In this study, we investigated the antimicrobial effect of 14 different herbal petal extracts on various foodborne and food spoilage bacteria. Herbal petal extracts were prepared with 70% ethanol followed by sequential hexane, chloroform, ethyl acetate, n-butanol, and water fractionation. Antimicrobial activity was highest in the ethanol fraction from roselle (Hibiscus sabdariffa L.) petals as determined by the paper disc method. The roselle ethanol extract retarded the growth of food spoilage bacteria in kimbap (rice rolled in dried laver). Foodborne microorganisms (e.g. Bacillus cereus and Clostridium perfringens), on the other hand, were most efficiently inhibited by the ethyl acetate fraction of the roselle petal extract as determined by growth inhibition curves. Our study shows that roselle petals harbor antimicrobial activity against foodborne and food spoilage microorganisms. The critical ingredient is highly enriched in the ethyl acetate fraction of the extract.
To search of more selective vasculogenic relaxation activity, the antioxidant activity of silkworm male pupae was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, and anticoagulant activity of them was measured clotting time in both activated partial thromboplastin time (aPTT). Because, most of cGMP-enhancing agent such as, sildenafil, promotes thrombin-induced platelet aggregation, developed unexplained thrombic conditions including heart attack. To search more suitable and safe drug for vasculogenic relaxation, we purified silkworm pupae male extract. The ethyl acetate extract of silkworm male pupae showed strong scavenging activity in both DPPH and aPTT anticoagulant activity. The antioxidant activity potential of the individual fraction was in order of ethyl acetate > n-butanol > chloroform > n-hexane. The ethyl acetate soluble fraction exhibiting strong anti-oxidant and anticoagulant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Cholesterol was isolated as one of the active principles from ethyl acetate fraction, together with, minor portion, ${\beta}-sitosterol$.
In this study we investigated the biological activities of Crataegi Fructus, including antioxidative, fibrinolytic, $\alpha$-glucosidase inhibitory, and thrombin inhibitory activities. Crataegi Fructus, hot water extract was fractionated into hexane, $CHCLl_3$, ethyl acetate, butanol, and water fractions, and each of these was assayed individually. The water fraction showed the highest extraction yield at 4.08% (w/w). The antioxidative activities of the water, ethyl acetate, and butanol fractions were 31.07%, 45.87%, 50.28%, and 91.74%, respectively. Assays for fibrinolytic activity indicated that only the butanol fraction has significant efficacy at 1.93 plasmin units/ml. Thrombin inhibitory assays indicated that the 10-fold dilutions of the $CHCLl_3$, ethyl acetate, and butanol fractions had inhibitory activities of 34.97%, 41.43%, and 58.10%, respectively. The 10-fold dilutions of the only ethyl acetate fraction demonstrated $\alpha$-glucosidase inhibitory activities of 75.07%. From the above results, we propose that extracts of Crataegi Fructus can be used as a material for the development of biofunctional foods.
This study compared the antimicrobial activity and chemical composition of two plants used as "Injin" (Korean herbal medicine), namely, Injinho (Artemisia capillaris Thunberg) and Haninjin (Artemisia iwayomogi Kitamura). The ethyl acetate and ether fractions of crude methanol extracts from A. capillaris and A. iwayomogi were tested against three grampositive bacteria (Bacillus cereus, Bacillus subtilis, Staphylococcus aureus), two gram-negative bacteria (Escherichia coli, Pseudomonas fluorescens), and a yeast (Saccharomyceus cerevisiae). The antimicrobial activity of the ethyl acetate and ether fraction of both plants was strong, but that of A. iwayomogi extracts was higher than that of A. capillaris extract for the microbes tested. The minimum inhibitory concentration of the ether and ethyl acetate fraction of A. iwayomogi was highest for P. fluorescens and lowest for S. aureus and E. coli. We analyzed the chemical composition of the ethyl acetate fraction of A. capillaris and A. iwayomogi using gas chromatography-mass spectrometry. The main components of A. capillaris and A. iwayomogi were escoparone (86.82%) and scopoletin (20.47%), respectively.
The antihistaminic action of eighteen herbal medicines was investigated by the radioligand binding and functional assays. The hexane fractions of Trichosanthis radix, Mori cortex radicis and Evodiae fructus dosedependently inhibited [$^3$H] mepyramine binding to H$_1$, receptor in guinea-pig brain homogenates and histamine-induced contraction of isolated guinea-pig ileum. Antihistaminic action of the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus was more potent than their antimuscarinic action evaluated from the inhibition of [$^3$H]QNB binding and carbachol response. The ethyl acetate and chloroform fractions from Scutellariae radix also inhibited histamine-induced contraction, but antihistaminic potencies of these fractions were almost identical with their antimuscarinic potencies. The hexane fractions of Mori cortex radicis and Evodiae fructus inhibited selectively the increase of histamine-induced cutaneous vascular Permeability in the rat dorsal skins. However, the ethyl acetate fraction from Scutellariae radix inhibited eqipotently the effects of histamine and serotonin on the vascular permeability. These results demonstrate that the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus have the selective histamine H$_1$receptor blocking activity.
We screened for antibacterial substances against methicillin-resistant Staphylococcus aureus (MRSA). Methanolic extract of Eisenia bicyclis exhibited anti-MRSA activity according to a disk diffusion assay. To identify the active compound(s), the methanolic extract was further fractionated using hexane, dichloromethane, ethyl acetate, and n-butanol. The ethyl acetate-soluble fraction showed both the greatest anti-MRSA activity and the highest polyphenol content. The minimum inhibitory concentrations of the ethyl acetate fraction ranged from 32 to 64 ${\mu}g$ per mL against methicillin-susceptible S. aureus and MRSA strains. High-performance liquid chromatography analysis revealed that both the methanolic extract and the ethyl acetate soluble fraction contained sizeable quantities of dieckol, which is a known anti-MRSA compound. Thus, these data strongly suggest that the anti-MRSA activity of E. bicyclis may be mediated by phlorotannins such as dieckol.

To evaluate the longevity property of Allium hookeri (Liliaceae) leaves, this study was performed. Ethanol extract of A. hookeri was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions, and lifespan-extending effects of the fractions were checked using Caenorhabditis elegans model system. The most active ethyl acetate-soluble fraction elevated heat stress tolerance, and increased expression of stress resistance protein. Further studies were performed to investigate several aging-related factors such as reproduction, food intake, growth and movement of C. elegans. The results revealed that there were no significant changes in aging-related factors including reproduction and food intake, however, ethyl acetate-soluble fraction treatment led to up-regulation of locomotory ability and growth of aged worms, suggesting ethyl acetate-soluble fraction affected healthspan as well as lifespan of nematode.
Heated onion juice was partitioned using the solvents hexane, chloroform, ethyl acetate, and butanol. The ethyl acetate fraction showed the strongest scavenging effect on the ABTS radical. The antioxidant activities of the ethyl acetate fraction from raw and heated onion (120, 130, and $140^{\circ}C$) were evaluated using radical scavenging assays. Radical and nitrite scavenging activities were higher in heated onion than raw onion, and the higher the temperature of heat treatment, the greater the radical and nitrite scavenging activities. Heated onion ($140^{\circ}C$, 2 hr) was more effective than raw onion, having higher DPPH radical scavenging (5.7-fold), hydroxyl radical scavenging (6.4-fold), superoxide radical scavenging (2.3-fold), hydrogen peroxide scavenging (11.8-fold), and nitrite scavenging (4.3-fold) activities. Onion increased its physiologically active materials after heating, and in this regard, heated onion can be used as biological material for the manufacture of health foods and supplements.

To enhance the utilization of chestnut husk discarded in the processing company antioxidative activities and compounds were investigated. Antioxidative activities of solvent fractions from chestnut husk were examined by benzoic acid hydroxylation method ferric thiocyanate method and DPPH test. Ethyl acetate fraction showed strong antioxidative activities comparable to BHA. Active compounds were isolated and purified from ethyl acetate fraction by Sephadex LH-20 column chromatography and preparative HPLC. A major active compound gallic acid was identified by $^{1}$H and $^{13}$ C-NMR. The phenolic acid contents was analyzed by GC and ellagic acid (172.22 mg%) and gallic acid (107.39 mg%) were major phenolic acid of chestnut husk.
Based on the following tests with fractions extracted from organic solvents such as benzene, chloroform, ethyl acetate and methanol, this study is to analyse the antimutagenicity of Lonicerae Flos. When carrying out Ames test with Salmonella typhimurium strain, it seemed that there was stronger antimutagenicity in TA 100 treated by MNNG than did one in TA 98 by NPD, and that there was stronger antimutagenicity through base pair exchange than one through frame shift. In the umu test, each fraction tended to inhibit the activity of ${\beta}-galactosidase$ induced by AF-2. As shown in these above tests, the ethyl acetate fraction was the strongest among four fractions. On the other hand, its component consisted of luteolin, apigenin, chlorogenic acid and five unknown ingredients. Of these unknown ingredients, E, F strongly tended to inhibit the activity of ${\beta}-galacosidase$. In addition, there was also the decrease in its activity of apigenin, luteolin and chlorogenic acid.
