Antioxidaive Activity of Lotus Root(Nelumbo nucifera G.) Extracts. (연근 추출물의 항산화 효과)

  • This study was investigated on the antioxidant capacity of 80% ethanol extract of lotus root in vitro. The ex-traction yields of 80 % ethanol extract was 9.14%. Lotus root ethanol extract was fractionated by the following: n-hexane, chloroform, ethylacetate and n-butanol. n-Butanol fraction showed the highest ex-traction yield of all fractions. Antioxidative activities of different fractions were examined by l.l-diphenyl-2-picrylhydrazyl(DPPH) radical generation, the Rancimat test, the nitrite scavenging ac-tivity and the thiobarbituric acid(TBA) method, and compared with the properties of the commercial antioxidant butylated hydroxytoluene(BHT). The antioxidative capacity of the ethylacetate fraction was the highest among fractions and its fraction showed higher contents of total polyphenol. Furthermore, the antioxidative capacity of the ethylacetate fraction was similar to that of BHT. In conclusion, these results suggest that lotus root may be a good candidate as a natural antioxidant source.

Antimicrobial and Antioxidative Activities from Moutan Cortex Extract (목단피 추출물의 항균 및 항산화 작용)

  • Methanol extract and various solvent fractions from Moutan Cortex were tested for their antimicrobial activities, free radical scavenging activities and antioxidative activities, and phenolic compounds in ethylacetate fraction were analyzed by GC and HPLC. In the antimicrobial activities test, the ethylacetate fraction of methanol extract showed stronger than other fractions. The antimirobial activities were more effective against Gram positive bacteria than Gram negative bacteria. Minimum inhibitory concentration(MIC) of ethylacetate fraction showed 156-1250$\mu\textrm{g}$/ml against Cram positive bacteria and 2500-5000$\mu\textrm{g}$/mg against Gram negative bacteria. The free radical scavenging activities and antioxidative activities using linoleic acid were higher in ethylacetate fraction. The antioxidative activity of ethylacetate fraction was similar to ${\alpha}$-tocopherol. The 3 major phenolic compounds were analyzed by GC and HPLC and these content were determined. The content of p-hydroxybenzoic acid, methyl gallate and gallic acid were 1.35%, 14.61% and 4.01%, respectively.

The Effects of Different Extracts of Ostericum koreanum on the Production of Inflammatory Mediators in LPS-stimulated RAW264.7 Cells (강활의 RAW264.7 세포에서 LPS에 의해 유도되는 염증물질 생성에 대한 효과)

  • Objectives : In this study, the effects of ethylacetate extract of Ostericum koreanum on inflammation in RAW264.7 cells were investigated. Methods : Dried roots of Ostericum koreanum was extracted with 80% methanol for 24 h, and then fractionated with n-butanol, n-hexan and ethylacetate. RAW264.7 cells, a mouse macrophage line were incubated with different concentrations of the extract for 30 min and then stimulated with LPS at indicated times. Cell toxicity was determined by MTT assay. The concentrations of nitric oxide (NO) and prostaglandin E2 ($PGE_2$) were measured by Griess assay and enzyme immunoassay (EIA), respectively. The expressions of inducible nitric oxide synthease (iNOS) and cyclooxyganase (COX) -2 mRNA and protein were determined by RT-PCR and Western blot. Results : The methanol extract of Ostericum koreanuman and its fractions were significantly inhibited the NO and PGE2 productions in LPS-stimulated RAW264.7 cells. Among the fractions of Ostericum koreanuman the ethylacetate fraction was more strongly inhibited NO and $PGE_2$ productions compared with other fractions. The ethylacetate fraction was also suppressed LPS-induced mRNA expressions of iNOS and its protein levels in RAW264.7 cells. Conclusions : This study suggests that the ethylacetate fraction of Ostericum koreanum may have an anti-inflammatory property through suppressing inflammatory mediator productions in activated macrophages, suggesting have a therapeutic potential for the treatment of various inflammatory diseases.

Biological Activities of the Root of Cichorium intybus

  • Several biological activities of extracts from roots of Cichorium intybus Linne (Compositae) were studied in this paper. The antiinflammatory activity of the methanol extract of this root was investigated against carrageenin induced edema in rat‘s hind paw. Significant inhibitory effects were observed at the dose of 1,000 mg/kg and were compared with aspirin as a control. The hepatoprotective activities of the methanol extract, ethylacetate and butanol fraction were studied on mice whose livers are damaged by $CCl_4$. The serum transaminase activities (ALT, AST) were reduced at the dose of 1,000 mg/kg of the methanol extract, 500 mg/kg of ethylacetate and butanol fraction, respectively. The bile juice secretion was also increased significantly from each fraction. The antidiabetic activity was examined on strepto-zotocin-induced diabetic rats with methanol extract. Methanol extract gave a significant reduction of blood glucose levels in 1 week and 3 weeks.

  • The ethanol extract of leaf mustard(Brassica juncea) exhibiting high antimicrobial activities was fractionated in the order of hexane, chloroform, ethylacetate and butanol fractions to test antimicrobial activity. The highest antimicrobial activity for the bacteria tested was found in the ethylacetate fraction, but a lesser extent in the butanol fraction. In contrast to antimicrobial activity for the bacteria, both ethylacetate and butanol fractions showed weak antimicrobial activity for yeasts. Unknown compound A in the ethylacetate fraction which exhibited a strong antimicrobial activity was isolated by silica gel column chromatography and HPLC, and exhibited 9 times more antimicrobial activity than the ethylacetate fraction.

Inhibitory Effect of Enteromorpha linza on the Melanogenesis in B16 Melanoma Cells (파래 추출물의 멜라닌 생성 억제 효과)

  • Melanin is a polymer of phenol which produces hyperpigmentation in the skin. Melanin synthesis is catalyzed by the enzyme tyrosinase. To investigate the whitening activity of the fractions from the ethanol extract of Enteromorpha linza, we studied the inhibitory effect on the tyrosinase activity and melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from E. linza was as strong as that of kojic acid, a positive control. Also, the melanin production was significantly inhibited by the ethylacetate fraction in a dose dependent manner. The ethylacetate fraction showed the highest activity in the fractions and as strong activity as that of arbutin, a positive control. From these results, we suggest that the E. linza extract might be able to apply to cosmetic and medical fields.

Effect of Ailanthi Radicis Cortex Extracts on Melanogenesis

  • Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. Melanogenesis is controlled by the intra- and extracellular environments. In the present study, to develop a new whitening agent, it was investigated the antioxidant activity and the inhibitory effect of Ailanthi Radicis Cortex extract on tyrosinase activity and on melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from Ailanthi Radicis Cortex was higher than that of arbutin. The ethylacetate fraction showed scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radicals in a dose dependent manner. The highest inhibitory activity of melanogenesis was also in ethylacetate fraction ($40.0{\pm}5%$ at the concentration of $400{\mu}g/ml$). This study demonstrates that the Ailanthi Radicis Cortex extract might be used to be a potential agent for skin whitening.

ANTICANCER EFFECTS OF CAESALPINIA SAPPAN EXTRACTS ON ORAL CARCINOMA AND OSTEOSARCOMA CELLS (소목 추출물의 구강암 및 골육종 세포주에 대한 항암작용에 관한 연구)

  • Anticancer effect of methanol extract of Caesalpinia sappan L. on oral carcinoma(KB) and osteosarcoma(HOS) cells were investigated in this study. In order to elucidate the anticancer mechanism of Caesalpinia sappan L, we analyzed telomerase inhibitory effect of the methanol extract of Caesalpinia sappan L. In addition, we prepared 5 fraction samples according to its polarity differences and analyzed anticancer effects on oral carcinoma and osteosarcoma cells. Following results are obtained in this study. 1. 50% cell proliferation inhibitory value($IC_{50}$) of the methanol extract of Caesalpinia sappan L. against oral carcinoma(KB) cells and osteosarco ma(HOS) cells were $9.0{\mu}g/ml\;and\;10.9{\mu}g/ml$, respectively. 2. The methanol extract of Caesalpinia sappan L. showed inhibitory effect of telomerase which is required for cancer cell immortality. Therefore, it seems that the anticancer effect of methanol extract of Caesalpinia sappan is at least partially due to telomerase inhibitory effect. 3. Five fraction samples were prepared according to its polarity and 88.7% of ingredient of total methanol extract was transferred to ethylacetate fraction. Thin layer chromatography analysis showed that dichloromethane fraction contained ingredient with relatively high polarity and ethylacetate fraction contained similar ingredient found in total methanol extract. 4. Anticancer effect was observed in n-hexane, dichloromethane, and ethylacetate fractions. The highest anticancer effect was found in dichloromethane fraction which had $IC_{50}$ value of $4.4{\mu}g/ml$ and > $4.0{\mu}g/ml$ against oral carcinoma(KB) cells and osteosarcoma(HOS) cells, respectively.

  • This study was performed to investigate the antioxidant effect of 80% ethanol extracts from Pimpinella brachycarpa in vitro. The extraction yields of 80% ethanol extract was 9.23 g/100 g. The extract was further fractionated subsequently by n-hexane, chloroform, ethylacetate, n-butanol and water. Water fraction showed the highest extraction yield among fractions. Antioxidative activities of different fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical generation, Rancimat test, thiobarbituric acid (TBA) value, nitrite scavenging activity, inhibition of lipid peroxidation and peroxide value in linoleic acid in comparison with the commercial antioxidant butylated hydroxytoluene (BHT). Antioxidant activities of n-hexane fraction of Pimpinella brachycarpa ethanol extract were the highest among fractions. Furthermore, the antioxidative capacity of the n-hexane fraction was similar to that of BHT.

Screening of Functional Materials from Solvent Fractions of Apple Flower Leaf Extract (사과꽃잎 추출물의 용매 분획으로부터 기능성 소재의 탐색)

  • Fractional solvent extraction by organic solvents such as hexane, chloroform, ethylacetate, and butanol was carried out using 70% ethanol extract of apple flower leaves. Biological activities including antioxidant, whitening, antimicrobial and anti-wrinkle activities were investigated and bio-active materials of the extracts were identified using GC/MSD. Among the tested solvent fractions, ethylacetate fraction showed the highest total polyphenol content (1218.94 ${\mu}g/mL$), and flavonoid (140 ${\mu}g/mL$). The DPPH radical scavenging activities was over 80% at a dry matterbased concentration of 200 ${\mu}g/{\mu}L$ and SOD-like activity was over 90% at 50 ${\mu}g/mL$ concentration in ethylacetate fraction that was slightly lower than of ascorbic aicd. Tyrosinase inhibition activity related to skin-whitening was over 60% by ethylacetate fraction of 100 ${\mu}g/mL$. As an anti-aging effect, elastase inhibitory activity was about 45% in ethylacetate fraction. Also, it showed a significantly antimicrobial activity against P. acenes. From GC/MSD analysis, a characteristic peak of high content in ethylacetate fraction was identified as kaempferol, which has been reported as a bioactive compound.
