Selective Estrogen Receptor Modulation by Larrea nitida on MCF-7 Cell Proliferation and Immature Rat Uterus

  • Larrea nitida is a plant that belongs to the Zygophyllaceae family and is widely used in South America to treat inflammatory diseases, tumors and menstrual pain. However, its pharmacological activity remains unclear. In this study we evaluated the property of selective estrogen receptor modulator (SERM) of Larrea nitida extracts (LNE) as a phytoestrogen that can mimic, modulate or disrupt the actions of endogenous estrogens, depending on the tissue and relative amount of other SERMs. To investigate the property of SERM of LNE, we performed MCF-7 cell proliferation assays, estrogen response element (ERE)-luciferase reporter gene assay, human estrogen receptor (hER) binding assays and in vivo uterotrophic assay. To gain insight into the active principles, we performed a bioassay-guided analysis of LNE employing solvents of various polarities and using classical column chromatography, which yielded 16 fractions (LNs). LNE showed high binding affinities for $hER{\alpha}$ and $hER{\beta}$ with $IC_{50}$ values of $1.20{\times}10^{-7}$ g/ml and $1.00{\times}10^{-7}$ g/ml, respectively. LNE induced $17{\beta}$-estradiol (E2)-induced MCF-7 cell proliferation, however, it reduced the proliferation in the presence of E2. Furthermore, LNE had an atrophic effect in the uterus of immature rats through reducing the expression level of progesterone receptor (PR) proteins. LN08 and LN10 had more potent affinities for binding on $hER{\alpha}$ and ${\beta}$ than other fractions. Our results indicate that LNE had higher binding affinities for $hER{\beta}$ than $hER{\alpha}$, and showed SERM properties in MCF-7 breast cancer cells and the rat uterus. LNE may be useful for the treatment of estrogen-related conditions, such as female cancers and menopause.

프로게스테론의 비유전자 수준 작용 : 포유류 난소에서의 신호 전달 경로를 중심으로 (Non-Genomic Actions of Progesterone : Focussed on the Signaling Pathways in the Mammalian Ovary)

  • 본 논문은 포유동물 난소에서의 신속한 프로게스테론(P4) 선호 전달경로에 관해 현재 통용되는 지식을 요약하였다. P4는 안드로겐과 에스트로겐 합성 과정에서의 중요한 중간 산물이면서 그 자체로도 배란, 난포폐쇄(atresia), 황체형성과정(luteinization)에서 결정적인 역할을 하며, 모든 포유동물의 초기 임신 유지에 필수적이다. 이와 같은 생리적인 중요성에도 불구하고 포유동물 난소에서의 정확한 P4 작용기작은 아직까지도 완전히 알려져 있지 않다. 이러한 관점에서 볼 때, 비유전자 수준이면서 전사와 무관한 P4의 세포내 작용을 매개하는 수용체의 실체에 관해 오래 동안 계속된 의문과 논란은 과학적인 흥미를 유발하는 생식생리학의 주요 관심사이다. 포유류 난소에서 P4는 1) 잘 알려진 유전자 수준의 경로(genomic pathway)인 호르몬이 소위 고전적인 세포 내의 수용체에 결합하고, 이어 리간드-호르몬 복합체가 전사조절물질로 작용하여 표적 유전자 발현을 조절하거나, 2) 유전자에 직접 작용하지 않기 때문에 비유전자 수준이라 불리우는 경로(non-genomic pathway)로 작용한다. P4의 비유전자 수준 작용의 주요한 특징은 (i) 신속하고, (ii) 전사억제제에 반응하지 않고, (iii) 세포막과 연관된 물질들에 의해 신호가 전달된다. 아마도 난소에서 P4의 비유전자 수준 작용은 (a) 세포막 또는 그 근처에 위치한 고전적인 P4 수용체(PGR), (b) 세포막 프로게스틴 수용체(membrane progestin receptors; MPR $\alpha$, MPR $\beta$ and MPR $\gamma$) 패밀리, (c) progesterone receptor membrane component I(PGRMC1), 그리고 (d) serpine I mRNA binding protein(SERBP1)의 세포막 복합체에 의해 매개되는 것으로 추정된다. 포유류 난소에서의 P4 작용에 대한 완전한 이해를 위해서는 향후 많은 연구가 필요할 것이다.

이란산 흑석류 농축액과 그 제품의 성분 및 함유된 Phyto 에스트로겐류에 관한 연구 (Proximate Compositions and Selected Phytoestrogens of Iranian Black Pomegranate Extract and Its Products)

  • 천연 호르몬 보충 제제의 개발을 위한 기초 연구로 이란산 흑석류 과즙 농축액과 이를 이용한 제품의 화학성분들을 분석하였다. 석류 농축액의 일반성분은 수분 39.3%, 조지방 0.4%, 조단백질 0.9%, 조회분 1.4%, 그리고 탄수화물은 42.0%이었다. 아미노산 함량은 글루탐산이 1310.0ppm, 아스파르트산이 896.2ppm, 아르기닌이 877.7ppm, 페닐알라닌이 57.5ppm순으로 무기 성분들은 철분 6640.0ppm, 염소 3464.0ppm, 칼륨 2550.8ppm, 인 150.0ppm, 칼슘 80.0ppm순으로 많이 함유되어 있었다. 비타민은 5가지의 수용성 비타민이 함유되어 있었으며 그 중 비타민 C(20mg/100g)를 제외한 나머지 비타민들이 아주 적은 양이 있었다. 주요 지방산은 palmitic acid(8.3%)와 stearic acid(69.4%)로 전체 지방산의 약 60~80%를 차지하고 있었다. 또한 6종의 Phyto 및 에스트로겐류가 들어 있었으며 각각 daidzein 23.72ppm, quercetin 9,75ppm, catechin 1.48ppm, genistein 0.29ppm, 2,3-di-MeO-estradiol이 0.04ppm, 그리고 17$\beta$-에스트라디올이 0.15ppm이 함유되어 있었다. 이상의 결과들을 석류 농축액으로 제조한 제품 포에버 120과 칡과 대두 isoflavon 농축분말과 비교하였다.

Association of Cytochrome-17 (MspA1) Gene Polymorphism with Risk of Gall Bladder Stones and Cancer in North India

  • Background: Cholelithiasis is associated in 54%-98% of patients with carcinoma of the gallbladder, and a high incidence among females suggests a role of female hormones in the etiology of the disease. Cytochrome $P450C17{\alpha}$ (CYP-17) is a key enzyme involved in estrogen metabolism and polymorphisms in CYP-17 are associated with altered serum levels of estrogens. Thus, we investigated whether the CYP-17 MspA1 gene polymorphism might impact on risk of gall bladder cancers or gallstones, as well as to determine if this gene polymorphism might be linked with estrogen serum levels and lipid profile among the North Indian gall bladder cancer or gallstone patients. Materials and Methods: CYP-17 gene polymorphisms (MspA1) were genotyped with PCR-RFLP in cancer patients (n=96), stone patients (n=102), cancer + stone patients (n=52) and age/sex matched control subjects (n= 256). Lipid profile was estimated using a commercial kit and serum estrogen was measured using ELISA. Results: The majority of the patients in all groups were females. The lipid profile and estrogen level were significantly higher among the study as compared to control groups. The frequency of mutant allele A2 of CYP17 MspA1 gene polymorphism was higher among cancer (OR=5.13, 95% CI+3.10-8.51, p=0.0001), stone (OR=5.69, 95%CI=3.46-9.37, p=0.0001) and cancer + stone (OR=3.54, 95%CI=1.90-6.60, p=0.0001) when compared with the control group. However there was no significant association between genotypes of CYP17 MspA1 gene polymorphism and circulating serum level of estrogen and lipid profile. Conclusions: A higher frequency of mutant genotype A1A2 as well as mutant allele A2 of CYP-17 gene polymorphism is significantly associated with risk of gallbladder cancer and stones. Elevated levels of estrogen and an altered lipid profile can be used as predictors ofgall bladder stones and cancer in post menopausal females in India.

Construction of High Sensitive Detection System for Endocrine Disruptors with Yeast n-Alkane-assimilating Yarrowia lipolytica

  • To construct a highly sensitive detection system for endocrine disruptors (EDs), we have compared the activity of promoters with the n-alkane-inducible cytochrome P450 gene (ALK1), isocitrate lyase gene (ICL1), ribosomal protein S7 gene (RPS7), and the translation elongation factor-1${\alpha}$ gene (TEF1) for the heterologous gene in Yarrowia lipolytica. The promoters were introduced into the upstream of the lacZ or hERa reporter genes, respectively, and the activity was evaluated by ${\beta}$-galactosidase assay for lacZ and Western blot analysis for hER${\alpha}$. The expression analysis revealed that the ALK1 and ICL1 promoters were induced by n-decane and by EtOH, respectively. The constitutive promoter of RPS7 and TEF1 showed mostly a high level of expression in the presence of glucose and glycerol, respectively. In particular, the TEF1 promoter showed the highest ${\beta}$-galactosidase activity and a significant signal by Western blotting with the anti-estrogen receptor, compared with the other promoters. Moreover, the detection system was constructed with promoters linked to the upstream of the expression vector for the hER${\alpha}$ gene transformed into the Y. lipolytica with a chromosome-integrated lacZ reporter gene under the control of estrogen response elements (EREs). It was indicated that a combination of pTEF1p-hER${\alpha}$ and CXAU1-2XERE was the most effective system for the $E_2$-dependent induction of the ${\beta}$-galactosidase activity. This system showed the highest ${\beta}$-galactosidase activity at $10^{-6}\;M\;E_2$, and the activity could be detected at even the concentration of $10^{-10}\;M\;E_2$. As a result, we have constructed a strongly sensitive detection system with Y. lipolitica to evaluate recognized/suspected ED chemicals, such as natural/synthetic hormones, pesticides, and commercial chemicals. The results demonstrate the utility, sensitivity, and reproducibility of the system for identifying and characterizing environmental estrogens.

유방암 환자에 있어서 폐경상태에 따른 위험인자의 상관성 연구 (A Study on Relationship to Risk Factors according to Menopausal Status in Breast Cancer)

  • It is important to identify modifiable risk factors for breast cancer, because the breast cancer is one of the major causs of mortality among women. Some reported that obesity is a risk factor for breast cancer, but the results are not constant. Many risk factors are related to the duration of estrogenic stimulation of the breast. In general, early menarche and late menopause are positive risk factors. Human breast cancer has different characteristics according to the status of menopause(premenopause and postmenopause). In premenopausal women, about 60% of circulating estrogen is from the ovaries in the form of estradiol, and the remaining 40% is estrogen formed primarily in the adipose(fat) tissue via aromatization of androstenedion from the adrenal glands. After menopause this adipose cell production of estrone is the main source of estrogens and the level of estrone is maintained approximately at premenopausal levels. This study was undertaken to determine the role of body size and body mass index by status of menopause in development of breast cancer using retrospective case/control study. From March 1991 to February 1997 at the Wonkwang University Hospital, the breast cancer cases(n=72) and controls(n=86) were selected. By statistical analysis method, regression analysis, paired T-test and multiple logistic regression were done to estimate the influenced factors same as height, weight, BMI, age at menarche and age at menopause. The following results were obtained : 1. In premenopausal women, age at menarche was showed comparatively high correlation coefficients and BMI was described prominently highly in postmenopause. 2. At the results of multiple regression analysis, age at monarch, BMI and weight were showed as significant variables. In this method, critical factor ($R^2$) was 0.054. 3. Paired samples T-test was undertaken to test mean difference between two groups of cases and controls. The result of test performance showed a significant difference. 4. In comparison with women whose weight less than 50 kg, the ORs for the upper 5th group was 1.82(95% confidence interval). The heaviest women had a higher risk(OR=1.14, 95% confidence interval $1.12{\sim}1.31$, p=0.005). Higher body mass index was significantly associated with increased risk of premenopausal breast cancer (OR=1.01, 95% confidence interval $1.08{\sim}l.18$, p=0.05).

  • Estrogen과 estrogenic endocrine disrupting chemicals(EDCs)는 주로 스테로이드 수용체와 작용하여 다양한 표적 단백질 유전자의 전사를 조절한다. 구조적으로 estrogen 수용체와 유사한 estrogen 수용체형 수용체(estrogen receptorrelated receptor, ERR)는 포유동물에서 배발생 후기에 외배엽 형성과 관련되어 있다고 알려진 고아핵수용체(orphan nuclear receptor)이다. 본 연구에서는 해양무척추동물인 둥근성게(Strongylocentrotus nudus)의 발생배를 재료로 estradiol-$17{\beta}(E_2)$과 EDCs의 일종인 nonylphenol(NP)이 발생과정의 형태학적 변화와 $ERR{\beta}$ like 1의 mRNA발현에 미치는 영향을 조사하였다. $E_2$와 NP가 처리된 발생배는 발생속도가 지연되었으며, 초기 유생기에 가까운 후기배의 비정상적인 발달형태가 관찰되었다. 수정란부터 초기 유생기까지 측정한 결과, 이들 화학물질에 의해 $ERR{\beta}$ like 1 mRNA는 포배기에 급격히 감소하는 패턴을 보였다. 이상의 결과는 둥근성게의 초기 배가 $E_2$와 NP에 의해 비정상적으로 발생되며, $ERR{\beta}$ like 1의 감소가 이 비정상적 배발생과 관련되어 있음을 시사한다.

조피볼락, Sebastes schlegeli의 성분화 기간 중 Cytochrome P450 Aromatase 유전자의 발현 (Expression of Cytochrome P450 Aromatase Genes during Sex Differentiation in Korean Rockfish, Sebastes schlegeli)

  • 자웅이체형 어류의 성결정 및 성분화는 일반적으로 각 개체의 유전형을 따른다. 자연환경에서는 자신이 가진 유전정보의 조절에 따라 성분화 시기에 아로마테이즈 유전자의 발현이 증가하거나 감소하고, 그 결과 스테로이드 호르몬들의 조성이 결정되어 각기 다른 방향으로 성분화하는 것으로 알려져 있다. 본 연구에서는 해산 태생 어류인 조피볼락(Sebasts schlegeli)의 ovarian type aromatase (P450aromA)와 brain type aromatase (P450aromB)의 유전자를 부분적으로 클로닝하여, 이들의 염기서열을 밝혔으며, 각 유전자에 대한 primer를 제작한 후 성분화 기간 중 이 유전자들의 발현을 조사하였다. 조피볼락 아로마테이즈 유전자들은 조사를 시작한 출산 후 35일째에 여러 개체의 머리와 몸에서 각각 발현되었으나, 52일째에는 아로마테이즈 유전자들의 발현 개체수가 현저히 감소하였다. 그리고 출산 59일째에는 발현 개체 수가 다시 증가하였다. P450aromA과 P450aromB의 발현 양상은 전반적으로 유사하였으나, 45일째에는 P450aromB의 발현 개체수가 P450aromA 발현 개체 수보다 훨씬 많았다. 조직학적 분석을 통해 제시된 이 종의 성분화 시기는 지금까지 출산 $50{\sim}65$일 전후로 알려져 왔으나, 본 연구에서 나타난 아로마테이즈 유전자들의 발현 결과를 볼 때, 이 종의 실질적인 성분화는 알려진 것보다 최소한 $1{\sim}2$주 정도 빨리 진행될 가능성이 있다. 본 연구의 결과는 또한 태생어류의 성분화도 난생어류의 성분화에서 보고된 것과 마찬가지로 아로마테이즈의 작용과 밀접하게 연관되어 있다는 사실을 시사한다.

Adverse Reproductive Effects on Plasma Vitellogenin and Sex Steroid Levels, and Gonadosomatic Index in Juvenile Common Carps (Cyprinus carpio) Exposed to 17$\beta$-Estradiol and D-2-Ethylhexyl Phthalate

  • Environmental estrogens are natural or synthetic substances present in the aquatic environment, especially in effluent from sewage treatment. However, the adverse effects of these estrogenic substances on fish reproduction are unknown. Di-2-ethylhexyl phthalate (DEHP) is the most common phthalate, which Ps used as a plasticizer in polyvinylchloride (PVC), and it is widespread in the environment and has been found in aquatic organisms and sediments. Therefore, juvenile common carps (Cyprinus carpio) were exposed to nominal concentrations of 17$\beta$-estradiol (E2) (0.5, 5, 50 $\mu\textrm{g}$/L) and DEHP (10, 100, 500 $\mu\textrm{g}$/L) for 21 days, to determine the adverse reproductive effects of these compounds on plasma vitellogenin (VTG) induction, sex steroid level, and gonad weight. Electrophoresis (SDS-PAGE) revealed that much of VTG was induced in fish exposed to 5 and 50 E$_2$ $\mu\textrm{g}$/L, but none of DEHP exposure showed induction. Enzyme-linked immunosorbent assay (ELISA) revealed that VTG was significantly induced in fish exposed to 5 and 50 E$_2$ $\mu\textrm{g}$/L, and combination of 50 E$_2$ $\mu\textrm{g}$/L with 10 and 500 DEHP $\mu\textrm{g}$/L, but none of DEHP exposure showed induction. Analysis of sex steroid levels in some fish revealed that testosterone (T) was detected in both male and female fish of the control and DEHP exposures, but none of fish exposed to 22 concentrations had detectable testosterone level. On the other hand, E$_2$ exposure induced 17$\beta$-estradiol in plasma of male fish, but there was no induction of 17$\beta$-estradiol in plasma of male fish exposed to DEHP. Comparison of gonadosomatic index (GSI) revealed that maximal E$_2$ exposure inhibited ovarian growth, but maximal DEHP exposure stimulated testicular growth. The results indicated that those comparisons can be a useful bio-indicator for determining adverse reproductive effect of endocrine disrupting chemicals (EDCs).

파일럿 규모의 하수 처리 공정별 E-screen Assay에 의한 에스트로겐 활성과 내분비계 장애물질의 농도 평가 (Estimation of Estrogenic Activity by E-screen Assay and Stepwise Analysis of Endocrine Disruptors in Pilot Sewage Treatment Plant)

  • 물 재이용 목적으로 설계한 pilot scale의 하수 처리공정에서 배출되는 방류수 중 내분비계 장애물질을 GC/MS로 분석하였고, 처리 공정별로 제거율을 비교하였다. 각 처리 공정별 방류수에서 nonylphenol이 주로 검출되었고, 평균 $0.36{\sim}0.94$ ${\mu}g/L$으로 높게 검출되었으나, E2와 EE2는 처리수에서 정량 이하로 검출되었다. 내분비계 장애물질은 처리 공정별로 $50{\sim}100%$의 제거율을 보여주었다. E-screen assay에 의해 얻어진 양-반응 곡선에서 E2의 EC50값은 $9.0{\times}10^{-3}$ M로 bisphenol A와 p-octylphenol의 EC50값인 $2.736{\times}10^{-5}$ M, $9.760{\times}10^{-6}$ M에 비해 매우 높았다. 이는 알킬페놀류가 E2에 대한 상대적인 에스트로겐 활성도가 매우 낮음을 보여주었다. 환경 호르몬 농도와 이 물질의 상대적인 에스트로겐 활성도에 근거하여 계산된 에스트로겐 활성도(ng-EEQ/L)는 E-screen assay에 의해 실측한 총 에스트로겐 활성도(ng-EEQ/L)에 비해 평균 2배의 높은 활성도를 보여 주었다. 각 처리 공정별 방류수의 에스트로겐 활성도는 1 ng-EEQ/L 이하의 매우 낮은 활성도를 보여주었다.
