Effect of Enzyme-Treated Radish Leaves on Lipid Metabolism in Rats Fed a High-Fat Diet

  • The purpose of this study was to investigate the effect of enzyme-treated radish (Raphanus sativus L.) leaves on lipid metabolism in rats fed a high fat diet. Rats were divided into four experimental groups which were composed of a high fat diet group (HF group), a high fat diet with 10% radish leaf powder-supplemented group (MA group), a high fat diet with 5% enzyme-treated radish leaf powder-supplemented group (MB group) and a high fat diet with 10% enzyme-treated radish leaf powder-supplemented group (MC group). Total dietary fiber content of enzyme-treated radish leaves were greater than untreated radish leaves. Body weight gain and food efficiency ratio (FER) of the HF group increased compared to the MA, MB and MC groups. The serum total cholesterol, LDL-cholesterol and atherogenic index contents in the radish leaf powder-supplemented groups were lower than that of the HF group, while those values for the MB and MC groups were significantly lower than that of the HF group. The serum HDL-cholesterol contents of the MB and MC groups increased compared to the HF group. The hepatic triglyceride contents of the MA, MB and MC groups decreased compared to the HF group. In fact, the hepatic triglyceride contents of the MB and MC groups were significantly lower than the MA group. The hepatic total cholesterol contents of the MB and MC groups significantly decreased compared to those of the HF group. The fecal total cholesterol contents of the MA, MB and MC groups significantly increased compared to those of the HF group. These results indicate that supplementation with enzyme-treated radish leaves increase the useful fiber contents. Furthermore, it may have a pronounced impact on lipid metabolism of serum and liver in rats fed a high fat diet.

알팔파 사포닌이 흰쥐 혈청 콜레스테롤 농도에 미치는 영향 (Effect of Alfalfa Saponin on the Serum Cholesterol Level in Rats)

  • 알팔파 뿌리에서 사포닌을 추출, 단리하여서, 콜레스테롤 무첨가식이에 0.2% 및 0.4% 첨가군과 콜레스테롤 첨가식이에 0.4% 군을 설정, 80g 전후의 Wistar계 흰쥐수컷에 21일간 급여하였다. 그 결과 혈청지질과 분중지질 배설량을 측정한 결과 다음과 같다. 1. 알팔파 사포닌중 Medicagenic acid함량은 36.4%이었다. 2. 알팔파 사포닌 급여에 의하여 분중 콜레스테롤 배설량은 증가하였으나, 담즙산은 증가하지 않았다. 3. 알팔파 사포닌은 콜레스테롤 첨가, 무첨가식이에서 0.4%첨가에 의해 혈청콜레스테롤 저하작용을 나타내었지만, 0.2% 첨가에서는 관찰되지 않았다. 4. 사포닌의 혈청 콜레스테롤 저하작용은 소화관내에서의 콜레스테롤 흡수저하에 기인되는 것으로 여겨진다.

  • mechanisms for the hypocholesterolemic effects of $\beta$-glucan remain unclear. Rats were divided into 3 groups ; normal control group, atherogenic group(oral administration of cholesterol 40 mg/kg/day plus vit. $D_2$ 320,000 IU/kg/day), $\beta$-glucan treatment group(atherogenic treatment plus $\beta$-glucan 0.135 g/kg/day). The $\beta$-glucan treatment group showed moderate increases of serum lipids concentration compared with atherogenic group. In histopathological examination, aortas showed no critical lesions. The total fecal neutral sterols and bile acids excreted for 6 days was increased compared with both normal and atherogenic group. To compare effects of soluble fiber and insoluble fiber extracted from barley on postprandial lipemia, 5 healthy male adults ingested on separate days a low-fiber(total dietary fiber 2.61g) control meal or dietary fiber-enriched(12.61g) meals. Fasting and postprandial blood samples were obtained for 6.5h and serum lipids were analyzed. The serum total lipids, total cholesterols, LDL & VLDL-cholesterol were markedly reduced with soluble fiber-enriched meals, but no decrease with insoluble fiber-enriched meals. These results suggest that mechanisms for the hypocholesterolemic effect of $\beta$-glucan on rats were due to the inhibition of cholesterol absorption in the intestinal lumen and acceleration of cholesterol catabolism in the liver. And the soluble dietary fiber($\beta$-glucan) has the hypocholesterolemic effect by dropping serum LDL & VLDL-cholesterol in the clinical study.

Effect of Onion Powder Supplementation on Lipid Metabolism in High Fat-cholesterol Fed SD Rats

  • This study was performed to examine the effects of onion powder supplementation on lipid metabolism in male SD rats fed a high fat and high cholesterol diet. Experimental groups were control (C), high fat fed group (HF), high fat+onion powder intake (OP), high fat+quercetin intake (Q). The HF diet contained 1% cholesterol, 4% lard, 0.3% Na-taurocholate, and quercetin supplementation level was 0.1 g/kg diet. The OP group showed lower body weight gains compared to the control, while there was no significant difference in food efficiency ratio efficiency. When the proportion of fecal bile acids per total lipids was calculated, there was a significant decrease in the HF group compared to the control group, while the levels of the control group was same as that of the OP group. There was no significant difference between the HF and Q groups in bile acid/total lipid in feces. The amount of total cholesterol in liver increased significantly in HF group compared to the control group, while total cholesterol decreased significantly in the OP group compared to the HF group. There was a significant decrease in GOT (glutamic oxaloacetic transaminase) activity in OP and Q groups compared to the HF group. In conclusion, feeding onion powder to hyperlipidemic rates appeared to control weight gain, significantly lower the level of total cholesterol in the liver, and recover GOT activity. We also demonstrated that onion powder intake was more effective than quercetin intake.

The effect of seamustard on blood lipid profiles and glucose level of rats fed diet with different energy composition

  • Recently, Korean people are consuming seaweeds almost 3.5 times more now than three decades ago. It is well known that seaweeds contain lots of soluble dietary fiber in addition to micronutrients such as ${\beta}$-carotene, iodine and some bioactive components. Seaweeds are considered to be effective for preventing chronic diseases including obesity, diabetes mellitus, atherosclerosis, cancer or constipation. This study was conducted to investigate the effect of seamustard intake on body weight gain, blood glucose level and lipid profiles in rats fed diets with different energy nutrient composition. Male Sprague-Dawley rats (average initial weight 103.7g) were divided into groups for two experiments as follows; Control, M2.5 & M5 groups (Exp. I) and M5, M10, HCM5, HCM10, HFM5 & HFM10 groups (Exp. II). The rats were fed diet and water ad libitum for 4 weeks. In general, there was no significant difference in blood glucose and triglyceride concentration among groups. In Exp. I, serum LDL-cholesterol level of rats fed diet with 5% seamustard powder (M5) was significantly lower than that of control group, while HDL-cholesterol level, TC/LDL ratio and weight of adrenal gland were higher. In Exp. II, food intake, body weight gain and EER of high fat diet with 10% seamustard group (HFM10) were the lowest among groups. Except gastrocnemius muscle, all organ weights of HFM10 group were the lowest. Fecal cholesterol excretion and serum LDL-cholesterol concentration of HFM10 group were the highest, while serum HDL-cholesterol level was the lowest among groups. Interestingly, HDL-cholesterol concentration was the highest in HCM5 group among groups. From these results, it was suggested that seamustard intake might be more effective for body weight control, but not for improving blood lipid profiles in high fat diet than in high carbohydrate diet.

토양분석을 통한 고고학적 해석-익산 왕궁리 수혈유구 토양을 대상으로 (The Analysis of the Archaeological Soils excavated at Wanggung-ri)

  • Coprostanol is a metabolic product of cholesterol, formed by microbial action in the mammalian gut. This chemical compound is the major sterol in human and has been routinely studied as a biomarker of sewage pollution in marine and lacustrinesediments. This has led to the search for coprostanol as a biomarker in archaeologicalsoils, in order to detect the presence of fecal material. In this study, five samples of archaeological soils excavated at Wanggung ri, Iksancity, were used to assess the possibility of using coprostanol as indicators of ancient human activity in archaeological areas. The sampled soils were analyzed MXRD,EDXRF for their physical and chemical properties. And coprostanol was analysed byGC/MSD, using SIM method to detect and quantify specific compound. The results showed the soils were composed of quartz and feldspars, inorganicelement such as $SiO_2$, $Al_2O_3$, $Fe_2O_3$ etc. Moreover, the result from the analysis wasindicated that the specific compound is coprostanol. The coprostanol was determined at $0.16~1.01\mug$/g in the range of concentrations. This finding indicate that clear promise exists for the exploitation of coprostanol as biomarker of ancient human activity inarchaeological survey. Therefore such studies can serve to increase the confidence we place on biomarker-based methodologies for assessing fecal pollution. The application of this methodology has proved a simple and effective way of searching for that pattern in successively more aged deposits either known or suspected to contain fecal material. And the more scientific analysis of the soils will be showed to utility of the area ancient dietary life style, ancient environment.

고콜레스테롤 함유 청국장 식이 섭취가 흰쥐의 지질대사에 미치는 영향 (Effects of Cheonggukjang on Lipid Metabolism in Female Rats Fed Cholesterol Diet)

  • 콜레스테롤식이에 수준별 청국장 첨가 식이가 흰쥐의 지질대사에 미치는 영향을 조사하고자, 성숙한 흰쥐 암컷을 표준군, 대조군(콜레스테롤식이), 5% 및 10% 청국장군(대조식이에 5% 및 10% 청국장 분말을 첨가한 식이) 등 4군으로 나누어 4주간 사육한 결과는 다음과 같다. 실험동물의 체중증가량, 식이섭취량, 식이효율 및 자궁주위 지방 무게는 청국장군들이 대조군 보다 유의하게 감소되었다. 간의 콜레스테롤과 중성지질 농도는 대조군에 비하여 10% 청국장군이 유의하게 감소되었다. 혈청의 중성지질, 총 콜레스테롤, LDL+VLDL-콜레스테롤 농도 및 동맥경화지수는 대조군에 비해 10% 청국장군이 유의하게 감소되었고, 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율은 대조군에 비해 10% 청국장군이 유의하게 증가되었다. 변의 총 지질 배설량은 5% 및 10% 청국장군들이 대조군 보다 유의하게 증가 된 것으로 나타났다. 이상의 결과로 보아 콜레스테롤 식이에 10% 수준으로 청국장분말 첨가 섭취시 흰쥐의 체중 감소와 혈청과 간의 중성 지질, 총 콜레스테롤과 LDL+VLDL-콜레스테롤 농도 및 동맥경화지수를 낮추고, 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율이 증가되어 지질대사의 개선 효과가 있는 것으로 나타났다.

콜레스테롤 식이를 섭취한 흰쥐에 새송이버섯이 지질대사 및 효소활성에 미치는 영향 (Effects of Pleurotus eryngii on Lipid Levels and Enzyme Activity in Male Rats Fed High Cholesterol Diets)

  • 새송이버섯이 고콜레스테롤 식이를 섭취한 흰쥐의 지질대사에 미치는 영향을 조사하고자, 생후 8주령 숫쥐를 정상식이군, 대조식이군(5% 옥수수기름, 5% 돈지 및 0.5% 콜레스테롤을 첨가한 식이) 및 대조식이에 새송이버섯 분말을 3% 및 5%씩 첨가한 식이군 (3% 및 5% 새송이버섯군) 등 4군으로 나누어 4주간 사육한 결과는 다음과 같다. 실험동물의 체중증가량 및 식이효율은 대조군에 비하여 3% 및 5% 새송이버섯군이 유의하게 감소되었다. 간과 부고환지방의 무게는 대조군에 비해 5% 새송이버섯군이 유의하게 감소되었다. 간의 콜레스테롤 및 중성지질 농도는 대조군에 비하여 5% 새송이버섯군이 유의하게 감소되었다. 혈청의 중성지 질, 총 콜레스테롤과 LDL+VLDL-콜레스테롤 농도 및 동맥경화지수는 대조군에 비해 새송이버섯군들이 유의하게 감소되었다. 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율은 대조군에 비해 새송이버섯군들이 유의하게 증가되었다. 변의 지질배설량은 5% 새송이버섯군이 대조군 보다 유의하게 증가된 것으로 나타났다 이상의 결과로 보아 고콜레스테롤 식이에 새송이버섯 분말 첨가 급여시 흰쥐의 일부 지질대사를 개선시키는 것으로 나타났다.

영지가 알코올 섭취한 흰쥐의 간기능 및 지질대사에 미치는 영향 (Effect of Ganoderma Lucidum on the Liver Function and Lipid Metabolism in Alcohol-Consuming Rats)

  • The effects of Ganoderma Lucidum consumption (GL) on the liver function and lipid metabolic disorder induced by alcohol consuming were studied in rats using GL powder(1%, 2%, 4%) and 10% ethly alcohol. Thirty-five rats(Sprague-Dawley, male) were divided into five groups and fed experimental diets for seven weeks. The concentrations of serum cholesterol, HDL-cholesterol and triglyceride were the highest in the alcohol group, while the levels were apparently reduced by in groups fed GL. The atherogenic indices(AI) also tended to be higher in the alcohol group. Glutamic oxaloacetic transaminase(GOT) activities were significantly lower in body alcohol + 2% GL and alcohol + 4% GL groups, and glutamic pyruvic transaminase(GPT)activities lower only in the alcohol + 2% GL compared with the alcohol group. Alkaline phosphatase(ALP) activities were not significantly different among the groups. The elevated levels of liver cholesterol and triglyceride due to alcohol consumption, were apparently reduced by GL supplementation. On microscopic observation of liver tissues, fat droplets appeared extensively on the liver-lobule in the alcohol group, while fat droplets appeared only on the central vein in the alcohol + 1% GL group. However, in groups administered 2% and 4% GL, fat droplets appeared similar to the alcohol group. The concentrations of fecal bile acid were significantly increased in groups fed GL. These results indicate that GL exerted some beneficial effects on lipid metabolic disorder caused by long-term alcohol consumption. Thus, it seems that GL may be effective in preventing or curing some aspects of alcohol toxicity. More detailed studies are needed in order to determine proper dietary levels of GL for combating alcohol toxicity.

Enhancement of Lipid Metabolism and Antioxidant Defense Status in Mice Fed with High Fat Diet Supplemented with Antheraea pernyi Silk Fibroin Powder

  • The effect of diet supplementation of Antheraea pernyi (A. pernyi) silk fibroin on the lipid metabolism and antioxidant defense status in high fat-fed mice was investigated. The animals were given normal control diet (NC group), high fat diet (HF group), or high fat diet supplemented with A. pernyi silk fibroin powder (HFS group) for 7 weeks. After the experimental period, the HF group showed significant increase in body weight, plasma and hepatic total cholesterol levels, and hepatic triglyceride concentration, and decreased activities of hepatic antioxidant enzymes relative to NC group. However, the HFS group exhibited marked reduction in body weight, plasma cholesterol and hepatic triglyceride levels, hepatic lipogenic enzyme activities, and lipid peroxidation rate, and higher high-density lipoprotein (HDL)-cholesterol level, fecal triglyceride content, and antioxidant enzymes activities compared with that of HF group. These findings demonstrate that dietary feeding of A. pernyi silk fibroin could improve the lipid metabolism and antioxidant defense system via regulation of hepatic antioxidant and lipogenic enzymes activities. Hence, this silk fibroin may be beneficial as a functional biomaterial for the development of therapeutic agent against high fat diet-induced hyperlipidemia and its related diseases.
