The Effects of GamiTakliSodocyum on Wound Healing (加味托里消毒飮이 瘡傷 治癒에 미치는 影響)

  • In order to investigate the effects of GamiTakliSodocyum(GTS) on wound healing, migration of epidermis, formation of granulation tissue and number of capillary within the granulation tissue were measured in diabetic mice by local application and NZW rabbits by local application and prescription of medicine in vivo, and proliferation of human epidermal keratinocytes and human dermal fibroblasts and composition of extracellular matrix were measured in vitro. The results were summerized as follows. 1. $2\%,\;10\%$ GTS remarkably increased migration of epidermis in diabetic mice by local application. 2. $2\%,\;10\%$ GTS remarkably increased formation of granulation tissue, number of neovascularization within the granulation tissue in diabetic mice by local application. 3. 5\%,\;10\%$ GTS remarkably increased migration of epidermis in NZW rabbits by local application. 4. $5\%,\;10\%$ GTS remarkably increased fonnation of granulation tissue, number of neovascularization within the granulation tissue in NZW rabbits by local application. 5. $5\%,\;10\%$ GTS increased migration of epidennis in NZW rabbits by prescription of medicine. 6. $5\%,\;10\%$ GTS increased formation of granulation tissue, number of neovascularization within the granulation tissue in NZW rabbits by prescription of medicine. 7. GTS didn't show effect on the proliferation of human epidermal keratinocytes. 8. GTS increased the proliferation of cultured human dermal fibroblasts. 9. GTS increased the expression of procoliagen ${\alpha}1(I) mRNA in cultured human dermal fibroblasts. 10. GTS increased the expression of fibronectin mRNA in cultured human dennal fibroblasts according to dosage of GTS using northern blot hybridization but didn't increase, using RT-PCR. From the above results, it is conclude that GTS might use on wound healing.

Effects of Simulated Acid Rain on the Shoot Growth and Internal Tissue of Arabidopsis thaliana (애기장대의 shoot 생장과 내부조직에 미치는 인공산성비의 영향)

  • This experiment was carried out to investigate the effects of simulated acid rain (SAR) in the shoot growth and internal structure of Arabidopsis thaliana. In the shoot growth, the plants treated with SAR (pH 3.0) for 15 days showed no morphological change compared to the control plants. Some change was observed in the internal structure of the stems: the epidermis and cortex tissues of the stems treated with SAR were partly damaged. The plants treated with SAR showed no noticeable difference compared to the control plants, but morphological changes were observed in the leaf. The leaves of the plants treated with SAR showed many white necrotic spots on the part of upper epidermis. A light microscopic examination of the leaves with necrotic spots showed that the upper epidermis was severely compressed with the damaged cuticle layer and the mesophyll cells were also damaged and compressed. However, noticeable structural change of vascular bundle cells was not observed.

  • Anatomy of Pinus koraiensis needles and Ginkgo biloba leaves was investigated after exposure to simulated acid rain, drought and salt solution. All of the stresses applied to the needle of Pinus koraiensis caused collapse of mesophyll tissues; epidermis was collapsed due to acid rain, severe compress and transformation of epidermis, mesophyll, and endodermis to drought, and collapse of phloem and transfusion tissue to salt solution. Spongy mesophyll tissues, however, collapsed in the leaves of Ginkgo biloba; epidermis and palisade tissues collapsed due to acid rain, compress and transformation of spongy mesophyll tissues to drought, and hypertrophy and collapse of endodermis and transfusion tissues to salt solution.

An experimental study for the development of prescription on atopic dermatitis (아토피 피부염 외치치료제 처방 개발을 위한 실험적 연구)

  • Objectives : Atotang was composed of 10 kinds of traditional medicinal herb. This research was performed to examine biological effects of Atotang for the development of prescription on atopic dermatitis. Methods : Atotang was extracted with 80% EtOH. Free radical scavenging assay has tested for anti-oxidative activity as well as the contents of total polyphenol. We observed the production of ROS, nitric oxide(NO) and the inflammatory cytokines such as interleukin-1beta(IL-${\beta}$), IL-6, tumor necrosis factor-alpha(TNF-${\alpha}$), Prostaglandin E2($PGE_2$) in Raw 264.7 cells stimulated by LPS. We used Disc diffusion method to investigate antibacterial activity on Candida albicans, Staphylococcus aureus and Staphylococcus epidermis. Result : Content of total phenolic compound of Atotang was 36.3 mg/g ext. DPPH and ABTS scavenging activities were 77% and 46% at 200 ug/ml respectively, showing dose-dependent increase. The amounts of ROS and NO in RAW 264.7 cells were decreased by 30% and 19% at 200 ug/ml, respectively, showing dose-dependent decrease. The prodcution of IL-1beta, IL-6 and TNF-alpha in RAW 264.7 cells were decreased dose-dependently by 81%, 67%, and 20% at 200 ug/ml, respectively. Atotang was reduced LPS-stimulated production of $PGE_2$ by 33%. Atotang on C. albicans, S. aureus and S. epidermis was selected by a disc diffusion method and inhibition effect of the Atotang on the growth of S. epidermis was the greatest. Conclusion : The results indicated that Atotang showed biological activities showing anti-oxidant, anti-inflammatory and antibacterial effects. Based on these results, it is concluded that Atotang can be applied to the prescription on atopic dermatitis.

Diagnosis of Subclinical Mastitis-Causing Pathogens Using MALDI-TOF Mass Spectrometry in a Certified Organic Dairy Farm in Korea

  • We identified mastitis-causing pathogens using matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) in an organic dairy farm and evaluated the effects of antimicrobial restriction on antimicrobial susceptibility. A total of 43 Holstein cows without any clinical sign of mastitis were used in this study, and 172 quarter milk samples were cultured on blood agar plates for 24 hours at 37℃. Subsequently, bacterial species were identified and antimicrobial susceptibility tests were performed. The subclinical mastitis infection rates in the cows and quarters were 58.1% (25/43) and 25.6% (44/172), respectively. In the species identification, Staphylococcus aureus (40.9%) was the most prominent isolate, followed by S. chromogenes (22.7%), S. epidermis (18.2%), S. simulans (11.4%), S. haemolyticus (2.3%), S. muscae (2.3%), and S. xylosus (2.3%). In the antimicrobial susceptibility test, all isolates were 100% susceptible to 24 of 28 antibiotics, except for benzylpenicillin, cefalotin, cefpodoxime, and trimethoprim/sulfamethoxazole. The resistance rates of S. aureus, S. chromogenes, and S. muscae isolates to trimethoprim/sulfamethoxazole were 27.8%, 10%, and 100%, respectively, and the resistance rates of S. epidermis and S. xylosus to benzylpenicillin were 50% and 100%, respectively. S. chromogenes, S. epidermis, S. simulans, S. haemolyticus, and S. xylosus were resistant to cefalotin and cefpodoxime. In conclusion, restrictions on antimicrobial use for organic dairy farm certification have resulted in a high Staphylococcus spp. infection rate. Therefore, our study indicates the importance of mastitis management strategies implemented by farmers together with veterinary practitioners, even if mastitis does not appear clinically in organic dairy farms.

Changes of Fruit Structure and Sugar Contents during the Fruit Growth and Development in 'Yumyeong' Peach [Prunus persica (L.) Batsch] (과실생육시기별 '유명' 복숭아의 과실조직 발달과 당 변화)

  • 'Yumyeong' is one of the most popular peach varieties in Korea. This study was conducted to monitor the developments of cells and tissues, and the changes in sugar contents during fruit growth and development. At bloom, there were two rows of vascular tissues, and the number and the position of internal vascular bundles were consistent during the fruit growth, however, the number of vascular tissues were increased and the distribution was irregular in the flesh tissues. The tissues between the inner integument and the internal vascular bundles showed different development characteristics from other parenchyma cells which were consisted of small and dense cells containing tannins. Such observation suggested that the stone of peach was consisted of inner epidermis and cells in the internal vascular tissues. The outer epidermis consisted of single layer cells at bloom differrentiated into 1-2 layers by horizontal cell divisions on 14 days after full bloom. On 30 days after full bloom, the epidermis was consisted of 5-6 layers by vertical cell divisions. The cell layers consisting the outer epidermis were gradually decreased to 1-2 layers at maturity. The observations on the changes in the epidermis confirmed that some of the cells consisting the hypodermis of peach fruit were originated from the cells of outer epidermis. Tylosis was observed from 35 days after full bloom and the size and number of tylosis were increased until full fruit maturity. The sucrose content showed the tendency of sharp increase from 50 days to 120 days after full bloom then decrease slightly. After when stone hardening ended, other solids showed a gradual decrease tendency from 80 days after full bloom.

Epidermal Changes of the Adhesive Disks During Wall Attachment in Parthenocissus tricuspidata (착생에 따른 담쟁이덩굴 흡착근 표피조직의 변화)

  • The present study examined the epidermal changes of adhesive disks which occur during attachment in Parthenocissus tricuspidata using scanning and transmission electron microscopy. Several adhesive disks, each covered with a bract, develop from the shoot apical meristem during early development. In the initial stage, the adhesive disks are club-shaped and their upper and lower epidermis are indistinguishable. However, in the actively growing stage, they become spherical and both epidermis are clearly differentiated into the adventitious roots. Prior to wall attachment, the adhesive disks exhibit adaxial convex and abaxial concave shapes, and electron-dense substances are abundant in the vacuoles of epidermal cells. The peripheral area of the adhesive disk is adhered first to the wall surface, while the central area is drawn inward in a vacuum-like state during attachment. As the attachment progresses and the electron-dense substances continue to discharge, the upper and lower epidermis rapidly undergo deterioration and the disks shrink considerably. At this stage, structural changes of the lower epidermis occur much faster than in the upper one. The discharged substance is accumulated on the wall surface, and this aids the attachment of adhesive disks on the wall for long periods. In this manner, the shape and structure of the adhesive disk epidermis change drastically from initial growth to the mature stage. Further, the role of electron-dense substance and shrinkage of the disk during attachment has been discussed in Parthenocissus tricuspidata.

Effects of Dermal Cell Combination on the Formation of Basement membrane and Epidermis in Skin Equivalents (진피세포의 조성이 인공피부의 기저막과 표피형성에 미치는 영향)

  • European Union prohibited the marketing of cosmetic products containing constituents that have been examined through animal experiments. Thus, non-animal test models are needed to replace animal experiments. The reconstructed skin models are important as a test system for cosmetic, pharmaceutical, and medical device safety testing. In the present study, we tried to develop an optimal skin equivalent model containing basement membrane and epidermis. For this purpose, we used mesenchymal stem cells (MSCs) and/or preadipocytes as well as fibroblasts as the dermal matrix cells. The formation of basement membrane and epidermis was verified by immunohistochemical stains. Among various models, the epidermis was thickest when MSCs were used in the dermal matrix. Furthermore, PCNA and involucrin distribution showed that dermal matrix with MSCs resembled human skin. Therefore, skin equivalents with MSCs could be developed as a non-animal test model to replace animal experiments.

Seasonal Change of Skin Mucus Cells of Misgurnus mizolepis (Cobitidae) Dwelling in a Natural Stream in Korea (자연하천에 서식하는 미꾸라지 Misgurnus mizolepis 피부 점액세포의 계절변화)

  • Histological observation on the seasonal variation of mucus cells of the mud loach Misgurnus mizolepis inhabiting a natural stream was carried out on three skin regions (dorsal, lateral and occiput) from March 2008 to February 2009. Our results showed no differences in general morphology by season, but the mucus cells of the epidermis showed significant seasonal change in their size and number as the water temperature changed. The ratio of surface area of the mucus cell layer and mucus cells, and the number of mucus cells in surface area of the epidermis were the greatest in the cold winter and the least in the hot summer in all regions of the epidermis. In particular, the occiput seemed to be a very sensitive region in response to environmental change, showing wide fluctuations in the size of mucus cells throughout the year and a great change in between seasons, especially from late autumn to early winter when the temperature decreased. As the temperature became colder, a small and spherical-shaped mucus cell was transformed into a large and elongated columnar form with a lot of secreted mucus material in a superficial layer of the epidermis. From our results, we can safely surmise that cold temperature is an important environmental factor having a close relationship with the modification of mucus cells of M. mizolepis in winter.

Experimental Studies on the Hair Growth Activity of Trimix Extracts of Mylabris Phalerata Pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata in C57BL/6N mice (반모(斑蝥), 남성(南星), 반하(半夏)(생(生))추출 혼합물의 발모효과에 대한 실험적 연구)

  • To determine whether topical application of trimix extracts of Mylabris phalerata Pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata lead to affects on the hair growth activity in C57BL/6N mice. To examine the hair growth activity of the extracts of Mylabris phalerata Pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata gross, microscopic, and immunohistochemical method were performed. In order to examine the mRNA expression of hair growth related substance, RT-PCR method was performed. Experimental group I on day 14, The most extensive hair growth activity was observed in whole skin area of all the mice whose hair had been clipped. Brdu immunoreactive cells of all the experimental groups were more heavily stained in epidermis, bulge, outer root sheath, inner root sheath, subcutaneous tissue, hair bulb and cutaneous trunci muscles than that of control group on day 12 of hair growing cycle in C57BL/6N mice. VEGF immunoreactive density of all the experimental groups was more heavily stained in epidermis, bulge and cutaneous trunci muscles than that of control group on day 12. FGF and c-kit immunoreactive cells of all the experimental groups were heavily stained in epidermis, outer root sheath, inner root sheath and cutaneous trunci muscles on day 12. PKC-$\alpha$ immunoreactive density of all the experimental groups was mildly stained in epidermis and cutaneous trunci muscles than that of control group on day 12. On day 12, the expression of bFGF (138%, 119%, 120%), VEGF (146%, 144%, 133%), IGF-1 (165%, 141%, 119%) and PLI (121%, 116%, 123&) in each experimental groups was more increased than that of control group. On day 16, The expression of IGF-1 (126%, 149%, 151%) in all the experimental group was more increased than that of control group (100%). The expression of bFGF (92%, 94%) and VEGF (101%, 97%), PL1 (102%, 109%) in all the experimental group was more decreased than that of experimental group I, II on day 12. But the expression of bFGF (109%) and VEGF (127%), and PL1 (105%) in each experimental group III was more increased than that of control group (100%). These experiments suggest that trimix extracts of Mylabris phalerata pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata may stimulate the topical hair growth activity and its experimental group I can be useful for treatment of alopecia areata.
