Antimicrobial Activity of Fruit of Crataegus pinnatifida Bunge against Multidrug Resistant Pathogenic Pseudomonas aeruginosa and Candida sp. (항생제 다제내성 Pseudomonas aeruginosa 및 Candida 균주에 대한 산사자의 항균 활성)

  • The fructus of Crataegus pinnatifida Bunge (CBF) has been used as medicinal and food source in worldwide. In this study, antimicrobial activity of the methanol extract and its sequential organic solvent fractions of CBF against different pathogenic bacteria and fungi, including multidrug resistant Pseudomonas aeruginosa and Candida sp., were investigated. The methanol extract of CBF was active against various gram-positive and gram-negative bacteria, and the ethylacetate and butanol fractions of CBF showed strong antibacterial activity against Listeria monocytogenes, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Proteus vulgaris, Escherichia coli and various multidrug resistant Pseudomonas aeruginosa with minimal inhibitory concentration of 1.0~7.5 mg/mL. Also the fractions showed anti-Candida activity against C. albicans, C. kruseis and C. geochares. The methanol extract of CBF and its solvent fractions, except n-hexane fraction, did not show any hemolytic activity against human red blood cell up to $500\;{\mu}g/mL$, respectively. The hemolysis in n-hexane fraction at $500\;{\mu}g/mL$ was less than 9.9%. Our results suggest that the CBF could be developed as a potent antibacterial agent, especially for multidrug resistant Pseudomonas aeruginosa.

Nutritional Composition and Antioxidant Activity of the White Lotus (Nelumbo nucifera Gaertn) Leaf (백련(白蓮) 잎의 영양 성분 및 항산화 활성)

  • The nutritional composition of the white lotus(Nelumbo nucifera Gaertn) leaf are moisture($1.87{\pm}0.06%$), crude ash($9.57{\pm}0.07%$), crude protein($15.53{\pm}0.4%$) and crude lipid($2.42{\pm}0.1%$) per 100g. The highest mineral content was K and Ca. The principal free sugars in the white lotus leaf was sucrose, fructose and glucose. The total phenolics compounds evidenced maximal levels of $8,842.73{\pm}20.57\;g/100g$. DPPH and hydroxyl radical of scavenging ability of each sample tended to increase with increasing degrees of the sample concentration. The $IC_{50}$ values of the ethanol extract, n-hexane, chloroform, ethylacetate, butanol and water fraction from the white lotus leaf to DPPH radicals were 5.65, 8.5, 8.27, 2.03, 2.3 and $13.7\;{\mu}g/mL$ respectively. The $IC_{50}$ values of the hydroxyl radical scavenging ability of the ethanol extract, ethylacetate, butanol and water fraction were 390, 574, 327 and $378\;{\mu}g/mL$ respectively.

Antioxidant and α-Glucosidase Inhibitory Activities of the Extract from Sparganium stoloniferum Buch.-Ham. Root and Its Constituent Compounds

  • Three compounds, vanillic acid, p-hydroxylcinnamic acid, p-hydroxybenzaldehyde have been isolated from the ethylacetate extract of Sparganium stoloniferum Buch.-Ham roots using silica gel open column chromatography, preparative thin-layer chromatography (pTLC) and reverse phase high performance liquid chromatography. The structures of the compounds were established on the basis of IR, extensive 1D NMR, and MS analyses. The ethylacetate (EtOAc) extract, vanillic acid, and p-hydroxybenzaldehyde showed $\alpha$-glucosidase inhibition activity of 72.71%, 20.13%, and 30.42%, at the concentration of 10 ${\mu}g/mL$, respectively. The EtOAc extract exhibited strong antioxidant activity with an $IC_50$ value of 24.37 ${\mu}g/mL$ against DPPH radical scavenging activity, the vanillic acid, p-hydroxylcinnamic acid, and p-hydroxybenzaldehyde with an $IC_50$ value of 2.10 ${\mu}M$, 1.59 ${\mu}M$, and 2.72 ${\mu}M$ against DPPH, respectively.

Anti-tumor Activity of the Extract of $Alpinia$ $officinarum$ using Hollow Fiber Assay (Hollow FiberAssay을 이용한 고량강 추출물의 항종양 효과)

  • The purpose of this investigation was to evaluate anti-tumor activity and detect what compounds affect its activity form $Alpinia$ $officinarum$ Hance. Two fractions, methanol and ethylacetate, were isolated by Amberlite XAD-2 resin column chromatography from methanol extract of the rhizomes of $Alpinia$ $officinarum$. In hollow fiber assay, the methanol extract and methanol fraction were found to inhibit the tumor growth against colon tumor cell lines such as Colo-320, HCT116 and WiDr. Three diarylheptanoids [5-hydroxy-1,7-diphenyl-3-heptanone, 5-hydroxy-7-(4"-hydroxy-3"-methoxyphenyl)-1-phenyl-3-heptanone and 3,5-dihydroxy-1,7-diphenylheptane] and two flavonoids [galangin and kaempheride] were isolated and identified from the methanol fraction, which is higher activity than ethylacetate fraction. Among these diarylheptanoids and flavonoids, 3,5-dihydroxy-1,7-diphenylheptane, galangin and kaempheride as active components on anti-tumor activity were mainly posited in methanol fraction.

  • These experiments were carried out to develop a novel, simple, and rapid method to determine urinary sulfonamides using fluorescamine by spectrofluorometry. To get optimal conditions for the sulfonamide-fluorescamine reaction, sulfonamides such as sulfamethazine, sulfamerazine, sulfadimethoxine and sulfamonomethoxine, dissolved in buffers with various pH ranges were reacted with various concentrations of fluorescamine. and then, the fluorescence intensity and stability of the fluorophore were measured. To eliminate the interfering substances in urine, the fluorophore in buffers and urine with a definite pH range was extracted with some organic solvents. After then the fluorescence intensity was measured in organic and aquous phases. The results obtained were summarized as follows: 1. The maximal fluorescence of sulfonamides was presented in acidic state, pH 4.5~5.0, at 30 minutes after reaction. 2. The optimal concentration ratio of sulfamethazine and ffuorescamine was more than 1 : 40 in mole. 3. In pH 4.0, the intensity was maximal but was time-dependent, whereas in pH 8.0, the intensity was time-independent. 4. Sulfamethazine-fluorescamine conjugate could be dissolved in some of organic solvents in acidic state such as chloroform, n-butanol, and ethylacetate. 5. Sulfamethazine-flnorescamine conjugate in swine urine coule be extracted with ethylacetate in acidic state, pH 4.0~5.0.

A Study on the removal characteristics of VOCs and Odors with Biofilter (Biofilter에 의한 VOCs 및 악취물질의 제거특성에 관한 연구)

  • The objective of this research was to investigate the removal efficiencies ofVOCs and odors with newly developed biofilter which was designed to sustain the biofilm constantly on the packed media. Initially, four types of media, for example, fiber, activated carbon, ceramic and the mixture of activated carbon and ceramic(AIC mixture J, were used for packed materials of biofilter. When ethylalcohol was selected as a test gas for media efficiency, fiber and AIC mixture had better removal efficiencies of ethyl alcohol than others. Removal efficiencies for acetaldehyde, ethyl alcohol, butylalcohol, ethylacetate and diethylamine in biofilter with fiber and AIC mixture as packed media were increased as the residence time increased. Butylalcohol, especially, showed the maximum removal efficiency among all used VOCs and odors. In case of ethyl acetate, the difference of removal efficiencies between low and high residence times was wide remarkably.

Antioxidant, α-Glucosidase Inhibitory and Antimicrobial Activities of Extracts from Maesa japonica (Thunb.) (빌레나무 추출물의 항산화, α-글루코시다아제 억제 및 항미생물 활성)

  • Maesa japonica (Thunb.) is an evergreen shrub belonged to the Myrsinaceae family, which was discovered in 2006 in South Korea. And, its biological functions have not been well studied. In this study, we determined the antioxidant activities, ${\alpha}$-glucosidase inhibitory effects and antimicrobial activities of methanol extract and the solvent fractions of M. japonica leaves and twigs. The highest antioxidant activity obtained by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay and reducing power assay was found in the ethylacetate fraction of twigs methanol extract, which contained the highest level of total phenolic compounds compared to the other fractions. In addition, ethylacetate fraction of twigs extract exhibited higher inhibitory activities against ${\alpha}$-glucosidase ($IC_{50}=0.8{\mu}g/m{\ell}$) compared to the $IC_{50}$ of the buthanol fraction ($IC_{50}=16{\mu}g/m{\ell}$) of leaves extract. It showed antimicrobial activities against Bacillus atrophaeus and Bacillus subtilis subsp. Spizizenii. Although the data is too limited, the current study is the first report on biological functions of M. japonica.

Research on antifungal and insecticidal activity of Natural extracts for Protecting Organic Cultural heritages (유기질문화재 보존을 위한 천연 추출물의 항균·살충활성 연구)

  • This study evaluated the activity of biocidal compounds isolated from natural products and confirmed applicable possibility as conservative agents for orgainic cultural heritages damaged by biological degradation. we used methanol extracts of 11 Korean trees and 17 medicinal plants as natural resources. Among these extracts, Phellodendron amurense was investigated on antifungal activity against Coniophora puteana, Lentinus lepideus, Tyromyces palustris and Aspergillus niger. Asarum sieboldii was investigated on insecticidal activity against adult of Lasioderma serricorne. First, the extract of Phellodendron amurense was partitioned into hexane, dichloromethan and ethylacetate fraction. Among them, the ethylacetate fraction had the most significant antifungal activity. In addition, Asarum sieboldii essential oil showed an effective antifungal and insecticidal activity. As a result, the extract of Phellodendron amurense and Asarum sieboldii essential oil could be useful for conservation of organic cultural heritage against biological deterioration by wood rot fungi and Lasioderma serricorne.

A Study on the Screening of Antimicrobial Activity and the Isoflavonoids in Korean Arrowroot (칡뿌리의 항균활성과 Isoflavonoid 화합물에 관한 연구)

  • The experiments were carried out to investigate the antimicrobial activity and the content of isoflavonoids each fraction of the methanol extract from Korean arrowroots. The strongest antimicrobical fraction of the methanol extract was the etylacetate fraction and the other fraction was not detected antimicrobial activity. The yield of the ethylacetate fractation was 7~9% of the methanol extract of Korean arrowroot TLC was carried out using silical gel plate and CH$_2$C1$_2$- $CH_3$OH(7 : 3) and puerarin, daizin were detected by spraying 10% H$_2$SO$_4$ and Rf was 0.60 and 0.69, respectively. Puerarin, daidzin and daidzein contents were determined of the ethylacetate fraction of the sample Puerarin and daidzin contents were abundant in the samples of November, while daidzein content was abundant in the sample of Februery.

Anti-allergic Effects of Petasites japonicum (머위(Petasites japonicum) 추출물의 항알레르기 효과)

  • It is well known that the Petasites japonicum have been used for a long time in traditional medicine for the treatment of allergic diseases such as lacquer poisoning and asthma. Anti-allergic actions of Petasites japonicum extracts were asessed by testing their effects on the degranulation of mast cells. For this, hexosaminidase released (degranulation marker) from RBL-2H3 cells(mast cell line) was used. At the concentration of 300 $\mu\textrm{g}$/mL of the methanol, ethylacetate and hot water extract, the degranulation of RBL-2H3 cells were inhibited 83.33, 69.75 and 35.4%, respectively. These results suggest that the Petasites japonicum could be provide a effective resource for the control of allergic diseases.
