  • The methanolic extract of Lysimachiae christinae Herba, which has been used for diuretic, calculi remover and jaundice remedy in oriental countries, was found to possess analgesic and antiinflammatory effect in Freund's complete adjuvant treated rat. From ethyl acetate fraction of the herb a phenyl propanoid isoferulic acid was isolated as an active principle.

Functional Chemical Components and Their Biological Activities of Houttuynia cordata and Lespedeza cuneata (어성초와 야관문의 기능성 성분 분석과 항산화, 항고혈압, 및 항당뇨 활성)

  • For this study, we prepared organic solvent fractions from methanol extracts of Houttuynia cordata and Lespedeza cuneate, and analyzed their chemical components and various biological functions such as anti-oxidation, angiotensin-converting enzyme (ACE) inhibition, and α-glucosidase inhibitory activities. We found that DPPH radical scavenging activity was highest in the ethyl acetate fractions of Houttuynia cordata (90.8%) and Lespedeza cuneata (91.2%), whereas ABTS radical scavenging activity was highest in the ethyl acetate fractions of Houttuynia cordata (86.1%) and the chloroform fractions of Lespedeza cuneata (95.6%). FRAP activity was highest in the ethyl acetate fraction of Houttuynia cordata (360.1 mg TE/g) and Lespedeza cuneata (239.2 mg TE/g). ACE inhibitory activity was highest in the chloroform fraction of Houttuynia cordata (13.2%) and Lespedeza cuneata (35.2%). And, α-glucosidase inhibitory activity was highest in the ethyl acetate fraction of Houttuynia cordata (56.3%), and the water residue of Lespedeza cuneata (93.6%). Finally, we investigated the DPPH radical scavenging activity of 20 types of pure compounds identified in Houttuynia cordata and Lespedeza cuneate. The results show that quercetin demonstrates the highest DPPH radical scavenging activity. Overall, these results help us to understand the functional chemical components of Houttuynia cordata and Lespedeza cuneate and the biological effects of these components.

In vitro Inhibitory Activity on Rat Intestinal Mucosa ${\alpha}-glucosidase$ by Rice Hull Extract (왕겨추출물의 쥐소장점막 ${\alpha}-glucosidase$에 대한 in vitro에서의 저해효과)

  • In order to search for the way to utilize rice hull as a renewable resource, the inhibition on ${\alpha}-glucosidase$ and the fractionation of rice hull extract was investigated. An ethanol extract of rice hull from Japonica-type rice seeds exhibited 30% inhibitory activity on rat intestinal brush border ${\alpha}-glucosidase$ (1.4 mU/mL) in vitro at the concentration of 0.8 mg/mL using 6 mM p-nitrophenyl ${\alpha}-D-glucopyranoside$ as a substrate $(IC_{50}\;162\;mg/mL)$. Among the fractions obtained by partitioning the ethanol extract successively with solvents, the ethyl acetate fraction at the concentration of 0.8 mg/mL was found to exhibit the most potent inhibitory activity i.e. 65% inhibition of ${\alpha}-glucosidase\;(IC_{50}\;0.14\;mg/mL)$. Silica gel column chromatography of the ethyl acetate fraction exhibited slightly higher (90%) inhibitory activity, and its subsequent fractionation by Sephadex LH-20 column chromatography did not improve inhibitory activity. Considering the inhibitory activity and yield, the ethyl acetate fraction obtained by the solvent-partitioning process would be a candidate for the hypoglycemic food if it has in vivo effectiveness.

Ethyl Acetate Fraction of Cnidium monnieri(L). Cussion Suppresses PAM plus A23187-induced Inflammation Reaction through Blockade of NF-κB and MAPK activation (벌사상자 Ethyl Acetate 분획물의 항염증활성연구)

  • Cnidium monnieri (L). Cussion is used as a tonic agent in traditional oriental medicine. However, the molecular mechanism of mast cell-mediated anti-inflammatory modulation has not been fully understood. The aim of the present study was to demonstrate the effects of Cnidium monnieri (L). Cussion eathyl acetate fraction on the expression of pro-inflammatory cytokines, as well as to elucidate its mechanism of action in the human mast cell line (HMC-1). Cells were stimulated with phorbol 12-myristate 13-acetate (PMA) plus A23187 in the presence or absence of Cnidium monnieri (L). Cussion eathyl acetate fraction. Cnidium monnieri (L). Cussion eathyl acetate fraction significantly inhibited the PMA plus A23187-induction of inflammatory cytokines such as tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-6 and IL-8. Moreover, EtOAc fraction attenuated cyclooxygenase (COX)-2 expression. In activated HMC-1 cells, phosphorylation of extra-signal response kinase (ERK) 1/2 decreased after treatment with EtOAc fraction. Moreover EtOAc fraction inhibited PMA plus A23187-induced nuclear factor (NF)-${\kappa}B$ activation, $I{\kappa}B$ degradation. EtOAc fraction suppressed the expression of TNF-$\alpha$, IL-6, IL-8 through a decrease in the ERK 1/2, as well as activation of NF-${\kappa}B$. These results indicated that Cnidium monnieri (L). Cussion EtOAc fraction exerted a regulatory effect on inflammatory reactions mediated by mast cells.

Antimicrobial and Antioxidative Effects of Roselle (Hibiscus sabdariffa L.) Flower Extract and Its Fractions on Skin Microorganisms and Oxidation

  • Antimicrobial and antioxidative effects of 14 different herbal flower extracts on skin microorganisms and oxidation were tested in this research. Herbal flower extracts were prepared with 70% ethanol. Among the herbal flower extracts, roselle (Hibiscus sabdariffa L.) flower extract showed the highest antimicrobial activity against Staphylococcus epidermidis as determined by a paper disc method. The seventy % ethanol extract of roselle flower was fractionated by sequential hexane, chloroform, ethyl acetate, n-butanol, and water fractionation. The growth of S. epidermidis, Streptomyces collinus, Streptomyces coeruleoprunus, Salmonella enteritidis, Vibrio parahaemolyticus, and Malassezia pachydermatis was most efficiently inhibited by ethyl acetate fraction of roselle flower extract as determined by a paper disc method and growth inhibition curves. In addition, the ethyl acetate fraction, water fraction and butanol fraction showed free radical scavenging and DNA cleavage inhibition activities. These results demonstrate that roselle flowers hold antimicrobial and antioxidative activities against skin microorganisms and oxidants.

  • Tyrosinase catalyzes the rate-limiting steps in melanin biosynthesis which is involved in skin-coloring and local hyperpigmentation of human beings, and unfavorable darkening of food products, Inhibitory effects on 3,4-dihydroxy-phenylalanine (dopa) oxidase activity of tyrosinase by 594 kinds of herbal extracts prepared from herbal medicines and wild plants in Korea were estimated. Two herbal extracts prepared from radicis cortex of Morus alba and rhizoma of Curcuma longa were selected as those exhibiting potent inhibitory effects on the enzyme activity. These herbal extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol, and polar residue. The inhibitory effects on dopa oxidase activity of tyrosinase were shown in ethyl acetate fraction of Morus alba, and in methylene chloride fraction of Curcuma langa. The ethyl acetate fraction of Marus alba exhibited 50% of inhibition on dopa oxidase activity of tyrosinase at the concentration of 12 ${\mu}g/ml$, and methylene chloride fraction of Curcuma langa at 51 ${\mu}g/ml$.

Antimicrobial Activities and Phenolic Compounds of Pyroligneous Liquor (목초액의 항균활성과 페놀화합물의 함량)

  • Antimicrobial activities of pyroligneous liquor were investigated by determining Minimal Inhibitory Concentration (MIC). The solvent extracts of pyroligneous liquor, which were extracted by using solvents with different polarities such as hexane, ethylacetate, or butanol. The activities were examined by disc diffusion method using MIC against 7 food poisoning microbe type strains. Antimicrobial activities were shown in hexane, ethylacetate, butanol, and aqueous fractions of pyroligneous liquor. Among the four fractions, ethylacetate fraction showed the highest inhibitory effect on the microorganism such as Shigella sonnei, and Yersinia enterocolitica at the concentration of 2.0 mg/disc. The purified P-1 and P-2 fractions isolated by silica gel column chromatography from ethylacetate fraction of pyroligneous liquor had the highest antimicrobial activity. The total phenolic compounds content in ethylacetate, hexane, butanol, and aqueous fraction was 488.3 mg/g, 403.8 mg/g, 83.6mg/g, and 74.5 mg/g, respectively. Taken together, these results suggest that the ethyl acetate fraction could be suitable for the development of isolation and identification of antimicrobial compound from pyroligneous liquor, resulting from the above antimicrobial activity.

Inhibitory Effects of Paeonia suffruticosa Extracts on Maltase and Sucrase

  • This study was carried out to investigate inhibitory effect of extracts from root cortex of Paeonia suffruticosa on ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) and postprandial hyperglycemia. Methanol extract and organic solvent (n-hexane, ethyl acetate, butanol, aqueous) fractions from the crude drug were determined for the inhibitory activities against maltase, sucrase and ${\alpha}-amylase$. The methanol extract from the crude drug strongly inhibited maltase (72%) and sucrase (76%) at the concentration of $100\;{\mu}g/ml$. Among the fractions examined, the ethyl acetate fraction from the natural plant drug showed potent inhibitory effects on maltase (85%) and sucrase (81%) at the concentration of $100\;{\mu}g/ml$. The ethyl acetate fraction from root cortex of Paeonia suffruticosa also exhibited significant reductions (21%) of blood glucose elevation in mice loaded with maltose.

  • The antimicrobial activities of three parts of Korean sword bean , such as whole seed, dotyledon and hull were investigated . Yields were higher hull other parts in water and methanol extract. Methanol extract in seed parts was fractionated with different solvents, such as hexane, chloroform, ethyl acetate, butanol and water, yields of water fractino showed 3.3% , 2.5% and 0.3% in whole seed, 채쇼ledon and hull, respectively. In solvent extracts using methanol, hexane, chloroform, and water , methanol extract showed the most effective antimicrobial activities. Antimicrobial activities of ethyl acetate fraction of methanol extractwas higher than those of other fractions.

  • The methanolic extract of fruiting body of Paecilomyces tenuipes DGUM 32001 showed significant cytotoxicity against human cancer cells: HepG2 and MCF-7. The methanolic extract was further fractionated with organic solvents such as chloroform and ethyl acetate in that order. Among the fractions tested, the ethyl acetate fraction showed the highest cytotoxicity against the carcinoma tested. The $IC_{50}$ values of ethyl acetate fraction against HepG and MCF-7 were 40 and 9.6 ${\mu}g/ml$, respectively.
