Evaluation of the Cytotoxicity of Root Canal Disinfectants and Root Canal Sealers on L Cells in Vitro (수종(數種) 근관소독제(根管消毒劑) 및 근관충전재(根管充塡材)의 세포독성(細胞毒性)에 관(關)한 실험적(實驗的) 연구(硏究))

  • This study was to evaluate the cytotoxic effect of three root canal disinfectants (formocresol, camphorated phenol and eugenol) and ten root canal sealers(Cavitec, Hypo-cal, Vitapex, AH26, Canals, Mynol, $N_2$, $N_2$-Medical, Z. O. E. and Calvital) in vitro. The experiments were performed in four differrent modes. In the first and second experiment, the "long-distance" cytotoxicity of three root canal disinfectants were tested on L cells. In the third exeriment, ten root canal sealers were tested for cytotoxicity by means of the tissue culture-agar overlay method immediately, 4 and 24 hours after the experiment. In the fourth experiment, the study with radioactively labeled L cells were employed to determine the relative cytotoxicity of ten root canal sealers. The results were as follows; 1. Every vapors from disinfectants showed more or less cytotoxicity. Of the three disinfectats, formocresol appeared to be the highest cytotoxic effect and camphorated phenol was the lowest. 2. Root canal sealers tested in tbis study showed cytotoxicity at every stage of time intervals. 3. The highest cytotoxic effect was freshly mixed $N_2$ meaical and $N_2$ also revealed the highest cytotoxic effect after 4 or 24 hours among these materials. Vitapex was found the lowest cytotoxic effect at all experimental stage. 4. Root canal sealers except N2 and Mynol showed cytotoxic effect were decreased cytotoxicity according to the time elapsed.

  • This experimental study was made to investigate the effect of the "Hi-Pol" composite resin on the human dental pulp. 38 cavities of healthy permanent teeth were divided into 5 groups which were used as experimental materials. Group 1: Zinc Oxide-Euginol paste was applied to the cavities as controls $\cdots\;\cdots8$ cases Group 2: "Hi-Pol"*filling with Dycal** base $\cdots\;\cdots9$ cases Group 3: "Hi-Pol" filling without Dycal base $\cdots\;\cdots9$ cases Groud 4: Adaptic*** filling with Dycal base $\cdots\;\cdots6$ cases Group 5: Adaptic filling without Dycal base $\cdots\;\cdots6$ cases The treated teeth were extracted after 1 week, 2 weeks, 3 weeks and 4 weeks and processed for histological study. The results obtained from this experimental study were as follows; 1. The controls applied zinc oxide-eugenol showed the minimal pulp response and group 3 and group 5 showed the most severe pulp response. 2. In group 3 and group 5, the severity of pulp response increased in intensity according to the time elapsed. 3.   • The purpose of this study was to investigate the antibacterial effect of zinc oxide eugenol(ZOE), zinc phosphate cement(ZPC), glass ionomer cement, resin, and Vitapex to S. muntans, S. sanguis, S. fecalis and E. coli by agar diffusion method. Four wells were punctured in mitis-salivarius agar plate per each group and each wells were filled with restorative matetials. The width of inhibition zones produced in mitis - salivarius agar were measured as the parameter of the antibacterial effect after 16 hours and 40 hours. In S. mutans and S. sanguis, the largest inhibition zone was produced on ZOE, followed by glass ionomer cement, and ZPC. Inhibition zones was not observed in resin and Vitapex. In S. fecalis, ZOE and glass ionomer cement showed wider inhibition zone than ZPC. In E. coli, ZOE showed wider inhibition zone than ZPC, but no inhibition zone was observed on glass ionomer cement.

  • The present study was designed to help elucidate the effect of glass ionomer cements on the exposed dental pulp by means of histologic examination. A total of 40 cavities of class V were prepared on the teeth of 4 dogs with exposure of 1mm in diameter on the bases of them. 20 cavities were filled with glass ionomer cement as the experimental group and the other 20 cavities were filled with zinc oxide eugenol cement as the control group. The dogs were sacrificed at one, two, three, and four weeks after filling, and the specimens were routinely prepared and stained with Hematoxylin-Eosin. The obtained microscopic findings were as follows: Inflammatory cell infiltrations were observed in control in 1 week, which decreased markedly with time. In all control groups, hemorrhage around exposed pulp tissue and coagulation change of pulp were observed. Secondary dentin formation and thickened predentin were observed in 4 week cases, and the recovery of pulp tissue was favorable on the whole. Inflammatory cell infiltration was observed in all GIC groups. Proliferation of blood vessel and congestion were observed with coagulation changes around the exposed pulp tissue. Secondary dentin formation and thickened predentin were observed in 3 weeks. In the experimental 4 week case, secondary dentin formation was evident. On the whole, pulpal irritation of glass ionomer cement was relatively severe. Recovery of pulp tissue in GIC groups was less favorable compared with that of ZOE groups.

  • Both immune reaction and hypersensitivity reaction are occurred by the same mechanism, the antigen and antibody reaction. The favorable result of this reaction towards a host is called clinically an immune reaction and the opposite results is called an hypersensitivity reaction. Type IV hypersensitivity reaction is a delayed type which is related to the cellular immune reaction and a contact hypersensitivity is included in this type. Various dental materials such as metal (mercury, nickel, chrome, cobalt), resin and eugenol are etiologic substances. Patch test kit is composed of test substance with a controlled concentration which respond only to a susceptible patient and an aluminum chamber, and etiologic substances for hypersensitivity can be easily and comfortably found just by applying the kit to the patient's skin. In this case report, the patch test was performed to a patients with oral lichen planus and the allergen, restorative material was found. After removal of the matching restoration from the patient's mouth, the symptom was improved.

Flavoral Essential Oil Components in the Stems of Agastache rugosa for Aromatherapy (아로마테라피용 배초향(Agastache rugosa) 줄기의 방향성 정유 성분)

  • This study was carried out to investigate the flavoral essential oil components in the stems of Agastache rugosa. These components were analyzed using gas chromatography-mass selective detector (GC-MSD). The stems of Agastache rugosa were contained alcohols, aldehydes, ketones, fatty acid esters, and terpenoids. The peak area (%) of estragole was highest among its oil components and the next were pulegone and menthone. The terpenoid alcohols found were 1-octen-3-ol, chavicol, spatulenol, 3-hexen-1-ol, 2-cyclohexen-1-ol, methyl eugenol, and octaethyllene glycol. The stems also contained ketones such as pulegone, menthone, cis-isopulegone, 2-cyclohexene-1-one, 3-octanone, 1-cyclohexanone, isoindole-1-one, t-ionone, inden-2-one, as well as the aldehydes of 4-methoxycinnam and benzaldehyde. The following esters were also detected 1-isopulegone-3-yl acetate, caryophyllene oxide, acetate and benzendicarboxylic acid ester. The terpenoids in the stems were identified as caryophyllene, limonene, cyclohexasiloxane-D, germacrene-D, anethole, cadinene, muurolene, and bourbonene. Overall Agastache rugosa contained several functional oil components including phenylpropanoids and terpenoids as flavoral essential oil components for natural aromatherapy.

Antibacterial Activity of Clove Oil against Foodborne Pathogenic Bacteria and Sensory Attributes in Clove Oil-Enriched Dairy Products: A Preliminary Study

  • This study was conducted to evaluate the antibacterial activity against Cronobacter sakazakii and Salmonella enteritidis as well as the sensory attributes of milk products supplemented with various concentrations (control, 0.5%, 1.0%, 1.5%, and 2.0%) of clove oil. In this study, clove oil was shown to have strong antibacterial activities. In addition, all the samples were assessed by ten researchers trained in five sensory attributes, namely, taste, flavor, color, texture, and overall acceptability. Compared to the control, 5% clove oil supplemented was the best in market milk, while in yogurt and kefir, 1.0% supplementation was the best. In terms of sensory attributes, the low score of color and flavor of market milk, yogurt, and kefir is attributed to the characteristics of the supplemented clove oil. Consequently, this study presents the possibility of producing bio-functional milk products supplemented with clove oil, and for controlling the growth of foodborne pathogenic bacteria in milk products using clove oil.

A Study on the Brain Activation Components in Herbs -For the Spearmint, Sweetbasil, Rosemary- (허브의 두뇌 활성화성분에 관한 연구 -스피아민트, 스위트바질, 로즈마리를 대상으로-)

  • Mentha spicata, Ocimum basilium L., and Rosemarinus officinalis L. were selected as subjects for the study, and analysis of rosmarinic acid contents by HPLC analysis and the fragrance components of each herb were compared by GC-MS analysis. It was prepared and cultivated in a herb farm located in Sacheonjin-ri, Gangneung-si in 2022 (Fig. 1) before flowering, collected in the summer, stored at minus 20℃, and dried 50 g each. The analysis was requested to the NICEM analysis room of Seoul National University to obtain the test results. Through the above study, HPLC showed that the content of rosmarinic acid was 555.53 mg/Kg, which was absolute, 27.12% in rosemary for 1,8-cineol in GC-MS, and 32.44% in linalool in sweet basil. For eugenol, 23.11% in sweet basil and 19.92% in rosemary were found in α-pinene. Through this study, it was verified that spearmint, sweet basil, and rosemary contain a large amount of major ingredients that act on concentration or brain activation.

The utilization of fungicide and insecticide from medicinal plants for conservation of cultural properties (천연약재로부터 문화재보존용 방충방균제 개발연구)

  • The germicidal and insecticidal properties of volatile components extracted from star anise(Illicium verum Hooker filius) and clove (Eugenia caryophyllata THUNBERG)were evaluated against five microorganisms and three insects for the purpose of developing biocidal active substances from medicinal plants. The volatile components of star anise and clove showed strong antimicrobial effect against Aspergillus niger, Penicillium funiculosum, Mucor hiemalis, Trichoderma viride, and Aureobasidium pullulans. The extracts of each medicine also showed insecticidal effects against Sitophilusoryzae L., Lyctus linearis GOZE, and Reticulitermes spertus kyushuensis Morimoto. Fumigant toxicities to adult insects were determined. In the case of fumiganttoxicity, the extract of star anise showed 100% mortality against R. spertus, S.oryzae, and L. linearis at rates of $2.5\mu\ell$, $50\mu\ell$, $250\mu\ell$/filter paper, respectively but showed no killing effects by clove. The volatile components of star anise and clove were investigated by means of GC/MS. The main constitute, anethole among 20components from star anise and eugenol among 9 components from clove were identified. The mixture of star anise and clove as the volume ratio of 2 : 1 showed higher properties for antimicrobial and insecticidal effect than each volatile component. A. niger was inhibited by the mixture(125ml/$m^3$) for up to 10 days of exposure. Also, from the result of observing state change of organic materials by volatile extracts of star anise and clove, volatile extracts effects have no effect on natural organic materials of organic cultural properties and can be used as biological control agent. As research contents as above, the insecticidal and germicidal agents from star anise and clove and the mixture of them were more efficient and high level to prevent biological damage for conservation of organic cultural properties. So they may be used in new development of biologicalinsecticidal and germicidal agents for conservation of cultural properties.

Application of Volatile Antifungal Plant Essential Oils for Controlling Pepper Fruit Anthracnose by Colletotrichum gloeosporioides

  • Anthracnose caused by Colletotrichum gloeosporioides has been destructive during pepper fruit production in outdoor fields in Korea. In vitro antifungal activities of 15 different plant essential oils or its components were evaluated during conidial germination and mycelial growth of C. gloeosporioides. In vitro conidial germination was most drastically inhibited by vapour treatments with carvacrol, cinnamon oil, trans-cinnamaldehyde, citral, p-cymene and linalool. Inhibition of the mycelial growth by indirect vapour treatment with essential oils was also demonstrated compared with untreated control. Carvacrol, cinnamon oil, trans-cinnamaldehyde, citral and eugenol were among the most inhibitory plant essential oils by the indirect antifungal efficacies. Plant protection efficacies of the plant essential oils were demonstrated by reduced lesion diameter on the C. gloeosporioides-inoculated immature green pepper fruits compared to the inoculated control fruits without any plant essential oil treatment. In planta test showed that all plant essential oils tested in this study demonstrated plant protection efficacies against pepper fruit anthracnose with similar levels. Thus, application of different plant essential oils can be used for ecofriendly disease management of anthracnose during pepper fruit production.
