Selective miRNA Expression Profile in Chronic Myeloid Leukemia K562 Cell-derived Exosomes

  • Background: Chronic myeloid leukemia (CML) is a myeloproliferative disorder of hematopoietic stem cell scarrying the Philadelphia (Ph) chromosome and an oncogenic BCR-ABL1 fusion gene. The tyrosine kinase inhibitor (TKI) of BCR-ABL1 kinase is a treatment of choice for control of CML. Objective: Recent studies have demonstrated that miRNAs within exosomes from cancer cells play crucial roles in initiation and progression. This study was performed to assess miRNAs within exosomes of K562 cells. Methods: miRNA microarray analysis of K562 cells and K562 cell-derived exosomes was conducted with the 6th generation miRCURYTM LNA Array (v.16.0). Gene ontology and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were also carried out. GO terms and signaling pathways were categorized into 66 classes (including homophilic cell adhesion, negative regulation of apoptotic process, cell adhesion) and 26 signaling pathways (such as Wnt). Results: In exosomes, 49 miRNAs were up regulated as compared to K562 cells, and two of them were further confirmed by quantitative real-time PCR. There are differentially expressed miRNAs between K562 cell derived-exosomes and K562 cells. Conclusion: Selectively expressed miRNAs in exosomes may promote the development of CML via effects on interactions (e.g. adhesion) of CML cells with their microenvironment.

  • Communication between endothelial cells (ECs) and smooth muscle cells (SMCs) via miR-143/145 clusters is vital to vascular stability. Previous research demonstrates that miR-143/145 released from ECs can regulate SMC proliferation and migration. In addition, a recent study has found that SMCs also have the capability of manipulating EC function via miR-143/145. In the present study, we artificially increased the expression of miR-143/145 in ECs, to mimic a similar change caused by miR-143/145 released by SMCs, and applied untargeted metabolomics analysis, aimed at investigating the consequential effect of miR-143/145 overexpression. Our results showed that miR-143/145 overexpression alters the levels of metabolites involved in energy production, DNA methylation, and oxidative stress. These changed metabolites indicate that metabolic pathways, such as the SAM cycle and TCA cycle, exhibit significant differences from the norm with miR-143/145 overexpression.

  • The profile of membrane currents was investigated in differentiated neuronal cells derived from human neural stem cells (hNSCs) that were obtained from aborted fetal cortex. Whole-cell voltage clamp recording revealed at least 4 different currents: a tetrodotoxin (TTX)-sensitive $Na^+$ current, a hyperpolarization-activated inward current, and A-type and delayed rectifier-type $K^+$ outward currents. Both types of $K^+$ outward currents were blocked by either 5 mM tetraethylammonium (TEA) or 5 mM 4-aminopyridine (4-AP). The hyperpolarization-activated current resembled the classical $K^+$ inward current in that it exhibited a voltage-dependent block in the presence of external $Ba^{2+}$ (30 ${\mu}$M) or $Cs^+$ (3${\mu}$M). However, the reversal potentials did not match well with the predicted $K^+$ equilibrium potentials, suggesting that it was not a classical $K^+$ inward rectifier current. The other $Na^+$ inward current resembled the classical $Na^+$ current observed in pharmacological studies. The expression of these channels may contribute to generation and repolarization of action potential and might be regarded as functional markers for hNSCs-derived neurons.

Saccharomyces cerevisiae Hsp30 is Necessary for Homeostasis of a Set of Thermal Stress Response Functions

  • Saccharomyces cerevisiae Hsp30 is a plasma membrane heat shock protein that is induced by various environmental stress conditions. However, the functional role of Hsp30 during diverse environmental stressors is not presently known. To gain insight into its function during thermal stress, we have constructed and characterized a ${\Delta}hsp30$ strain during heat stress. $BY4741{\Delta}hsp30$ cells were found to be more sensitive compared with BY4741 cells, when exposed to a lethal heat stress at $50^{\circ}C$. When budding yeast is exposed to either heat shock or weak organic acid, it inhibits Pma1p activity. In this study, we measured the levels of Pma1p in mutant and Wt cells both during optimal temperature and heat shock temperature. We observed that $BY4741{\Delta}hsp30$ cells showed constitutive reduction of Pma1p. To gain further insights into the role of Hsp30 during heat stress, we compared the total protein profile by 2D gel electrophoresis followed by identification of differentially expressed spots by LC-MS. We observed that contrary to that expected from thermal-stress-induced changes in gene expression, the ${\Delta}hsp30$ mutant maintained elevated levels of Pdc1p, Trx1p, and Nbp35p and reduced levels of Atp2p and Sod1p during heat shock. In conclusion, Hsp30 is necessary during lethal heat stress, for the maintenance of Pma1p and a set of thermal stress response functions.

  • In optical scanning holography, 3-D holographic information of an object is generated by 2-D active optical scanning. The optical scanning beam can be a time-dependent Gaussian apodized Fresnel zone plate. In this technique, the holographic information manifests itself as an electrical signal which can be sent to an electron-beam-addressed spatial light modulator for coherent image reconstruction. This technique can be applied to 3-D optical remote sensing especially for identifying flying objects. In this paper, we first briefly review optical scanning holography and analyze the resolution achievable with the system. We then present mathematical expression of real and virtual image which are responsible for holographic image reconstruction by using Gaussian beam profile.

Effects of Thujae Orientalis Folium (TOF) on Gene Expression of Human melanoma cells (SK-MEL-2) (측백엽(側柏葉)이 인간 유래 악성 흑색종 세포의 유전자 발현에 미치는 영향)

  • Objective : Thujae Orientalis Folium (TOF) can cool the blood and stop bleeding, eliminate phlegm and relieve cough in Oriental medicine. In addition, the fresh is used alone externally. Recently, TOF is known to have anti-tumor component. And also known to have tyrosinase inhibitory effect. Method : For these reasons, this study was designed to investigate anti-cancer and whitening activities of TOF. In this experiment, effects of TOF on proliferation rates of melanoma cells and on changes in genetic profiles were investigated. The genetic profile for the effect on human derived melanoma cell, SK-MEL-2, was measured using microarray technique, and the functional analysis on these genes was conducted. Results : Total 541 genes were up-regulated and 1,079 genes down-regulated in cells treated with TOF. Genes induced by TOF were mainly concerned with anti-cancer effects and apoptosis. Genes suppresed by TOF were related in extracellular signalling pathway. The network of total protein interactions was measured using cytoscape program, and some key molecules, such as THAP1, MAX1, STAM2, SMAD6, CYCS, PEX5, PSEN1, NONO, MAP2K7 and CREB1 that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conculusion : These results suggest possibility of TOF as anti-cancer drug for human melanoma. In addition, the present author also suggest that related mechanisms are involved in inhibition of several cancer pathway, activation of apoptosis pathway and suppression of general metabolic pathway.

Self-differentiation of University Students and their Responses to Kinetic House-Tree-Person Drawings (대학생의 자아분화와 동적 집-나무-사람 그림 반응특성 연구)

  • The purpose of this study was to verify the usefulness of kinetic House- Tree- Person drawing as a diagnostic measure for the degree of self-differentiation, which is an essential part of college students' development. Participants for the study were four hundred thirty five(272 male and 163 female) university freshmen enrolled in 4-year colleges located in Pusan. The Participants completed a scale of self-differentiation and a Kinetic House- Tree-Person drawings test. The evaluation system for kinetic House-Tree-Person drawings was established based on the indexes of Buck(1948) and Bums(1972) and used exiting literatures as reference. The data were analyzed using means, standard deviations, t-test, one-way ANOVA, and Scheffe's test. The results obtained from the study are as follows: First, the mean level of college students' self-differentiation was 2.81, which is about average. Some significant gender differences were found in the areas of self-intergration, family projection, and emotional separation. Male students scored higher on self-intergration, whereas female students score higher on family projection and emotional separation. Second. self-differentiation was higher when the student drew a house with smoke coming out of the chimney, a single-story house with flat roof, or with detailed description of curtains, roof and roof tiles. Third, self-differentiation seemed to be higher when branches and fruits were included, when there was no expression of roots, when large crowns and branch openings were presented, and when no slant lines or base lines appeared. Forth, self-differentiation showed ㅁ higher level when the portrait shows eyes, mouth and neck without omission, when it included the whole body instead of face only, and when there was no person with just a profile, a back, or with a stiff posture, and when there was no weak and thin lines. Individuality also marked higher if a person was in motion and than one person was added. Finally, self-differentiation showed significant difference according to the overall harmony of the drawing, ordering of contents, hand pressure, the kind and shape of tree, and presence of other persons. The more harmonious the picture was, and the more family members are added, the higher the level of self-differentiation was.

  • Subjected to seismic action causing large deformation of bridge columns, the plastic hinge region is commonly formed in the column end zone. The deformation capacity of a concrete column can be expressed by using plastic hinge length. The mechanical properties of high-strength reinforcing steel is different from that of normal-strength steel and the mechanical properties of steel will influence the plastic hinge formation. Therefore, in other to accurately predict the deformation of concrete column using high-strength steel, the plastic hinge length can be expressed as a function of the mechanical properties of steel such as the tensile to yield strength ratio and the strain at ultimate state. However, little research has been conducted into the effect of mechanical properties of steel on the plastic hinge length. It was difficult to measure the plastic hinge length from the test results. Therefore, the plastic hinge length of concrete columns was investigated from the curvature profile. A numerical approach was used to study the effect of various parameters on plastic hinge length. Based on the results of the numerical parametric study, a new expression for plastic hinge length was proposed.

Analysis of Hydrostatic Bulging of a Rectangular Diaphragm by Using the Energy Method (에너지법에 의한 직사각형 격막의 정수압벌징 해석)

  • The present study is concerned with the analysis of three-dimensional sheet metal forming process by the upper-bound method. For the analysis a systematic approach is necessary for the expression of geometric configuration of the deforming workpiece. In the present paper geometric configuration is constructed by three unit surfaces which are defined by sweeping the vertical section curves and boundary curve. The principal components of strain increment during the process is calculated directly from the change of geometric configuration for an arbitrary triangular element. The corresponding solution is found through optimization of the total energy consumption with respect to some parameters assumed in the velocity field and geometric profile. In order to verify the effectiveness of the present method, hydrostatic bulging of a rectangular disphragm is analyzed and the computation by the present method for the geometric shape renders the good result. From the comparison of the present results with the existing experimental results and elastic-plastic finite element solutions, good agreements have been obtained for the pressure curves, polar membrane strains and pressure distributions. The present method can thus be further applied to the analysis of other three-dimensional sheet metal forming processes.

Clinico-Morphological Profile and Receptor Status in Breast Cancer Patients in a South Indian Institution

  • Background: Breast cancer is the most common malignancy in women worldwide and the second most common cancer in females in India. Receptor status may be important for survival. Objective: To analyse and correlate the clinical and morphological parameters with receptor status in breast carcinoma patients in a tertiary care institution in Southern India. Materials and Methods: This retrospective study involved 320 patients of breast cancer diagnosed in an oncology hospital over a period of $3^{1/2}$ years. Data was analysed using SPSS Version 21. Results: Some 60.6% patients with breast carcinomas belonged to the age group of 40 to 60 years. The most common histological type was infiltrating ductal carcinoma, not otherwise specified, accounting for 84.4% of patients. On immunohistochemistry, estrogen receptor (ER) and progesterone receptor (PR) were expressed in 56.3% and 53.1% of cases, respectively. Conclusions: Breast cancers in India, a developing country, occur in younger women and tend to be more aggressive with lower rates of ER and PR expression and higher histological tumor grades. Both ER and PR status of the tumors had significant associations with the patient age, pathological TNM stage and histological tumor grade.
