Effect of various fractions of Bacopa monnieri Linn. aerial parts on ethanol-induced hepatotoxicity in rats

  • The ethyl acetate fraction (EAF) and n-butanol fraction (NBF) of ethanolic extract of Bacopa monnieri aerial parts were screened for hepatoprotective activity and in vivo antioxidant activity on ethanol-induced hepatotoxic rats. Ethyl acetate fraction was found to be more potent even though both the fractions were endowed with significant hepatoprotective activity. EAF and NBF were investigated for hepatoprotective activity in albino rats at 300 mg/kg, p.o. dose and compared with standard drug Silymarin (25 mg/kg, p.o.). Results show that both the fractions were effective in blunting ethanol-induced enhanced activities of serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, alkaline phosphatase, level of serum bilirubin (both total and direct), liver weight loss and was also effective in reducing ethanol-induced lipid peroxidation both in vitro and in vivo. Furthermore, the fractions could also enhance ethanol-induced suppressed activities of superoxide dismutase, catalase and decreased level of reduced glutathione. Results of hepatocellular damage caused by ethanol and its recovery by EAF and NBF, suggest that they might be considered as a potential source of natural hepatoprotective agents, which could be related to the free radical scavenging properties of saponins present in high concentration in the fractions.

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.

Effect of Saururus chinensis BAILL Extract for Pharmacopuncture on the melanogenesis in B16F10 cells (삼백초 약침액이 B16F10 흑색종세포의 멜라닌 합성에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the melanogenesis inhibition effect of Saururus chinensis BAILL (SC) on in B16F10 melanoma cells. Methods : SC was fractionated ethanol extract by the hexane, ethyl acetate, butanol and water. We confirmed the inhibitory effect of tyrosinase activity and melanogenesis of all fraction samples. Results : Hexane fraction of Saururus chinensis BAILL (HSC), ethyl acetate of SC (ESC), and butanol of SC (BSC) were discovered to inhibit tysoinase activity and melanogenesis in the absence or presence of ${\alpha}$-MSH. However, water fraction of SC (WSC) did not affect tyrosinase activity and melanogenesis. In addition, all fractions did not inhibit the catalytic activity of cell-free tyrosinase from B16F10 melanoma cell lines. Conclusions : These results suggest that HSC, ESC and BSC reduce pigmentation by indirectly regulating tyrosinase.

The study on collagen sysnthesis and collagenase inhibition assay in natural plants

  • Type I (collagen) and procollagen are reduced in aged human skin. This reduction could result from increased degration by metalloproteinases and from reduced procollagen synthesis and skin collagenase is required for initiation of the degration of type I collagen. In the present study, we study on assay the collagen and collagenase in natural plants using the fibroblast human skin cell. We select the 15 kind of plants used to herbal and 4 kind of fraction(by methylene chloride, ethyl acetate, n-butanol, water). Among these extract, the ethyl acetate fraction from benincasa hispida/prunus persica, trichosanthes kiriowii, trogopterus xanthipes and methylene chloride fractions from benincasa hispida/prunus persica, torilis japonica and n-butanol fraction from cnidium officinale, chrysanthemum sibiricum were selected for further experiments as they exhibited distinctive amount of collagen compared to other natural extracts. These extracts were again subjected to collagenase assay test. Benincasa hispida/prunus persica extract was shown to have exellent collagen synthesis activity from result of the collagen assay test and the other extract was shown to have over 130% of collagen synthesis activity. But, in the study of collagenase assay test just only trogopterus xanthipes extract was shown to have collagenase inhibition.

Antioxidant Effects of Solvent Fraction from Sanguisorbae officinalis L. with Acetone (오이풀 아세톤 추출물을 이용한 용매 분획물의 항산화 효과)

  • The solvent extracts of Sanguisorbae officinalis L. were investigated for the activities of antioxidants as a functional ingredient for cosmetic products. The electron donating effect of ethyl acetate layer and n-butyl alcohol layer was appeared similar activity with positive control butylated hydroxy anisole (BHA) at all concentrations. In addition, in 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging assay, ethyl acetate layer, n-butyl alcohol layer and water layer were over 99% effect at all concentrations and higher than that of BHA. Also in hydrogen peroxide scavenging assay, ethyl acetate layer and n-butyl alcohol layer were higher than that of positive control ascorbic acid. The measured superoxide dismutase (SOD)-like activity of n-butyl alcohol was more than 50% at concentration of 1,000 ${\mu}g/mL$ and superoxide anion radical scavenging ability showed more than 45% at 1,000 ${\mu}g/mL$ of n-butyl alcohol layer. All these findings suggested that ethyl acetate layer and n-butyl alcohol layer have a great potential as a cosmeceutical ingredient with an antioxidant effect.

Cellular Protective Effect and Component Analysis of Euphorbia humifusa Extracts (땅빈대 추출물의 세포 보호 효과 및 성분 분석에 관한 연구)

  • In this study, the cellular protective effect, antioxidative property and component analysis of Euphorbia humifusa extracts were investigated. The ethyl acetate fraction ($3.68\;{\mu}g/mL$) and aglycone fraction ($3.15\;{\mu}g/mL$) of Euphorbia humifusa extract showed prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of Euphorbia humifusa extract on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($0.43\;{\mu}g/mL$) and aglycone fraction ($0.35\;{\mu}g/mL$) of extract showed higher ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g/mL$). The cellular protective effects of fractions of Euphorbia humifusa extract on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of extract protected cellular membranes against ROS in a concentration dependent manner ($5{\sim}25\;{\mu}g/mL$), and was more effective than (+)-${\alpha}$-tocopherol, lipid peroxidation chain blocker. Aglycone fraction from Euphorbia humifusa extract showed 2 bands in TLC and 2 peaks in HPLC. In HPLC chromatogram of aglycone fraction, peak 1 and peak 2 were identified as quercetin and kaempferol, respectively. And these components are very effective as antioxidant. Thus, these results indicate that fractions of Euphorbia humifusa extracts can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Fractions of Euphorbia humifusa extracts can be applicable to new functional cosmetics for antioxidant.

  • Methanol extract of Lonicera Japonica (Caprifoiaceae) flower significantly inhibited serum GPT, GOT, LDH and AIP activities at the doses of 500 and 1,000mg/kg in $CCl_4$-intoxicated rats. As confirmed with the fiactions of hexane, chlorofom, ethyl acetate, butanol and water, the butanol fraction showed significant inhibition of increased activity of serum GPT, LDH and AIP in $CCl_4$-intoxicated rats, indicating that the most active components might be contained in the butanol fraction.

Inhibitory Effect of Cell Differentiation against 3T3-L1 Pre-Adipocytes and Angiotensin Converting Enzyme (ACE) Activity of Ice Plant (Mesembryanthemum crystallinum) (아이스플랜트(Mesembryanthemum crystallinum)의 3T3-L1 전지방세포 분화 및 Angiotensin-Converting Enzyme(ACE) 활성 억제)

  • Mesembryanthemum crystallinum (Family: Aizoaceae) is an annual plant consisting of ice crystal-shaped bladder cells, which is responsible for its common name ice plant. This study investigated biological activities according to general components and extraction solvent in order to examine the functionality of ice plant. The total content of free amino acids was 32.57 mg/g, including 4.64 mg/g of L-alanine as the most abundant and 2.60 mg/g of ${\gamma}$-aminobutyric acid. Regarding angiotensin converting enzyme inhibitory activities of solvent fractions of ice plant, ethyl acetate fraction and chloroform fraction showed activities of $33.17{\pm}3.20{\sim}88.19{\pm}3.20%$ and $23.72{\pm}2.89{\sim}86.78{\pm}2.24%$, respectively, similar to $Captopril^{(R)}$ ($19.51{\pm}3.44{\sim}84.72{\pm}1.06%$) and $Enalapril^{(R)}$ ($24.93{\pm}1.12{\sim}91.32{\pm}3.62%$) as positive control groups. Regarding inhibition of lipid droplet production in 3T3-L1 preadipocytes by ice plant, anti-adipogenic activities were $53.00{\pm}0.45{\sim}65.75{\pm}0.31%$ and $44.16{\pm}0.29{\sim}63.32{\pm}0.36%$ in the ethyl acetate fraction and butanol fraction, respectively, showing the lowest lipid droplet production. The chloroform fraction and hexane fraction showed activities of $38.33{\pm}0.09{\sim}56.55{\pm}0.50%$ and $31.17{\pm}0.50{\sim}55.10{\pm}1.93%$, respectively, whereas the water fraction showed activity of $26.32{\pm}2.27{\sim}49.48{\pm}0.05%$. Therefore, all solvent fractions inhibited fat accumulation of 3T3-L1 preadipocytes according to treatment concentration. According to the results above, it would be possible to utilize ice plant as a new health functional material.

Anti-oxidative and Anti-cancer Activities of Extracting of Yacon (Yacon 추출물의 항산화 및 항암 활성)

  • This study investigated the anti-oxidative and anti-cancer activities by extracting of Yacon by fraction with organic solvents. The content of total phenol was the highest by 45.53% in ethyl acetate fraction and the results of electron donating abilities were 65.20% ($100\;{\mu}g/ml$), 91.81% ($500\;{\mu}g/ml$) and 95.06% ($1000\;{\mu}g/ml$) and those of nitrite scavenging abilities were 11.71% ($100\;{\mu}g/ml$), 36.81% ($500\;{\mu}g/ml$), 59.70% ($1000\;{\mu}g/ml$) in ethyl acetate fraction, which were higher concentration than the control group. Xanthine oxidase inhibitory activities were 23.74% ($100\;{\mu}g/ml$) and 43.41% ($500\;{\mu}g/ml$) in low concentration, which showed higher activities than the control group. After measuring the growth inhibitory activities for cancer cells, it has been found that hexane fraction had the highest growth inhibitory activities by 23.75% ($10\;{\mu}g/ml$), 34.67% ($50\;{\mu}g/ml$), 54.21% ($100\;{\mu}g/ml$) for SNU-1. Meanwhile, hexane fraction demonstrated high growth inhibitory activities by 41.38% ($10\;{\mu}g/ml$), 50.53% ($50\;{\mu}g/ml$), 60.91% ($100\;{\mu}g/ml$) and butanol fraction had 17.05% ($10\;{\mu}g/ml$), 43.87% ($50\;{\mu}g/ml$), 62.99% ($100\;{\mu}g/ml$) for HeLa. Therefore, it has been believed that there were peculiar properties of anti-oxidative and anti-cancer activities depending on each fraction. Furthermore, it has been expected that further studies on the development of functional food and concrete components depending on the components of each fraction to be conducted.

A Study on the Stability and Moisturizing Effect for the Cream Containing Castanea crenata Leaf Extract (밤나무 잎 추출물 함유 크림의 안정성 및 피부 보습효과에 관한 연구 (2))

  • In the previous study, we evaluated and reported about the anti-oxidative activities of extract/fraction of Castanea crenata leaf. Extract/fraction of Castanea crenata leaf showed excellent free radical scavenging activity, cell protective activity and inhibitory activity on tyrosinase and elastase. In this study, in order to investigate the stability of cream containing 0.2 % Castanea crenata ethyl acetate fraction. pH, viscosity, and absorbance were measured under 4 different temperature ($4^{\circ}C$, $20^{\circ}C$, $37^{\circ}C$, $45^{\circ}C)$ and under the sun light at 2 weeks intervals for the 8 weeks. The variations on pH and viscosity of all experimental creams were similar to control cream. The absorbance variation of extract from experimental cream at 353 nm was in the order: under the sun > $45^{\circ}C$ > $37^{\circ}C$ > $20^{\circ}C$ > $4^{\circ}C$. It shows that ethyl acetate fraction in the cream can be oxidized under the sun. The bad smell and discoloration were not shown. Also, physical changes as creaming and cohesion were not shown. Also, transepidermal water loss (TEWL) and water contents in skin were measured. The cream containing Castanea cranata leaf extract was applied to the right lower arm. After 120 min, TEWL of parts was decreased as 29.7 % (experimental cream) and 5.4 % (control cream) respectively. And the water contents in skin were increased 22.6 % (experimental cream) and 24.7 % (control cream) respectively. It was confirmed that a cream containing ethyl acetate fraction of Castanea crenata leaf shows the superior moisturizing effect. The results showed that Castanea crenata leaf extract could be used as a new active ingredient for anti-aging cosmeceuticals.
