Exreaction of Thiosulfinates from Garlic Using Gas-in-Liquid-Dispersion

  • Extraction yields of thiosulfinates from garlic were studied by using gas-in-liquis-dispersion(GLD) system to maxmize the thiosulfinate extraction. Extracted thiosulfinates were spectrophotometically quantified using synthesized dially1 thiosulfinate. The conditions for maximum extraction were optimized by response surface methodology. The volatile components extracted by various methods, such as hexane extraction, simultaneous steam distillation and GLD system, were compared by using gas chromatography. The results indicated that the thiosulfinate yield was increased by incresaing temperature and nitrogen gas flow rate, while the effects of bubble sizes on thiosulfinate extraction were not significant at the ranges tested. Application of GLD system resulted in extraction of more volatile components than other extraction methods, Therfore, it was suggested that GLD system was one of the efficient extraction methods among the ever introduced ones, for thiosulfinate extraciton.

염료추출 및 분석 조건에 따른 꼭두서니의 색소성분 분리 거동 (Separation of Chromophoric Substance from Madder Plant under Different Extraction and Analytical Conditions)

  • This research was aimed to establish the standard extraction and analytical procedures for examining the chromophoric substance in madder root with the ultimate goal of identifying the dyes in badly faded textiles of archaeological origin. The separation temperature of gas chromatography, pH and other extraction conditions were tested. The results were as follows: The suitable separation temperature for the GC cappillary column was 50∼305$^{\circ}C$, and methanol was a good GC solvent for both standard alizarin and madder extraction. The best extraction of madder was achieved by 90 min soaking in room temperature followed by filtration and the actual heat extraction procedure. The best pH for extracting alizarin was pH 3 and above pH 5 alizarin was not detectible. Only alizarin and no purpurin was found in the extraction of the currently used madder plant.

  • Based on solid phase extraction, gas chromatography and mass spectrometry procedure for determining phenol and its derivatives in natural water was presented. In solid phase extraction, three types of techniques using solid phase adsorption material were treated with acid and salt, and converted second portion of acetyl derivatives. Under the these condition, extraction efficiency and detection ability dependent on extraction methods were discussed. Obtained results using optimized solid phase extraction techniques showed more convenience, simplifier and lower cost than the conventional analytical methods with holding wide dynamic range and lower detection limits.

디스크타입 고상추출장치 및 컬럼커플링을 이용한 물시료의 PCDD/Fs 간이분석법 (Fast Analytical Method of PCDD/Fs in Water by Combination of Disk Type Solid Phase Extraction and Column Coupling Chromatography)

  • The analytical methods of polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans (PCDD/Fs) for water sample with extremely low concentration was developed to extract large volume and to improve fast chromatography for clean up. Semi-automated solid phase extraction (SPE) system and column coupling chromatography using elution pump was optimized and applied to each processes of treatment plant. Results of disk type SPE indicated that this system was applicable below 40 L of aqueous sample with a flow rate of 0.08 ~ 0.2 L/min. Average recoveries of SPE using labeled sampling spike of $^{37}Cl$-2,3,7,8-TCDD was 97%. Column coupling method resulted in reduced clean up time, solvent volume, increased average recoveries with constant elution rate. The combined methods were applied to the monitoring of drinking water treatment plant. Limits of detection (LOD) of each process were calculated. For example, LOD of raw and treated water ranged 0.094~0.968 pg/L and 0.028~0.364 pg/L, respectively. Combined methods of extraction and cleanup techniques provided fast analysis of PCDD/Fs with high accuracy and low LODs for water samples.

  • Mugwort (Artemisia montana), which is a perennial plant mainly distributed throughout Northeast Asian regions, has been used as a preferred source of various foods and traditional medicines in Korea. In particular, as essential oils extracted from mugwort were reported to be biologically active, its steam distillate has been widely used to treat various conditions, such as itching, hemorrhoids, and gynecological inflammation. Therefore, efforts have been devoted to develop effective methods for the collection of bioactive essential oils from mugwort. In this study, five mugwort extracts were obtained using different extraction conditions, namely, 6 % ethanol at room temperature and at 80 ℃, pure ethanol, n-hexane, and an adsorbent resin. To evaluate the five extracts of mugwort, area-under-the-curve values (AUCs), chemical profiles, and major bioactive essential oil contents were investigated using gas chromatography-mass spectrometry (GC-MS). An overall assessment of the volatile components, including essential oils, in the five extracts was conducted using AUCs, and the individual essential oil in each extract was identified. Furthermore, the four major essential oils (1,8-cineole, camphor, borneol, and α-terpineol), which are known to possess anti-microbial and anti-inflammatory activities, were quantified using authentic chemical standards. Based on the evaluation results, pure ethanol was the best extractant out of the five used in this study. This study provides evaluation results for the five different mugwort extracts and would be helpful for developing extraction methods to efficiently collect the bioactive oil components for medical purposes using chemical profiles of the extracts.

한우 Lactoferrin의 생화학적 특성 및 항균 활성 (Biochemical Properties and Antibacterial Activity of Lactoferrin from Korean Native Cow)

  • 우리나라 재래종인 한우의 초유로부터 lactoferrin(Lf)을 분리하기 위해서 batch extraction, ion exchange chromatography, gel filtration chromatography, affinity chromatography 등의 정제과정을 실시하였다. 각 정제 단계에 의해 한우 Lf과 결합되어 있던 여러 물질들이 제거되었으며 한우 초유 1 liter에서 정제과정을 통하여 회수한 Lf의 양은 65mg로 회수율이 29.4%였다. SDS-PAGE와 HPLC를 이용해 확인한 결과, 정제 단계를 거침에 따라 순도가 높아지는 것 알 수 있었다. 정제된 한우 Lf은 분자량이 81 kDa 이고 등전점은 pI 9였으며, 철 함량이 0.56mg/g으로 철 포화도는 약 40.6%로 측정되었다. 한우 Lf과 젖소 Lf은 아미노산 조성과 ${\alpha}$-helix 함량에서 서로 다르게 나타났다. E. coli O111에 대한 항균성은 젖소 Lf이 한우 Lf 보다 높았으며, 최소저해농도(MIC)는 젖소 Lf이 1.5mg/ml, 한우 Lf은 2.75mg/ml로 젖소 Lf의 항균 활성이 더 높은 것으로 나타났다.

  • Kyonin(Armeniacae Semen)is the herb medicine that contains amygdalin as a major ingredient. Amygdalin in water is decomposed into benzaldehyde. HCN. and glucose by emulsin. a hydrolysis enzyme in kyonin. A useful and practical method for the optimum extraction condition of amygdalin without enzymatic hydrolysis is required. The extraction yield of amygdalin of natural formula kyonin was 0.5% from crude powers. 0.7% from small pieces. 1.2% from half pieces and 2.7% from whole pieces. (omitted)

고성능 액체 크로마토그래피를 이용한 고삼 중의 oxymatrine 분석에 관한 연구 (Determination of oxymatrine in Sophora Radix by High Performance Liquid Chromatography)

  • 고성능 액체 크로마토그래피를 이용하여 고삼 중의 oxymatrine을 분석하였다. acetonitrile을 포함하는 완충용액과 $C_{18}$컬럼을 이용한 역상 크로마토그래피를 수행하였다. Oxymatrine의 분리는 215 nm에서 UV흡수가 관측되었다. 고삼 중의 oxymatrine의 추출은 다양한 용매와 다양한 추출방법을 사용한 결과, 마쇄한 고삼의 최적의 추출 효율은 $80^{\circ}C$에서 50% ethanol로 5시간 동안 환류가열 하였을 때 가장 높은 효율을 보였다. 대부분의 추출 방법은 복잡한 전처리를 필요로 하지만, 본 연구에서는 승화 추출법으로 복잡한 전처리를 제외시킨 추출법을 고안하였다. 승화 추출은 높은 진공 ($1{\times}10^{-3}$ torr) 과 높은 온도 ($200^{\circ}C$)에서 수행되었다. 승화 추출법을 사용한 추출 효율은 다른 추출 방법들과 비교해서 전처리 과정을 거치지 않은 장점은 있었으나 추출 효율이 좀 낮았다.

기체크로마토그래피/질량분석법에 의한 물시료 중 Polycyclic Aromatic Hydrocarbons의 분리 및 추출에 관한 연구 (A Study on the Separation and Extraction of Polycyclic Aromatic Hydrocarbons in Water Sample by Gas Chromatography/Mass Spectrometry)

  • 기체크로마토그래피/질량분석법(GC/MS)을 이용하여 물시료 중에 미량으로 함유된 19종의 여러고리방향족탄화수소류에 대한 액체-액체 추출법, disk추출법 및 고체상 추출법을 이용한 시료 매트릭스로부터 분리, 추출하는 방법을 조사하였다. GC/MS의 오븐온도프로그램을 $80{\sim}310^{\circ}C$까지 단계적으로 변화시킨 결과 19종의 PAH류를 분리할 수 있었다. 액체-액체 추출법에 의한 물시료 중 PAH류의 회수율은 71.3~109.5% 범위로 나타났고, filtration에 의한 $C_{18}$ disk추출법의 회수율은 80.7~94.9% 범위로 높게 나타났다. 그러나 $C_{18}$ Sep-Pak을 사용한 고체상 추출법의 회수율은 51.8~77.9%범위이었다. 이상의 추출법에 대한 추출 회수율, 재현성 및 실험의 편리성 등 장단점을 비교한 결과, $C_{18}$ disk를 사용한 거르기법이 PAH류의 추출에 가장 적합한 방법임을 확인하였다. 각 추출법의 검출한계(S/N=5)는 액체-액체 추출법 및 고체상 추출법의 경우 0.25~6.25 ppb, disk추출법은 0.05~1.25 ppb 범위이었다.

Effect of Microwave Assisted Water Extraction on Insoluble Phenolic Compounds through Bioactivity of Fagopyrum esculentum

  • Fagopyrum esculentum (Buckwheat) is a globally used alternative crop that contains several useful substances with various effects; however, many of these substances (rutin, quercetin, etc.) are water insoluble. To extract these substances, alcohols is required, which is inconvenient because these solvents cause diverse problems. Many studies are underway to achieve effective extraction of these substances with water. Among of these studies, microwave assisted water extraction (MAE) has been performed extensively. In this study, we performed the extraction in various solvents and/or microwave from Fagopyrum esculentum. The analysis of the content of useful substances and the bioactivity were performed and shown to increase in MAE. Liquid chromatography-mass was performed in order to identify of the useful water-insoluble substances. Catechin, quercetin, and rutin, which are all insoluble in water, were hardly extracted with water even on heating (4.4 ppb, 3.9 ppb and 60.3 ppb, respectively). However, MAE was found to extract much more of these substances than water (1204 ppb, 110.8 ppb and 2946 ppb, respectively). Although less efficient than alcohols, MAE showed much higher efficiency than simple water extraction. These results indicate that water extraction using microwave technology is effective in cases where it is difficult to extract useful substances using water.
