Effect of Kamicheonmagudeungeum on Hypertension (가미천마구등음이 고혈압 병태모델에 미치는 영향)

  • Kamicheonmagudeungeum(KCGE), a traditional herbal medicine, has been used as a therapeutic agent for the treatments of acute stage of hypertension. We evaluated the effects of KCGE on hypertension induced by DOCA-salt in rats. The systolic blood pressure and pulse rate significantly lowered by oral administration with KCGE. The levels of plasma hormones including dopamine, epinephrine and norepinephrine, and serum electrolyte were also reduced in KCGE treated rats. In addition, the production of reactive oxigen species (ROS) were greatly decreased by the treatment with KCGE in cultured bovine endothelial cells and angiotensine converting enzyme (ACE) activites were also inhibited by KCGE in a dose dependent manner. This study indicated that KCGE is a safe and effective treatment for hypertension.

Effect of Tungstate Supplemented Diet on the Toluene Metabolism in Rats

  • To evaluate an effect of oxygen free radical on the toluence metabolism, the rats were fed on a tungstate sup-plemented diet(0.75g of tungstate included in 1kg of standard diet) or a standard diet. To the present xanthine oxidase deficient animal model, toluene(0.15ml/100g of body weight) was injected and then the animals were sacrificed after 24 hrs to determine the toluene metabolizing enzyme activities and toluene metabolite, hippuric acid concentration. The increasing rate of urinary hippuric acid concentration was significantly(p<0.01) higher in tungstate fed animals than in standard diet fed ones. Hepatic cytochrome P_450 contents were significantly higher(p<0.01) in tungstate fed animals than in standard diet fed ones. And tungstate fed animals showed a ten-dency of higher activities of benzylalcohol dehydrogenase while a significantly higher activites of benzaldehyde dehydrogenase (p<0.01) than standard diet fed animals. In conclusion, the more possibly reduced oxygen free radical in toluene-treated rats fed with a tungstate supplemented diet than in those fed with a standard diet would be responsible for the enhancement of toluene metabolism.

  • The effects of insecticides on biochemical precesses in soil were studied by determining the effects of the chemical structure of each insecticides on enzyme activities, pesticide residue and total number of bacteria revealed when soil treated with urea was incubated at $28{\pm}1^{\circ}$ for 56 days. The inhibition effects of insectides on enzyme activites in soil decreased in the order: dithiophosphoric acid > thiophosphhoric acid > phosphoric acid > carbamate insecticides for urease and phosphatase, thiophosphoric acid > dithiophosphoric acid > phosphoric acid > carbamate insecticides for L-glutaminase and protease. The inhibition effects of organophophorus insecticides on enzyme activities in soil were maintained longer than those of carbamate insecticides. Carbamate insecticides increased the activities of protease and L-glutaminase at 56 days. When insecticides were treated in soil together with urea, the degradation of insecticides was accelerated. By treatment of insecticides, the total number of bacteria was decreased at the early stage of treatment but thereafter increased according to phosphoric acid and carbamate insecticides.

Effect of Giant Embryonic Rice Supplementation on the Lipid Peroxide Levels and Antioxidative Enzyme Activities in the Plasma and Liver of Streptozotocin-induced Diabetic Rats (거대배아미 식이가 streptozotocin 유도 당뇨 흰쥐의 혈장과 간 조직 중 지질과산화물 농도와 항산화 효소 활성에 미치는 효과)

  • Effects on the feeds of streptozotocin-induced diabetic rats with a giant embryonic rice on lipid peroxides level and antioxidative enzyme activites in plasma and liver tissues were investigated. Along with the experimental periods, all animals in diabetic groups had a lower increase rate in body weight than the normal control group. A giant embryonic rice-fed group showed a inhibition in the decrease of body weight, and a increase in feed intake compared to the normal control group. The organ weights of the diabetic control group were heavier than those of the normal control while rice-fed groups including the giant embryonic rice-fed group were found to have lower organ weights, and its blood sugar level was found to be lower than those of the normal group. Lipid peroxides of the giant embryonic rice-fed animals showed a lower lipid peroxidation values compared to that of the diabetic control group. Plasma vitamin A and E concentrations of the diabetic control group were significantly decreased compared to the normal control while those of the giant embryonic rice-fed group were found to be significantly higher than those of the diabetic control. Of the hepatic antioxidative enzymes, SOD activity of the giant embryonic rice-fed group was higher than that of the diabetic control group. Taken these together, low lipid peroxidation values and, in contrast, high antioxidative enzyme activities were thought to be a cause for decreasing hepatic oxidative damages.

Efficacy of Uniconazole as a Phytoprotectant Against $SO_2$ Injury in Snap Bean (강남콩에 대한 $SO_2$ 피해경감제로서 uniconazole의 효과에 관한 연구)

  • This study was conducted to determine the efficacy of using uniconazole,[(E)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazole-1-yl)-1-penten-3-ol)] as a phytoprotectant against $SO_2$ injury in snap been (Phaseolus vulgaris L. 'Strike'). Thirteen days prior to $SO_2$ fumigation, plants were given a 100 ml soil drench of uniconazole solution at concentrations of 0.02, 0.10, 0.25 and 0.50 mg/pot. All four uniconazole concentrations were significantly effective in providing protection against $SO_2$ exposure(3 h at 1.5 ppm), but uniconazole treatment above 0.02 mg/pot severely reduced stem elongation, leaf enlargement, flowering date and pod number and weight. Uniconazole treatment had little or no effect on stomatal conductance but reduced transpiration rate on a whole plant basis by nearly 40%. This may reflect an alteration in canopy structure by reducing stem elongation and leaf enlargement. Although uniconazole did not increase the activities of superoxide dismutase(SOD) and peroxidase(POD) in non-$SO_2$-fumigated plants, it significantly increased those enzyme activities in $SO_2$-fumigated plants. Chlorophyll concentration on the basis of unit area was increased 50-60% by uniconazole. However, the difference was not detected on the basis of dry weight. $SO_2$ increased variable chlorophyll fluorescence (Fv) 48% after 1.5 h of exposure in non-uniconazole treated plants but decreased Fv in the plants after 3 h of exposure. By appliing uniconazole, it was possible to maintain high Fv values in the latter group of plants. These results suggest that the phytoprotective effects of uniconazole are related to its growth-retarding properties as an anti-gibberellin as well as the increase of activites of free radical scavengers such as SOD and POD.

Effects of Green Tea Catechin on the Superoxide Dismutase, Glutathione Peroxidase and Xanthine Oxidase Activites of Kidney in Diabetic Rats (녹차 Catechin이 당뇨쥐 신장조직의 Superoxide Dismutase, Glutathione Peroxidase 및 Xanthine Oxidase)

  • The purpose of this study was to investigate the effect of green tea catechins on the antioxidative defense enzyme activity of kidney in diabetic rats. Sprague-Dawley male rats weighting 100$\pm$10g were randomly assigned to one normal and three STZ-induced diabetic groups; catechin free diet(DM-0C group), 0.25% catechin diet(DM-0.25C group) and 0.5% catechin diet(DM-0.5C group). Diabetes was induced by intravenous of 55mg/Kg body weight of STZ in sodium citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Rats were sacrified at the 6th day of diabetic states. Superoxide dismutase(SOD) activity in kidney was decreased by 25% and 20% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. Glutathione peroxidase(GSHpx) activity in kidney was were no significant differences the diabetic groups compared to normal group. Xanthin oxidase(XOD) activity was increased by 110% and 63% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. The contents of superoxide radical(O$_2$)in kindney were 116% and 33%, respectively, higher in DM-0C and DM-0.25C groups than normal group. DM-0.5C group and normal groups were similar levels in their superoxide radical contents of kidneys. Levels of TBARS(thiobarbituric acid reactive substance) in kidney were increased by 62% in DM-0C group, when compared with normal group, but those of DM-0.5C group were similar to that of normal groups. These results indicate that free radical generation system was weakened and free radical scavenger system was enhance in kidney of STZ-induced diabetics rats by dietary catechin. Thereby it may reduce renal disorders such as oxidative damage and aging of tissue.

Anti-Inflammatory and Anti-Oxidative Effect of Pinus koraiensis Cone Shell Extracts (잣피 추출물들의 항산화 및 항염증에 미치는 영향)

  • The present study examined the anti-inflammatory and anti-oxidative effects of Pinus koraiensis (PK) cone shell extracts in vitro. Anti-inflammatory and anti-oxidative effects of PK cone shell extracted with hot water, 20% ethanol (EtOH), or 50% EtOH were examined using 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging assay, superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activites, as well as nitric oxide (NO) and anti-inflammatory cytokine measurements. The 20% EtOH extract of the PK cone shell decreased the NO and inflammatory cytokines secretion, and increased the ABTS radical scavenging, SOD, CAT, and GPx activities. This indicates that the 20% EtOH extract of the PK cone shell would be helpful in inflammation and oxidation systems. Therefore, the 20% EtOH extract of PK cone shell has great potential as a useful health food.

Effects of Dietary Fats on Plasma Lipids and the Level of Lipid Peroxidation and Antioxidant Enzymes in Rats Treated with Dimethylhydrazing (Dimethylhydrazine을 투여한 쥐에서 식이 지방이 혈장 지질 조성과 조직의 과산화물형성 및 항산화효소 수준에 미치는 영향)

  • This study was designed to compare the effect of different dietary fats on plasma lipids, the degree of lipid peroxidation and the activity of antioxidant enzymes in RBC and liver rats treated with or wighout 1, 2-dimethylhydrazing (DMH). Male Sprague Dawley rats, at 7 weeks-old, were divided into control and DMH-treated grous, and each group was again subdivided into four were perilla oil (PO), blend fat (BF) containing ten different kinds of dietary oil, beef tallow (BT), corn oil (CO). At the same time, each rat was injected intramusculary with saline(for control) or DMH twice a week for 6 weeks to give total dose of 180 mg/kg body weight. Compared with BT feeding, BF reduced plasma total choesterol level and PO and Co reduced plasma TG levels (p<0.05). DMH injection decreased plasma cholesterol in all dietary groups. However, PO decreased tocopherol levels and increased TBARS levels in RBC compared to BT. The degree of hemolysis in PO group was higher than that of BT group (p<0.05 only in control group. Fatty acid composition of hepatic microsome was reflected by dietary fatty acid profile. The peroxidizability index and TBARS level in hepatic micorsome were significantly increased but tocopherol level was lowered in PO group compared to BT group. Activites of superoxide dismutase and glutathione peroxidase in RBC and hepatic cytosol were not influenced y dietary fats and DMH treatment(p<0.05). Overall, perilla oil rich in $\omega$3 $\alpha$-linolenic acid could be a very important dietary source in reducing plasma lipids and blend fat was also good dietary oil mixture in reducing plasma cholesterol. However, the degree of lipid peroxidation was greater in tissue by perilla oil feeding and it is very difficult to use only perilla oil as oil source for meal preparation, so that it could be suggested to use more perilla oil and fish to give an equal effect of blend fat in order to reduce the risk factors against cardiovascular disease.

  • Traditional herbal drugs of Korea, namely, Atractylodes Rhizome(蒼朮) and Atractylodes Rhizome White(白朮) which are derived from and old-grown rhizome and a newly-grown rhizome of Atractylodes japonica Koidz.(Compositae), respectively, were investigated their potential liver-protective activities against hepatotoxicity induced by $CCl_4$ intoxication in mice. Each herbal drug was extracted separately with chloroform, methanol and butanol. No extract of Atractylodes Rhizome showed a significant reduction of the duration of hypnosis produced by hexobarital after $CCl_4$ intoxication in mice. In addition, the the treatments of Atractylodes Rhizome extracts produced no decrease of aspartate aminotransferase(EC 2.6.1.1) activity in serum. However, the methanol extract of Atractylodes Rhizome White exhibited a marked protection from hepatotoxicity induced by $CCl_4$ intoxication in mice. It produced significant reductions of the duration of hypnosis and serum enzyme activity, but no other extract showed liver-protective activity against $CCl_4-induced$ hepatotoxicity.

Effect of Sea Tangle, Laminaria japonicus, Extract on The Activities of Glucokinase and Hexokinase in Alloxan-Induced Diabetic Mellitus Mice (Alloxan 처리 당뇨병 마우스의 췌장 glucokinase 및 hexokinase에 대한 다시마 열수추출물의 효과)

  • The study was undertaken to determine the effect of the aqeuous extract of sea tangle, Laminaria japonicus, on the activites of glucokinase and hexokinase in the pancreas of diabetic mellitus mice which were induced by alloxan. After one week of alloxan injection, the levels of serum glucose and insulin secretion were dramatically increased, however, the insulin secretion was decreased with administration of sea tangle extract. Alloxan injection allowed the serum glucose level to increase and level was decreased by sea tangle extract administration. Furthermore, it was observed the sea tangle extract was effective in recovering the levels of insulin secretion. Enzyme activities of the glucokinase and hexokinase were decreased by alloxan treatment. In contrast, sea tangle extract administration to the mice allowed to increase proportionally. These results suggested that sea tangle extract is highly effective in treatment of diabetic mellitus induced by alloxan.
