Production of Spirulina Extract by Enzymatic Hydrolysis (효소 가수분해 방법을 이용한 스피루리나 추출물의 제조)

  • An efficient production method of spirulina extract was developed by enzymatic treatment using cell lytic and proteolytic enzymes. The suitable dosage of Tunicase, a cell lytic enzyme, was found to be 2.0% (w/w). Proteolytic enzymes were screened to obtain high solid recovery and spirulina extraction (SE) index, which indicates nucleic acid-related substances content. Among the seven tested proteases, Esperase was selected and optimal dosage of this enzyme was 2.0% (w/w). The solid recovery and SE index of simultaneous treatment and co-treatment using optimal dosages of Tunicase and Esperase were greatly similar, respectively. However, co-treatment had the effect of shortening total hydrolysis time. The SE index and solid recovery of co-treatment were significantly enhanced by 75% $(11.4{\rightarrow}20.0)$ and 45% $(45.2%{\rightarrow}65.3%)$, respectively, than those of the non-treated extracts.

Candida rugosa Lipase-Catalyzed Production of Optically Pure S-(+)-Ketoprofen (Candida rugosa 리파제를 이용한 광학적으로 순수한 S-(+)-Ketoprofen의 생산)

  • Enzymatic resolution reactions were investigated using Candida rugosa lipase for the production of potically pure S-(+)-Ketoprofen. When the enzymatic hydroysis (and esterification) of recemic ketoprofen esters (and recemic ketoprofen with alcohol) was investigated comparatively, aqueous media was more specific for S-enantiomer than organic media. In the enzymatic hydrolysis of racemic ketoprofen ethyl ester in aqueous media, optimal temperature and pH for enantioselectivity were $37^{\circ}C$ and 4, respectively. The stereoselectivity of the enzyme was enhanced by adding dialcohols such as ethylene glycol and propylene glycol. The enantiomeric ratio obtained in the 40 %(v/v) ethylene glycol was 2-fold higher than that without the additive. By adding $CH_2Cl_2$, $CHCl_3$ and $CCl_4$ (5%,v/v), the enantioselectivity was reversed. A dramatic increase in the stereoselectivity was achieved using lipase purified by anion exchange chromatography. The type A lipase(the first eluted lipase fraction) showed an enantiomeric ratio of >100, whereas the type B lipase(the second eluted lipase fraction) exhibited enantimomer ratio of 9.0 in the hydrolysis of racemic ketoprofen ethyl ester.

  • Enzymatic conversion of pyruvic acid produced by microorganism to tryptophan was investigated. A luminescent bacteria. Beneckea sp., was used for the production of pyruvic acid. As a source of tryptophanase which synthesizes tryptophan from pyruvic acid, indole and ammonia, whole cells of Enterobacter aerogenes ATCC 10031 were used directly in the reaction mixture. To increase the production of tryptophan, nonionic detergents and nonaqeous organic solvents were used ms reserviors of indole in the reaction mixture. In the case of nonionic detergents, TritonX-100 was very effective. When 1.5% of Triton X-100 was used, 7.7g/$\ell$ of tryptophan was produced at 37$^{\circ}C$ for 48hr. In the case of nonaqueous solvents, 8.7g/$\ell$ of tryptophan was produced at 37$^{\circ}C$ for 48 hr, when 10% of benzene was used. This amount of tryptophan corresponds to conversion of 48% of Indole and 36% of pyruvic acid, respectively.

Effect of SAA Pretreatment on SSF at Low Temperature to Bioethanol Production from Rice Straw (암모니아수 침지 전처리 공정을 이용한 볏짚의 저온 동시당화발효)

  • Physical and chemical barriers, caused by the close association of the main components of cellulosic biomass, hinder the hydrolysis of cellulose to fermentable sugars. Since the main goal of pretreatment is to increase the enzyme accessibility improving digestibility of cellulose, development of an effective pretreatment process has been considered to be important. In this study, SAA (Soaking in Aqueous Ammonia) was chosen as pretreatment because this is the simple and low-cost method. Rice straw of which the production is outstandingly high in domestic agriculture residues in Korea was chosen as raw material. SSA pretreatment with various reaction time of 3 h to 72 h was tested. The enzymatic hydrolysis and SSF (Simultaneous Saccharification and Fermentation) were performed at three different temperature (30, 40 and $50^{\circ}C$) to investigate performance of SSF upon various pretreatment conditions. As a result, this SAA treated-rice straw was found to have great potential for effective enzymatic hydrolysis and SSF with lower enzyme dosage at lower temperature ($30^{\circ}C$) than its conventional SSF. In SAA addition, SAA reduced fermentation time to 24 h owing to increase the initial hydrolysis rate substantially.

Optimization of ultrasonic-assisted enzymatic hydrolysis conditions for the production of antioxidant hydrolysates from porcine liver by using response surface methodology

  • Objective: The objective of this study was to optimize ultrasonic-assisted enzymatic hydrolysis conditions, including enzyme-to-substrate (E/S) ratio, pH, and temperature, for producing porcine liver hydrolysates (PLHs) with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity by using response surface methodology (RSM). Methods: The study used RSM to determine the combination of hydrolysis parameters that maximized the antioxidant activity of our PLHs. Temperature ($40^{\circ}C$, $54^{\circ}C$, and $68^{\circ}C$), pH (8.5, 9.5, and 10.5), and E/S ratio (0.1%, 2.1%, and 4.1%) were selected as the independent variables and analyzed according to the preliminary experiment results, whereas DPPH free radical scavenging activity was selected as the dependent variable. Results: Analysis of variance showed that E/S ratio, pH, and temperature significantly affected the hydrolysis process (p<0.01). The optimal conditions for producing PLHs with the highest scavenging activity were as follows: E/S ratio, 1.4% (v/w); temperature, $55.5^{\circ}C$; and initial pH, 10.15. Under these conditions, the degree of hydrolysis, DPPH free radical scavenging activity, ferrous ion chelating ability, and reducing power of PLHs were 24.12%, 79%, 98.18%, and 0.601 absorbance unit, respectively. The molecular weight of most PLHs produced under these optimal conditions was less than 5,400 Da and contained 45.7% hydrophobic amino acids. Conclusion: Ultrasonic-assisted enzymatic hydrolysis can be applied to obtain favorable antioxidant hydrolysates from porcine liver with potential applications in food products for preventing lipid oxidation.

Enzymatic Hydrolysis of Marine Algae Hizikia fusiforme (해조류 톳 (Hizikia fusiforme)의 효소 가수분해)

  • In this study, we investigated the effect of reaction factors on enzymatic hydrolysis of Hizikia fusiforme, which is brown algae in marine biomass resource, using commercial enzymes. The composition of H. fusiforme is 38.9% of reducing sugar, 4.8% of moisture, 17.8% of ash, and 38.5% of others. In the condition of 1-5% substrate, the increase of substrate concentration enhanced the increase of reducing sugar formation; however, the hydrolysis yield did not increase after 24 h. After reaction of 75 h, conversion yield of reducing sugar were obtained to 16.45%, 17.99%, and 14.55% at 1, 2.5, and 5% substrate, respectively. As a result of effect of enzyme amount, the formation of reducing sugar did not show considerable change at 1% substrate. However, in the condition of 2.5% substrate, the great change of reducing sugar formation was observed by the increase of enzyme amount. The conversion yields of reducing sugar were obtained to 18.77% and 22.83% at 1% and 2.5% substrate with 30% enzyme, respectively. As a result of heat treatment of biomass, the high yield was obtained in 2.5% substrate and the yields were increased to 0.06-7.2% by the heat treatment. This result will provide the basic information for production process of biofuels and chemicals from marine biomass H. fusiforme.

Effect of Malonic Acid-Catalyzed Pretreatment on the Hydrolysis of Gracilaria verrucosa (Malonic acid를 이용한 전처리가 꼬시레기의 가수분해에 미치는 영향)

  • In this study, the effects of malonic acid-catalyzed pretreatment on the subsequent enzymatic hydrolysis of red macro-algae Gracilaria verrucosa for production of biosugar (total reducing sugar) were investigated. In the hydrothermal pretreatment condition of 300 mM malonic acid, 1:20 solid-to-liquid ratio at $130^{\circ}C$ for 60 min, a 49.2% biosugar yield was achieved. Moreover, by subsequent enzymatic hydrolysis after pretreatment, maximum yield of 64.5% was achieved.

pH-Controlled Synthesis of Cephalexin by a Purified Acetobacter turbidans Ampicillin Acylase

  • It has been known that, in enzymatic synthesis of cephalexin, the conversion yield was reduced by high loading of ampicillin acylase. In order to elucidate this phenomena, pH-controlled synthesis of cephalexin was examined using a purified Acetobacter turbidans acylase. When the pH of the reaction mixture was maintained at $6.20{\pm}0.04$, the reduction of the maximal conversion rate was not observed even with high enzyme loading. The kinetic parameters also suggest that pH drop during the enzymatic synthesis of cephalexin was mainly attributed to the rapid hydrolysis of D-${\alpha}$-phenylglycine methyl ester to D-${\alpha}$-phenylglycine, rather than the disappearance of 7-amino-3-deacetoxycephalosporanic acid for cephalexin synthesis. At higher molar ratio of two substrates, [D-${\alpha}$-phenylglycine methyl ester]/[7-amino-3-deacetoxycephalosporanic acid], the conversion rate was also elevated under pH-controlled enzymatic synthesis, which implies that the main reason for the pH drop is due to the production of D-${\alpha}$-phenylglycine methyl easter, the effect of a water-methanol cosolvent system on the ester, the effect of a water-methanol cosolvent system on the conversion profile was also examined. Even the though the conversion rate was increased in 10% methanol solution, a higher than 16% methanol in the reaction mixture caused an inactivation of enzyme.

In vitro-growth and Gene Expression of Porcine Preantral Follicles Retrieved by Different Protocols

  • This study was conducted to determine how the isolation method of the porcine preantral follicles influenced the following follicular growth in vitro. Mechanical and enzymatical isolations were used for retrieving the follicles from prepubertal porcine ovaries, and in vitro-growth of the follicles and the expression of folliculogenesis-related genes were subsequently monitored. The enzymatic retrieval with collagenase treatment returned more follicles than the mechanical retrieval, while the percentage of morphologically normal follicles was higher with mechanical retrieval than with enzymatic retrieval. After 4 days of culture, mechanically retrieved, preantral follicles yielded more follicles with normal morphology than enzymatically retrieved follicles, which resulted in improved follicular growth. The mRNA expression of FSHR, LHR Cx43, DNMT1 and FGFR2 genes was significantly higher after culture of the follicles retrieved mechanically. These results suggest that mechanical isolation is a better method of isolating porcine preantral follicles that will develop into competent oocytes in in vitro culture.

Inhibition Patterns of Dopachrome Formation as Influenced by Sulfur Dioxide ($SO_{2}$에 의한 dopachrome 형성 억제 패턴)

  • Inhibiting action of sulfur dioxide $(SO_{2})$ on enzymatic discoloration was investigated with crude enzyme preparations from homogenized tissues or sliced disks of raw potato tubers. $SO_{2}$ appeared to inhibit the formation of dopachrome in a competitive manner. At insufficient concentrations of $SO_{2}, the formation of dopachrome was reinitiated as time elapsed. The present results suggested that $SO_{2}$ would form an additional compounds with certain intermediate during the course of enzymatic browning and delay the enzymatic discoloration. To inhibit the production of dopachrome in sliced disks of raw potato tubers, much higher concentrations of $SO_{2}$ were required than homogenized tissues.
