Lipase Inhibitory Mode of Dieckol Isolated from Eisenia bicyclis Ethanol Extract (Eisenia bicyclis 에탄올 추출물로부터 분리한 Dieckol의 Lipase 저해 Mode)

  • This study was performed to investigate the possible use of Eisenia bicyclis (EB) ethanol extract to inhibit activity against lipase. In tests, the lipase inhibitory activity of EB ethanol extract was noted as being 43, 27, and 24% at concentrations of 5, 2.5, and 1 mg/ml, respectively. Isolation was carried out by liquid and liquid extraction, silica-gel column chromatography, and HPLC. The results showed that the lipase inhibitory activity of the ethyl acetate (EA) fraction from EB ethanol extract exhibited the strongest lipase inhibitory activity with an $IC_{50}$ value of 1.31 mg/ml. The EA fraction was separated using silica-gel column chromatography and we obtained 22 sub-fractions. Amongst them, the EA1 fraction showed the highest lipase inhibitory activity with an $IC_{50}$ value of 0.54 mg/ml. Eight peaks were obtained from the EA1 fraction by HPLC. Fraction 5 also showed a strong lipase inhibitory activity with an $IC_{50}$ value of 0.37 mg/ml. The fraction 5 was identified as dieckol and the inhibition pattern analyzed from Lineweaver-Burk plots revealed a non-competitive inhibitor. These results suggest that EB has potential as a natural anti-obesity agent.

Antioxidant Activity of Solvent Fraction from Broccoli Sprouts Cultivated at the Plant Factory System (식물공장에서 재배한 브로콜리 새싹 용매 분획물의 항산화 활성)

  • This study was designed to determine the antioxidant activity of solvent fractions of broccoli sprouts grown by controlling the growing environment at the plant factory system. Fractionation was achieved with chloroform, n-hexane, ethyl acetate, butanol, water by 70% EtOH extract of the broccoli sprouts. Each solvent fraction was put through TLC and HPLC to separate active components. Higher antioxidant activities were observed for the butanol and ethyl acetate layers. Further evaluation of each of the 5 layers (LH1 to LH5) of the butanol fraction showed that the refined LH3 extract had a high antioxidant effect. Components with similar Rf values from TLC had the same retention times and peaks in the HPLC analysis. It was also determined that the sulforaphane content was high at the chloroform and butanol layers and the sulforaphane was responsible for, the high antioxidant activity. Thus, to use for functional materials, the butanol extract/layer of broccoli sprouts is recommended as the most effective.

Antibacterial Activity and Nitric Oxide Production Inhibitory Activity of the Extract and its Fractions from the Leaves of Prunus sargentii (산벚나무 잎 추출물 및 분획물의 항균활성과 Nitric Oxide 생성억제 활성)

  • This study was carried out to investigate the antibacterial activity against pathogens of acne and the anti-inflammatory effect of 75% ethanol extract and its fractions from the leaves of Prunus sargentii. In the antibacterial activity by the disc diffusion assay, the extract showed the highest effect against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis in 5 mg/disc. However, the ethyl acetate fraction showed the highest antibacterial activity in 1 mg/disc. On the other hand, the hexane and chloroform fraction showed strong nitric oxide (NO) production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability of Raw 264.7 by MTT assay, the extract and all fractions were exhibited normal viabilities as nontoxic result. Consequently, the extract from the leaves of P. sargentii and its ethyl acetate fraction could be applicable to functional materials for antibacterial activity related fields. Moreover, the hexane and chloroform fraction could be applicable to candidate materials as anti-inflammatory agent.

Antimicrobial and Antioxidant Activities of Various Solvent Extract from Different Parts of Loquat (Eriobotrya japonica, Lindl.) (비파 부위별 용매추출물의 항균 및 항산화 활성)

  • Antimicrobial and antioxidative activities of various solvent extracts from different parts of the loquat were investigated to process it as the functional food. The extraction yield showed higher in butanol fraction of 4.95% in peel and 4.42% in seed than others, but water fraction showed high extraction yield of 3.89% in leafs 23.6% in the seedless fruit and 21.1% in fresh, In the antimicrobial activity test, ethyl acetate fraction that leaf and fruit excluded seed inhibited higher 19, 15 mm and 16, 15 m in clear zone far Staphylococcus aureus and Escherichia coli in each. Ethyl acetate fraction that extracted from a series of polar and nonpolar solvent fractionation of methanol extracts showed better results in the hydrogen donating activity of 82% in leaf, 74% in seedless fruit, 68% in seed, 52% in peel and 30% in fresh.

Identification and Bioassay of Bioactive Compounds Isolated from Phytolacca americana (미국자리공 (Phytolacca americana)에서 추출한 생물활성 물질의 동정 및 생물검정에 관 하여)

  • This study was carried out for the identification and bioassay of bioactive compounds which isolated from Phyutolacca americana L. obtained results are summarized as follows: 1. Two biological active compounds were found from crude extracts of P. americana roots, using the systematic solvent fractionations, such as ethyl acetate fraction and butanol fraction. One biological compound was isolated from the ethyl acetate fraction with silicagel column chromatography, which was identified as a ${\alpha}-spinasterol$ by spectral analysis of IR, H-NMR, C-NMR and MS. The other one is isolated from butanol fraction which was identified as phytolaccoside E by spectral analysis of IR, H-NMR, C-NMR and MS. 2. The ${\alpha}-spinasterol$ and phytolaccoside E induced necrosis of primary root and resulted in death of the tested plant. These two compounds strongly inhibited to the growth of Mucor racemous and Phytophthora infestants but did not inhibited the growth of Colletotricum lagenarium and Fusarium oxisporum. 3. Cytotoxicity of the two biological active compound was exammined to the two different animal cancer cell lines (L1210, K562). The phytolaccoside E has some cytotoxical activity to the growth of cancer cell lines (L1210, K562) but $\alpha$-spinasterol has not cytotoxical effect.

Study of Gagamtongsung-San about cytotoxicity in L1210 (가감통선산(加減通聖散)의 여러 가지 분획에 따른 L1210 암(癌) 세포주(細胞株) 억제(抑制) 효과(效果))

  • Objectives : The purpose of this study was to investigate inhibitory effect of Gagamtongsung-San(GTS) on the cancer. Methods : This study estimated the cytotoxicity of GTS about L1210 and NIH3T3. We used GTS extract distilled with water, n-Hexane, Ethyl acetate and Butanol. The cytotoxicitys of GTS about cancer cells and normal cells were tested using a colorimetric tetrazoliun assay(MTT assay). Results : The results of this study were obtained as follow ; l. Cytotoxicity of water extract of GTS in L1210 cell lines was significantly increased, compared with NIH3T3. 2. n-Hexane fraction of GTS had similar cytotoxicity between L1210 and NIH3T3, and that have similar $IC_{50}$ of water extract of GTS at 276 ${\mu}g/ml$ 3. Ethyl acetate fraction of GTS had low degree cytotoxicity both L1210 and NIH3T3 cell lines. 4. Butanol fraction of GTS had cytotoxicity between L1210 and NIH3T3. Significantly, Cytotoxicity of GTS in L1210 cell lines was significant increased. 5. $H_2O$ fraction of GTS had no cytotoxicity both L1210 and NIH3T3.

Inhibitory Effects of Ixeris Dentata on the Mutagenicity of Aflatoxin $B_1$, N-methyl-N연-nitro-N-nitrosoguanidine and the Growth of MG-63 Human Osteosarcoma Cells (씀바귀 추출물들의 돌연변이 유발 억제 및 MG-63 암세포 성장 저해 효과)

  • Ixeris dentata was extracted with methanol and then the methanol extract was further fractionated to hexane, chloroform, ethyl acetate, butanol and aqueous fraction. The methanol extract of lxeris dentata had the strong antimutagenic effect on the aflatoxin B1(AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Ames mutagenicity test and SOS chromotest. Among the solvent extracted fractions from the methanol extract, the chloroform fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity of AFB1 with inhibition rate of 74 percent. The methanol extract of Ixeris dentata also revealed the inhibitory effect on the growth of MG-63 human osteosarcoma cells after 6 days of breeding at 37℃. The chloroform fraction and the ethyl acetate fraction from the methanol extract of lxeris dentata were most effective and inhibited the growth of MG-63 cells by 97 and 93 percent, respectively. It is suggested that the inhibitory effects of lxeris dentata on the mutagenicity and the growth of MG-63 human osteosarcoma cells are strong in the lipid soluble fractions.

Inhibitory Effect of Methanol Extract and Its Fractions from Grape Seeds on Mushroom Tyrosinase (포도씨 추출물과 분획물의 Tyrosinase 저해활성)

  • The objectives of this study were to determine inhibitory effect on tyrosinase activity and antioxidant activity of the methanolic extract from grape seeds and to investigate relationships between tyrosinase inhibitory activity and antioxidant activity of the extract. The 80% methanol extracts of grape seeds were fractionated subsequently with hexane, chloroform, ethyl acetate and water. The methanolic extract and fractions from grape seeds inhibited tyrosinase activity in a concentration dependent manner. The methanolic extracts showed the highest inhibitory effects on tyrosinase activity. The inhibitory effects of the ethyl acetate fraction from the methanolic extract on tyrosinase activity was higher than other fractions. The ethyl acetate fraction from methanolic extracts showed higher antioxidant activity and contained higher polyphenolic and flavonoid contents compared to other fractions. The correlation coefficients among the polyphenoilc content of methanolic extracts, ABTS radical cation scavenging activity and inhibitory effect of tyrosinase were relatively high. These results suggest that grape seeds may have potential as a depigmentation agent for cosmetics and functional food products.

Screening and Characterization of Anticholesterogenic Substances from Edible Plant Extracts (식용식물 추출물로부터 콜레스테롤 합성 저해제의 검색 및 분리)

  • Inhibitory effects of several edible plant extracts against 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase, a rate-limiting enzyme in the biosynthesis of cholesterol, were screened. Inhibition rates of $10{\sim}15%$ were observed with hot water extracts of Allium fistulosum, Allium sativum and Cucurbita maxima. Methanol extracts of Aster scaber, Allium sativum, Zingiber officinale, Oenanthe javanica and Angelica keiskei effectively reduced the enzyme activity with inhibition rates of $29{\sim}51%$. The methanol extract of Angelica keiskei was fractionated sequentially with chloroform, ethyl acetate and n-butanol. Of the fractions ethyl acetate fraction showed the highest inhibition against the enzyme. $Luteolin-7-O-{\beta}-D-glucoside$ and hyperoside isolated from the ethyl acetate fraction of Angelica keiskei inhibited the enzyme activity by 65.5% and 14.8%, respectively, at the concentration of $30{\mu}M$.

Isolation and Identification of Antioxidant Substances from the Stems of Butterbur (Petasites japonicus) (머위(Petasites japonicus) 엽병으로부터 항산화 물질의 분리 및 동정)

  • The stems of P etasites japonicus were extracted with ethanol and then partitioned with hexane, chloroform, ethyl acetate, n-butanol and water, successively. The antioxidant potency of five crude fractions were determined using (1) 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, (2) thiobarbituric acid reactive substances (TBARS) assay in the linoleic acid model system, and (3) lipoxygenase inhibition assay. Among the crude fractions, the ethyl acetate fraction exhibited the most potent antioxidant effect. By activity-guided fractionation, compound PJ-4 was isolated from the ethyl acetate fraction through the repeated silica gel open column chromatography. The chemical structure of the isolated compound was determined as kaempferol by $^1H-$ and $^{13}C$-NMR analysis and its antioxidative capacity was further investigated. DPPH radical scavenging activity of the compound was 65.76% at the concentration of $100 \;{\mu}g/mL$. The inhibitory activity of the compound against lipid peroxidation and lipoxygenase exhibited 43.47% and 58.60%, respectively at the concentration of $100\;{\mu}g/mL$. The result suggests that the compound may serve as a useful natural antioxidant and furthermore indicates the possibility of developing the stems of Petasites japonicus as a natural antioxidant source.
