Nutritional Component and Anticancer Properties of Various Extracts from Haesongi Mushroom (Hypsizigus marmoreus) (해송이버섯(Hypsizigus marmoreus)의 영양성분과 추출용매에 따른 암세포 생장억제 효과)

  • This study was aimed to analyze the nutritional components and anticancer properties of Haesongi mushroom (Hypsizigus marmoreus), which has been recently available in Korea, to estimate its nutritional and functional values. Fruit body of Haesongi mushroom was investigated for its proximate components and mineral contents. Its water and ethanol extracts were compared for nutritional components such as $\beta$-glucan, protein, and total sugar. Anticancer effects of both extracts were measured against human cancer cell lines in vitro. This mushroom contained high protein (22.63%), total dietary fiber (30.80%), and K (3383.3 mg/100 g). The water extract contained more nutritional components such as $\beta$-glucan (9.32 mg/g), protein (17.71%), and total sugar (39.93%), compared with the ethanol extract. Moreover the extraction yield of the water extract was higher than the ethanol extract. The growth inhibitory effects of the water extract (5 mg/mL) on AGS, HepG2, and SW480 human cancer cells were 90.61, 75.43, and 58.49%, respectively. However, the ethanol extract showed 81.79, 49.90, and 25.71% growth inhibition, respectively. In this study, it is demonstrated that water is a more efficient solvent than ethanol for extracting nutritional and functional components from Haesongi mushroom.

Optimization of Production Yield for Neohesperidin by Response Surface Methodology (반응표면 분석법을 이용한 neohesperidin 생산 수율의 최적화)

  • Neohesperidin is a natural new nutrition sweetener, widely existing in plants of dry citrus peel, which can be derived from extraction. Since the sweetness is 1,300-1,500 times greater than that of sugar, neohesperidin are widely used in fruit juices, wines, beverages, bakeries and pharmaceutical formulations, and are particularly suitable for consumption by diabetic patients. However, the yield of extraction from citrus peel waste is very low. In this study optimal yield conditions were determinedusing response surface methodology (RSM) in order to increase the neohesperidin extraction yield. The critical factors for maximum extraction yield were selected extraction pressure ($x_1$), extraction time ($x_2$), and concentration of ethanol ($x_3$). As a result, the extraction yield was improved when the extracting pressure increased. The extraction yield also increased in a time-dependent manner. When adding ethanol as an assistance solvent to the supercritical carbon dioxide, extraction yield was increased as more ethanol concentration was added. Finally, the extraction yield of neohesperidin was improved to about 162.22% compared to ethanol extraction as a conventional method.

In spite of about 213.000 million won of domestic market in 1993. it seems like that the ginseng market of the future has not a good prospects The total market sale of white ginseng in Korea has been continuously decreased since 1991 And 963 tons of white ginseng was consumed in domestic market in 1993 The domestic market sales of white ginseng in origina1 root shave. was 90, 000 million won in 1993 and market price of the fine root used as a source of processed products has not been changed in these ten years. The total market sale of red ginseng and its processed products was 58, 000 million won in 1993 9.800 mi11ion won of red ginseng in original root shape and 48.000mi11ion of processed red ginseng product. Ginseng products such as extracts, drinks, teas and tonics etc atre mostly exported to south-east Asia. And the total exports of ginseng pi.oducts (extracts, drinks teas) decreased to 54 million dollars in 1994, compared with 85 million dollars in 1992. Despite of extensive knowledge about ginseng little is still known about the development of new processed ginseng pl.oducts because of "Know-How". Some papars have presented the effects of extracting method(amounts of solvent. time. temperature, equipment. etc.) on the quality and yields of ginseng extr acts. Also. some researchers have carried out a few studies on the poriflcation of the extracts and the amounts of precipitation in the drink at variotas pH during the storage for preventinly drink from precipitation. Extract powder by irradiation and ozone treatment have been reported by some researcher for the improvement hygienic quality of ginseng products There are also some reports about the effects of ginseng components on the acid production by lactic acid bacteria or acetic acid bacteria. and alcohol production by yeast for the development of new ginseng products processed by fermentation. To make ginseng more able to contribute to the health of mankind in the future. consistent and considerable efforts should be focussed on improving the taste of ginseng and developing various new product as a health food or a function food.tion food.

Functional evaluation of marine micro-algae Amphidinium carterae extract (해양 미세조류 Amphidinium carterae 추출물의 기능성 평가)

  • In this study, the antimicrobial, antioxidant activities and ${\alpha}$-glucosidase inhibitory activities of Amphidinium carterae ethanol extract (AE) was evaluated for using as a functional food ingredient. Chlorella ethanol extract (CE) was used to the comparison as a control. Anticancer activities of the AE and CE were analyzed by HepG2 and HT-29 human cancer cell. The AE showed antimicrobial activities for all tested bacterial strains. Whereas, CE showed antimicrobial activities for several tested bacterial strains only. The CE showed higher total phenolics contents, DPPH and ABTS radical-scavenging activities (47.36 mg/g, 22.42% and 28.58%, respectively) than those of AE (8.88 mg/g, 20.16% and 17.69%, respectively). AE showed anti-diabetic effect on ${\alpha}$-glucosidase inhibitory activity with dose-dependantly manner. The cell viability of AE ($125{\mu}g/mL$) on HepG2 and HT-29 human cancer cells were 38.12% and 11.27%, respectively. It was demonstrated that ethanol was efficient solvent for extracting functional components from A. carterae. These results indicated that AE can be described as a good candidate for using as a functional food ingredient.

Bioactive Compounds and Antioxidant Activity of Jeju Camellia Mistletoe (Korthalsella japonica Engl.) (제주 동백나무 겨우살이의 용매별 기능성 성분 및 항산화 작용)

  • Mistletoes are hemi-parasitic plant growing on different host tree and shrubs. They are traditionally used in folkloric medicine for the treatment of diarrhea, cough, diabetes, hypertension, cancer and skin infection. The purpose of this study was to determine the contents of phenolics and antioxidant activity of 70% ethanol, 100% methanol and hot water extracts of Jeju camellia mistletoe (Korthalsella japonica Engl.). Ethanol was most effective in extracting total phenols (7,427 mg gallic acid equivalent (GAE)/100 g) and flavonoid (1,777 mg rutin equivalent (RE)/100 g). The free radical scavenging activity, 1,1-diphenyl-2-picryl hydrazyl (DPPH) (EC50 = 7.8 mg/ml) and hydrogen peroxide (H2O2) (EC50 = 1.4 mg/ml), and the capacity for chelating metal ions (EC50 = 8.0 mg/ml) and reducing power (EC50 = 14.9 mg/ml) of the samples also higher in ethanolic extracts. The strong correlation (r2 = −0.996~−0.881) between antioxidant capacities and the phenolic contents implied that phenolic compounds are a major contributor to the antioxidant activity of the ethanolic extracts of Jeju camellia mistletoe. As conclusions, Jeju camellia mistletoe contains bioactive substances with a potential for reducing the physiological as well as oxidative stress and this could explain the suggested cancer preventive effect of these plants as well as their protective role on other major diseases.

Comparative Studies of Seven Solvents for the Extraction of Neutral Lipid in Corn Embryo (옥수수 배의 중성 지질의 추출을 위한 7종 용매의 비교 연구)

  • The comopositon of lipids extracted from corn embryo with various solvents were analyzed. The solvents for the extraction were benzene(BZ), n-hexane(HX), pet. ether(PE), trichlorethylene (TE), chloroform-methoanol(2:1, v/v) (CM), dichlormethane - methanol(2:1, v/v)(DM) and hexanediethyl ether(5:1, v/v)(HD). The lipids were than fractinated by silicis acid column chromatography(SACC) into three lipid fractions. The Neutral lipid fractons were further separated by thin layer chromatography(TLC) and the individual lipid spots were quantitatived by TLC scanner. And then the fatty acid compositions of total lipids and neutral lipids were determined by gas chromatography(GC). Crude oil contents of corn embryo were most efficient by using DM, CM and neutral lipid was extracted much HX, BZ, HD systems than did PE, DM, DM an CM were most efficient solvent systems for extracting glycolipid and phospholipid. The major component of the neutral lipid fractions was found to be triglyceride, and it was superior DM to PE. Linoleic acid was the predominant fatty acid in the total lipids, and it was most efficient with BZ and TE. The major fatty acid in neutral lipids was also linoleic acid and it was superior BZ to PE, CM, HD and oleic acid was similar to seven solvents and palmitic acid was found much superior in using CM.

Screening of Antimicrobial Activity for Medicinal Herb Extracts (한약재 추출물의 항균효과 검색)

  • Twenty kinds of medicinal herbs were extracted by water and 95% ethanol and then antimicrobial activity of the extracts was investigated against various kinds of microorganisms. Water extracts of Gardeniae fructus (Gardenia jasminoides), Lycii fructus (Lycium chinense) and Schizandrae fructus (Schizandra chinensis) showed inhibitory effects on the growth of most of the bacteria. In the case of ethanol extracts, the 3 kinds of the samples such as Gardeniae fructus, Schizandrae fructus and Lithospermi radix (Lithospermum erythrorhizon) showed inhibitory effects on the growth of almost all bacteria. In particular, ethanol extract from Phellodendri cortex (Phellodendron amurense) showed the best inhibitory effect on the growth of S. aureus in the concentration of 0.01%. By the way, inhibitory effects of water extracts from these medicinal herbs were not so good on the growth of fungi but those of ethanol extracts were better and ethanol extracts of Phellodendri cortex showed best. Antimicrobial activity was variable according to the used extracting solvent. For example, inhibitory effets of ethanol ext-racts were $2{\sim}100$ times better than those of water extracts. Ethanol extract of Lithospermi radix was the most effective not only bactericidal effects but also sensory evaluation tests for tastes.

Extraction of Intracellular Lipids from Recombinant E. coli for Improving Long-chain Fatty Acid Production (긴 사슬 지방산 생산을 위해 재조합된 E. coli로부터의 세포 내 지질 추출)

  • Recently, biohydrocarbons are gathering an interest as a new bioenergy due to the versatile applicability. In the present work, a process is proposed for the recovery of lipids from Recombinant E. coli MG1655 which provides longer chain fatty acids. After the growth of the recombinant E. coli, the cells were disrupted by high pressure homogenizer for obtaining intracellular lipids and the resulting solutions were centrifuged and extracted. For the efficient cell disruption with high pressure homogenizer, the pressure higher than 5,000 psi was required. In addition, under the conditions of applied pressure 5,000 to 20,000 psi, 1~3 pass homogenizing was enough for the more than 90% cell disruption. As organic solvents for extraction of lipid, hexane/isopropyl alcohol and ethyl acetate/ethanol systems showed excellent extracting power. With these solvent systems, the 60% lipid could be recovered. Moreover it was found that the extracted lipids contained long-chain fatty acids such as $C_{12}$, $C_{14}$, $C_{16}$ and $C_{18}$.

Antioxidative and Biological Activities of Santalum album Extracts by Extracting Methods (추출방법에 따른 백단향의 항산화 및 생리활성)

  • Santalum album has been used as a folk medicine for treatment of skin diseases, inflammation, gonorrhea, gleet, and cystitis in India and other Asian countries. In a search for possible bioactive agents from natural sources, we found that the various solvent extracts of S. album showed significant antioxidative effect in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity test and moderate other biological functions submitted to the several bioassay systems for whitening and cytotoxicity evaluations. Among the tested extracts displayed DPPH radical scavenging activity, and the 70% acetone extract showed the most potent activity with an $IC_{50}$ value of $18.6\;{\mu}g/ml$, more potent than a positive control, L-ascorbic acid ($IC_{50}$, $28.7\;{\mu}g/mL$). Also, anti-lipid peroxidation, tyrosinase inhibitory, and cytotoxic effects were determined in each experiment. Total phenolic content of 70% acetone extract was found to be 117.1 mg equivalent of gallic acid per g of extract. Previous phytochemical investigation reveals the presence of phenolic compounds. The results indicate that S. album possess potential antioxidant activity and phenolic constituents are responsible for this capacity.

Investigation of Antimicrobial Activity and Stability of Ethanol Extracts of Licorice Root (Glycyrrhiza glabra) (감초의 에탄올 추출물의 항균활성 및 안정성 조사)

  • Conditions for extraction of antimicrobial materials from licorice root, Glycyrrhiza glabra, were optimized. Among solvents tested, 95% ethanol gave highest antimicrobial activity, and was chosen as optimal extracting solvent. Extraction temperature and time were optimal at room temperature and for 12 hr, respectively. Minimal inhibitory concentration (MIC) of 95% ethanol extracts was determined against 14 microorganisms. Reference microorganisms included 6 Gram(-) bacteria, 4 Gram(+) bacteria, and 4 yeast strains. Ethanol extract exerted very strong growth inhibition on Gram(+) bacteria, while was moderately effective for Gram(-) bacteria and yeasts. Treatment at $180^{\circ}C$ for 30 min or extreme pHs merely destroyed antimicrobial activity of ethanol extract. These findings suggest ethanol extract of G glabra may be useful as natural preservative.
