Enhanced Production of Anticancer Agent Camptothecin by Double Elicitors in Suspension cultures of Camptotheca acuminata (희수[Camptotheca acuminata] 현탁세포배양에서 복합 Elicitor에 의한 항암제 Camptothecin 생산 촉진)

  • Cell cultures of Camptotheca acuminata, which is known to produce the anticancer indole alkaloid camptothecin and its dervatives, were made to enhance the productivity of camptothecin. In suspension cultures, the maximum cell growth rate in exponential growth phase was $0.269day^{-1}$ which was correlated to 2.58days of cell doubling time. The production of camptothecin was non-growth associated. The camptothecin production was the highest at 11th day form inoculation and then it decreased. Various elicitors were applied to enhance the production of camptothecin. Both of jasmonic acid and cellulase increased the production of camptothecin. The optimal dosing time of elicitor was the beginning of the cultures. The combination of two elicitors was more effective to produce camptothecin than single applications. $20.42{\times}10^{-3}mg/\ell$ of camptothecin was obtained with combined application of two elicitors in two days.

Antagonistic Potential of Native Trichoderma viride Strain against Potent Tea Fungal Pathogens in North East India

  • Indigenous strains of Trichoderma species isolated from rhizosphere soils of Tea gardens of Assam, north eastern state of India were assessed for in vitro antagonism against two important tea fungal pathogens namely Pestalotia theae and Fusarium solani. A potent antagonist against both tea pathogenic fungi, designated as SDRLIN1, was selected and identified as Trichoderma viride. The strain also showed substantial antifungal activity against five standard phytopathogenic fungi. Culture filtrate collected from stationary growth phase of the antagonist demonstrated a significantly higher degree of inhibitory activity against all the test fungi, demonstrating the presence of an optimal blend of extracellular antifungal metabolites. Moreover, quantitative enzyme assay of exponential and stationary culture filtrates revealed that the activity of cellulase, ${\beta}$-1,3-glucanase, pectinase, and amylase was highest in the exponential phase, whereas the activity of proteases and chitinase was noted highest in the stationary phase. Morphological changes such as hyphal swelling and distortion were also observed in the fungal pathogen grown on potato dextrose agar containing stationary phase culture filtrate. Moreover, the antifungal activity of the filtrate was significantly reduced but not entirely after heat or proteinase K treatment, demonstrating substantial role of certain unknown thermostable antifungal compound(s) in the inhibitory activity.

Function of Cell-Bound and Released Exopolysaccharides Produced by Lactobacillus rhamnosus ATCC 9595

  • The physiological characteristics and function of the exopolysaccharide (EPS) produced by Lactobacillus rhamnosus ATCC 9595 were determined. The total quantity of EPS was rapidly increased to 496$\pm$20 mg/l during the exponential phase, and then maintained steadily during the stationary phase. During the exponential phase (18 h), the total EPS consisted of 61% cell-bound EPS (cb-EPS) and 39% released EPS (r-EPS), whereas the relative proportion of EPS during the stationary phase (48 h) was convered to 23% cb-EPS and 77% r-EPS. On gel permeation chromatography, cb-EPS was fractionated as a single peak of 8.6$\times10^6$ Da, whereas r-EPS was fractionated as two peaks with average molecular weights of 4.3$\times$10$^4$ and 8.6$\times10^6$ Da. Interestingly, both EPS species exhibited anticancer properties and cholera toxin-binding activities. Our results suggest that the EPS generated by L. rhamnosus ATCC 9595 might be suitable for use as a functional food or food supplement.

  • The paper considers the computation method in the performance evaluation of cellular network in the phase-type distribution assumptions that the channel holding times induced from mobility are modeled by well-fitted distributions to reflect an actual situation. When ww consider a phase-type distribution model instead of exponential distribution, the complexity of the computation increase exponential even though the accuracy is improved. We consider an efficient numerical algorithm to compute the performance evaluations in cellular networks such as a handoff call dropping probability, new call blocking probability, and handoff arrival rate. Numerical experiment shows that numerical analysis results are well approximated to the results of simulation.

  • The effects of GroEL/ES chaperone on the production of soluble form of B. macerans cyclodextrin glucanotransferase (CGTase) in recombinant E. coli were investigated. The cgt gene and groEL/ES genes are under the control of T7 promoter and Pzt-1 promoter, respectively. The optimal concentrations of inducers, IPTG and tetracycline, were found to be 1.0 mM and 10 ng/ml, respectively. When tetracycline and IPTG were added at the early exponential phase (2h) and exponential phase (3h) of growth, respectively, about 1.5-fold increase of soluble CGTase activity and 1.6-fold increase of soluble CGTase protein were obtained. An SDS-PAGE analysis revealed that about $37.2\%$ of total CGTase protein was in the soluble fraction when GroEL/ES chaperone was overexpressed.

  • The probability distribution of the repair process should be determined to choose the optimal spare level of a weapon system with a queueing model. Though most weapon systems have a multi-step repair process, previous studies use the exponential distribution for the multi-step repair process. But the PH distribution is more appropriate for this case. We utilize the PH distribution on a queueing model and solve it with MGM(Matrix Geometric Method). We derive the optimal spare level using the PH distribution and show the difference of results between the PH and exponential distribution.

Monitoring of Bioluminescent Lactobacillus plantarum in a Complex Food Matrix

  • A bioluminescent Lactobacillus plantarum (pLuc2) strain was constructed. The luminescent signal started to increase during the early exponential phase and reached its maximum in the mid-exponential phase in a batch culture of the strain. The signal detection sensitivity of the strain was the highest in PBS (phosphate buffered saline), followed by milk and MRS broth, indicating that the sensitivity was influenced by the matrix effect. The strain was used in millet seed fermentation which has a complex matrix and native lactic acid bacteria (LAB). The luminescent signal was gradually increased until 9 h during fermentation and abolished at 24 h, indicating that the strain could be specifically tracked in the complex matrix and microflora. Therefore, the bioluminescent labeling system can be used for monitoring LAB in food and dairy sciences and industries.

  • A laser interferometry of very high spatial resolution was built to study the solid phase epitaxial regrowth of furnace-annealed silicon wafers. As boron concentration increases up to $1.0E20/cm^3$, regrowth rate increases, but pre-exponential term and activation energy are decreased.

Eu(Ⅲ) Luminescence Phase-Modulation Spectroscopy as a Site-Selective Probe of Y Zeolite

  • Phase shift spectroscopy is applied to Eu(Ⅲ) luminescence from $Eu^{3+}$-exchanged Y zeolite. The phase shift and intensity modulation of luminescence following intensity-modulated excitation are measured as a function of modulation frequency and they are fitted into a double exponential decay. The fast decay component, compared with the slow one, has narrower spectral bandwidth and is emitted from the $Eu^{3+}$ that has more polar and definite environment with higher symmetry and that interacts more easily with hydrated water molecules. The fast decay component is attributed to $Eu^{3+}$ at site Ⅱ' while the slow one to $Eu^{3+}$ at sites Ⅰ' and Ⅰ.

  • In order to develope a protoplast fusion of the genus Cellulomonas having high assimilibility of cellulose, the optimum conditions for the protoplast formation of Cellulomonas flavigena NCIB 12901 was investigated and observed by means of Scanning Electron Microscope. The results suggested that the susceptibility of the cell wall by lysozyme treatment on protoplast formation was considerably depend on the cultural periods of the cells. Cells of C. flavigena at mid exponential phase could more efficiently convert to protoplast cells than those at late exponential phase did. The rate of the protoplast formation was 95%, even though the rate was over 99.9% on counting by indirect method after osmotic shock treatment, when cells of the organism at mid exponential phase were treated with lysozyme (400$\mu\textrm{g}$/$m{\ell}$) for 6 hours and observed by SEM. In the evaluation of protoplast formation of the genus Cellulomonas, direct method of the observation under Scanning Electron Microscope was much more reliable than the counting method of protplasts after osmotic shock treatment. Because defferences between the number of spheroplast and protoplast were not able to be figured out on counting the number of protoplast after osmotic shock treatment.
