Enzymatic Hydrolysis of Beef Tallow (효소에 의한 우지의 가수분해 반응)

  • Reef tallow was hydrolyzed with lipase under the conditions of liquid state and solid state. Lipase OF 360 was used for that purpose, and the lipase had the maximum activity when the olive oil was used as a substrate at pH 6 and $37^{\circ}C$. Beef tallow was dispersed by an agitator to perform a liquid enzymatic reaction. Water content, reaction temperature, and enzyme amount were varied as parameters affecting hydrolysis percentage. Ninety three percents of tallow were hydrolyzed at the following conditions: water content 80% w/w, temperature $37^{\circ}C$, and enzyme amount 200 unitlg tallow. In order to conduct a solid phase enzymatic reaction, sonication was employed for pretreating tallow with the enzyme solution. Molten tallow was sonified with the enzyme solution, and solidified by lowering temperature. And then hydrolysis reaction proceeded at $30^{\circ}C$. Sonication intensity and time were varied to control hydrolysis percentage. Optimum values of the intensity and the time were found to exist since the hydrolysis percentage did not increase further according to the increases of the intensity and the time.

Preparation of Diacylglycerol from Lard by Enzymatic Glycerolysis and Its Compositional Characteristics

  • The aim of this study was to prepare diacylglycerol (DAG) by enzymatic glycerolysis of lard. The effects of reaction parameters such as lipase type, reaction temperature, enzyme amount, substrate molar ratio (lard/glycerol), reaction time, and magnetic stirring speed were investigated. Lipozyme RMIM was found to be a more active biocatalyst than Novozym 435, and the optimal reaction conditions were 14:100 (W/W) of enzyme to lard substrate ratio, 1:1 of lard to glycerol molar ratio, and 500 rpm magnetic stirring speed. The reaction mixture was first incubated at $65^{\circ}C$ for 2 h and then transferred to $45^{\circ}C$ for 8 h. At the optimum reaction conditions, the conversion rate of triacylglycerol (TAG) and the content of DAG in the reaction mixture reached 76.26% and 61.76%, respectively, and the DAG content in purified glycerolized lard was 82.03% by molecular distillation. The distribution of fatty acids and Fourier transform infrared spectra in glycerolized lard samples were similar to those in lard samples. The results revealed that enzymatic glycerolysis and molecular distillation can be used to prepare more highly purified DAG from lard.

Biochemical Studies of an Endoglucanase from Marine Rotifer, Brachionus plicatilis

  • Cellulase was purified from marine rotifer, Brachionus plicatilis, to homogeneity by using chromatographic methods. Purified enzyme is an endo-${\beta}$-1,4 glucanase and shows a strong hydrolytic activity against carboxymethyl (CM) -cellulose. The physicochemical parameters of enzyme activity were determined. The molecular weight of the purified protein was approximately 62 kDa as determined by SDS-polyacrylamide gel electrophoresis. The enzymatic capability to digest cellulose of Chlorella cell wall was compared with that of other well known cellulases from Thermomonospora fusca. Experiments involving Chlorella digestion indicated that CM-cellulase from marine rotifer, Brachionus plicatilis, could digest Chlorella very efficiently while cellulase purified from Thermomonospora fusca did not. From the result here, we propose that the cellulolytic system from marine rotifer is responsible for the hydrolysis of cellulosic wall of Chlorella, probing that rotifer digests Chlorella as a major live food.

The Effects of Phenolic Radical Carriers on the Enzymatic. Bleaching of Kraft Pulp (Kraft펄프의 효소표백반응에 미치는 페놀라디칼 전달체의 영향)

  • The effects of enzymatic pretreatments on the bleaching of kraft pulp were studied. The kappa number of pulp samples which represents the lignin content of pulp decreased by 25.2% by the pretreatments with xylanase(EC 3.2.1.8, Pulpzyme HB) while it decreased by 13.7% without enzyme pretreatments after the extraction of the pretreated pulp samples in 1N NaOH. To enhance the effects of enzymatic pretreatment on the bleaching of kraft pulp, phenols were used as radical carriers with the simultaneous use of peroxidase(EC 1.11.1.7, Novozyme 502), $H_2O_2$, and xylanase. Guaiacol (1mM) was most effective by decreasing the kappa number by 29.6% when a low initial concentration of $H_2O_2$ (0.1mM) was used. The use of either a higher initial concentration of $H_2O_2$ or phenols lacking electron donating substituents such as phenol and p-chloyophenol, however, decreased the efficiency of enzymatic pretreatment indicating that the production rate and the stability of phenolic radicals are important parameters.

Non-enzymatic Antioxidant Status and Biochemical Parameters in the Consumers of Pan Masala Containing Tobacco

  • Background: Tobacco consumption is one of the leading causes of oral submucous fibrosis, oral cancer and even premature death. The present study was designed to compare the biochemical parameters and non-enzymatic antioxidant status and the lipid peroxidation products in pan masala tobacco users as compared with age-matched non-user controls. Methods: Pan masala and tobacco users of age $33.2{\pm}9.94$ years and age-matched controls ($31.2{\pm}4.73$ years) were enrolled for the study. Plasma levels of vitamin E, vitamin C, albumin, bilirubin, uric acid, glucose, urea, creatinine, aspartate amino transferase (AST), alanine amino transferase (ALT) were measured by standard methods. Serum malondialdehyde (MDA) levels were estimated as a measure of lipid peroxidation. Results: In the pan masala tobacco users, as compared to the controls, the level of vitamin C ($68.5{\pm}5.9$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.05$) vitamin E ($18.4{\pm}5.3$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.001$), albumin ($37.5{\pm}7.01$ vs $44.3{\pm}9.99g/L$, $p{\leq}0.001$), and malondialdehyde ($10.8{\pm}1.29$ vs $1.72{\pm}1.15nmol/ml$, $p{\leq}0.001$) were found to be significantly altered. Malondialdehyde was significantly correlated with vitamin E (r=1.00, p<0.001) and vitamin C (r=1.00, p<0.001) in pan masala tobacco users. Serum levels of AST ($31.0{\pm}16.77$ IU) and ALT ($36.7{\pm}31.3$ IU) in the pan masala tobacco users were significantly raised as compared to the controls (AST, $25.2{\pm}9.51$ IU, p=0.038; ALT, $26.2{\pm}17.9$ IU, p=0.038). Conclusion: These findings suggest that pan masala tobacco users are in a state of oxidative stress promoting cellular damage. Non-enzymatic antioxidants are depleted in pan masala tobacco users with subsequent alteration in the biochemical parameters. Supplementation of antioxidants may prevent oxidative damage in pan masala tobacco users.

  • Saururus Chinensis Baill (Saururaceae) has been used as folk medicine for analgesics, beriberi, edema, hepatitis, and icterus, etc. Hepatoprotective effects of Saururus chinensis Baill (SCB) administration on function of the biochemical parameters in liver of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) treated rats were investigated. After 7 days from TCDD(1$\mu\textrm{g}$/kg) injection, SCB(200mg/kg) was administered into rats intraperitoneally for 4 week.s We examined the antioxidative enzymatic activity by measuring the level of AST and ALT in serum and SOD, Catalase, GPx, GSH and GSSG in liver tissue of rats. Optimization of ultrasonic-assisted enzymatic hydrolysis conditions for the production of antioxidant hydrolysates from porcine liver by using response surface methodology

  • Objective: The objective of this study was to optimize ultrasonic-assisted enzymatic hydrolysis conditions, including enzyme-to-substrate (E/S) ratio, pH, and temperature, for producing porcine liver hydrolysates (PLHs) with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity by using response surface methodology (RSM). Methods: The study used RSM to determine the combination of hydrolysis parameters that maximized the antioxidant activity of our PLHs. Temperature ($40^{\circ}C$, $54^{\circ}C$, and $68^{\circ}C$), pH (8.5, 9.5, and 10.5), and E/S ratio (0.1%, 2.1%, and 4.1%) were selected as the independent variables and analyzed according to the preliminary experiment results, whereas DPPH free radical scavenging activity was selected as the dependent variable. Results: Analysis of variance showed that E/S ratio, pH, and temperature significantly affected the hydrolysis process (p<0.01). The optimal conditions for producing PLHs with the highest scavenging activity were as follows: E/S ratio, 1.4% (v/w); temperature, $55.5^{\circ}C$; and initial pH, 10.15. Under these conditions, the degree of hydrolysis, DPPH free radical scavenging activity, ferrous ion chelating ability, and reducing power of PLHs were 24.12%, 79%, 98.18%, and 0.601 absorbance unit, respectively. The molecular weight of most PLHs produced under these optimal conditions was less than 5,400 Da and contained 45.7% hydrophobic amino acids. Conclusion: Ultrasonic-assisted enzymatic hydrolysis can be applied to obtain favorable antioxidant hydrolysates from porcine liver with potential applications in food products for preventing lipid oxidation.

Texture Characteristics of Horse Meat for the Elderly Based on the Enzyme Treatment

  • Horse meat is nutritionally adequate to the elderly, but it has a comparatively hard texture in contrast to most of the food. In practice, the meat intake in the elderly is generally bated because the relatively difficult texture of the meat can diminish mastication. Thus, strategies are being developed to produce meat products remanding detracted mastication exertion and possibly exalt ingestion and nutritional stand, in the elderly. Hence, the effects of enzymes on textural characteristics of horse meat were studied, because they have well-known favorable efficacy on the meat tenderness by causing important demotion of the myo-fibrillar protein and collagen. Four treatments namely, papain, bromelin, pepsin, and pancreatin, alongside one control were invoked to the horse meat. Their effects on the texture parameters were determined. All the above enzymatic treatments significantly reduced hardness and resilience (p<0.001). These results present opportunities to produce essential fatty acids fortified horse meat with soft texture and satisfied technological characteristics. The intake of the essential fatty acids intensified horse meat could aid the elderly to get their aimed essential fatty acid demands. Results also suggest that horse meat tenderized through enzymatic processing stand for auspicious options for the comprehension of texture-revised diets in the elderly population.

  • The hydrolytic kinetics of biodegradable poly(l-lactide) (PLLA) have been studied by using two model systems, solution-grown single crystal (SC) and Langmuir monolayer techniques, for elucidating the mechanism for both alkaline and enzymatic degradations. The present study investigated the parameters such as degradation medium and time. The Langmuir mono layers of PLLA showed faster rates of hydrolysis when they were exposed to a basic subphase rather than they did when exposed to neutral subphase. Both degradation mediums had moderate concentrations to show a maximized activity, depending on their sizes. An alkaline degradation of SCs of PLLA showed the decrease of molecular weight of the remained crystals due to the erosion of chain-folding surface. However, the enzymatic degradation of SCs of PLLA occurred in the crystal edges thus the molecular weight of remained crystals was not changed. This behavior might be attributed to the size of enzymes which is much larger than that of alkaline ions; that is, the enzymes need larger contact area with monolayers to be activated.

Antihypertensive effect of an enzymatic hydrolysate from Styela clava flesh tissue in type 2 diabetic patients with hypertension

  • BACGROUND/OBJECTIVES: In this randomized, placebo-controlled, double-blind study, we evaluated the antihypertensive effects of enzymatic hydrolysate from Styela clava flesh tissue in patients with type 2 diabetes mellitus (T2DM) and hypertension. SUBJECTS/METHODS: S. clava flesh tissue hydrolysate (SFTH) (n = 34) and placebo (n = 22) were randomly allocated to the study subjects. Each subject ingested two test capsules (500 mg) containing powdered SFTH (SFTH group) or placebo capsules (placebo group) during four weeks. RESULTS: In the SFTH group, systolic and diastolic blood pressure decreased significantly 4 weeks after ingestion by 9.9 mmHg (P < 0.01) and 7.8 mmHg (P < 0.01), respectively. In addition, the SFTH group exhibited a significant decrease in hemoglobin $A_{1c}$ with a tendency toward improvement in homeostasis model assessment of insulin resistance, triglyceride, apolipoprotein B and plasma insulin levels after 4 weeks. No adverse effects were observed in other indexes, including biochemical and hematological parameters in both groups. CONCLUSION: The results of our study suggested that SFTH exerts a regulatory, antihypertensive effect in patients with T2DM and hypertension.
