In Vitro Test on Allelopathic Effects of Leaf Extracts from Phytolacca americana and Armoracia rusticana (미국자리공 및 겨자무 잎 추출물의 Allelopathy 효과 기내 검정)

  • Allelopathic effects on some crops(rice, barley, wheat, Chinese cabbage, leaf mustard, onion, welsh onion, tobacco, red pepper) for extracts of Phytolacca americana and Armoracia rusticana were investigated in MS solid medium. Germination percentage, radicle length, hypocotyl length, plant height, fresh weight, dry weight, and seedling vigor were rapidly inhibited by methanol extract from Phytolacca americana according to increase of the concentration in test plants. Autotoxicity of Phytolacca americana began to appear at concentrations greater than 50$\mu$l methanol extracts per 20ml medium. The fresh weight and dry weight of Phytolacca americana were also decreased by its own methanol extract according to increase of the concentration. Initial amounts of growth of all crops were also inhibited by methanol extracts from Armoracia rusticana, but the inhibitory effects were lowered than those of methanol extracts of Phytolacca americana. Of four fractions, ethyl acetate, petroleum ether, $1^{st}H_2O, 2^{nd}H_2O$, reextracted from methanol extracts of Phytolacca americana, the ethyl acetate fraction showed the highest allelopathic effects on germination percentage and initial amounts of growth. Chlorophyll contents of rice, barley and Chinese cabbage were more inhibited in the ethyl acetate fraction than in the other fractions. Free proline content of Chinese cabbage was increased 31.2 times in the 100$\mu l$ ethyl acetate fraction, and the contents of rice and barley were also increased according to concentration levels in the ethyl acetate fraction.

  • The Cytotoxic activity of the ethyl acetate soluble fraction of Perilla frutescens on human oral epithelioid carcinoma cell lines was evaluated by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyl-2H-tetrazalium bromide (MTT) colorimetric method. The light microscopic study was carried out to observe the morphological changes of cultured human oral epitheloid carcinoma cell lines (KB) These results were obtained as follows; 1. subfraction 1 of the ethyl acetate soluble fraction of perilla frutescens was shown significant cytotoxic activity (p<0.001) at 10-2 mg/ml concentration, this concentration was decreased the cytotoxic activity of 70.3% . 2. The comparison of IC50 values of various subfractions in human oral epithelioid carcinoma cell lines was exhibited that their susceptibility to these subfractions decreased in the following order ; subfraction 5 > subfraction 4 > subfraction 3 > subfraction 2 > subfraction 1 by MTT assay. In light microscopy, the subfraction 1 of the ethyl acetate soluble fraction of Perilla frutescens showed the highest cytotoxic activity. These findings suggest that subfraction 1 possessed the most cytotoxic constituents.

  • Antimicrobial activity of UlGeum (Curcuma longa L.) was investigated. Methanol extract of dried UlGeum was fractionated to hexane, chloroform, ethyl acetate, butanol and water fraction. The antimicrobial activity of five crude fractions were examined using impregnated paper disk agar diffusion. Ethyl acetate fraction showed the highest inhibitory effect on the microorganisms such as B. subtilis, S. aureus, E. coli, L. monocytogenes and V. parahaemolyticus at 1,000 ${\mu}g$/disc. Ethyl acetate fraction was further fractionated by silica gel column and thin layer chromatography (TLC). The antimicrobial compound was isolated from their fractions and its chemical structure was identified as a 2,3-dihydrobenzofuran by GC-MS and $^1H$-NMR.

Antioxidant Activities of Ulva lactuca Extracts with Different Solvents (갈파래(Ulva lactuca) 용매별 분획의 항산화활성)

  • The fractions of Ulva lactuca were studied to verify the antioxidant activities. The fractions from the ethanol extract of U. lactuca were prepared by the systematic extraction procedure with the solvents such as hexane, ethyl ether, ethyl acetate, butanol and $H_2O$. Furthermore, ethyl ether, ethyl acetate and aqueous fractions of U. lactuca were purified using HPLC. The antioxidant activities of purified samples from ethyl ether, ethyl acetate, and aqueous fractions were investigated using 1,1-diphenyl-2-picryl-hydrazil (DPPH). L-ascorbic acid, a positive control showed the highest DPPH radical scavenging activity. In addition, purified sample from aqueous fraction also showed relatively high activity. Purified sample from ethyl acetate fraction showed moderate activity, but purified sample from ethyl ether fraction showed the lowest activity. Dose dependent patterns were observed on all three samples tested. The lipid peroxidation inhibition activities of these three purified samples were also investigated. Purified sample from ethyl ether fraction of U. lactuca showed the highest activity and as strong activity as that of $\alpha-tocopherol$, a positive control. These results suggest that U. lactuca may be a useful candidate for a natural antioxidant agent.

Effects of Antioxidative, DPPH Radical Scavenging Activity and Antithrombogenic by the Extract of Sancho (Zanthoxylum Schinilolium) (산초(Zanthoxylum Schinifolium) 뿌리, 줄기 및 잎 추출물의 항산화, DDPH Radical 소거 작용 및 항혈전 효과)

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. Chemopreventive Potential of an Ethyl Acetate Fraction from Curcuma Longa is Associated with Upregulation of p57kip2 and Rad9 in the PC-3M Prostate Cancer Cell Line

  • Background: Turmeric ($Curcuma$ $longa$) has been shown to possess anti-inflammatory, antioxidant and antitumor properties. However, despite the progress in research with $C.$ $longa$, there is still a big lacuna in the information on the active principles and their molecular targets. More particularly very little is known about the role of cell cycle genes $p57^{kip2}$ and Rad9 during chemoprevention by turmeric and its derivatives especially in prostate cancer cell lines. Methods: Accordingly, in this study, we have examined the antitumor effect of several extracts of $C.$ $longa$ rhizomes by successive fractionation in clonogenic assays using highly metastatic PC-3M prostate cancer cell line. Results: A mixture of isopropyl alcohol: acetone: water: chloroform: and methanol extract of $C.$ $longa$ showed significant bioactivity. Further partition of this extract showed that bioactivity resides in the dichloromethane soluble fraction. Column chromatography of this fraction showed presence of biological activity only in ethyl acetate eluted fraction. HPLC, UV-Vis and Mass spectra studies showed presence three curcuminoids in this fraction besides few unidentified components. Conclusions: From these observations it was concluded that the ethyl acetate fraction showed not only inhibition of colony forming ability of PC-3M cells but also up-regulated cell cycle genes $p57^{kip2}$ and Rad9 and further reduced the migration and invasive ability of prostate cancer cells.

Antioxidant and Metalloproteinase Inhibitory Activities of Ethanol Extracts from Lespedeza cuneata G. Don (야관문 에탄올 추출물의 항산화 및 Metalloproteinase 저해 활성)

  • BACKGROUND: Lespedeza cuneata G. Don is a well-known medicinal plant. In this study, the biological activities of L. cuneata extracts were investigated. METHODS AND RESULTS: L. cuneata shoot was extracted with 30% ethanol and further fractionated with organic solvents. Total phenolic and flavonoid content, antioxidant activity, and matrix metalloproteinase inhibition effect of the extract and fractions were measured. Among the tested extract and fractions, the highest contents of total phenolic and flavonoids were found in ethyl acetate fraction (117.8 mg GAE/g and 35.9 mg QE/g, respectively). Ethyl acetate fraction showed the highest DPPH and ABTS radical scavenging activity, and the antioxidant activity of the other fractions followed the order n-hexane fraction>ethanol extract>methyl chloroform>n-butanol fraction. Inhibitory effect on the expression of matrix metalloproteinases (MMP1 and MMP3) was highest in the fraction of ethyl acetate, and n-butanol fraction also significantly inhibited the expression of MMP3. Antioxidant activities of L. cuneata extracts were significantly positively related to their phenolic and flavonoid content. CONCLUSION: Ethyl acetate fraction of L. cuneata ethanol extract showed potent antioxidant and matrix metalloproteinases inhibitory activities. Those activities might be related to the high total phenolic and flavonoid content of the extract.

Cytotoxic activities of various fractions extracted from some pharmaceutical insect relatives

  • This research was performed to screen the cytotoxic activities of some pharmaceutical insect relatives. Cytotoxic activities of total extract and fractions of hexane, ethyl acetate, methanol, water and boiling water were extracted from four pharmaceutical insect relatives: the Chinese gall, the cicada slough, the hornet nest and the batryticated silkworm. These extracts were investigated against the cancer cell lines of L1210, P388 and SNU-1 in vitro tests. Results showed that, ED, , against the cancer cell lines of L1210, P388 and SNU-1 were 0.55, 0.50, and $0.83{\mu}g/ml$ in the ethyl acetate fraction from the Chinese gall; 1.07, 2.19, and $2.24{\mu}g/ml$in the ethyl acetate fraction, 1.51, 1.26, and $1.45{\mu}g/ml$ in the water fraction and 1.48, 2.29, and $1.29{\mu}g/ml$in the boiling water fraction from the cicada slough; 3.31, 2.00, and $6.61\mug/ml$ in the water fraction from the hornet nest and 13.80, 19.95, and $31.62{\mu}g/ml$in the hexane fraction and 33.88, 21.88, and $25.12{\mu}g/ml$in the ethyl acetate fraction from the batryticated silkworm, respectively. All of the fractions mentioned above showed high cytotoxic activities and could be suggested for further studies in vivo tests.

Antioxidant Activity of Solvent Fraction from Black Garlic (흑마늘 용매 분획물의 항산화 활성)

  • To confirm antioxidant activity of black garlic, methanol extract of black garlic was fractioned by hexane, chloroform, ethyl acetate, buthanol and water. Antioxidant activities of solvent fractions were assayed in 100, 250, 500 and $1,000\;{\mu}g/mL$ concentrations. The contents of total phenol and flavonoids were significantly higher 5.5~11.6 times in chloroform, ethyl acetate and hexane fraction than other fractions. Antioxidant activities of solvent fractions were increased by higher sample concentrations and their activities were significantly higher in chloroform and ethyl acetate fractions than others. DPPH radical scavenging activity was over 50% in $1,000\;{\mu}g/mL$ concentration, except butanol and water fraction. In the same concentration, reducing power was also significantly lower in butanol and water fraction. ABTS radical scavenging activity was higher in hexane, chloroform and ethyl acetate fractions and was over 70% at $1,000\;{\mu}g/mL$ concentration. In $1,000\;{\mu}g/mL$ concentration, the range of hydroxy radical scavenging activity was 50.27~81.02% and SOD-like ability was 26.73~47.64%. Antioxidant activity in linoleic acid reaction system was significantly higher when storage time was longer and sample concentration was higher in non-polar solvent fractions. Nitrite scavenging activity was relatively higher than antioxidant activity and the activity in $100\;{\mu}g/mL$ concentration was over than 50%, except butanol fraction.

Antibaterial Activity and Skin Moisturizing Effect of Cedrela sinensis A. Juss Shoots Extracts (참죽나무 새순 추출물의 항균 활성과 피부 보습효과에 관한 연구)

  • In this study, we investigated the antibacterial activity and the skin moisturizing effect of cream contaning Cedrela sinensis A. Juss shoots extracts. MIC values of ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts on P. acnes and S. aureus were 0.13 %, 0.03% respectively and P. ovale and E. coli were 0.25 % equally. The results showed that the antibacterial activity of the ethyl acetate fraction on P. acnes and S. aureus were higher than the methyl paraben. The cream containing the ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts was formulated and its skin moisturizing effect was evaluated in a clinical trial. The skin hydration and transepidermal water loss (TEWL) were investigated after topical application of the cream. The skin hydrating effect of the cream containing Cedrela sinensis A. Juss shoots extract was 10 ~ 15 % higher than the placebo cream, and transepidermal water loss was decreased to 6.7 g/$m^2h$ in the subjects applied with cream containing the ethyl acetate fraction of Cedrela sinensis A. Juss shoots extracts. These results indicate that ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts could be applicable to new functional antibacterial cosmetics.
