Overexpression of Recombinant Arylsulfatase Cloned from Pseudoalteromonas carrageenovora

  • Arylsulfatase cloned from a marine bacterium, Pseudoalteromonas carrageenovora, was over-expressed in Escherichia coli. Most of the recombinant arylsulfatase was found in the cell lysate with induction up to $10{\mu}M$ IPTG. However, enzyme activity was observed both in the culture supernatant and cell lysate by induction with IPTG concentration of $50-5,000{\mu}M$. Most of the recombinant enzyme was localized in the periplasmic space with $10{\mu}M$ IPTG induction, while half of the enzyme was distributed in the periplasmic space with $50{\mu}M$ IPTG induction. Cell growth and arylsulfatase activity did not change with the induction time, and the level of recombinant arylsulfatase expression was maintained at 4-5 U/mL after 6 to 14 hr of culture.

  • In the 1970's the Interior Design Associations was established and the activity of youth designers who werecalled the "first generation of Korean interior designers" were created. This study is aimed to analyze characteristics and trends of commercial interior design in the 1970's. The design methods include the documentary research and the actual proof research conducted. The frames of analysis were made by the background theories about Korean interior design, and the annual case studies were analyzed and estimated according to the design types. The design types were analyzed by the geometrical simplicity research, the romantic emotional expression, the Korean identity expression, the machine technical asthetic expression and the eclectic style with western classics. In the early 1970's, the abstract, brief, and simple expression were presented most frequently by the geometrical form and the repetition of the pattern. From the mid-1970's the romantic and emotional atmosphere of the youth culture that was popular at that time were expressed as vernacular design by the rough finishing of the natural materials such as plaster, brick, and wood floorings etc. The space such as a Korean food restaurant relates to the Korean traditional culture aims to be different through the expression by the Korean traditional patterns, furniture, and materials. In the late 1970's the metals and glass were used for the expression of the machine aesthetic form but was not popular because of the rare application. The type that revived the past western traditional form was presented by using the arch, dome, and the curved and luxurious moldings.

  • New media art moves out of place and time constraints, sublimates the benefits of technology into art, and presents a new way of communication with the audience. This paper analyses the tendency of nonverbal communication methods by analysing examples of facial expressions and sound used in new media art from early times. As a result, it can be seen that the digital paradigm in the new media art has a nonlinear thinking, which makes a perceptual reduction of immersion and dispersion. The facial expression in new media art made it possible not only to overcome the limit of space and time of various expressions through 'visual distortions, enlargement, and virtualisation', but also to enable new ways of communication to display facial parts combined or separated in the digital environment. The sound in new media art does not stay in auditory sense, but pursues multi-sensory and synesthesia by cooperating with visual and tactile, evolves by revealing characteristics of space expansion and sensibility and interaction of audience.

  • Wind forces on structures are usually schematized by the sum of their mean static part and a nil mean fluctuation generally treated as a stationary process randomly varying in space and time. The multi-variate and multi-dimensional nature of such a process requires a considerable quantity of numerical procedures to carry out the dynamic analysis of the structural response. With the aim of drastically reducing the above computational burden, this paper introduces a method by means of which the external fluctuating wind forces on slender structures and structural elements are schematized by an equivalent process identically coherent in space. This process is identified by a power spectral density function, called the Generalized Equivalent Spectrum, whose expression is given in closed form.

  • This paper presents a new temporal reasoning scheme based on explicit expression of time intervals. The proposed scheme deals with the general problem of temporal knowledge representation and temporal reasoning and may be used in rule-based systems and qualitative models. Time intervals, not time points, are defined in terms of orders and/or numbers in a quantity space. As a result, the system behavior is represented in the form of partially ordered networks. Such explicit and qualitative description of temporal quantities enables both reduction of ambiguity and parsimonious used of temporal information. Based on the proposed temporal reasoning scheme, a new rule-based qualitative simulation system is being built and evaluated.

Characteristics of Relational Representation of Light and Architectural Space Elements in Luis Barragan's Houses (루이스 바라간 주택공간에 나타난 빛과 건축공간요소의 상관적 표현 특성)

  • Luis Barragan is a representative Mexican architect. He designed architectural space as a local architect in consideration of the Mexico's harsh Mediterranean climate. His manifestation of the regionality can be found mainly in his precise expression of light. Such method and concept of using light in Barragan's architecture made a huge impact on later architects but detailed studies on this issue have not been made actively. Hence, this paper intended to suggest a starting point or an orientation by illustrating the characteristics of using and expressing light in architecture through his cases of housing works. The study method was as follows. First, the general characteristics of light were identified by analyzing the elements of architectural space that dealt with light and the characteristics of expressing light shown in the architectural space. Second, the architectural background of Luis Barragan and the architectural space elements of the light expressed in his architectural space were extracted to form a hypothetical ground. Third, five of his works on the houses in his prime (after the year of 1945) were analyzed to explore the characteristics of relational representation between light and the architectural space elements in his architecture. Through this process, it was learned that, in his architectural works, light was the embodiment of the location-related, time-based, daily and illusionary experience into architecture. In addition, meditative light could be experienced by contrasting the soft and dispersed light and shadow of the inside space of Barragan's houses with the strong external light. Therefore, through the process above, this study is hoped to provide an opportunity for the contemporary architects of our era to rethink the fundamental correlation between architecture and light.

  • The use of colors plays an important role in the field of space design. The selection of proper colors influences on people who use space by making the most of the psychological and physical functions of colors. Especially, the medical space is relatively more important than other indoor spaces in the selection of colors. The medical space has to be something to do with diversity and functions, and at the same time with stability and quietude, as well as the space with general convenient facilities. Color is not only related to the expression but also to the distinction of things and the existence. It also functions as a visual language. Human beings depend heavily on the function of color as a visual language. According to previous studies, human beings depend on color by 80% of the whole range of eyesight. Thus, color is an important factor in recognizing things and space. For example, in hospital, it Is very important to apply proper colors to various medical treatment spaces for the sake of patients' rest and healing. Various colors which do not consider the function of patients' rest and healing may make the space confusing against the change of space and structure. It is also important to apply colors to the change of space and structure. Another key point that I would like to make is that color has visual, direct and physical influence on the human body. For example, the color red with long wavelength serves to regulate body temperature. Research on the color scheme for hospitals should consider both physical and psychological factors. That is why the research should be carried out in depth through continuous clinical demonstration. The present research aims to outline color planning.

  • This paper is studied on the korean traditional landscape in the view of the meaning of the contemporary landscape. The landscape architecture is expanding in the various research field of landscape. The seven concept of landscape can be divided as a regional concept, a land- form concept, an ecological concept, a land use concept, a heritage concept, a scenery concept, and a park and garden concept. This paper is deal with 985 kyoung(景) in 104 places and 16 Gok(曲) in 16 places as the korean traditional landscape. The object of landscape is generally focus on a reality as well as an unreality. In kyoung and Gok, the landscape is organized as an order of a small universe. The expression of the structure of korean traditional landscape is the fifth dimensional space which is organized by the third dimensional space with a time, an image and a feeling.

  • Characteristics of 154 traditional fairy tales (62 Asian, 34 European, 40 American, and 18 African fairy tales) were investigated according to their corresponding cultural areas. The results showed that male adults appeared more frequently in the traditional fairy tales than other characters. The expression of time, such as 'long ago','once upon a time', and 'one day' was provided, and there was an extreme contrast, fixation, and reiteration of literary styles in the traditional fairy tales, regardless of the cultural areas. Also, the characteristics of time and space that appeared in African fairy tales had a significant difference from other cultures. Also in accordance to the cultural areas, these fairy tales illustrated a significant difference in the pattern of problem solving.
