Poly(3-hydroxybutyrate) (PHB) production by fermentation was examined under both restricted- and ample-oxygen supply conditions in a single fed-batch fermentation. Recombinant Escherichia coli transformed with the PHB production plasmid pSYLl07 was grown to reach high cell density (227 g/l dry cell weight) with a high PHB content (78% of dry cell weight), using a glucose-based minimal medium. A simple flux model containing 12 fluxes was developed and applied to the fermentation data. A superior closure (95%) of the carbon mass balance was achieved. When the data were put into use, the results demonstrated a surprisingly large excretion of formate and lactate. Even though periods of severe oxygen limitation coincided with rapid acetate and lactate excretion, PHB productivity and carbon utilization efficiency were not significantly impaired. These results are very positive in reducing oxygen demand in an industrial PHA fermentation without sacrificing its PHA productivity, thereby reducing overall production costs.
Pichia pastoris, a methylotrophic yeast widely used for the production of heterologous proteins, was used to produce Hepatitis B surface antigen (HBsAg) under the control of the strong, tightly-regulated alcohol oxidase promoter. It is highly induced during the growth on methanol, but the presence of non-methanol carbon sources such as glycerol and glucose repressed fully the expression of alcohol oxidase. In this study, glycerol and sorbitol feedings for the expression of the recombinant HBsAg were compared to examine the potential of sorbitol as a less repressive carbon source in fed-batch fermentation. The sorbitol feeding enhanced the production yield by 12% compared to that in glycerol feeding, although the cell concentration was lower.
Poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] microspheres were prepared using solvent evaporation technique. P(3HB-co-4HB) with 3.9 mol% 4HB was synthesized by fed-batch culture of Ralstonia eutropha. The effects of concentration and type of surfactant (Tween 80, sodium dodecylsulfate, and polyvinyl alcohol), addition of dispersion stabilizer (Acacia), concentration of polymer and model drug (bovine serum albumin) on particle size of the microspheres and their in vitro drug release characteristics were investigated. The average particle size of the microspheres decreased with the addition of dispersion stabilizer and increased with the concentration of surfactant, drug and polymer. Amount of drug release increased with decreasing particle size of the microspheres.
A carriersupported mycelial growth of Sreptomyces erythreus was applied to erythromycin fermentation sistem using celite as a support material. Hyphal growth through the pore matrices of the materials showed anchorages and provided a stable biofilm growth. When the phospate concentration was limited to 0.8g corn steep liquor/L(corresponding to 40mg KH2PO4/L), the specific production rate of erythromycin was increased from 557$\mu$g/g-cell.hr under unlimited condition to 2, 898 $\mu$g/g-cell.hr. A fluidized-bed bioreactor was operated for erythromycin production by a repeated fed-batch mode. The control of free mycelial concentration and the extension of production phase were considered important to maintain the reactor productivity at a desired level. The erythromycin production under phosphate-limited condition could be maintained for at least 600hrs.
A synthetic wastewater composed of powdered baby food and inorganic nutrients was treated by five 3.6L anaerobic reactors in order to test the relationship between solids retention time (SRT) and media surface ratio, and the removal efficiencies of organic substrate. Of the five reactors, four were semi-continuously fed stirred-tank reactors and one continuously-stirred batch reactor. The influent was 7430mg/L in COD, 7120 mg/L $BOD_L$ and 6350mg/L in $BOD_5$, respectively. Operating temperature was $35{\pm}1^{\circ}C$ and pH in the range 6.9 to 7.2. In this experimental study it was found that a linear relationship existed, within the experiment range, between SRT and media surface ratio, and that SRT and removal efficiency increased with increasing media surface ratio. The substrate removal efficiencies were 82.7 to $88.2\%$ in COD, 82.9 to $88.4\%$ in $BOD_L$ and 83.3 to $88.7\%$ in $BOD_5$, respectively.

On the basis of high osmotic tolerance and xylitol production, a novel yeast strain was screened from soils of rice farming. The isolated strain HY200 was systematically characterized by using general approaches of Biolog Microlog$^{TM}$ and 18S rRNA sequence analyses, and consequently was designated as Candida tropicalis HY200. Under formulated culture conditions, relatively high xylitol yield ($77\%$) and productivity (2.57 g/l$\codt$h) were obtained, in practice, when 200 g/l of xylose was supplemented. In the utilization of nitrogen, inorganic compounds could not serve as nitrogen sources. As a promising phenotype, HY200 steadily flocculated during and/or after growing in the formulated medium. The extent of flocculation was partly affected by nitrogen sources. However, regardless of the kinds of carbon source fed, the flocculent cells were always observed at the end of the exponential growth phase. These observations strongly suggest that the strain HY200 could effectively be used as a potential candidate for the production of xylitol from xylose, especially in repeated batch mode, because of its flocculation ability and tolerance to high substrate concentrations.
Gibberellic acid (GA$_3$) is a commercially important plant growth hormone, which is gaining much more attention all over the world due to its effective use in agriculture and brewing industry. Industrially it is produced by submerged fermentation technique using Ascomycetous fungus Gibberella fujikuroi. Solid state and immobilized cell fermentation techniques had also been developed as an alternative to obtain higher yield of GA$_3$. This review summarizes the problems of GA$_3$ fermentation such as production of co-secondary metabolites along with GA$_3$, substrate inhibition and degradation of GA$_3$ to biologically inert compound gibberellenic acid, which limits the yield of GA$_3$ in the fermentation medium. These problems can be overcome by various bioprocessing strategies e.g. two - stage and fed batch cultivation processes. Further research on bioreactor operation strategies such as continuous and / or extractive fermentation with or without cell recycle / retention system need to be investigated for improvement in yield and productivity. Down stream processing for GA$_3$ isolation is also a challenge and procedures available for the same have been critically evaluated.
The conditions for ${\beta}$-galactosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at $50^{\circ}C$ for 5 days with fed-batch culture in the optimal medium containing 1.5% lactose, 0.6% yeast extract 0.15% $K_2HP0_4$and initial pH 9.5, and then final enzyme activity under the above conditions was 5200 unit/ml of cell free extract.
A packed bed reactor (PBR) was fed with nitrate containing synthetic wastewater or effluent from a sequencing batch reactor used for nitrification. The C source introduced into the PBR consisted of volatile fatty acids (VFAs) produced from anaerobic acidogenesis of food wastes. When nitrate loading rates ranged from $0.50\;to\;1.01\;kg\;N/m^{3}{\cdot}d$, the PBR exhibited $100{\sim}98.8%\;NO_{3}^{-}-N$ removal efficiencies and nitrite concentrations in the effluent ranged from $0\;to\;0.6\;NO_{2}^{-}-N\;mg/L$. When the PBR was further investigated to determine nitrate removal activity along the bed height using a nitrate loading rate less than $1.01\;kg\;N/m^{3}{\cdot}d$, 100% nitrate removal efficiency was observed. Approximately 83.2% nitrate removal efficiency was observed in the lower 50% of the packed-bed height. When reactor performance at a C/N ratio of 4 and a C/N ratio of 5 was compared, the PBR showed better removal efficiency (96.5%) of nitrate and less nitrite concentration in the effluent at the C/N ratio of 5. VFAs were found to be a good alternative to methanol as a carbon source for denitrification of a municipal wastewater containing 40 mg-N/L.
Using crude soybean oil as the sole carbon source, the lincomycin production from Streptomyces lincolnensis LC 345 was investigated in the air lift bioreactor. When 30 g/L of crude soybean oil was used, the maximum lincomycin concentration reached 0.89 g/L, after 5 days of culture. When CSL concentration was increased from 10 to 30 g/L, Lincomycin concentration was increased from 0.6 to 1.2. On the other hand, when CSL concentration was increased from 40 to 60 g/L, it was decreased from 1.15 to 0.7 g/L. Using these results, fed batch cultures for comparing the use of crude soybean oil and glucose as a conventional carbon source were carried out in a 5 L air lift bioreactor. When crude soybean oil was used as the sole carbon source, the maximum lincomycin concentration was 2.0 g/L, which was about 2.0 fold higher than that of glucose medium after 7 day of culture. The product yield from olive oil was 0.042 g/g consumed carbon source, which was about 3.8 fold higher than that of glucose.
