matter (DM) and neutral detergent fiber (NDF) intakes tended to linearly increase with increasing dietary CP levels. Intakes and digestibility of crude protein increased linearly with increasing dietary CP level (p<0.01). Crude protein digestibility of the 10.5% CP diet was lower (p<0.05) than that in diets with higher levels of CP, while there were no significant differences among the other three levels of CP (12.5, 13.7 and 14.4%). Daily milk yield tended to increase with increased CP from 10.5 to 14.4%. Income over feed in terms of US$/kg of milk increased with increased CP from 10.5 to 13.7% and decreased when the CP level was higher than 13.7% (quadratic effect p<0.09). Milk composition was not significantly affected by increasing level of CP, however there were relatively high contents of protein and fat among treatments. The proportion of milk-urea N (MUN), ammonia-N ($NH_3$-N) and bloodurea N (BUN) were closely correlated and increased linearly with increasing CP levels (p<0.01). Balanced diet was found in diet containing 12.5 and 13.7% CP of the rations when BUN and MUN were used as indicators of the protein to energy ratio in the diet. Conclusions can be made that increasing dietary CP levels from 10.5 to 13.7% using cottonseed meal as the main source to completely replace soybean meal was beneficial to cows consuming rice straw and cassava chips based-diets. Increasing the CP level above 13.7% of total ration did not additionally improve milk yield and composition or net income.
