To develop the safe and natural antimicrobial agents, the 68 ethanol extracts from the 61 different kinds of oriental herbal medicine were prepared and their antimicrobial activities were evaluated. The herbal medicine used were from China (46 kinds), South Korea (14 kinds), North Korea (5 kinds) and Vietnam (3 kinds), respectively, and the root (27 species) was popular part in this study. The average water content and extraction ratio for ethanol were 7.10% and 6.75%, respectively. Determination of antimicrobial activity by disc-diffusion assay at 0.5 mg/disc concentration showed that the extract of Angelica tenuissima Nakai (china), Illicium verum, Junci medulla, Rhus javanica L., Salvia miltiorrhiza Bunge and Syzygium aromaticum has strong antimicrobial activities against different food spoilage and pathogenic bacteria and fungi. Determination of MIC and MBC/MFC further showed that the extract of Syzygium aromaticum has MIC of 1.25 mg/mL and MBC/MFC of 1.25~5.00 mg/mL against Listeria monocytogenes, Bacillus subtilis, Staphylococcus epidermidis, Staphylococcus aureus, Salmonella typhimurium, Proteus vulgaris, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Saccharomyces cerevisiae. And, the extract of Junci medulla, Rhus javanica L. and Salvia miltiorrhiza Bunge showed strong antibacterial activities with MIC of 0.08~0.63 mg/mL and MBC/MFC of 0.08~2.50 mg/mL against the tested bacteria except E. coli and P. aeruginosa. In a while, the results of hemolytic activity of 68 different herbal extracts against human red blood cells showed that the extract of Angelica tenuissima Nakai has hemolytic activity at 0.5 mg/mL concentration. Therefore, Illicium verum, Junci medulla, Rhus javanica L., Salvia miltiorrhiza Bunge and Syzygium aromaticum were finally selected for natural antimicrobial resources. Further research on active substances and the mode of action of the selected herbal medicine is necessary.

This study was conducted to investigate the anti-oxydant and anti-winkle efficacy as cosmetics ingredient of Lysimachia christinae Hance. Recently, the study of wrinkle improvement of natural products has received continuous interest. so we looked at relationship between reactive oxygen species (ROS) generation and pro-collagen synthesis and matrix metalloproteinases (MMPs) through this study. L. christinae Hance were extracted with 70% ethanol (LcHE) and distilled water (LcHW), respectively, and the experiment was conducted. LcHE had better ROS inhibition effect than LcHW and showed no toxicity up to 250 ㎍/mL concentration as a result of MTT assay in HaCaT cells, so we selected LcHE and conducted the wrinkle improvement material study. We confirmed that the synthesis of type-1 pro-collagen reduced by UVB is activated through pro-collagen synthesis assay. we confirmed that LcHE inhibited the increase in MMP-1 -3 -9 of MMPs induced by UVB in skin cells through western blot and we also performed real-time PCR to confirm the effect of the extract with dependence of concentration at mRNA levels. Therefore, it is expected that Lysimachia christinae Hance is used as a natural material for cosmetics that can effectively prevent wrinkles and skin aging by UVB.

We produced the Red ginseng residue water(RGW), ethanol(RGE), 1,3-butylene glycol(RGB) extract from Red ginseng residues, analyzed the components of the extracts by HPLC, and evaluated the cell viability on B16F10, antioxidant and anti-wrinkle effects for application of cosmetics. As a result, RGW, RGE, RGB have various ginsenoside and its content of RGB were higher than RGW, RGE as component analysis by using high performance liquid chromatography(HPLC). RGW showed similar with RGB in cell viability on B16F10 which were higher than RGE. DPPH radical scavenging activity increased according to the RGE>RGB>RGW. SOD-like activity increased according to the RGB>RGE>RGW. Also, elastase inhibition effect increased according to the RGW>RGB>RGE. These results suggested that RGB and RGW may have potential for the application of antioxidant and anti-wrinkle effects for cosmetics.
Anti-diabetic effects of extracts and fractions of Sasa borealis (SB), white lotus roots (LR) and leaves (LL), and their mixture were determined in 3T3-L1 adipocytes and Min6 cells by investigating insulin-sensitizing activity and glucose-stimulated insulin secretion, respectively. SB, LR, LL, and mixture of SB, LR, and LL (3 : 2 : 3) were extracted using 70% ethanol, and m mixture extract was fractionated by XAD-4 column chromatography with serial mixture solvents of methanol and water. Fractional extractions were utilized for anti-diabetic effect assay. SB and LR extracts increased insulin-stimulated glucose uptake, but not as much as mixture of SB, LR, and LL. Significant insulin-sensitizing activities of 20 and 80% methanol fractions of SB, LR, and LL mixture extract were observed in 3T3-L1 adipocytes, giving 0.5 or $5\;{\mu}g/mL$ each fraction with 0.2 nM insulin to attain glucose uptake level similar to that attained by 10 nM insulin alone. Similar to pioglitazone, peroxisome proliferators-activated $receptor-{\gamma}\;(PPAR-{\gamma})$ agonist, 20 and 80% methanol fractions increased adipocytes by stimulating differentiation from fibroblasts and triglyceride synthesis. LL extract and 20, 60, and 80% methanol fractions of the mixture suppressed ${\alpha}-amylase$ activity, but did not modulate insulin secretion capacity of Min6 cells in both low and high glucose media. These data suggest 20 and 80% methanol tractions contain potential insulin sensitizers with functions similar to that of $PPAR-{\gamma}$ agonist. Crude extract of SB, LR, and LL mixture possibly improves glucose utilization by enhancing insulin-stimulated glucose uptake and inhibiting carbohydrate digestion without affecting insulin secretion in vivo.
This study aimed to investigate the suppressive effect of acorn extracts, by evaluating 70% ethanol extract (AE) and hot water extract (AW) using 3T3-L1 preadipocytes. We applied various levels (0, 100, 200, 300 and $500 {\mu}g/mL$) of AE and AW to 3T3-L1 preadipocytes. The cell viability of the 3T3-L1 preadipocytes was not affected by up $300 {\mu}g/mL$ of extracts, but was suppressed by level $500{\mu}g/mL$ of both AE and AW by 20% and 9% respectively. The accumulation of lipid droplets in differentiated 3T3-L1 preadipocytes was dose-dependently suppressed by AE and AW. Especially, at high concentrations ($300{\mu}g/mL$), AE (42%) was more effective than AW (41%). Reactive oxygen species (ROS) was also dose-dependently suppressed by treatment with AE (58%) and AW (52%). With regard to the mRNA related to differentiated 3T3-L1 preadipocytes, $PPAR-{\gamma}$ and aP2 were suppressed by treatment with AE (54 and 40%) and AW (38 and 18%). From our results, acorn extract (AE) has more suppressive effects than AW in differentiated 3T3-L1 preadipocytes. We therefore concluded that acorn has suppressive effects against obesity in differentiated 3T3-L1 cells due to antioxidation.

Quality characteristics of white lotus leaf tea (LLT) fermented with or without mycelial Paecilomyces japonica were investigated. Extraction yield and browning index of hot water extract from non fermented and fermented LLTs were higher than those of ethanol extract (p<0.05). In all LLTs, nutritional components such as total free sugar, free amino acids and minerals of hot water extracts were higher than those of ethanol extracts except for total organic acids (p<0.05). Contents of total free sugar and organic acids were markedly increased through fermentation process of mycelial Paecilomyces japonica. in the same solvent extracts (p<0.05). Contents of most taste components of fermented LLT were increased by mycelial solid fermentation (p<0.05), but total free amino acids of two extracts were decreased in the range of $37.1{\sim}67.2%$ as compared to non-fermented LLT. Fifty-nine volatile compounds were identified by GC and GC-MS, including 11 aldehydes, 14 alcohols, 11 ketones, 11 hydrocarbones and 12 acids. Aldehyde and ketone compounds were more identified in fermented LLT than in non-fermented LLT being abundant alcohol compounds by simultaneous steam distillation and extraction. The most abundant compounds of LLT identified in this study were curcumene followed by 2,6-bis(1,1-dimethylethyl)-4-methyl-phenol and cyclohexen. Main compounds of fermented LLT were 2,6-bis(1,1-dimethylethyl)-4-methyl-phenol, butanoic acid, furfural, benzaldehyde, hexanoic acid and 2(3H)-furanone.
The wild simulated ginseng (WSG) has been effectively used in folk medicine as a remedy against hepatic disease, hypertension and arthritic disease. However, there is still lack of scientific proof about its antioxidant capability. The present study has been conducted to evaluate the protective role of the WSG ethanol extract in the CCl4-induced oxidative stress and resultant hepatic disfunction in ICR mice. The electron donating abilities and IC50 of WSG etnanol extract were 76.86 ${\pm}$ 1.06% and 33.3 ${\mu}g$/mL (that of ascobic acid was 16.5 ${\mu}g$/mL), respectively. Total antioxidant status of WSG extract was 2.13 ${\pm}$ 0.06 mmoL/mg, while the values of ascorbic acid and BHT were 3.63 ${\pm}$ 0.06 and 3.12 ${\pm}$ 0.02, respectively. ICR mice (aged 3weeks) were fed for 4 weeks on AIN-93M diet and had free access to food and water. The animals were divided into three groups: normal group (intraperitoneally (i.p) injected with PBS at 100 ${\mu}L$/mouse), group C; CCl4-induced and without any treatment. (i.p injected only PBS, 100 ${\mu}L$ /mice), group G; CCl4-induced and treated with WSG (i.p injected with 5 mg WSG extract per mouse, suspended in 100 ${\mu}L$ phosphate buffer). After the i.p. injection of WSG or PBS (5 times for 7weeks), all mice were administered CCl4 in olive oil at the last day of the experiment, except for normal group. The normal group was administered only olive oil. Determination of plasma triglyceride, total cholersterol, fasting glucose and GPT activity was performed using automatic blood analyzer. To evaluate the protective effect against the oxidative stress, DNA fragmentation and TBARS were determined in blood leucocytes and RBC and hepatocyte, respectively. Body and organs weights and food intake did not show significant differences among the groups. Blood total cholesterol of group G was similar to that of normal group, which was the lowest in group C. The fasting blood glucose level was the highest in normal group (205.20 ${\pm}$ 135.24), which were decreased in group C (134.2 ${\pm}$ 79.31) and group G (126.48 ${\pm}$ 77.05). TBARS values in a red blood cell and hepatic tisuue homogenate were lower in group G comparing to the group C. DNA% in tail, tail length (TL) and tail moment (TM) of blood leucoocytes showed the highest values in group C (20.11 ${\pm}$ 2.47, 17.36 ${\pm}$ 2.58, 94.11 ${\pm}$ 12.29) and they were significantly diminished in group G (9.63 ${\pm}$ 1.19, 7.04 ${\pm}$ 1.50, 38.64 ${\pm}$ 7.60). In conclusion, wild simulated ginseng might be a protective agent against the oxidative stress.
This study was conducted to investigate effect of complex extract of various mushrooms and sharp toothed eel on the skin conditions with atopic dermatitis and acne symptoms. The total phenol concentration was increased in order of hot water> ethanol> ethyl acetate> petroleum ether> chloroform extract. Especially, when the hot water extract was used, it was about 2-3 fold higher than that of ethyl acetate, petroleumether, and chloroform extract. When the complex extract concentration was increased from 1.0 to 50 mg/L, the DPPH scavenging rate increased from 10.1 to 81.4%. The reduction power was sharply increased from 0.05 to 0.27 (700 nm) when the complex extract concentration was increased from 25 to 75 mg/L. However, above 100 mg/L, it was not decreased. In the case of SOD-like activity, it was 45.7% at 100 mg/L. Total numbers of patients with atopic dermatitis were 15 and 5 patients with severe acne symptoms. According to photos taken before and after the treatment and questionnaire results, considerable improvements in skin conditions are observed in the patients with atopic dermatitis and acne. For atopic patients, erythema and edema have been improved but the degree of effect was dependent on the individual's constitution. Concerning acne, the effect of coating of the extracts was prominent for first week and the degree decreased with time till 4 weeks. The complex extracts wereefficient in soothing rash and maturation. Side effects such as a scar were not detected during the application and treatment.
The purpose of this study was to investigate the chemical composition of the freeze-dried stems and fruit of the cactus Cheonnyuncho. The analysis showed that powdered stems have the highest fat content (1.91%) and the powdered fruits have he highest protein content (2.62%). The K content of the fruits higher than that of the stems, while the Ca, Mg, Na and P contents of the stems were higher than those of the fruits. Both the stems and fruits powders contained high levels of the amino acids glutamic acid and aspartic acid. The free sugars such as sucrose, fructose, and glucose were detected in both the stems and fruits. The 75% ethanol (EtOH) extract showed a relatively high antioxidative activity compared to those of the water and 75% methanol (MeOH) extracts. Furthermore, the 75% EtOH extract of the stem powder exhibited a total polyphenol content of 3.60 g/100 g, and a total flavonoid content of 2.00 g/100 g. The antioxidant activities of the stem and fruit powder extracts, measured in DPPH radical scavenging experiments, were higher than that of the control group.

To investigate the anti-diabetic and antioxidant effects of Opuntia humifusa cladodes, O. humifusa cladodes powder was extracted with 75% ethanol and fractionated with various solvents. Among the extracts fractionated with various solvents, ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activities of O. humifusa cladodes were highest ($89.94{\pm}1.15%$ and $29.01{\pm}3.03%$, respectively) in the ethyl acetate fraction. Further, total phenolic and flavonoid contents of the ethyl acetate fraction were the highest ($196.02{\pm}5.26$ and $114.00{\pm}10.03{\mu}g/mg$, respectively). DPPH and ABTS radical scavenging activities increased according to the concentration of O. humifusa cladodes extract, and those of the ethyl acetate fraction were the highest. Ferric reducing antioxidant powers of chloroform and the ethyl acetate fraction were higher than those of other fractions. Overall, the ethyl acetate fraction of O. humifusa cladodes showed the highest anti-diabetic and antioxidant effects. Results indicate that O. humifusa cladodes powder has potential as a useful ingredient with anti-diabetic and antioxidant effects.
